Background: The composite flour (50% sweet potato, 30% corn, and 20% cowpea) had proven that it could substitute 100% wheat flour for biscuit making. Intake of antioxidants, dietary fiber, and protein, had been known to reduce plasma glucose level. Objective: To evaluate the effect of composite flour diet on plasma glucose levels, body weight and food consumption of streptozotocin (STZ)-diabetic rats.Method: Thirty male Sprague-Dawley rats aged ± 2 months (± 200 grams), were divided into three groups of ten rats. They were (1) non-diabetic standard diet (ND-STD), (2) diabetic (65 mg/kg of body weight by STZ induction) standard diet (D-STD), and (3) diabetic composite flour diet (1,5 g/100 g of body weight/day) (D-CFD), for 4 weeks, 3 days after STZ induction. The plasma glucose obtained from GOD-PAP method were determined before induction, after induction and every week during diet intervention.Results: Plasma glucose levels was significantly lower in D-CFD when compared with D-STD (p<0,05). The weight of D-STD was found to be significantly decreased as compared to ND-STD (p<0,05). There were no changes in food consumption of ND-STD, D-STD and D-CFD (p<0,05).Conclusion: The composite flour could reduce plasma glucose level in diabetic rats and did not give negative effects on body weight and food consumption.

Objective: The rats were randomly divided into 5 groups and each group consisted of 6 rats and the duration of treatment was 45 days. Body weight, fasting plasma glucose levels, SGOT, SGPT and ALP levels were measured. Methods: Albino Wistar male rats; 10- weeks old with a bodyweight ranged between 180-250 g were used. Anti-diabetic effect of plant Costus spicatus. In rhizome extract of ethanol extract. Result: The present study clearly indicates in the form of administration of C. spicatus rhizome extract towards glucose fed mice normalizes blood glucose level. Recent studies have revealed induced diabetic rats caused anti-diabetic effects. Histopathological studies of Pancreasin diabetic and treated groups are determined to show the protective action of the ECS. The administration of STZ resulted in a significant increase in plasma glucose level, SGOT, SGPT and ALP along with a reduction in body weight. Conclusion: All these effects were observed on 45th day. ECS and Glibenclamide of STZ induced diabetic animals restored the normal plasma glucose levels and SGOT, SGPT and ALP levels without damaging the pancreas. The aim of this study was to investigate if dietary administration of γ-aminobutyric acid (GABA)-producing Lactobacillus brevis DPC 6108 and pure GABA exert protective effects against the development of diabetes in streptozotocin (STZ)-induced diabetic Sprague Dawley rats. In a first experiment, healthy rats were divided in 3 groups (n=10/group) receiving placebo, 2.6 mg/kg body weight (bw) pure GABA or L. brevis DPC 6108 (~109microorganisms). In a second experiment, rats (n=15/group) were randomised to five groups and four of these received an injection of STZ to induce type 1 diabetes. Diabetic and non-diabetic controls received placebo [4% (w/v) yeast extract in dH2O], while the other three diabetic groups received one of the following dietary supplements: 2.6 mg/kg bw GABA (low GABA), 200 mg/kg bw GABA (high GABA) or ~109 L. brevis DPC 6108. L. brevis DPC 6108 supplementation was associated with increased serum insulin levels (P<0.05), but did not alter other metabolic markers in healthy rats. Diabetes induced by STZ injection decreased body weight (P<0.05), increased intestinal length (P<0.05) and stimulated water and food intake. Insulin was decreased (P<0.05), whereas glucose was increased (P<0.001) in all diabetic groups, compared with non-diabetic controls. A decrease (P<0.01) in glucose levels was observed in diabetic rats receiving L. brevis DPC 6108, compared with diabetic-controls. Both the composition and diversity of the intestinal microbiota were affected by diabetes. Microbial diversity in diabetic rats supplemented with low GABA was not reduced (P>0.05), compared with non-diabetic controls while all other diabetic groups displayed reduced diversity (P<0.05). L. brevis DPC 6108 attenuated hyperglycaemia induced by diabetes but additional studies are needed to understand the mechanisms involved in this reduction.

