The whole plant of the methanolic extract from Leucas cephalotes was screened for invitro antioxidant (using the DPPH method), invivo analgesic (using hot plate test in mice) and anti-inflammatory (using rat paw edema test) activities. The methanolic extract of Leucas cephalotes (MELC) scavenged the DPPH radicals in a dose-dependent manner. The IC50 value to scavenge DPPH radicals was found to be 421.3µg/ml. A significant (p<0.0005) analgesic activity was observed at 60 min with 200 mg/kg, and 400 mg/kg exhibited maximum activity. The maximum anti-inflammatory response was produced at 3 hr and 2 hr with doses of 200 and 400 mg/kg, respectively. These results suggest that the methanolic extract from Leucas cephalotes exerts significant analgesic and anti-inflammatory effects, which were comparable with standard drugs.

Many scientists study structure and charagterization of placenta and experimenting on their biological activity, due to develop new drugs and biopreparation for human and animal health. The present study aimed to isolate bovine placental lactogen from placenta by semi purification method and determine antioxidant, anti-inflamatory activity and effect on immune responses was also investigation. Semi purified protein was separated  by gel-chromatographic method using 3 methods of tissue homogenizations of placental tissue using distilled water, phosphate buffer saline and Tris-HCl, by use of 4 methods of protein extraction and precipitation with distilled water, sulfate ammonium salt and organic solvents, such as ethanol and acetone.Antioxidant activity of semi purified  protein was measured by DPPH method, effect on immunity was investigated by using MTT assay kit, and anti-inflammatory activity was measured by using 1 % carrageenan-induced rat paw edema models. Measurements of total proteins which were homogenized in PBS, followed by acetone precipitation is 6217.5 µg/ml is the highest and homogenized in PBS, followed by sulfate ammonium salt precipitation is 1477.4 µg/ml is the lowest concentration. Bovine placental extract DPPH radical scavenging activity was 55.1±1.2% and rutin was 67.1±1.2% but results of immunity experiment demonstrated splenocyte division in placental extract added medium for stimulation index 1.45 or splenocyte culture increased by 1.5 times than as compared to those in medium not added placental extract, as well as placental extract added in the medium stimulation index increased 1.08 or splenocyte division by 0.8 times or almost the same than those wells with culture treated with mitogen only. Semi-purification placental protein was inhibition on 1% carrageenan induced rat paw edema and it means bovine placental extract effectively reduce acute inflammation. ABSTRACT Geissospermum vellosii (Pao pereira) is a Brazilian tree whose stem barks are rich in indole alkaloids that present intense anticholinesterase activity. The present study evaluated the effects of a stem bark fraction (PPAC fraction) and ethanolic extract (EE) of Pao pereira in classic murine models of inflammation and pain. The EE and PPAC fraction, both at a dose of 30 mg/kg, significantly reduced mice abdominal constriction induced by acetic acid by 34.8% and 47.5%, respectively. In the formalin test, EE (30 mg/kg) and PPAC fraction (30 and 60 mg/kg) inhibited only the second phase, by 82.8%, 84.9% and 100%, respectively. Compared with indomethacin, similar doses of EE or PPAC fraction were approximately twice as effective in causing antinociception. PPAC fraction was not effective in the hot plate test but reduced the inflammatory response at the second (50.6%) and third (57.8%) hours of rat paw edema induced by carrageenan. Antihyperalgesic activity was observed within 30 min with a peak at 2 h (60.1%). These results demonstrate that compounds in PPAC fraction have anti-inflammatory and antinociceptive activity by a mechanism apparently unrelated to the opioid system. Regardless of similar responses to indomethacin, the effects of PPAC fraction are mainly attributed to acetylcholine actions.

Objective: The present investigation aims at development and evaluation of herbal anti-inflammatory gel using of methanolic extract from Emblica officinalis fruits (MEEOF) and Aegle marmelos fruits (MEAMF). Methods: The gels were prepared using Carbopol 934, various concentrations of MEEOF, MEAMF, propylene glycol 400, methylparaben, propylparaben, and required amount of distilled water. Then, skin pH (6.8-7) was maintained by dropwise addition of triethanolamine. Prepared formulations were evaluated for physical appearance, pH, spreadability, viscosity and homogeneity, skin irritation on animal model (rabbit), and anti-inflammatory activity using carrageenan-induced rat paw edema model on albino Wistar rats of either sex (150–200 g). Change in edema volume of the rat hind paw was measured, and percent inhibition was calculated. ICH guidelines have followed for stability studies. Results: Results reveal that gel showed good appearance, homogeneity, and spreadability. Viscosity is ranging between 4200 and 4500 centipoises. All formulations have shown no skin irritation to animals. Formulations F4 and F5 significantly inhibited the inflammation to the extent of 56.66%, 61.66% at 3 h and 59.21%, 63.15% at 4 h, respectively, while the reference drug reduced the inflammation by 66.66% at 3 h and 76.31% at 4 h. The preparation was stable under normal storage conditions and did not produce any skin irritation, i.e., erythema and edema when applied over the skin on storage. Conclusion: It was concluded that anti-inflammatory effect of F4 and F5 was comparable to standard.

