Abstract Background: The preventive role of muscular strength in the diminishing of a neuroinflammation is yet unknown. In this study, the role of the prophylactic muscular strength exercise was investigated, whether it would diminish cognitive alterations and modify the antioxidant intracellular scenery in an animal neuroinflammation model of the CA1 region of the hippocampus. Methods: The animals received muscular strength training for eight weeks, three times a week. Subsequently, the stereotaxic surgery, with intra-hippocampal infusion of either saline solution or l ipopolysaccharide (LPS) was performed. Next, behavioral tests were performed: objects and social recognition. At last, the animals were euthanized and the collect of the hippocampus and the prefrontal cortex were performed and, later, the dosage of the antioxidant activity was performed. Results: The results showed that the muscular strength exercises was capable of showing a beneficial prophylactic effect in the oxidative stress caused by an acute neuroinflammation. There was diminishing of the reduced glutathione concentration (GSH) and increase of the activity of the catalase enzyme (CAT) in the group (SE + LPS), regarding the control groups. In the prefrontal cortex, there was only an increase of the CAT activity in the group (SE + LPS), regarding the groups (CT) and (SE + SAL). As for the cognitive alterations there were found in the (SE + LPS) group, diminishing the mnemonic hazard of the discriminative and social memories, when compared to the control groups. Conclusion: We concluded, therefore, that the induction of a local inflammatory process in the hippocampus leads to mnemonic deficits in behavioral activities and increase of the GSH concentration, and that the muscular strength exercise performed prophylactically presents a protective effect capable of minimizing such mnemonic deficits and increasing the antioxidant defenses in mice that suffered a local neuroinflammatory process in the hippocampus.

Development of cardiovascular diseases (CVD) has been linked with changes to the lipid profile that can be observed during the postprandial period, a phenomenon known as postprandial lipemia (PL). Physical exercise is currently the number one non-pharmacological intervention employed for prevention and reduction of risk factors for the development of CVD. This in turn has created a growing interest in the effects of physical exercise on regulation and equilibrium of lipid metabolism. In this review we compare the results of studies that have investigated the beneficial effects of strength training on PL. We analyzed articles identified in the PubMed, Scopus and EBSCO databases published from 1975 to 2013 in international journals. Studies were selected for review if they covered at least two of four keywords. The results of these studies lead to the conclusion that strength training is effective for reduction of postprandial lipemia because it increases baseline energy expenditure. This type of training can be prescribed as an important element in strategies to treat chronic diseases, such as atherosclerosis.

Heme oxygenase (HO) family catalyzes the conversion of heme into free iron, carbon monoxide and biliverdin. It possesses two well-characterized isoforms: HO-1 and HO-2. Under brain physiological conditions, the expression of HO-2 is constitutive, abundant and ubiquitous, whereas HO-1 mRNA and protein are restricted to small populations of neurons and neuroglia. HO-1 is an inducible enzyme that has been shown to participate as an essential defensive mechanism for neurons exposed to oxidant challenges, being related to antioxidant defenses in certain neuropathological conditions. Considering that neurodegenerative diseases (Alzheimer’s Disease (AD), Parkinson’s Disease (PD) and Multiple Sclerosis (MS)) and neuropsychiatric disorders (depression, anxiety, Bipolar Disorder (BD) and schizophrenia) are associated with increased inflammatory markers, impaired redox homeostasis and oxidative stress, conditions that may be associated with alterations in HO-levels/activity, the purpose of this review is to present evidence on the possible role of HO-1 in these Central Nervous System (CNS) diseases. In addition, the possible therapeutic potential of targeting brain HO-1 is explored in this review.

Alcohol use disorder is a chronically relapsing disorder that involves (1) compulsivity to seek and take alcohol, (2) difficulty in limiting alcohol intake, and (3) emergence of a negative emotional state (e.g., dysphoria, anxiety, irritability) in the absence of alcohol. Alcohol addiction encompasses a three-stage cycle that becomes more intense as alcohol use progresses: binge/intoxication, withdrawal/negative affect, and preoccupation/anticipation. These stages engage neuroadaptations in brain circuits that involve the basal ganglia (reward hypofunction), extended amygdala (stress sensitization), and prefrontal cortex (executive function disorder). This chapter discusses key neuroadaptations in the hypothalamic and extrahypothalamic stress systems and the critical role of glucocorticoid receptors. These neuroadaptations contribute to negative emotional states that powerfully drive compulsive alcohol drinking and seeking. These changes in association with a disruption of prefrontal cortex function that lead to cognitive deficits and poor decision making contribute to the chronic relapsing nature of alcohol dependence.

