The objects of the study were Mature dried fruits of Chinese birch (Lycium chinense Mill., this. Solanaceae (Solanaceae)). Four samples were examined: No. 1 (Gifts of Pamir, Russia), No. 2 (Gullin Tianhe Pharmaceutical, China), No. 3 (A. J. Alliance, Russia), No. 4 (Globaltorg, Russia). The purpose of this work is to determine the amount of carotenoids in the fruits of Chinese by spectrophotometry. The quantitative determination of the amount of carotenoids in the raw material of Chinese Dereza was carried out. Thin-layer chromatography was used to confirm the presence of carotenoids in the fruits of Dereza. The expediency of using the spectrophotometric method for the detection and quantitative determination of carotenoids in extracts from the fruits of Chinese birch is substantiated. The electron spectra of hexane, acetone and acetone-hexane solutions in the wavelength range 400–500 nm have a maximum optical density at 450±2 nm, characteristic of β-carotene. The optimal conditions of extraction of carotenoids from the raw materials of this plant (extractant – hexane; ratio "raw material–extractant" – 1 : 5; extraction time – 90 minutes; the degree of grinding of raw materials – 0.5 mm). It was determined that the error of a single determination of the content of carotenoids in the fruits of Chinese Dereza with a confidence probability of 95% is ±3,49%. It was revealed that the content of carotenoids in the raw material of Chinese Dereza varies in the range of 33–39 mg%.

Abstract A single laboratory validation (SLV) study was conducted for a liquid chromatography (LC) method for the determination of total and all-trans-β-carotene in a variety of dietary supplements, including multivitamin tablets, softgels, capsules, and beadlet raw materials. Extraction variants were developed for the different types of supplements tested based upon the supplement type and level of β-carotene. Water dispersible formulations such as powders, emulsions, tablets, and capsules were enzymatically digested with protease and extracted with dichloromethane–ethanol. Oily suspensions were directly dissolved in dichloromethane–ethanol. After appropriate dilution or concentration, the extracts were chromatographed by using either a reversed-phase C18 column or, in products containing high amounts of α-carotene, a reversed-phase C30 column. The LC systems provided linear responses in the range of 0.1–50 μg β-carotene/mL. The main geometrical isomers of β-carotene (all-trans, 9-cis, 13-cis, and 15-cis) were well separated from each other and from other carotenoids such as α-carotene, cryptoxanthin, lutein, lycopene, and zeaxanthin. Duplicate determinations of total β-carotene performed by 2 technicians in 8 different test materials on 5 different days resulted in relative standard deviations of 1.2–4.4%. Recoveries determined for supplements and beadlet raw material spiked with β-carotene levels of 10 μg to 100 mg/test portion and 0.2–40%, respectively, ranged from 97.5 to 102.1%. On the basis of the accuracy, precision, and recovery results from the SLV study, the method is suggested for a collaborative study on the determination of total and all-trans-β-carotene in dietary supplements.

Rheumatic diseases include more than 1000 nosological forms, among which the highest incidence is osteoarthritis, rheumatoid arthritis and osteoporosis. The predominantly advanced age, the presence of comorbid diseases, the duration of the course of the disease and the need for concomitant treatment complicate the conduct of anti-inflammatory and analgesic therapy in such patients. Herbal medicines, in particular in the form of doses, due to the mildness of the action and the absence of significant side effects, can be used in the complex therapy of the acute period of rheumatic diseases, as well as for the prevention of relapse. The aim of the presented work was to develop the composition, technology and determination of the quality indices of herbal collection, which reveals analgesic and anti-inflammatory properties and can be used for the prophylaxis and auxiliary therapy of a number of conditions in rheumatology. Materials and methods. In the course of research, the methods used in the State Pharmacopoeia of Ukraine 2.0 (SPHU) were used. Results. As a result of the analysis of literary data and our own experimental studies on the chemical composition and pharmacological activity of certain types of medicinal plant material, we have developed herbal collection that is recommended for the treatment and prevention of rheumatic diseases of the joints. Studies have been conducted on the development of quality collection indicators, which guarantee the effectiveness, safety, reproducibility of the imposed collection. The basic technological parameters of the raw material are determined: specific, bulk and volume mass, porosity, degree, degree of shredding. In order to prevent uneven mixing, an optimal degree of grinding of the raw material of the investigated collection, which ranges from 0.20 mm to 0.35 mm, is established, which allows obtaining a homogeneous mixture and contributes to the maximum yield of extractive and active substances. A series of experimental works aimed at studying the influence of the method of infusion (for 15, 30, 45, 60 minutes and until complete cooling) on the quality of the collected water extracts from the collection was conducted. The optimal method for setting the collection for 60 minutes is the best, since the content of extractives and flavonoids in the aqueous extract was highest and amounted to 49.57±0.52 % and 0.52±0.03 % respectively. Conclusions. The composition of the herbal drug was developed: Harpagophyti radiх, Sophoraе alabastra, Valvae fructus Phaseoli vulgaris, Polygoni avicularis herba with the following ratio of components 50: 20: 20:10. The technological parameters of the raw material are studied: specific, volumetric and bulk mass, porosity, singularity of raw materials. The optimum degree of shredding of raw material is established, which helps to maximally remove biologically active substances and extractives. Experimental way is determined the optimal mode of collection, which can be recommended to the consumer to obtain the maximum therapeutic effect.

