Penicillium digitatum is one of the most important causes of postharvest decay of Mexican lime fruit. The first stage of this study dealt with examining the effect of savory (Satureja hortensis) essential oil on P. digitatum mycelial growth in vitro. Savory essential oil (SEO) applied at concentrations of 0, 200, 400, 600, 800, 1000, and 1200 μL·L−1 to potato dextrose agar (PDA) medium. The results revealed that the application of SEO at concentrations of 1000 and 1200 μL·L−1 completely prevented the growth of P. digitatum. Gas chromatography–mass spectrometry results indicated that the dominant components of SEO were carvacrol (55.67%) and γ-terpinene (31.98%). In the second phase of the experiment, in vivo assays were conducted to evaluate the efficiency of SEO (800 and 1000 μL·L−1), hot water (40 and 50 °C), and gum arabic coating (2.5% and 5%) in restricting the fungi activity on Mexican lime fruit. The Mexican lime fruit were immersed in the aqueous solutions of SEO and gum arabic or in the hot water for 5 minutes, and then stored at 8 °C for 30 days. Savory essential oil at the concentration of 800 μL·L−1 proved to be the most effective treatment in conserving bioactive compounds of the fruits such as total phenols. This treatment also optimally maintained antioxidant activity and suppressed the activity of polyphenol oxidase (PPO) in the fruit peel. Moreover, hot water at 40 °C caused the least physicochemical changes and the highest appearance quality during storage.

The chemical composition of winter savory, peppermint, and anise essential oils, and in vitro and in vivo phytotoxic activity against weeds (Portulaca oleracea, Lolium multiflorum, and Echinochloa crus-galli) and food crops (maize, rice, and tomato), have been studied. Sixty-four compounds accounting for between 97.67–99.66% of the total essential oils were identified by Gas Chromatography-Mass Spectrometry analysis. Winter savory with carvacrol (43.34%) and thymol (23.20%) as the main compounds produced a total inhibitory effect against the seed germination of tested weed. Menthol (48.23%), menthone (23.33%), and iso-menthone (16.33%) from peppermint only showed total seed germination inhibition on L. multiflorum, whereas no significant effects were observed with trans-anethole (99.46%) from anise at all concentrations (0.125–1 µL/mL). Low doses of peppermint essential oil could be used as a sustainable alternative to synthetic agrochemicals to control L. multiflorum. The results corroborate that in vivo assays with a commercial emulsifiable concentrate need higher doses of the essential oils to reproduce previous in vitro trials. The higher in vivo phytotoxicity of winter savory essential oil constitutes an eco-friendly and less pernicious alternative to weed control. It is possible to achieve a greater in vivo phytotoxicity if less active essential oil like peppermint is included with other active excipients.

The present study investigated the capability of an essential oil mix (MO: 1% and 3%) in ameliorating amnesia and brain oxidative stress in a rat model of scopolamine (Sco) and tried to explore the underlying mechanism. The MO was administered by inhalation to rats once daily for 21 days, while Sco (0.7 mg/kg) treatment was delivered 30 min before behavioral tests. Donepezil (DP: 5 mg/kg) was used as a positive reference drug. The cognitive-enhancing effects of the MO in the Sco rat model were assessed in the Y-maze, radial arm maze (RAM), and novel object recognition (NOR) tests. As identified by gas chromatography–mass spectrometry (GC–MS), the chemical composition of the MO is comprised by limonene (91.11%), followed by γ-terpinene (2.02%), β-myrcene (1.92%), β-pinene (1.76%), α-pinene (1.01%), sabinene (0.67%), linalool (0.55%), cymene (0.53%), and valencene (0.43%). Molecular interactions of limonene as the major compound in MO with the active site of butyrylcholinesterase (BChE) was explored via molecular docking experiments, and Van der Waals (vdW) contacts were observed between limonene and the active site residues SER198, HIS438, LEU286, VAL288, and PHE329. The brain oxidative status and acetylcholinesterase (AChE) and BChE inhibitory activities were also determined. MO reversed Sco-induced memory deficits and brain oxidative stress, along with cholinesterase inhibitory effects, which is an important mechanism in the anti-amnesia effect. Our present findings suggest that MO ameliorated memory impairment induced by Sco via restoration of the cholinergic system activity and brain antioxidant status.

Background: Trichomoniasis is one of the most common nonviral sexually transmitted infections worldwide which drug-resistant cases of the infection are rising. The aim of the study was to assessment the in vitro activity of Foeniculum vulgare and its main essential oil component on Trichomonas vaginalis. Also phytochemical investigation of F. vulgare essential oil was performed. Methods: Five T. vaginalis isolates subjected to susceptibility testing against essential oil and extracts of F. vulgare and anethole using microtiter plate method. The minimum lethal concentration (MLC) of the natural products was assessed in comparison with metronidazole. Gas chromatography-mass spectrometry and gas chromatography-flame ionization detector was applied for chemical investigation of the essential oil. Results: After 48 hours incubation, the most potent antitrichomonal agents were the methanolic and hexanic extract with MLC of 360 µg/ml and followed by the essential oil and anethole (1600 µg/ml). The isolates were sensitive to metronidazole with a mean MLC of 13.7 µg/ml. E-Anethole (88.41 %) was the major constituent of F. vulgare essential oil. Conclusion: The results suggested in vitro antiprotozoal properties of F. vulgare and anethole against T. vaginalis. Therefore further studies are needed to evaluate their in vivo effects and toxicity.

