Abstract The spread of drug-resistant bacteria via food has contributed to the dissemination of resistant bacteria among humans. However, the status of food contamination with resistant bacteria, particularly the quantitative level of resistant bacteria in food, has not yet been well elucidated. In this study, the abundance of colistin-resistant Escherichia coli in meat samples was quantified to understand the origin of the contamination of meat available in local Vietnamese markets. Fifteen samples each of chicken and pork meat purchased from local Vietnamese markets were assessed for the presence of colistin-resistant E. coli with the mobile colistin resistance gene mcr. The results showed that 40% (6/15) and 66% (10/15) of the pork and chicken meat samples, respectively, were contaminated with colistin-resistant E. coli. The median quantitative levels of colistin-resistant E. coli in the contaminated pork and chicken samples were 1.8 × 104 and 4.2 × 103 CFU/g, respectively. The results of phylogenetic analysis of isolates from a chicken meat sample showed that the contaminated colistin-resistant E. coli were a mix of multiple phylogenetical clones of bacteria that may have multiplied during sale. This is the first study to quantify the abundance of colistin-resistant E. coli in meat samples.

Abstract Although the spread of plasmid-mediated antibiotic-resistant bacteria is a public health concern, food contamination with plasmid-mediated antibiotic-resistant Escherichia coli has not been well investigated in Vietnam. The aim of this study was to describe the prevalence of colistin-resistant, carbapenem-resistant and endemic blaCTX−M in extended-spectrum β-lactamase (ESBL)-producing E. coli isolates. Colistin- and carbapenem-resistant ESBL-producing E. coli were isolated from chickens in Vietnam and Japan. The results showed that 52% and 93% of Vietnamese chicken was isolated with colistin-resistant and AmpC/ESBL-producing E. coli, respectively, while 52.7% of Japanese chickens were isolated with AmpC/ESBL-producing E. coli. Carbapenem-resistant E. coli has not been isolated in Vietnam or Japan. Genotyping revealed that colistin-resistant E. coli harboured mcr-1, and most of the AmpC/ESBL-related genes were blaCTX−M−55 and blaCTX−M−65 together with blaTEM in Vietnamese chickens, and blaCMY−2 in Japanese chickens. Multidrug resistance analysis showed that ESBL-producing E. coli isolates were more resistant to quinolones, streptomycin, and chloramphenicol compared with colistin-resistant E. coli isolates from Vietnam, suggesting selection in ESBL-producing E. coli for multiple antibiotic resistance genes. In conclusion, colistin-resistant E. coli was detected in about half of the chicken meat samples, the majority of which were found to harbour mcr-1. The high prevalence of ESBL-producing E. coli has remained constant across the last five years, and the predominant blaCTX−M for ESBL-producing E. coli was found to be blaCTX−M−55 or blaCTX−M−65, with the coexistence of blaTEM in Vietnam. Our results can be implemented in monitoring systems to combat the development of antimicrobial resistance.

The dissemination of antimicrobial resistance (AMR) bacteria has been associated with the inappropriate use of antibiotics in both humans and animals and with the consumption of food contaminated with resistant bacteria. In particular, the use of antibiotics as prophylactic and growth promotion purposes in food-producing animals has rendered many of the antibiotics ineffective. The increased global prevalence of AMR poses a significant threat to the safety of the world’s food supply. Objectives: This study aims at determining the prevalence of antibiotic-resistant Escherichia coli (E. coli) isolated from local and imported retail chicken meat in Qatar. Methodology: A total of 270 whole chicken carcasses were obtained from three different hypermarket stores in Qatar. A total of 216 E. coli were isolated and subjected to antibiotic susceptibility testing against 18 relevant antibiotics using disc diffusion and micro- dilution methods. Furthermore, extended-spectrum β-lactamase (ESBL) production was determined via a double-disc synergetic test. Isolates harboring colistin resistance were confirmed using multiplex-PCR and DNA sequencing. Results: Nearly 89% (192/216) of the isolates were resistant to at least one antibiotics. In general, isolates showed relatively higher resistance to sulfamethoxazole (62%), tetracycline (59.7%), ampicillin and trimethoprim (52.3%), ciprofloxacin (47.7%), cephalothin, and colistin (31.9%). On the other hand, less resistance was recorded against amoxicillin/clavulanic acid (6%), ceftriaxone (5.1%), nitrofurantoin (4.2%) and piperacillin/tazobactam (4.2%), cefepime (2.3%), meropenem (1.4%), ertapenem (0.9%), and amikacin (0.9%). Nine isolates (4.2%) were ESBL producers. Furthermore, 63.4% were multidrug-resistant (MDR). The percentage of MDR, ESBL producers, and colistin-resistant isolates was significantly higher among local isolates compared to imported chicken samples. Conclusion: We reported a remarkably high percentage of the antibiotic-resistant E. coli in chicken meat sold at retail in Qatar. The high percentage of MDR and colistin isolates is troublesome to the food safety of raw chicken meat and the potential of antibiotic resistance spread to public health. Our findings support the need for the implementation of one health approach to address the spread of antimicrobial resistance and the need for a collaborative solution.

