The aim of the study was to investigate the distribution and content of neuroglobin in the pyramidal neurons of the frontal and parietal cortex of white rats during simulated cholestasis.Material and methods. The study included 60 outbred white male rats weighed 200–250 g. Cholestasis was simulated by ligation of the common bile duct in the porta hepatis area (main group, n=30). Animals of the control group (n=30) were performed a false operation preserving physiological bile outflow. Sections of the frontal and parietal cortex of white rats were selected for investigation. The content and distribution of neuroglobin was detected immunohistochemically on paraffin sections using mouse monoclonal primary antibodies Anti- Neuroglobin antibody (Abcam). Cytophotometric analysis was used to perform quantitative assessment of the content of the studied molecular marker; after that statistical analysis was performed.Results. It was found that the content of neuroglobin in the pericarions of neurons of the frontal and parietal cortex modified in a wave-like mode. After ligation of the common bile duct, the content of neuroglobin significantly decreased in 2, 10 and 45 days (minimum on the 10th day) in the frontal cortex and in 90 days in the parietal cortex, and increased in 5 and 20 days of the experiment (maximum on the 20th day).Conclusion. In cholestasis, the content of neuroglobin in the pericarions of neurons of the frontal and parietal cortex modifies in waves: a decrease is observed on the 2nd, 10th, 45th day; an increase is observed on the 5th and 20th day; a recovery is observed on the 90th day.

The morphological condition of the bile ducts remains one of the most important problems of modern medical science. In order to obtain an analgesic effect in patients with acute cholangitis, opioids are often used. However, information on the effectiveness of opioids in the treatment of pathological conditions of the bile ducts is contradictory. The rapidly progressive destruction of the intrahepatic bile ducts associated with the use of narcotic agents has been described. Further study of the effect of opioids on the structural organization of the common bile duct is relevant. In order to establish the morphological state of the common bile duct under conditions of long-term opioid exposure, a study was performed on 24 sexually mature white male rats, aged 3.5-5.0 months and weighing 180-200 g, which were injected intramuscularly with Nalbuphine for 6 weeks. The study material is represented by histological specimens of the common bile duct of white rats. The “Aver Media” computer system was used to photograph microspecimens. The “ImageJ” computer program was used to measure the diameter of the lumen and the wall thickness of the common bile duct. After 2 weeks of Nalbuphine administration to white rats, plethora of wall microvessels and a significant increase in the longitudinal diameter of the lumen of the common bile duct were observed. After 4 weeks of the experiment, the common bile duct was dilated, the transverse and longitudinal diameters of its lumen almost doubled, pathological changes in its wall had all the signs of inflammation. In the later stages of the experiment (introduction of Nalbuphine for 6 weeks), the pathological changes increased and manifested by destructuring the wall of the common bile duct, disorganization of cholangiocytes, thinning of the cell layer due to detachment of cholangiocytes, polymorphism of their nuclei, destruction of intercellular junctions, stratification of its own plate, vacuolar dystrophy of the muscular membrane “varicose” expansion of venules, significant smooth muscle hyperplasia of arterioles, the presence of perivascular lymphocytic infiltrates in the duct wall.

Cholestasis (disturbance of the liver bile outflow into the duodenum) is a frequent complication of cholelithiasis and other pathologies of the hepatobiliary system. In this case, all metabolism types are disturbances and all body organs and systems, including the nervous system, are affected. The objective of the study was to establish changes in the c-fos immunoreactivity in the cerebellum neurons of rats at different time after modeling subhepatic cholestasis. In this work, we used a material of 60 white male rats 200–250 g in weight. In experimental animals, the ligation of the common bile duct was carried out, in control animals – a false operation while maintaining a physiological bile flow in the duodenum throughout the experiment. Subhepatic cholestasis in rats is accompanied by an increase in the cortex cerebellum on the 2–20th days after the ligation of the common bile duct of the number of neurons with the increased c-fos-immunoreactivity, with the maximum one on the 10th day of the experiment. In surviving animals, after the removal of cholestasis in the long term (45–90 days), the number of neurons with the increased c-fos immunoreactivity, as well as the expression of this protein in neurons is normalized.

The aim of this study was to compare the efficacy and safety of laparoscopic primary closure of the common bile duct (CBD) combined with percutaneous transhepatic cholangiographic drainage (PTCD) and laparoscopic choledocholithotomy with T-tube placement for the treatment of CBD stones. Between January 1991 and July 2002, 50 patients with choledocholithiasis and a CBD diameter larger than or equal to 1 cm underwent laparoscopic CBD explorations. The study group consisted of 10 patients undergoing laparoscopic primary closure of the CBD combined with PTCD. The control group consisted of 40 patients undergoing laparoscopic choledocholithotomy with T-tube placement. Parameters were compared statistically. The study group showed higher female/male ratio (6/4 vs 8/32, P = 0.02), less stone numbers (1.90 ± 0.88 vs 3.40 ± 1.65, P = 0.0078), shorter operation time (138 ± 37 minutes vs 191 ± 75 minutes, P = 0.014), and shorter postoperative stays (7 ± 3 days vs 10 ± 3 days, P = 0.0013). It seems that laparoscopic primary closure of the CBD combined with PTCD can shorten the operation time and postoperative stays as compared with laparoscopic choledocholithotomy with T-tube placement for the treatment of CBD stones.