Diabetes mellitus is a metabolic disease with special characteristic which increased blood glucose level (hiperglicemic), its happens because of the abnormality of insulin secretion, insulin mechanisme or both of them. Belimbing wuluh and red guava extract can become alternative functional drink that contain rich of vitamin C to help controlling blood glucose level. The aimed of this study was to determined the effect of belimbing wuluh and red guava extract on blood glucose levels of Rattus novergicus strain Wistar diabetic model. This research was an true experimental study with a randomized post-test control group design. Twentyfour (24) male wistar strain rats weighing 200-250 grams, aged 2-3 months, were divided into 4 groups. The control group (K-) non-diabetic rats, the control group (K+) diabetic rats given standard diet, and the treatment group diabetic rats given standard diet and belimbing wuluh and red guava extract intervetion doses 10,7 ml/day (P1) and 21ml/day (P2). Measurement of pretest posttest fasting blood glucose levels was carried out using biolyzer. The data were analyzed by One Way Anova test. The results showed that there were significant differences in delta pretest and posttest fasting blood glucose levels between all groups. Blood glucose levels decreased at 55.6%. In conclusion, belimbing wuluh and red guava extract 21ml/day most effectively decreased fasting blood glucose levels in diabetic wistar rats moldel.   Keywords: belimbing wuluh, blood glucose level, diabetes mellitus, red guava

The present study explored the assessment of the antidiabetic potential of Tinospora cordifolia & Juglans regia composite extract in STZ induced diabetes in wistar rats. As streptozotocin -associated infiltrations of increase glucose level has been reported to be responsible for diabetes. We evaluated the glucose lowering potential of Tinospora cordifolia & Juglans regia on the basis of its anti-diabetic property. Rats were administered streptozotocin (55 mg/kg i.p., once) with nicotinamide (120mg/kg) to induce experimental toxicity. The development of diabetes was assessed biochemically as well as histologically 72 hours after induction of diabetes. Body weight and blood glucose levels were determined in (0, 7th, 14th, 21st, 28th) days. Serum lipid profile and enzyme estimated, (kidney, liver, pancreas) tissue was measured at the end of the experimental period. Treatment with composite extracts TCAE high dose (350 gm/kg b.w.) & JRAE high dose (800 mg/kg b.w.) and TCHE high dose (350 gm/kg b.w.) & JRHE high dose (800 mg/kg b.w.) were noted to be more effective against the streptozotocin- induced toxicity as compared to Glibenclamide (5 mg/kg b.w.). it may be concluded that streptozotocin-induced glucose may be accountable for the induction of diabetes toxicity in rats. Interestingly, improvement in body weight, glucose level, lipid profiles, biochemical parameters and histopathological changes in kidney, liver and pancreas was observed following herbal treatment in STZ induced diabetic rats. Furthermore, composite extract of TCAE (350mg/kg b.w.) & JRAE (800mg/kg b.w.) was found to be efficacious than the composite extract of TCHE (350mg/kg b.w.) & JRHE (800mg/kg b.w.).

Diabetic complications occur as a result of increased reactive oxygen species (ROS) due to long term hyperglycaemia. Honey and ginger have been shown to exhibit antioxidant activity which can scavenge ROS. The main aim of this study was to evaluate the antioxidant and antidiabetic effects of gelam honey, ginger, and their combination. Sprague-Dawley rats were divided into 2 major groups which consisted of diabetic and nondiabetic rats. Diabetes was induced with streptozotocin intramuscularly (55 mg/kg body weight). Each group was further divided into 4 smaller groups according to the supplements administered: distilled water, honey (2 g/kg body weight), ginger (60 mg/kg body weight), and honey + ginger. Body weight and glucose levels were recorded weekly, while blood from the orbital sinus was obtained after 3 weeks of supplementation for the estimation of metabolic profile: glucose, triglyceride (TG), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH): oxidized glutathione (GSSG), and malondialdehyde (MDA). The combination of gelam honey and ginger did not show hypoglycaemic potential; however, the combination treatment reduced significantly (P<0.05) SOD and CAT activities as well as MDA level, while GSH level and GSH/GSSG ratio were significantly elevated (P<0.05) in STZ-induced diabetic rats compared to diabetic control rats.

We examined the effects of handling, cage transportation, anaesthesia and repeated bleeding on plasma glucose levels in mice and rats. Plasma glucose was determined using a compact glucose analyser Antsense®, which provides a quick and accurate method without the necessity for special specimen preparation. In mice, plasma glucose was significantly elevated after primary handling or cage transportation. Anaesthesia increased plasma glucose levels two-fold, whilst repeated bleeding induced a rapid but transient increase. However, it was found to be possible to sample plasma glucose levels at one-hour intervals without any apparent effect on plasma glucose level as a result of stress from the sampling procedure. In contrast, the same set of procedures i.e. handling, cage transportation and anaesthesia, when performed on rats, seemed to have small or no observable effect on levels of plasma glucose. These results show the importance of the sampling procedure when determining plasma glucose in mice. It is recommended that the procedure which causes the least influence on endogenous glucose levels should be the method of choice and that animals should be acclimatized to the procedure, by appropriate handling, prior to sampling.