Due to the content of phytosterols, extractive preparations of Urtica dioica roots are able to show antiandrogenic effect in the case of external therapy of men and women with androgenic alopecia. Oil extracts (OE) are characterized by several advantages when applied to the skin of the scalp compared to water-alcohol extracts. For the development of OE technology from Urtica dioica roots, it is important to choose the optimal extraction parameters, which are based on the quantitative determination of phytosterols in the extractant and the studied samples of extracts. The aim of the work is to choose the optimal parameters for obtaining OE from Urtica dioica roots based on quantitative determination of phytosterols content in experimental samples of OE by gas capillary chromatography. Materials and methods. Objects of the research – Urtica dioica root, refined corn oil, refined sunflower oil, samples of oil extracts. Determination of phytosterol content in experimental samples was carried out by gas capillary chromatography (chromatograph “Crystal 2000”, manufacturer – research and production company “Analytics”). Results. 5 different compounds of steroid structure (stigmasterol, β-sitosterol, etc.) were identified in sunflower oil by gas liquid chromatography, and 10 (campesterol, 2-α stigmasterol, β-sitosterol, Δ5-avenosterol, etc.) were identified in sunflower oil. The quantitative content of β-sitosterol in the sum of sterols of corn oil was significantly higher compared to the content of this substance in sunflower oil and amounted to 59.33 %. Optimal technological parameters were established considering the peculiarities of extraction with oil extractant and quantitative determination of the amount of phytosterols and β-sitosterol in experimental samples of OE. The total content of plant sterols in OE, including considering their amount in the extractant, was in the range of 7880 mg/kg; the amount of β-sitosterol was 4638 mg/kg. Conclusion. The choice of optimal parameters for obtaining OE from UDR based on determination of phytosterol yield by gas capillary chromatography was experimentally substantiated, namely: extractant – corn oil, raw material-extract ratio – 1: 5, extraction time – 6 h, extraction method – maceration

Humans need vitamin A and obtain essential vitamin A by conversion of plant foods rich in provitamin A and/or absorption of preformed vitamin A from foods of animal origin. The determination of the vitamin A value of plant foods rich in provitamin A is important but has challenges. The aim of this paper is to review the progress over last 80 years following the discovery on the conversion of β-carotene to vitamin A and the various techniques including stable isotope technologies that have been developed to determine vitamin A values of plant provitamin A (mainly β-carotene). These include applications from using radioactive β-carotene and vitamin A, depletion-repletion with vitamin A and β-carotene, and measuring postprandial chylomicron fractions after feeding a β-carotene rich diet, to using stable isotopes as tracers to follow the absorption and conversion of plant food provitamin A carotenoids (mainly β-carotene) in humans. These approaches have greatly promoted our understanding of the absorption and conversion of β-carotene to vitamin A. Stable isotope labeled plant foods are useful for determining the overall bioavailability of provitamin A carotenoids from specific foods. Locally obtained plant foods can provide vitamin A and prevent deficiency of vitamin A, a remaining worldwide concern.