PURPOSE: To evaluate the effects of biliary stenting in rats through analysis of collagen and elastin deposition in the bile ducts. METHODS: Twenty male rats underwent midline laparotomy, duodenotomy and transampullary stenting of the common bile duct with a 22Fr plastic stent. Animals were randomized in 4 groups, with 5 components in each: (I) control, (II) biliary stenting for 7 days, (III) biliary stenting for 14 days, and (IV) biliary stenting for 30 days. Sections of the common bile duct were stained using Hematoxylin Eosin; Sirius Red to quantitate the amount of collagen present; and Weigert's Resorcin-fuchsin to quantitate the amount of elastin present. RESULTS: The percentage of area stained for collagen was 13.4; 21.5; 29.5 and 32.8, for groups I to IV, respectively. The percentage of area stained for elastin fibers was 7.0; 5.2; 4.0 and 2.9, for groups I to IV, respectively. Collagen/Elastin ratio was 2.4; 5.1; 11.0 and 14.4 for groups I to IV, respectively. CONCLUSIONS: The biliary stenting leads to collagen and elastin deposition in the bile ducts; and collagen deposition and collagen/elastin ratio are proportional to the period of stenting.

The purpose of this study was to determine the anti-inflammatory activity test of ethanol extract of lotus seeds in rats induced by carrageenan. The research method used was experimental. The object used in this study was male white rats. Observations were made for 6 hours and then the data were analyzed using the one way analysis of variance (ANOVA) test. The anti-inflammatory activity test was carried out on the test animals which were divided into 5 groups, 3 each with the suspension test group with a dose of 200, 300, 400 mg/kg bw, the negative control group was the CMC Na 1% suspension and the positive control was diclofenac Na 2, 25 mg/kg bw. The results showed that the ethanol extract of the seeds of lotus (Nelumbo nucifera G) was proven to have an anti-inflammatory effect on carrageenan-induced male white rats at doses of 200, 300 and 400 mg/kg BW with the percentage value of inflammation inhibition at the 6th hour was 65.79, 74.65 and 80.77%, with the most effective dose in reducing anti-inflammatory drugs being 400 mg/kg bw. The ethanol extract of the seeds of lotus (Nelumbo nucifera) has anti-inflammatory activity. Keywords: anti-inflammatory activity, Nelumbo nucifera, paw edema

Objective: This study aimed to observe the effect of the 70% ethanol extract of Orthosiphonis stamineus Benth leaves on the pharmacokineticparameters of furosemide in white male rats.Methods: 18 Sprague–Dawley male rats were divided into three groups: The normal control group was given only 1% carboxymethyl cellulose,the furosemide group was given 7.2 mg/200 g body weight (BW) suspension of furosemide, and the combination group was given 700 mg/kg BWsuspension of the 70% ethanolic extract of O. stamineus Benth leaves for 4 days followed by a 7.2 mg/200 g BW suspension of furosemide. On the4th day of treatment, we performed orbital sinus blood sampling on the eyes of the rats and analyzed the levels of furosemide in plasma using highperformanceliquid chromatography.Results: Therefore, the results showed that the administration of the 70% ethanol extract of O. stamineus Benth leaves improves the pharmacokineticparameters of furosemide on Cpmax and the area under the curve (p<0.05).Conclusion: This study concludes that the 70% ethanol extract of O. stamineus Benth leaves improves the pharmacokinetic parameters of furosemidein white male rats.

