Cocoa is one of the leading plantation commodities in Indonesia. The part of cocoa trees that can be found in abundance is the leaf. The leaves cut out of cocoa trees are utilized by some farmers in composting, when in fact cocoa leaves have a great potential to be processed into health care products. The present study aims to determine the effect of high altitude landscape on antibacterial activities in ethanolic extract of cocoa leaves (Theobroma cacao, L). For the experiment, we employed a Completely Randomized Design (CRD) that is divided into 3 treatments—Aqua Dest (Control), ethanolic extract of high-altitude cocoa leaves (P1), and ethanolic extract of low-altitude cocoa leaves (P2)—repeated 5 times. The results show that the largest resistance zones of Eschericia coli and staphylococcus aureus are created by ethanolic extract of high-altitude cocoa leaves with the size of 19.96 mm and 20.52 mm, respectively. Our ANOVA test shows that extracts of both high- and low-altitude cocoa leaves have an effect on the growth of S.aureus (P = 0,000) and E. coli (P = 0,000) bacteria. From Duncan’s test we can conclude that ethanolic extract of high-altitude cocoa leaves differs significantly (P<0.05) from its lower altitude counterpart in inhibiting the growth of E.coli and S.aureus bacteria.

AbstractCocoa (Theobroma cacao L.), one of the most important export commodities from Indonesia, is widely planted with current total area of 1.6 million Ha, producing 500.000 metric tons of dry bean  in 2011 . At the time of harvest, instead of seed approximately the same volume cacao husk is produced. The aim of the study was to assess the potential of cocoa husk extract as an antibacterial against Escherichia coli, Bacillus subtilis, and Staphylococcus aureus, and to determine the minimum inhibitory concentration (MIC) of cocoa husk extract to the three test bacteria. Extraction of cocoa husk conducted by maceration method using ethanol 96%. Analysis of antibacterial activity was done by paper disc diffusion method. Completely Randomized Design of single factor presentage that is extract concentration of 0; 1; 2; 4; 8; 16; 32; and 64% (g/mL) with three replicans were applied.The results showed that the extract of cocoa pod husk has antibacterial activity against S. aureus, B. subtilis, and E. coli with the MIC are 8% (g/ mL), 16% (g/ mL), and 32% (g/ mL) respectively.AbstrakKakao (Theobroma cacao L.), salah satu komoditi ekspor terpenting Indonesia, ditanam secara luas dengan total luasan 1,6 juta Ha, menghasilkan 500.000 ton biji kering pada tahun 2011. Penelitian ini bertujuan untuk mengkaji potensi ekstrak kulit buah kakao sebagai antibakteri terhadap Escherichia coli, Bacillus subtilis, dan Staphylococcus aureusserta menentukan konsentrasi hambat minimum (KHM) ekstrak kulit buah kakao terhadap ketiga bakteri uji. Ekstraksi kulit buah kakao dilakukan dengan metode Maserasi menggunakan pelarut etanol 96%. Analisis aktivitas antibakteri dilakukan dengan metode difusi cakram  kertas. Penelitian  ini  menggunakan  Rancangan Acak  Lengkap (RAL) dengan faktor tunggal  konsen-trasi ekstrak, yaitu 0; 1; 2; 4; 8; 16; 32; dan 64% (g/mL), masing-masing dengan tiga kali ulangan. Hasil penelitian menunjukkan bahwa ekstrak kulit buah kakao berpotensi sebagai antibakteri terhadap S. aureus, B. subtilis dan  E. coli, dengan KHM berturut-turut adalah 8% (g/mL), 16% (g/mL), dan 32% (g/mL).

Contamination of meats and meat products with foodborne pathogenic bacteria raises serious safety issues in the food industry. The antibacterial activities of phosphorous-fluorine co-doped TiO2 nanoparticles (PF-TiO2) were investigated against seven foodborne pathogenic bacteria: Campylobacter jejuni, Salmonella Typhimurium, Enterohaemorrhagic E. coli, Yersinia enterocolitica, Shewanella putrefaciens, Listeria monocytogenes and Staphylococcus aureus. PF-TiO2 NPs were synthesized hydrothermally at 250 °C for 1, 3, 6 or 12 h, and then tested at three different concentrations (500 μg/mL, 100 μg/mL, 20 μg/mL) for the inactivation of foodborne bacteria under UVA irradiation, daylight exposure or dark conditions. The antibacterial efficacies were compared after 30 min of exposure to light. Distinct differences in the antibacterial activities of the PF-TiO2 NPs, and the susceptibilities of tested foodborne pathogenic bacterium species were found. PF-TiO2/3 h and PF-TiO2/6 h showed the highest antibacterial activity by decreasing the living bacterial cell number from ~106 by ~5 log (L. monocytogenes), ~4 log (EHEC), ~3 log (Y. enterolcolitca, S. putrefaciens) and ~2.5 log (S. aureus), along with complete eradication of C. jejuni and S. Typhimurium. Efficacy of PF-TiO2/1 h and PF-TiO2/12 h NPs was lower, typically causing a ~2–4 log decrease in colony forming units depending on the tested bacterium while the effect of PF-TiO2/0 h was comparable to P25 TiO2, a commercial TiO2 with high photocatalytic activity. Our results show that PF-co-doping of TiO2 NPs enhanced the antibacterial action against foodborne pathogenic bacteria and are potential candidates for use in the food industry as active surface components, potentially contributing to the production of meats that are safe for consumption.