Abstract Background: The dissemination of colistin-resistant bacteria carrying the colistin-resistant mobile gene, mcr-1 threatens medical care worldwide. In particular, contamination of food with colistin-resistant bacteria accelerates the community dissemination of colistin-resistant bacteria. Therefore, monitoring of colistin-resistant bacteria in food is important for controlling resistant bacteria. Unfortunately, the conventional culture methods for detecting colistin-resistant bacteria are not practical for monitoring food saftey. Therefore, development of a simple and rapid method to detect food contamination with colistin-resistant bacteria is desirable as an effective means for preventing the dissemination of resistant bacteria, particularly colistin-resistant bacteria.Findings: We developed a simple and rapid method for detecting Escherichia coli harboring the mcr-1 colistin resistance gene using a high-speed real-time polymerase chain reaction (PCR). The entire procedure, from sample processing to finals results, was performed within one hour. The practical utility of this method was verified by analyzing 27 retail meat samples for the presence of colistin-resistant bacteria. The results of the developed method were in agreement with the results of culturing colistin-resistant E. coli from the meat samples, demonstrating its efficacy and usefulness.Conclusions: A simple and rapid real-time PCR-based screening method was developed for detecting E. coli harboring mcr-1 in food samples. The practical utility of the procedure was confirmed using retail meat samples, indicating its potential as a convenient and rapid method to detect bacterial contamination of food items, especially in developing communities.

Introduction: Antimicrobial resistance is becoming a problem in public health. Zoonotic food-borne bacteria is infectious agent that can be transferred from animal to human through the food-producing animal we consume. Nowadays, antibiotic used for human and animal is not only to cure infection but also to aim animal’s growth promotion. It is known as non therapeutic antimicrobial agent (NTA) leading to antibiotic resistance. The third generation cephalosporins, cefotaxime, and also cefoxitin are included as important antibiotic for human. This study aims to identify the presence of cefotaxime-resistant Escherichia coli and cefoxitin-resistant Staphylococcus aureus isolated from chicken meat of both traditional and modern market in Surabaya.Methods: This is descriptive post test only experimental research. We used 8 samples of chicken meat from 4 different market using purposive sampling technique. We cultured Escherichia coli and Staphylococcus aureus from the chicken meat. Sensitivity test was done using Kirby-bauer disk-diffusion method.Results: All chicken meat sample bought from traditional market in Surabaya are contaminated by cefotaxime-sensitive Escherichia coli (n=4/4) while chicken meat sample bought from modern market are not contaminated by Escherichia coli (n=0/4). All chicken meat sample bought from traditional (n=4/4) are also contaminated by cefoxitin-sensitive Staphylococcus aureus. Half of chicken meat sample bought from modern market (n=2/4) are contaminated by cefoxitin-sensitive Staphylococcus aureus, while the other half (n=2/4) are contaminated by cefoxitin-resistant Staphylococcus aureus.Conclusion: Antibiotic resistance is found and all chicken meat samples have been highly contaminated with bacteria therefore food-processing should be done correctly.

