Four-year-old American ginseng (Panax quinquefolium L.) plants and soil samples were collected from nine ginseng gardens. Soil and leaf mineral contents were determined and six major ginsenosides, Rb1, Rb2, Rc, Rd, Re, and Rg1, were extracted from leaves and roots and quantified by high-performance liquid chromatography (HPLC). Correlation coefficients were more significant for soil nutrient levels vs. ginsenoside contents of leaves than of roots, suggesting that soil nutrient levels may play a major role in the synthesis of leaf ginsenosides. Minor elements in the leaf were also better correlated with ginsenoside contents of the root than that of the leaf. Iron content in the leaves exhibited highly significant correlations with the levels of Rb1, Rb2, Rc, and Rd, but calcium and copper contents were negatively correlated with Rg1 in the roots.

American ginseng (Panax quinquefolium) has long been cultivated in China for the function food and medicine. Here, ultra-high performance liquid chromatography was coupled with electrospray ionization and triple quadrupole mass spectrometry (UPLC-ESI−-TQ-MS) for simultaneous detection of 22 ginsenosides in American ginseng cultivated in Mudanjiang district of Heilongjiang. The extraction conditions also were optimized by a Box Behnken design experiment. The optimized result was 31.8 mL/g as ratio of liquid to raw materials, 20.3 min of extraction time, and 235.0 W of extraction powers. The quantitative MS parameters for these 22 compounds were rapidly optimized by single factor experiments employing UPLC-ESI−-multiple reaction monitoring or multiple ion monitoring (MRM/MIM) scans. Furthermore, the established UPLC-ESI−-MRM-MS method showed good linear relationships (R2 > 0.99), repeatability (RSD < 3.86%), precision (RSD < 2.74%), and recovery (94–104%). This method determined 22 bioactive ginsenosides in different parts of the plant (main roots, hairy roots, rhizomes, leaves, and stems) and growth years (one year to four years) of P. quinquefolium. The highest total content of the 22 analytes was in the hairy roots (1.3 × 105 µg/g) followed by rhizomes (7.1 × 104 µg/g), main roots (6.5 × 104 µg/g), leaves (4.2 × 104 µg/g), and stems (2.4 × 104 µg/g). Finally, chemometric methods, hierarchical clustering analysis (HCA) and partial least squares discrimination analysis (PLS-DA), were successfully used to classify and differentiate American ginseng attributed to different growth years. The proposed UPLC-ESI−-MRM-MS coupled with HCA and PLS-DA methods was elucidated to be a simple and reliable method for quality evaluation of American ginseng.

The extracted seeds of African breadfruit are identified to be extremely healthy whenever it is correctly processed. Therefore, the aim of the present study was to evaluate the effects of processing methods on the nutritional quality of African breadfruit seed. A qualitative phytochemical analysis including: Alkaloid, Flavonoid, Saponin, Tannin, Anthraquinone, Terpenoids, Steroid, and Cardiac Glycosides for the different fraction of African breadfruit seed was performed using a standard method. The result revealed the presence and greater amount of phytochemical for the raw fraction; seven in eight, six in eight for steamed fraction, and four in eight for boiled and roasted respectively. Anti-nutrient, Proximate, and Mineral Content were also conducted using standard methods. The amino acid composition was determined using High-Performance Liquid Chromatography (HPLC). The results of the present study revealed that anti-nutrients including Phytate, Tannins, and Oxalate were significantly p<0.05 reduced in the boiled fraction 5.47±0.15, 3.42±0.02 and 6.89±0.05, and highest in the raw fraction 7.77±0.01, 5.09±0.03 and 9.34±0.14. The proximate composition including; percentage crude fat, Ash, Carbohydrate, Fatty acid, and Energy value were significantly lower p<0.05 in the boiled fraction relative to the other fractions. Mineral contents; calcium, magnesium, sodium, potassium, and phosphorus were also significantly p<0.05 elevated in the boiled fraction relative to the raw, steamed, and roasted fraction. The amino acid composition was highest in the roasted and boiled fraction 57.350 and 56.978, and lowest in the steamed and raw fraction 35.754 and 28.748 respectively. Therefore, boiling (cooking) is encouraged for the preparation of African breadfruit seed.