ABSTRACT The aim of this study was to investigate the efficacy of navel orange, Citrus sinensis (L.) Osbeck, peel essential oil (NOPEO) for inhibiting spoilage fungi in potato slices. Sixteen different components accounting for 99.79% of the headspace components of NOPEO were identified by gas chromatography–mass spectrometry. d-Limonene was the major component of NOPEO. Antifungal activity of NOPEO was tested in vitro and in vivo against four foodborne fungi. A MIC of NOPEO against the four fungal species was 9.40 μL/mLair. NOPEO provided about 74, 74, 73, and 69% protection against Aspergillus niger, Mucor wutungkiao, Penicillium funiculosum, and Rhizopus oryzae at 2.00 μL/mLair concentration, respectively. NOPEO has been demonstrated to significantly improve the microbiological quality of potato slices.

Summary The nematicidal activity of the essential oil of Cymbopogon citratus and its main components were studied. During in vitro assays with Meloidogyne incognita second-stage juveniles (J2), the oil showed an LC50 of 166 μg ml−1. In vivo assays that exposed J2 to the LC50 oil concentration significantly reduced galls and eggs on tomato roots. According to gas chromatography-mass spectrometry analyses, the main components of the essential oil were geranial (42.7%) and neral (33.0%). The combination of these two isomers, known as citral, had an enhanced nematicidal activity at low concentrations as shown by in vitro tests through a synergistic interaction with undecan-2-one. In assays with tomato plants inoculated with M. incognita J2, the combination of citral (600 μg ml−1) and undecan-2-one (300 μg ml−1) showed a high reduction in the number of galls (63.4%) and eggs (56.3%) in relation to Tween 80® control. The citral/undecan-2-one combination showed a synergistic nematicidal activity against M. incognita during in vitro and in vivo assays.

Syzygium fruticosum (SF), a valuable Bangladeshi fruit, is considered an alternative therapeutic agent. Mainly, seeds are used as nutritional phytotherapy to ease physical and mental status by preventing chronic diseases. Here, we scrutinized the S. fruticosum seed’s fundamental importance in traditional medicine by following an integrated approach combining in vivo, in vitro, and in silico studies. The SF was fractionated with different solvents, and the ethyl acetate fraction of SF (EaF-SF) was further studied. Mice treated with EaF-SF (200 and 400 mg/kg) manifested anxiolysis evidenced by higher exploration in elevated plus maze and hole board tests. Similarly, a dose-dependent drop of immobility time in a forced swimming test ensured significant anti-depressant activity. Moreover, higher dose treatment exposed reduced exploratory behaviour resembling decreased movement and prolonged sleeping latency with a quick onset of sleep during the open field and thiopental-induced sleeping tests, respectively. In parallel, EaF-SF significantly (p < 0.001) and dose-dependently suppressed acetic acid and formalin-induced pain in mice. Also, a noteworthy anti-inflammatory activity and a substantial (p < 0.01) clot lysis activity (thrombolytic) was observed. Gas chromatography-mass spectrometry (GC–MS) analysis resulted in 49 bioactive compounds. Among them, 12 bioactive compounds with Lipinski’s rule and safety confirmation showed strong binding affinity (molecular docking) against the receptors of each model used. To conclude, the S. fruticosum seed is a prospective source of health-promoting effects that can be an excellent candidate for preventing degenerative diseases.

Chitosan is receiving increasing attention from the food industry for being a biodegradable, non-toxic, antimicrobial biopolymer able to extend the shelf life of, and preserve the quality of, fresh food. However, few studies have investigated the ability of chitosan-based coatings to allow the diffusion of bioactive compounds into the food matrix to improve its nutraceutical quality. This research is aimed at testing whether a hydrophilic molecule (tyrosol) could diffuse from the chitosan-tyrosol coating and cross the tomato peel. To this end, in vitro permeation tests using excised tomato peel and an in vivo application of chitosan-tyrosol coating on tomato fruit, followed by tyrosol quantification in intact fruit, peel and flesh during a seven-day storage at room temperature, were performed. Both approaches demonstrated the ability of tyrosol to permeate across the fruit peel. Along with a decreased tyrosol content in the peel, its concentration within the flesh was increased, indicating an active transfer of tyrosol into this tissue. This finding, together with the maintenance of constant tyrosol levels during the seven-day storage period, is very promising for the use of chitosan formulations to produce functional tomato fruit.

Abstract Background Silexan, a special essential oil from flowering tops of lavandula angustifolia, is used to treat subsyndromal anxiety disorders. In a recent clinical trial, Silexan also showed antidepressant effects in patients suffering from mixed anxiety-depression (ICD-10 F41.2). Since preclinical data explaining antidepressant properties of Silexan are missing, we decided to investigate if Silexan also shows antidepressant-like effects in vitro as well as in vivo models. Methods We used the forced swimming test (FST) in rats as a simple behavioral test indicative of antidepressant activity in vivo. As environmental events and other risk factors contribute to depression through converging molecular and cellular mechanisms that disrupt neuronal function and morphology—resulting in dysfunction of the circuitry that is essential for mood regulation and cognitive function—we investigated the neurotrophic properties of Silexan in neuronal cell lines and primary hippocampal neurons. Results The antidepressant activity of Silexan (30 mg/kg BW) in the FST was comparable to the tricyclic antidepressant imipramine (20 mg/kg BW) after 9-day treatment. Silexan triggered neurite outgrowth and synaptogenesis in 2 different neuronal cell models and led to a significant increase in synaptogenesis in primary hippocampal neurons. Silexan led to a significant phosphorylation of protein kinase A and subsequent CREB phosphorylation. Conclusion Taken together, Silexan demonstrates antidepressant-like effects in cellular as well as animal models for antidepressant activity. Therefore, our data provides preclinical evidence for the clinical antidepressant effects of Silexan in patients with mixed depression and anxiety.