Mitochondria act as hubs of numerous metabolic pathways. Mitochondrial dysfunctions contribute to altering the redox balance and predispose to aging and metabolic alterations. The sirtuin family is composed of seven members and three of them, SIRT3-5, are housed in mitochondria. They catalyze NAD+-dependent deacylation and the ADP-ribosylation of mitochondrial proteins, thereby modulating gene expression and activities of enzymes involved in oxidative metabolism and stress responses. In this context, mitochondrial sirtuins (mtSIRTs) act in synergistic or antagonistic manners to protect from aging and aging-related metabolic abnormalities. In this review, we focus on the role of mtSIRTs in the biological competence of reproductive cells, organs, and embryos. Most studies are focused on SIRT3 in female reproduction, providing evidence that SIRT3 improves the competence of oocytes in humans and animal models. Moreover, SIRT3 protects oocytes, early embryos, and ovaries against stress conditions. The relationship between derangement of SIRT3 signaling and the imbalance of ROS and antioxidant defenses in testes has also been demonstrated. Very little is known about SIRT4 and SIRT5 functions in the reproductive system. The final goal of this work is to understand whether sirtuin-based signaling may be taken into account as potential targets for therapeutic applications in female and male infertility.

Abstract Background Anti-vascular endothelial growth factor (VEGF) has been used as a therapeutic drug for the treatment of some human diseases. However, no systematic evidence is performed for assessing the role of VEGF in periodontitis. We carried out a comprehensive analysis to explore the role of VEGF in patients with periodontitis. Methods Multiple databases were searched for eligible studies. The pooled standardized mean difference (SMD) and odds ratio (OR) with the corresponding 95% confidence interval (CI) were applied to evaluate the effect sizes. Clinical data validation from microarray analysis was used. Pathway and process enrichment analysis were also investigated. Results Finally, 16 studies were included in this analysis. Overall, there was a significantly higher level of VEGF expression in periodontitis than in healthy control groups (OR = 16.64, 95% CI = 6.01–46.06, P < 0.001; SMD = 2.25, 95% CI = 1.25–3.24, P < 0.001). Subgroup analysis of ethnicity showed that VEGF expression was still correlated with periodontitis in the Asian and European populations. No correlation was observed between VEGF expression and age, gender, and pathological type. A large clinical sample data (427 periodontitis patients and 136 healthy controls) further validated that VEGF expression was higher in periodontitis than in healthy control groups (P = 0.023). VEGF was involved in many functions such as blood vessel development, response to growth factor, cell proliferation, and cell adhesion. Conclusions High levels of VEGF were credible implications for the development of periodontitis. Anti-VEGF therapy may be valuable for the treatment of periodontitis in clinical management.

This work presents an analysis of the intonation competence in a group of Italian children with cochlear implant (CI). Early cochlear implantation plays a crucial role in language development for children who were born deaf in that it favours the acquisition of complex aspects of language, such as the intonation structure. A story-generation task, the Narrative Competence Task, was used to elicit children’s stories. Narrations produced by 8 early implanted children and by 16 children with typically hearing (TH) (8 one-to-one matched considering the chronological age, TH-CA, and 8 considering the hearing age, TH-HA) were analysed considering intonation features (pitch accent distribution, edge tones and inner breaks). Results show that children with CI produce intonation patterns that are similar to those of both TH-CA and TH-HA control groups. Few significant differences were found only between children with CI and children matched for TH-HA in the use of rising edge tones. These results are discussed in light of the role of cognitive development in using prosody and intonation and the importance of early CI implantation. This study shows for the first time that intonation use of early implanted children is not different from that of typically developing children with the same chronological age.

AIM, DIAPH2, PTPRD and HIC1 are the cell glycoprotein, which play an important role in the occurrence and development of tumors. This study was designed to assess the association between DIAPH2, PTPRD and HIC1 SNPs and laryngeal cancer risk. PATIENTS AND METHODS: This study including 267 patients with histologically confirmed laryngeal cancer and 157 controls. The relationship between genetic variations DIAPH2 (rs6620138), PTPRD (rs3765142) and HIC1 (rs9901806) and the onset of laryngeal cancer were investigated. Statistical analysis to calculate the relationship between DIAPH2, PTPRD and HIC1 genes polymorphism and pathogenesis of laryngeal cancer. RESULTS: The results showed that rs6620138 DIAPH2 polymorphism could increase the onset risk of laryngeal cancer. Statistically significant differences in allele distribution of rs6620138 DIAPH2 and rs9901806 HIC1 in the case and control groups subgroups. CONCLUSIONS: This study results suggested that genetic variation of rs6620138 DIAPH2 polymorphism is related to the susceptibility to laryngeal cancer. Our results provide a basis to begin basic research on the role of DIAPH2 gene in the pathogenesis of laryngeal cancer.