Abstract Objective: Shiga toxin producing Escherichia coli (STEC) has known as a crucial zoonotic food borne pathogen. A total of 257 row chicken meat samples were collected from different markets in Hamadan city from January 2016 to May 2017. Samples were cultured on selective and differential culture media, and the virulence genes of E. coli isolates were analyzed by PCR assay. The antibiotic resistance patterns of E. coli isolates were determined by disk diffusion method. The genetic relatedness of STEC isolates were analyzed by ERIC-PCR. Results: Totally, 93(36%) of isolates were identified as E. coli in this current study. According serological and microbiological tests, 5(5.3%), 31(33.3%) and 7(7.5%) of E. coli isolates, characterized as Enterohemorrhagic E. coli (EHEC), STEC and attaching and effacing E. coli (AEEC) strains, respectively. High level resistance to tetracycline (89.8), ampicillin (82.8%) and sulfametoxazole-trimotoprime (71%) were detected among E. coli isolates. Analysis of ERIC-PCR results showed five different ERIC types among EHEC isolates. Based on our findings, chicken meat identified as a sources of STEC strains, therefore, the controlling and checkup the chicken meats for the resistance and virulent strains of E. coli should be consider as a crucial issues in public health.

The lack of antibiotics with a novel mode of action associated with the spread of drug resistant bacteria make the fight against infectious diseases particularly challenging. A quinoline core is found in several anti-infectious drugs, such as mefloquine and bedaquiline. Two main objectives were set in this work. Firstly, we evaluated the anti-mycobacterial properties of the previous quinolines 3, which have been identified as good candidates against ESKAPEE (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter spp. and Escherichia coli) bacteria. Secondly, a new series 4 was designed and assessed against the same bacteria strains, taking the pair of enantiomers 3m/3n as the lead. More than twenty compounds 4 were prepared through a five-step asymmetric synthesis with good enantiomeric excesses (>90%). Interestingly, all compounds of series 3 were efficient on M. avium with MIC = 2–16 µg/mL, while series 4 was less active. Both series 3 and 4 were generally more active than mefloquine against the ESKAPEE bacteria. The quinolines 4 were either active against Gram-positive bacteria (MIC ≤ 4 µg/mL for 4c–4h and 4k/4l) or E. coli (MIC = 32–64 µg/mL for 4q–4v) according to the global lipophilicity of these compounds.

Based on serological and microbiological tests, 36 (38.7% ± 9.9), 7 (7.5% ± 5.35), and 12 (12.9% ± 6.81) of the E. coli isolates were characterized as STEC, enteropathogenic E. coli (EPEC), and attaching and effacing E. coli (AEEC) strains, respectively. A high level of resistance to nalidixic acid (91.4% ± 5.7), tetracycline (89.2% ± 6.31), ampicillin (82.8% ± 7.67), and trimotoprime-sulfametoxazole (71% ± 9.22) was detected among the E. coli isolates. The analysis of the ERIC-PCR results showed five different ERIC types among the E. coli O157 isolates. Conclusions. Based on our findings, control and check-up of poultry meats should be considered as a crucial issue for public health.

In this study, the occurrence of the ciprofloxacin-resistant (CR) Escherichia coli in chicken meat was determined, and their clonal relations were investigated by using pulsed-field gel electrophoresis (PFGE). Antimicrobial resistance patterns of E. coli isolates were determined by using disc diffusion assay, and minimum inhibitory concentration of ciprofloxacin was determined by E-test. Plasmid-mediated quinolone resistance (PMQR) and extended spectrum beta-lactamase (ESBL) resistance genes were also screened through polymerase chain reactions. Sixty chicken meat samples were collected from different supermarkets and butchers in Sivas, Turkey. CR E. coli strains were determined in 59 (98.3%) chicken meat samples. By analyzing PFGE fingerprint data, 34 different pulsotypes were determined. All E. coli strains were found to be resistant to nalidixic acid, enrofloxacin, and norfloxacin. In addition, isolates were resistant to levofloxacin (40.7%), ampicillin (94.9%), trimethoprim-sulfamethoxazole (76.3%), tetracycline (69.5%), and chloramphenicol (44.1%). However, isolates were susceptible to imipenem and colistin. In this study, 81.4% of CR E. coli isolates were observed to have a multidrug-resistant profile, which is defined as resistance to three or more classes of antibiotics. Through phenotypic confirmation tests, five isolates (8.3%) were determined to be ESBL-producing. The PMQR genes were not determined in any of the isolates. Two isolates (3.4%) possessed the blaCTX-M and blaCMY-2 genes, and 40 isolates (67.8%) had the blaTEM gene. Taken together, retail raw chicken meat is highly contaminated with CR E. coli. However, these isolates are not found to be carriers of the PMQR genes, indicating a low public health problem.
