Two cultivars of sweetpotato [Ipomoea batatas (L.) Lam.], Commensal and Salyboro, were subjected to salt stress using axillary bud cultures. The salt levels ranged from 0–150 mM. After 10 weeks of growth, plantlet shoot height, dry weight, number of nodes, levels of proline, soluble carbohydrate, and protein; and metal ions sodium, potassium, magnesium, and calcium, were measured. In both cultivars, proline accumulation was higher in the shoot. There was a positive correlation between the increase in soluble carbohydrates and proteins in `Commnesal', but not in `Salyboro'. More sodium accumulated in the shoots of `Salyboro' compared to `Commensal'. The accumulation of sodium reduced the calcium and potassium, but not magnesium levels. Increase in sodium levels correlated with the increase in soluble carbohydrate levels is `Salyboro', but not in `Commensal'. A similar trend was evident with praline and sodium accumulation. Based on these and previous results, the cultivar `Salyboro' appears to be more susceptible to salt stress.

Two cultivars of sweetpotato, `Commensal' and `Salyboro', were subjected to salt stress using axillary bud cultures. The salt levels ranged from 0 to 10 g·L-1. After the initial experiments, levels of calcium in the medium were changed from 3 mm in the MS medium to 1.5, 6, and 12 mm. After 10 weeks of growth, plantlet shoot height, dry weight, number of nodes, levels of proline, soluble carbohydrate, and protein were measured. `Commensal' was tolerant to salt levels up to 4 g·L-1, but `Salyboro' was sensitive to concentrations of salt even at lower concentrations as evidenced by the growth and dry weight. Proline accumulation was higher in the shoot than in the root. The protein: carbohydrate ratios did not change much in `Commensal', but levels of carbohydrates increased in `Salyboro'. Reduction in calcium levels had a synergistic affect on salt-stressed cultivars. Enhanced levels of calcium reduced the inhibitory affects of salt stress. This was more pronounced in `Salyboro', which was susceptible. Proline levels were higher in plants subjected to salt stress and higher levels of calcium than controls, but lower than the plants subjected to salt stress. These and other metabolic changes suggest that calcium can reduce the adverse affects of salt stress in these two sweetpotato cultivars.

Axillary bud cultures of sweetpotato Ipomoea batatus L. [cultivars comensal and salyboro] were propagated in vitro. Nodal cultures of these were grown at different temperatures ranging from 10 °C to 30 °C at 5 °C intervals from the time of axillary bud transfer to 10 weeks of growth in a controlled growth chamber. After 10 weeks of growth, morphological and physiological parameters were measured including shoot height, number of nodes and branches, levels of proline, soluble carbohydrate and protein. There was not much difference in the cultures grown at 25 and 30 °C but temperatures lower than 25 °C were inhibitory to both cultivars, though the effects were more pronounced in salyboro than comensal. Salyboro grew slower and exhibited leaf discoloration, weak stems, and poor root growth. Proline levels increased in both cultivars and the increase was higher in shoot than root due to low temperature stress. Soluble carbohydrates and proteins increased in comensal, maintaining the carbohydrate protein ratio, but decreased in salyboro. The results suggest that the cultivar comensal may tolerate cooler temperatures and the cultivar salyboro is susceptible to them.

Nowadays sweet violet (Viola odorata) is an ornamental-medical plant that considered as endangered and threatened species. On the other hand, biotic and abiotic stresses impose a major threat to agriculture. Here, we investigated the effects of salinity and drought stresses, based on polyethylene glycol (PEG; 1, 1.5, 2, 2.5, 3 and 4%) and NaCl (0, 50, 100 and 150 mM), on growth characteristics, physiological parameters and antioxidant defense system of sweet violet under in-vitro conditions. The influences of NaCl and PEG gradients in the culture media on plant height, green leaf percentage, root dry weight (DW), and electrolyte leakage (EL) was described by a linear or quadratic model. All measured parameters (except EL) decreased when NaCl or PEG concentration increased. In contrast, EL increased other traits. Moreover, with increasing in salinity and drought severity, shoot DW decreased, while antioxidant enzymes activity such as catalase (CAT), peroxidase (POX) and superoxide dismutase (SOD) and proline content increased. However, total soluble carbohydrates (TSC), at all drought levels, increased with increasing NaCl concentration up to 50 or 100 mM, and then decreased. Most variations in the shoot DW, CAT activity, proline and TSC contents due to salt stress occurred at low concentration of PEG. Overall, our findings highlight that the effect of combined drought and salt stress was more severe. However, the sensitivity of the plant to drought or salinity stress was higher in the absence of other stress.

