Compared to gutta-percha, the endodontic cements are used in small quantity to seal root canals, but are indispensable to achieve hermetically sealed margins, where its biocompatibility depends on the sum of responses of each cell present in the periapical region. The object of this study was to evaluate the cytotoxicity of two endodontic cements, one based on epoxy resin (Sealer 26) and the other containing zinc oxide eugenol (Endofill) by using cultured peritoneal macrophages from Swiss mice to measure the induced production of nitric oxide. After solidification and pulverization, aliquots of 100mul of suspension containing 18mg/mL of the respective cements were added to 96-well tissue culture plates containing the tissue culture of macrophages at a concentration of 5.0X10(6) cells/ml. In the positive control group the cell culture was treated with 10mg/mL of lipopolyssaccharide from Escherichia coli 026:B6 and the cell culture alone represented the negative control. After 48 hours of incubation, at 37ºC, in 5% CO2, the cultures were placed in an ELISA automatic reader to evaluate the release of nitric oxide. The production of nitric oxide for cement Sealer 26 was between 36.1 and 313.0 mumols, with a mean of 143.82±111.03mumols, while for the Endofill these values were significantly less (p=0.01), varying from 50.8 to 125.7mumols, with a mean of 80.33±28.42 mumols. In the positive and negative control groups the mean release of nitric oxide was of 162.75mumols and 4.42mumols, respectively. There was no significant difference between the positive control group and cement Sealer 26 (p>0.05). Therefore, the cement Sealer 26 caused significantly greater toxicity to the macrophages, possibly due to the components from the epoxy resin and formaldehyde release during polymerization.

Abstract Background and aims: Cardiovascular diseases are widespread and causes many deaths in the world. The concentration of acute phase protein: C-reactive protein (CRP) and fibrinogen will rise dramatically when inflammation happens, which that can be used as an early marker of cardiovascular disease risk. Tempeh gembus contains fiber, unsaturated fatty acids and isoflavones are believed to reduce the inflammatory reaction. The aim of the study was to determinate the effect of tempeh gembus variations to levels of hcCRP and levels of fibrinogen of Sprague Dawley rats with atherogenic diet. Material and methods: This study was quasi-experimental with posttest only randomized control group design using 35 Sprague Dawley mice. The rats were randomized into 5 groups: negative control group given the standard diet, the positive control group given standard diet and atherogenic diet, and three treatment groups were given the standard diet, atherogenic diet and variation of tempeh gembus (tempeh gembus, heated tempeh gembus and tempeh gembus with bromelain enzyme) for 28 days. Serum levels of hsCRP and fibrinogen examined using ELISA (Enzyme-linked Immunosorbent Assay). Results and conclusions: The administration of tempeh gembus with bromelain enzyme is the most effective treatment for hsCRP serum level indicated a significant difference (p=0.028) between the negative control group, positive control group and first group with the third group. Fibrinogen serum levels showed significant differences in all treatment groups (p =0.042), administration of tempeh gembus with bromelain enzyme is the most effective treatment is shown by a significant difference between the negative control group and the positive control group with third group. Doxorubicin is an effective drug used in cancer therapy but produces reactive oxygen species (ROS) that are toxic to heart cells. The purpose of this study was to determine the cardioprotective activity of Mengkudu fruit ethanol extract against the heart induced by doxorubicin. Mengkudu fruit ethanol extract was obtained by maceration. Cardioprotective activity test is done by measuring blood LDH and CK-MB levels as well as cardiac histology. Animals were induced with DOX 5 mg/kg BW on day 1,7,14 and 20th. Administration of Mengkudu extract 100 mg / kg bw, 300 mg / kg bw, and 500 mg / kg BW given from day 1 to day 20 and on the 21st day cardiac serum levels of CK-MB normal group had a significant difference (p <0.05) with a negative control treatment group, treatment group I, treatment group II, and treatment group III and did not have a significant difference (P> 0.05) with a positive control group with vitamin e supplementation and serum LDH levels the normal group had a significant difference (p <0.05) with the negative control group, the treatment group I, the treatment group II, and the treatment group II and did not have a significant difference (P> 0.05) with the positive control group with vitamin e supplementation. Cardiology histology of the Mengkudu extract 100 mg/kg bw + DOX and the Mengkudu extract 300 mg/kg bb + DOX, and the negative control group showed bleeding, fragmentation and myocytolysis, in the treatment of group III, the group normal, and the positive control group did not show heart muscle damage. Based on the description above it can be concluded that the ethanol extract of Mengkudu fruit containing flavonoids has cardioprotective activity by inhibiting the formation of ROS. The higher the dose of an extract, the greater the decrease in LDH and CKMB levels and increase protection against heart damage.
