Abstract Nephrolithiasis appear to be a major health crisis among the population with serious medical related consequences throughout the lifetime of patient. The aim of the study was to evaluate the preventive effect of the hydro-alcoholic extract of A. lanata roots on Urolithiasis rats. Thirty adults male wistar Albino rats weighing 200 – 250 g were divided into five groups comprising 6 rats in each. Group I Served as positive control with water ad libitum. Group II as negative control which is disease treated group receiving 0.75% ethylene glycol mixed with drinking water for 28 days. Group III chosen as standard group receiving ethylene glycol for first 14 days and Cystone 750 mg/Kg from day 15 till day 28. Group IV and V received ethylene glycol for first 14 days and treatment regimen of LD (400 mg/Kg) and HD 800 mg/Kg orally from day 15 till day 28. Invitro studies like Nucleation, Aggregation and Growth assays were performed. Urine volume and pH was collected and observed for change in appearance, pH, odour and turbidity. Extract was given by preparing suspension and stability was observed by measuring its parameters. On Day 29, the kidneys were dissected and histopathology was done to check tubular injury. There was Increase in urine volume, enhanced excretion of urinary constituents like citrate, oxalate etc. and improving clearance rate. Improvement in pH and antioxidant activity was observed in treated groups. The extract showed that it has prominent effect on nephrolithiasis and has better safety profile in the dose given.

This study was designed to evaluate the Lithotriptic potentials of Phyllantus fraternus methanol leaf extract on ethylene glycol-induced kidney calculi in albino rats. Ethylene glycol (1% v/v) was administered in their drinking water for a period of 28 days. The Treatment was done with the extract at 200 mg/kg, 400 mg/kg and 600 mg/kg body weights. Cystone® at 500 mg/kg body weight was also given for a period of 21 days to the standard control group. The serum parameters such as calcium, phosphates, magnesium and albumin were measured and evaluated. The results for the Lithotriptic activity, where the kidney homogenates were analyzed are described as thus, the phosphate concentrations when compared were significant (p<0.05) between the groups’ 600 mg/kg body weight (9.61 ± 1.17) and the normal control (5.67 ± 0.70). Significant differences (p<0.05) for phosphates were also observed between 600 mg/kg (9.61 ± 1.17) and 200 mg/kg body weights (12.06 ± 0.51); 400 mg/kg (7.64 ± 0.44) and 200 mg/kg body weights (12.06 ± 0.51) and the 200 mg/kg and standard control groups Cystone® (7.96 ± 0.56) respectively. Significant differences (p<0.05) were also observed for phosphates concentration, when the normal control (5.67 ± 0.70) was compared to the 400 mg/kg body weight (7.64 ± 0.44) and the standard control group Cystone® (7.96 ± 0.56). From this study, it can be deduced that, the presented data indicated that, the administration of Phyllanthus fraternus methanol leaf extract to rats with ethylene glycol-induced kidney calculi, reduced and prevented the growth of kidney calculi, supporting the folklore claim regarding its Lithotriptic activity.

Lawsonia inermis Linn., commonly called as Henna belongs to family Lythraceae. Traditionally, it has been reported to have therapeutic activity in bronchitis, diabetes, antimicrobial, antibacterial, trypsin inhibitory, cytotoxicity, wound healing, antioxidant, anti-inflammatory, analgesic. However, the antiurolithiatic activity of the bark extract of  L. inermis Linn. is not known. In this study, we investigated protective effect of the alcoholic extract of L. inermis bark against ethylene glycol induced urolithiasis and its possible underlying mechanisms using male wistar albino rats. Animals were divided into seven groups and urolithiasis was induced by ethylene glycol (0.75% v/v) in drinking water for 28 days. Methanolic extract of Lawsonia inermis (MELI) bark (300 & 500 mg/kg, p.o.) were administered once daily from 15th day to 28th day as curative regimen and from 1st day to 28th day as preventive regimen. Cystone (750 mg/kg, p.o.) was used as a standard drug. After 28 days, various biochemical parameters like urine volume, pH were measured. Calcium, phosphate and oxalate were measured in urine and kidney homogenate. Serum creatinine, uric acid and urea nitrogen were estimated. Histopathology of kidney also studied. Treatment with the MELI extract significantly restored all elevated parameters including calcium, phosphate and oxalate in urine and kidney homogenate; creatinine, uric acid and urea nitrogen in serum when compared to model control group. The histopathological study of the kidney also supported the above results. It can be concluded that methanolic extract of bark of Lawsonia inermis Linn. has significant antiurolithiatic effect in experimental rats.