Salinity significantly impacts the growth, development, and reproductive biology of various crops such as vegetables. The cultivable area is reduced due to the accumulation of salts and chemicals currently in use and is not amenable to a large extent to avoid such abiotic stress factors. The addition of microbes enriches the soil without any adverse effects. The effects of microbial consortia comprising Bacillus sp., Delftia sp., Enterobacter sp., Achromobacter sp., was evaluated on the growth and mineral uptake in tomatoes (Solanum Lycopersicum L.) under salt stress and normal soil conditions. Salinity treatments comprising Ec 0, 2, 5, and 8 dS/m were established by mixing soil with seawater until the desired Ec was achieved. The seedlings were transplanted in the pots of the respective pH and were inoculated with microbial consortia. After sufficient growth, these seedlings were transplanted in soil seedling trays. The measurement of soil minerals such as Na, K, Ca, Mg, Cu, Mn, and pH and the Ec were evaluated and compared with the control 0 days, 15 days, and 35 days after inoculation. The results were found to be non-significant for the soil parameters. In the uninoculated seedlings’ (control) seedling trays, salt treatment significantly affected leaf, shoot, root dry weight, shoot height, number of secondary roots, chlorophyll, and mineral contents. While bacterized seedlings sown under saline soil significantly increased leaf (105.17%), shoot (105.62%), root (109.06%) dry weight, leaf number (75.68%), shoot length (92.95%), root length (146.14%), secondary roots (91.23%), and chlorophyll content (−61.49%) as compared to the control (without consortia). The Na and K intake were higher even in the presence of the microbes, but the beneficial effect of the microbe helps plants sustain in the saline environment. The inoculation of microbial consortia produced more secondary roots, which accumulate more minerals and transport substances to the different parts of the plant; thus, it produced higher biomass and growth. Results of the present study revealed that the treatment with microbial consortia could alleviate the deleterious effects of salinity stress and improve the growth of tomato plants under salinity stress. Microbial consortia appear to be the best alternative and cost-effective and sustainable approach for managing soil salinity and improving plant growth under salt stress conditions.

The primary growth of eight varieties of three species–white clover (3), red clover (4) and lucerne (1)–was sampled at fortnightly intervals and the percentage dry matter, in vitro digestibility, crude protein, water soluble carbohydrates, P, Ca, K, Na and Mg were determined.Differences between species were nearly always significant and the general order of merit was white clover, red clover and lucerne. The exceptions were for dry-matter percentage where this order was reversed, and red clover had the lowest Na and highest Mg content.

Escamoles, ant eggs (Liometopum apiculatum M), are edible insects consumed in great numbers and appreciated for their sensory characteristics, but not for their nutritional value. In Mexico, they reproduce in arid and semi-arid zones of Mexico and several states (Puebla, Tlaxcala, Hidalgo). Samples were gathered to determine reproduction conditions and perform an insect analysis to generate data composition in macronutrients, on dry basis, according to AOAC methods. The obtained data were: proteins: 42.12-50.63%; lipids: 30.27-34.96%; minerals: 6.53-7.85%; fibres: 1.91-2.56%; and soluble carbohydrates: 6.80-18.27%. Entomological material was collected in spring 2014, 3 nests for each zone, all from different altitudes and agroclimatic regions. Samples were representative for the insect supply in the areas studied. Insects are high in proteins and lipids, compared with most of the commonly consumed food, due to their high content of fatty acids; they must be refrigerated for later consumption. Minerals are moderated. Fibre and soluble carbohydrate levels are low, but an excess of proteins can be converted into carbohydrates via gluconeogenesis. Escamoles reproduce once a year in the spring, but sometimes one nest can provide escamoles eggs twice in the same season, once at the beginning and again at the end, depending on the biotic and abiotic conditions of their environment. Escamoles are a good source of nutrients; however, some rural people sell them to obtain extra income rather than consume them to improve their nutritional condition and welfare.

Abstract The elevation of arsenic (As) content in soils is of considerable concern with respect to its uptake by plant and subsequent entry into wildlife and human food chains. The treatment of sorghum seedlings with As as NaH2As4O. 7H2O at various concentrations (A1 = 0, A2 = 20, A3 = 40 and A4 = 60 mg As kg−1 soil) and salinity at four different levels (S1 = 0, S2 = 3, S3 = 6 and S3 = 9 dS m−1) reduced fresh and dry weights of sorghum plants. The co-application of As and salinity increased the guaiacol peroxidase (GPX) activity in shoot and root tissues. The highest GPX activity in shoot and root tissues was obtained at S2A4 and S3A3 treatments, respectively. The activity of catalase (CAT) in shoot was not changed, but unlike the GPX activity, salinity and As decreased the CAT activity in root tissues. Concerning the photosynthesis pigments, salinity had no effect on the chlorophyll ‘a’, chlorophyll ‘b’ and carotenoid content in leaves, but the As treatment significantly decreased the content of both chlorophyll types. Salinity increased the anthocyanin content in leaves. There were negative correlation between soluble carbohydrates (r2 = −0.78**) and stomata conductance (r2 = −0.45**) and dry weight of the plant biomass in this study. By increasing the salinity and As concentration in root medium, soluble carbohydrate in leaves increased but salinity decreased the leaf stomata conductance.