Aim: The experimental investigation of current research work was to identify traditional rich claim of Stereospermum suaveolens DC leaves for anti-inflammatory and anti-arthritic potential action in animals. Study design: Ethyl acetate fraction of Stereospermum suaveolens DC (Bignoniaceae) methanolic extract of leaves evaluated at 125mg/kg, 250mg/kg and 500mg/kg (p.o.) doses for anti-inflammatory and anti-arthritic activity. Methodology: Ethyl acetate fraction of Stereospermum suaveolens DC (Bignoniaceae) methanolic extract of leaves was evaluated for phytochemical investigation for total flavonoid content using UV spectroscopy and TLC study. Carrageenan induced rat paw edema (Acute method) and Freund’s complete adjuvant (FCA) induced chronic arthritis in wistar rats were used as an animal models to claim Stereospermum suaveolens DC leaves for anti-inflammatory and anti-arthritic potential.  The rat paw volume and percentage inhibition of the paw edema were evaluated for anti-inflammatory activity. The assessments of arthritis in rats were measured by haematological values and radiological examinations. Result: Ethyl acetate fraction of Stereospermum suaveolens DC (Bignoniaceae) methanolic extract of leaves showed presence of total flavonoids and saponins. The significant inhibition in paw volume and edema (p < .01) obtained at 250mg/kg and 500mg/kg oral dose. These obtained results were established confirmation outcome for presence of rich flavonoid contents in Stereospermum suaveolens DC leaves and provides valuable source of bioactive phytocomponents.  Conclusion: Ethyl acetate fraction of Stereospermum suaveolens DC (Bignoniaceae) methanolic extract of leaves  showed significant inhibition of inflammatory reaction as compared to standard drug indomethacin Sterospermum Suaveolens DC leaves were showed potential therapeutic role in treatment of inflammation and arthritis cases.

Benzoxazolones are widely distributed in plants and are of increasing interest for a variety of pharmacological properties, such as detoxification, antibacterial, anti-inflammatory, and analgesic activities and tranquilizers (Michaelidou and Hadjipavlou-Litina, 2005; Doğruer et al., 1998). 4-Hydroxy-2-benzoxazolone (HBOA) is one of the major bioactive compounds in traditional Chinese herb drugAcanthus ilicifolius  (Peng and Long, 2006) which has obvious anti-inflammatory and analgesic activities (Huo et al., 2004; Mani Senthil Kumar et al., 2008; Babu et al., 2001). In this research, we used 2-nitroresorcinol as starting material to prepare HBOA with a novel “one-pot-way.” Derivatives of HBOA TC-2∼TC-4 were obtained via electrophilic reagents and reacted with corresponding primary amines to afford Schiff Base derivatives TC-5∼TC-10 for further study. Anti-inflammatory and analgesic activities of those derivatives were determined by using carrageenan-induced rat paw edema test. The analgesic activities of the derivatives were determined by the hot-plate test.

The oleanane-type triterpene chichipegenin and the sterols peniocerol and macdougallin, isolated from Myrtillocactus geometrizans, showed anti-inflammatory activities in both the 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse ear edema model and the carrageenan- induced rat paw edema model. All tested compounds inhibited the TPA-induced edema in a dose-dependent manner, with ED50 values less than or equal to that shown by indomethacin. Among them, peniocerol was the most active compound. However, only peniocerol and macdougallin reduced carrageenan-induced rat paw edema. On the other hand, peniocerol and macdougallin showed cytotoxicity against several human cancer cell lines. These results indicate that compounds isolated from M. geometrizans possess antiinflammatory and cytotoxic properties, and the presence of chichipegenin in the aerial parts could justify the medicinal uses attributed to the plant.

The present study evaluates the analgesic and anti-inflammatory activities of the methanolic extract from Jatropha curcas (Euphorbiaceae) leaves to provide experimental evidence for its traditional use. Investigations on the analgesic effects and anti-inflammatory activities of J. curcas were carried out by utilizing the different animal models. It has been shown that the methanolic extract from Jatropha curcas leaves significantly increased pain threshold and reduced writhing response as well as inhibited the increase in vascular permeability. Also, it significantly decreased the carrageenan-induced rat paw edema. The results show that the methanolic extract from Jatropha curcas leaves have both central and peripheral analgesic activities and as anti-inflammatory effects, supporting the traditional application of this herb in treating various diseases associated with inflammation and pain.

Tamarindus indica L. is highly valued in Indian medicine for management of painful inflammation, diabetes and constipation. In the present study, the anti-inflammatory potential was evaluated using carrageenan induced rat paw edema model. The coarse seed powder was extracted with distilled water, ethyl acetate and methanol and submitted to chemical tests which revealed the presence of alkaloids, tannins, flavonoids, glycosides, phenolic compounds and steroids. All the three extracts i.e. aqueous, ethyl acetate and methanolic extract of Tamarindus indica seeds at 500 mg/kg showed significant anti-inflammatory activity in the second phase of inflammation. For comparison purpose, we used indomethacin (10 mg/kg) as a reference drug. The results showed that methanolic extract exhibited more promising anti-inflammatory activity compared to aqueous and ethyl acetate extracts.