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for ochratoxin A (OTA) detection in green, roasted and instant coffees was developed using anti-OTA monoclonal antibody. Immunological reagents prepared were OTA-BSA (4.76 mg/mL), anti-OTA.7 MAb (2x10³-fold dilution) and HRP-anti IgG (10³-fold dilution). The detection limit was 3.73 ng OTA/g and correlation coefficients (r) between this immunoassay and high performance liquid chromatography were 0.98 for green coffee, 0.98 for roasted and 0.86 for instant. OTA levels detected by ic-ELISA were higher than by HPLC, with ELISA/HPLC ratio of 0.66 - 1.46 (green coffee), 0.96 - 1.11 (roasted) and 0.93 - 1.82 (instant). ELISA recoveries for OTA added to coffee (5 - 70 ng/g) were 81.53 % for green coffee, 46.73 % for roasted and 64.35 % for instant, while recoveries by HPLC were 80.54 %, 45.91 % and 55.15 %, respectively. Matrices interferences were minimized by samples dilution before carrying out the ELISA assay. The results indicate that MAb-based ic-ELISA could be a simple, sensitive and specific screening tool for OTA detection, contributing to quality and safety of coffee products.

PPARγis a nuclear hormone receptor that functions as a master regulator of adipocyte differentiation and development. Full PPARγagonists, such as the thiazolidinediones (TZDs), have been widely used to treat type 2 diabetes. However, they are characterized by undesirable side effects due to their strong agonist activities. Pseudoginsenoside F11 (p-F11) is an ocotillol-type ginsenoside isolated fromPanax quinquefolium L.(American ginseng). In this study, we found that p-F11 activates PPARγwith modest adipogenic activity. In addition, p-F11 promotes adiponectin oligomerization and secretion in 3T3-L1 adipocytes. We also found that p-F11 inhibits obesity-linked phosphorylation of PPARγat Ser-273 by Cdk5. Therefore, p-F11 is a novel partial PPARγagonist, which might have the potential to be developed as a new PPARγ-targeted therapeutics for type 2 diabetes.

The use of computers with outstanding performance has become a real necessity in order to achieve greater efficiency and sustainability for the accomplishment of various tasks. Therefore, with the development of information technology and increasing dynamism in the business environment, it is expected that these computers will be more intensively deployed. In this paper, research was conducted in Danube region countries: Austria, Bosnia and Herzegovina, Bulgaria, Croatia, Czech Republic, Germany, Hungary, Moldova, Montenegro, Romania, Serbia, Slovakia, Slovenia, and Ukraine. The aim of the research was to determine what criteria are most significant for the introduction of high-performance computing and the real situation in each of the countries. In addition, the aim was to establish the infrastructure needed to implement such a system. In order to determine the partial significance of each criterion and thus the possibility of implementing high-performance computing, a multi-criteria model in a fuzzy environment was applied. The weights of criteria and their rankings were performed using the Fuzzy PIvot Pairwise RElative Criteria Importance Assessment—fuzzy PIPRECIA method. The results indicate different values depend on decision-makers (DMs) in the countries. Spearman’s and Pearson’s correlation coefficients were calculated to verify the results obtained.

Radiocarbon dates confirm a chronological sequence for late Holocene beach ridges at Wangiti Beach on Bathurst Island. The vegetation on these beach ridges can be clearly related to topography and distance from the sea. Monsoon rainforest occurs on the fore-dunes where the nutrient levels of the young sediments are relatively high. The mid-dunes support woodland dominated by Melalueca viridiflora and have nutrient-poor acid soils. Vegetation with a high component of monsoon rainforest species occupies the rear dunes, which have a high concentration of seawater macronutrient cations despite their older age than more seaward dunes. The relatively low elevation of the rear dunes supports the suggestion that soil nutrient levels are enriched by sea water or marine sediments during infrequent inundation events such as those during cyclones.