Cocoa (Theobroma cacao L.) development using modern breeding techniques can be facilitated by propagation of planting material through somatic embryogenesis. Various factors that may affect embryogenesis are the composition of culture medium and culture condition. Hormone commonly used to initiate the formation of callus is auxin with type 2.4-D (2.4 Dichlorophenoxy acetic acid). The aim of this study was to determine the effect of the addition of 2.4 -D hormoneson the process of cocoa embryogenesis. The treatments were arragged in factorial combination in completely randomized design, which consisted of two factors. Thefirst factor was the concentration of auxin 2,4-D 25 %, 50 %, 75 %, and 100 %; and the second factor was cocoa clones; Sulawesi 01 and Sulawesi 02. The resultshowed that the addition of 2.4-D hormone up to 100% on somatic embryogenesis of cocoa for Sulawesi 01 clone was not significantly different from Sulawesi 02 clone for all parameters. While on the addition of 2.4-D, there was significant difference between Sulawesi 01 and 02. Cocoa embryogenic callus using the addition of 2.4-D (25%-100%) was significantly different from control. Increased concentrations of 2,4-D hormone which is applied onto media would inhibit the formation of the somatic embryo. This experiment was conducted in order to optimize and quantify the acceptability of mango jam enriched with pectin extracted from husk of cacao grown in the department of Sucre, Colombia. The experiment was performed using a two-factor, completely randomized design: pectin concentration (0.2%, 0.4%, 0.6%) and citric acid concentration (0.1%, 0.3%, 0.5%). All experiments were conducted in triplicate for each treatment, for a total of 27 experimental units. Variables such as fresh fruits (65.5%) and sugar (34.0%) were held constant. Data analysis was performed using an analysis of variance while the comparison of means was performed by the Tukey's range test. After 24 hours, a test for measuring product acceptability was conducted among 60 people using a 7-point hedonic scale. The survey yielded an acceptability value (I like moderately) of 6.7 for pectin concentrations of 0.2% and citric acid concentrations of 0.3%.

Objective: The objective of the study was to evaluate whether ethanolic extracts of Nelumbo nucifera (EENN) seedpod and quercetin (active component of NN) possess antibacterial proprieties against Gram (-) bacteria such as Escherichia coli and Pseudomonas aeruginosa and Gram (+) bacteria such as Staphylococcus aureus. Methods: Antibacterial activities of EENN seedpod and quercetin were investigated using disc diffusion method, minimum inhibitory concentration against E. coli and P. aeruginosa and Gram (+) bacteria such as S. aureus. Results: The antibacterial activity of both EENN seedpod and quercetin was found to be increased in dose-dependent manner. The maximum zone of inhibition was exhibited by both EENN seedpod and quercetin against E. coli (14 mm and 15 mm) and P. aeruginosa (13 mm and 15 mm). Gram-negative bacteria were more susceptible to the EENN seedpod extract and quercetin than Gram-positive bacteria.Conclusion: The results of the present study suggested that the effect of EENN seedpod and quercetin against the tested bacteria in vitro may contribute to the in vivo activities of the EENN seedpod and quercetin.

This research was conducted at the experimental farm "La Maria" State Technical University of Quevedo, located at Km 7.5 Vía Quevedo - El Empalme, with the overall objective to determine the effect of different organic substrates on seedling growth cocoa nursery conditions. The specific objectives were to evaluate the growth of cocoa plants in different substrates study, identify the substrate that allows greater root development of seedlings of cacao and perform economic analysis of each of the treatments studied. The experiment was conducted under a completely randomized design with 10 treatments in three replicates and treatment means were compared with Duncan test at 95% probability. The treatments were: T1: 60% Bokashi + 40% black earth, T2: 60% Bokashi + 40% sawdust raft, T3: 60% Bokashi + 40% chaff burnt rice, T4: 40% Bokashi + 20% sawdust balsa + 20% black soil + 20% chaff burnt rice, T5: 60% vermicompost + 40% black earth, T6: 60% vermicompost + 40% sawdust raft, S7: 60% vermicompost + 40% chaff burnt rice, T8: 40% vermicompost + 20% black soil + 20% chaff burn rice + 20% sawdust raft, T9: 30% bokashi + 30% vermicompost + 20% black earth + 10% sawdust + 10% balsa burning rice chaff, T10: 75% + 25% black earth rice husks. The main results of this research it was observed that the planted in treating 9 (30% Bokashi, 30% vermicompost, 20% black soil, 10% sawdust balsa and 10% chaff burnt rice), was recorded 100% germination, plant also produced in this substrate were greater height, stem diameter, presence of leaves and root length. Treatment 10 (75% of black earth and 25% rice husks) produced the highest returns with 59.68% of the production of 1000 seedlings, meaning that for every dollar invested a profit of $ 0.60 was obtained for every dollar reversed, however, with treatment 9 (30% bokashi, 30% vermicompost + 20% black soil + 10% sawdust raft + 10% chaff burnt rice) plants showed better characteristics that are the basis for acceptance by the buyer, generating a return of 34.23%, as a result of higher production cost.