Abstract Background Cholestasis is the obstruction or the reduction in bile flow that results in intrahepatic accumulation of bile constituents, which progresses to develop liver pathology. Common bile duct ligation (BDL) in rodents is an experimental model of cholestasis that has been carried out in research for many years. BDL model of cholestatic liver injury involves other mechanisms, including oxidative stress, inflammation, and fibrogenesis. Antioxidant, antiinflammatory or antiapoptotic properties gained much interest for the amelioration of liver dysfunction. Aim the aim of this study is to assess the possible protective effects of onion supplementation on hepatic structural and functional alterations induced by BDL in rats, which reflect the effects of cholestasis resulting from intrahepatic accumulation of bile. Methods Thirty adult female Wistar rats were randomly and equally allocated into three groups: (1) control group, (2) BDL group; subjected to ligation of the common bile duct and (3) Onion-supplemented BDL groups (O-BDL). Both control and BDL groups received distilled water (solvent for onion powder) daily by gavage for 4 weeks. Onion-supplemented BDL group (O-BDL); subjected to ligation of the common bile duct and then received 500 mg/kg of onion powder dissolved in distilled water, daily by gavage for 4 weeks. At the end of the experimental period, plasma levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), direct bilirubin, total proteins, total antioxidant capacity (TAC), tumor necrosis factor-α (TNF-α), and hepatic tissue level of malondialdehyde (MDA) and transforming growth factor-β (TGF-β) were measured for all groups. In addition, histopathological examination of liver tissue samples was performed for the three groups. Results Plasma levels of ALT, AST, ALP, direct bilirubin, TNF-α and hepatic tissue levels of MDA and TGF-β were significantly increased and TAC was significantly decreased in the BDL group compared to the control group. In addition, altered architecture was detected in hepatic tissue samples of BDL group. Onion supplementation significantly decreased the plasma levels of ALT, AST, ALP, direct bilirubin, TNF-α and hepatic tissue levels of MDA and TGF-β in the O-BDL group when compared to the BDL group. Total proteins level was not significantly different among all the studied groups. In addition in O-BDL group, histopathological examination of liver revealed near normal structure of hepatic tissue. Conclusion BDL induces hepatic structural alterations and functional disturbances. Onion supplementation inhibits inflammation and oxidative insults that associate BDL, and subsequently protects against BDL-induced liver injury.

Objective: To compare the number of apoptosis, necrosis and viability of germinal epithelial cells in the contralateral testicle (CT) and plasma testosterone (PT) levels in white male rats which administered Nifedipine after unilateral testicular torsion (TT) compared with control group. Material & Methods: This research was an experimental study using male white rats (Rattus Norvegicus, Wistar strain) aged 10-12 weeks and body weight 150-200 gram. A total of 30 rats were then randomly divided into 5 groups (n=6) which were negative control group (KN), positive control group (KP1 and KP2) and Nifedipine-administered group (N1 and N2). Each group performed unilateral left side torsion of testicular of 1080o anticlockwise except the KN group. There was 4-hour ischemic duration in the KP1 and N1 groups while 10 hours in the KP2 and N2 groups. Administration of Nifedipine 30 minutes before detorsion by intraperitoneal injection dosed 100 μg/kgBW. All groups performed right orchidectomy and plasma blood sampling. Measurement of apoptosis, necrosis and viability of germinal epithelial cells in the CT using flowcytometry. Measurement of PT levels using Enzyme-Linked Immunosorbent Assay (ELISA). Results: The number of apoptosis, necrosis and viability of contralateral testicular germinal epithelial cells and PT levels in the KN group compared with KP1 and KP2 groups were significantly different (p<0.05). There was no significant discrepancy in apoptosis (p>0.05) in KP1 group compared with N1 group, as well as in KP2 group compared with N2 groups. The number of necrosis, viability of germinal epithelial cells in the CT and PT level in KP1 group in compared with N1 group, as well as in KP2 group in compared with N2 group were significantly different (p<0.05). Conclusion: Nifedipine administration prior to testicular detorsion can maintain cell viability and decrease the amount of necrotic germinal epithelial cells in the CT and prevent the decrease in PT levels after unilateral TT.

Background. Photodynamic therapy of liver diseases involves the introduction of photosensitizers into the common bile duct. Objective – to evaluate the effect on animals of under pressure introduction of the photosensitizer “Photolon” into the common bile duct. Material and methods. The control group was injected with physiological solution under pressure, the experimental one – with “Photolon”. The vital signs of the animals, the laboratory data were studied, a histological examination of the liver and duct was carried out. Results. The blood parameters of the experimental animals and their postoperative condition were not statistically different from those of the control group (p≤0.05). The liver and common bile duct had normal histological structure. Conclusions. The under pressure introduction of the photosensitizer “Photolon” into the common bile duct causes no morphological changes in the liver tissues and common bile duct and has no toxic effect on animals.

Abstract The aim of this animal study was to evaluate the preventive effect of novel biodegradable stents (BS) on biliary stricture and fibrosis after duct-to-duct (DD) biliary anastomosis. Ten mini-pigs were allocated to the control group (n = 5) and or the stent group (n = 5). The common bile duct was exposed through surgical laparotomy, and then resected transversely. In the stent group, a 4 mm or 6 mm polydioxanone/magnesium sheath core BS was inserted according to the width of the bile duct, followed by DD biliary anastomosis. In the control group, DD biliary anastomosis was performed without BS insertion. In the stent group, stents were observed without deformity for up to four weeks in all animals. Eight weeks later, histopathologic examination revealed that the common bile duct of the anastomosis site was relatively narrower in circumference in the control group compared to the stent group. The degree of fibrosis in the control group was more marked than in the stent group (3.84 mm vs. 0.68 mm, respectively, p < 0.05). Our study showed that novel BS maintained their original shape and radial force for an adequate time and then disappeared without adverse events.