The effects of initial sucrose (suc) concentrations in the medium (S0) on the carbon balance and growth of sweetpotato [Ipomoea batatas (L.) Lam. `Beniazuma'] and tomato (Lycopersicon esculentum Mill. `HanaQueen') plantlets were studied under controlled environmental conditions. Plantlets were cultured with 0, 7.5, 15, or 30 g·L-1 of S0 under high photosynthetic photon flux (160 to 200 μmol·m-2·s-1) and CO2 enriched (1400 to 2050 μmol·mol-1) conditions. Net photosynthetic rate per leaf area (Pl) decreased and dry weight per plantlet (Wd) increased with increasing S0, but did not differ significantly between S0 of 7.5 to 30 g·L-1 for sweetpotato or 15 to 30 g·L-1 for tomato. Carbon influxes and effluxes of the plantlets by metabolism of medium suc and/or photosynthesis, and respiration were estimated based on measurements of in situ and steady state CO2 exchange rates and sugar uptake during culture. At S0 from 7.5 to 30 g·L-1, photosynthesis was responsible for 82% to 92% and 60% to 67% of carbohydrate assimilation for sweetpotato and tomato, respectively. Estimated carbon balances of plantlets based on the estimated and actual increases of moles of carbon in plant tissue demonstrated that in situ estimation of carbon balance was reasonably accurate for sweetpotato at S0 of 0 to 15 g·L-1 and for tomato at S0 of 0 g·L-1 and that the actual contribution of photosynthesis for tomato at high S0 might be lower than the values estimated in the present experiment. Results showed that initial suc concentration affected the relative contribution of photosynthesis on their carbon balances and that the responses were species specific. The failure of validation at S0 in a range specific to each species suggested the need for further study on carbon metabolism of in vitro plantlets cultured with sugar in the medium.

The aim of present research was to study the genetic diversity among Asparagus species and its cultivars using morphological characteristics and RAPD markers. In-vitro and field experiments of 14 germplasm sources of Asparagus species and its cultivars were conducted at the Kohat University of Science and Technology to estimate the comparative performance. Highest genotypic variance, phenotypic variance, genotypic and phenotypic coefficient of variance were observed in shoot height in shoot related traits were 318.40, 320.30, 939.34 and 944.96, respectively, whereas highest values for root related traits were observed in root length that were 21.84, 22.35, 141.60 and 144.91 respectively. Among the shoot related traits maximum heritability, genetic advance and genetic gain were calculated for shoot length, while in root related traits these characters were highest for root dry weight. RAPD markers were used to evaluate genetic diversity analysis of 14 germplasm sources of Asparagus species and its cultivars. RAPD markers generated a total of 247 bands, of these 239 bands were polymorphic with average of 34.1 bands per primer. Cluster analysis based on Neighbor Joining methods showed that wild species (A. adsendens, A. densiflorus, A. capitatus, A. gracelus, A. plumosus, A. racemosus and A. setaceus) were genetically distant from A. officinalis and its cultivars (Abril, Apollo, Gersengum, Huchel, Para seletion and Taranga). The results of the present investigations could be particularly used for authentic identification and would be useful for evaluation of genetic improvement of Asparagus species and its cultivars. J. SUMMARYBulk harvests of primary growth were made at intervals during the spring and summer of 2 consecutive years from an ungrazed area of lucerne (Medicago sativa) and the digestibility by sheep was measured. At the same times, shoots of known length representative of the sward were collected, divided into 7·5 cm lengths, and separated into leaf and stem for the determination of ash, nitrogen, cellulose, and in vitro organic matter digestibility.Digestibility and leaf: stem ratio of the bulk harvests decreased and dry matter increased as the season advanced. Leaf composition changed little, but stem increased in lignin and decreased in ash and in vitro digestibility. In vitro digestibility and leaf: stem ratio were highly correlated with sheep digestibility.Chemical composition of shoots of known heights was similar to that of the bulk harvests. Leaf weights/shoot tended to increase linearly with height of shoot, whereas stem increased nearly as the square of the height. Dry matter of leaf and stem were inversely related to leaf:stem ratio. Stem digestibility decreased with leaf:stem ratio and with increase in shoot height.Chemical components of shoot fractions were associated with each other and were largely dependent on the distance from the top of the shoot at which they had been taken, irrespective of shoot height or time of year. Bottom stems had lower ash, nitrogen and digestibility but higher cellulose, dry weight and dry matter than top stems, whose composition approached that of leaves. Leaf fractions showed little consistent trend with position on the shoot.Some of the difficulties in studying changes in plant composition under field conditions are discussed.