The aim of this study was to determine the effect of giberrelin (GA3) storage durationand treatment on the viability of cocoa (Theobroma cacao L.) seeds carried out at theLaboratory of Indonesian Center for Seedling and Plant Protection (BBPPTP) Surabayaon Mojoagung No. No. 52, Mojoagung Subdistrict, Jombang Regency in February toApril 2014. This study used a completely randomized design with factorial patterns with2 factors. The first factor is the storage time which consists of 4 stages P1 (7 days ofnatural storage of seeds), P2 (14 days of storage of natural seeds), P3 (22 days of storageof natural seeds), and P4 (29 days of storage of natural seeds). The second factor isinvigoration treatment which consists of 3 types without treatment (I0), GA3 10 ppm(I1), GA3 20 ppm (I2). The results showed a significant interaction between storage timeand invigoration treatment of plant height and germination capacity, storage durationtreatment affected the variable number of leaves, 7 days storage time (P1) produced theaverage number of leaves (3.75 strands) even though the result is the same as the storageperiod of 14 days (P2). While the invigoration treatment had no effect on the observationvariable of the number of leaves.

Cymodocea rotundata  merupakan salah satu jenis lamun yang berpotensi sebagai antibakteri. Senyawa bioaktif yang bersifat sebagai antibakteri adalah fenol, flavonoid dan tanin. Tujuan penelitian ini adalah untuk mengetahui perbedaan lama inkubasi dan konsentrasi ekstrak lamun C. rotundata yang berbeda terhadap aktivitas antibakteri S. aureus dan E. Coli. Metode penelitian yang digunakan yaitu experimental laboratories dengan menggunakan rancangan dasar penelitian Rancangan Acak Lengkap (RAL) dengan pola faktorial, pola terbagi oleh faktor lama inkubasi bakteri (24 jam, 48 jam dan 72 jam) dan perbedaan konsentrasi ekstrak lamun (5%, 10% dan 15%). Data dianalisis menggunakan SIDIK RAGAM dan dilakukan analisis lanjut Beda Nyata Jujur (BNJ), apabila terdapat perbedaan pada perlakuan. Ekstrak lamun C. rotundata efektif sebagai antibakteri dengan kategori sedang yaitu zona hambat berkisar antara 5-10 mm. Konsentrasi optimum untuk menghambat S.aureus dan E.coli adalah 15% dengan lama inkubasi 48 jam dengan zona hambat yang dihasilkan masing-masing sebesar 6, 123 mm. Dan 5, 833 mm.Cymodocea rotundata is a type of seagrass that has a potential as an antibacterial. Bioactive compounds which act as such as antibacterial phenols, flavonoids and tannins. The purpose of this study was to determine the effect of different concentration and time of incubation  of seagrass C.rotundata as antibacterial against  S. aureus and E. coli. The method used was experimental laboratories using the basic design of the study completely randomized design (CRD) with factorial pattern (2 factor). The first factor were different concentration (5%, 10% and 15% ) and second factor were time of incubations (24 hours, 48 hours and 72 hours). Data were analyzed using ANOVA and conducted a further test Honestly Significant Difference (HSD), if treatment gave significant effect. C. rotundata extracts had antibacterial activity with medium category which inhibition zone ranges from 5-10 mm. The optimum concentration for inhibiting S. aureus and e. coli were 15% with incubation time of 48 hours resulting inhibition zone 6,123 mm and 5,833 respectively.

The study aimed to evaluate the growth and quality of SJ-02 cocoa seedlings under different irrigation depths and cultivation container sizes. The experiment was arranged as a completely randomized design, in a factorial scheme (4x3), where the first factor consisted of the application of four different irrigation depths (6, 10, 12 and 14 mm d-1) and the second one was composed of three different cultivation containers (small, medium and large). Twelve treatments were evaluated with eight repetitions, totaling 96 plants in the experimental field. At 74 days after sowing, the leaf area, stem diameter, plant height, dry mass of the aerial part, dry mass of the root system, total dry mass and Dickson's quality index of all seedlings were analyzed. The 10.32 mm d-1 irrigation depth and the medium and large cultivation containers provided the highest quality in seedling production, being the most suitable ones. However, there was no interaction between the factors