✓ Previous studies indicate that hyperglycemia, particularly that induced by exogenous glucose administration, exacerbates neurological deficits in the rat spinal cord ischemic model. The effect of inhibition of glucose uptake (glucose transporter) and initial metabolism (hexokinase) on neurological outcome was evaluated in the present investigation using the competitive inhibitors 2-deoxyglucose (2-DG) and 3-O-methylglucose (3-OMG). Sprague-Dawley rats, weighing 200 to 300 gm each, received either 0.25, 1, or 2 gm/kg 2-DG; 2 gm/kg 3-OMG; 2 gm/kg glucose; or an equivalent volume of 0.9% saline intraperitoneally. Rats were intubated and ventilated with 1% to 1.5% halothane. The aortic arch was exposed and snares were placed on the right and left subclavian arteries and the aorta distal to the left subclavian artery. The three vessels were occluded for 10, 11, 12, or 13 minutes. Lower-extremity neurological deficits were evaluated at 1, 4, 18, and 24 hours postocclusion based on a 15-point scale (normal = 0, severe deficit =15). Lower-extremity neurological deficits were significantly less severe in the groups treated with 2-DG (0.25 and 1 gm/kg) at 18 and 24 hours postocclusion (p < 0.05 for 0.25 gm/kg and p < 0.005 for 1 gm/kg, Student's t-test with Bonferroni correction). The lower 2-DG dose of 0.25 gm/kg did not significantly increase the plasma glucose level, suggesting that the glucose transporter was not markedly inhibited, and that the improved neurological outcome was more likely due to inhibition of hexokinase. The higher 2-DG dose of 1 gm/kg afforded protection despite significantly increasing the plasma glucose level, implying a strong inhibition of both the glucose transporter and hexokinase. Administration of 3-OMG, which only inhibits glucose uptake and not hexokinase, actually worsened the neurological deficit in a manner similar to that observed in rats treated with glucose. The authors conclude that the activity of the glucose transporter by itself does not significantly contribute to hyperglycemic exacerbation of neurological deficits. In contrast, the hexokinase step, at least in combination with the transporter and possibly alone, plays a significant role in hyperglycemic exacerbation of the lower-extremity neurological deficit in the paraplegic rat.

Background: There is a concern that smokers are at increased risk of diabetes and this association has received surprisingly little attention. This study aims to study the relationship of smoking with three glycaemic variables (HbA1c, fasting and 2 hour post prandial plasma glucose levels) in current smokers and describe the potential impact of smoking in the context of screening for diabetes by using one of these glycaemic variables.Methods: 150 patients attending the OPD of Santosh Medical College were part of this Cross sectional population based study. One hundred were smokers and fifty non-smokers and non-diabetics. All participants were subjected to fasting plasma glucose level, 2 hour plasma glucose level and HbA1c testing.Results: Mean values of all three variables are substantially higher in smokers as compared to non-smokers. HbA1c levels show an increasing trend with increasing age among both groups. Mean plasma glucose levels show increasing values with increasing age but the trend is not uniform. Among females, only HbA1c levels whereas among males all three variables show a strong correlation with smoking. No correlation was observed between the number of cigarettes smoked daily or smoking index and the glycaemic variables. All variables show an increasing trend as the number of years of smoking increases.Conclusions: A higher prevalence of diabetes is seen in current smokers when we consider HbA1c levels indicating the beneficial effect of using HbA1c levels for identifying current smokers at risk of diabetes mellitus.

Diabetes Mellitus (DM) is the most common of the endocrine disorder and chronic hyperglycaemia due to relative or absolute lack of insulin. The aim of the study is to investigate the the body weight profile, decreased of body weight (%) and glucose level of the bitter melon  juice and boiled extract dietin alloxan induced diabetic rats. Therats were divided randomly into three groups with fed AIN-93, the first group is control with water diet only, whereas the second group was given bitter melon juice extract diet, and third group with bitter melon boiled extract diet.  The diet of bitter melon juice and boiled extract were done by oral administration of 2ml/200 g body weight of rats for 8 days. The result showed that diet with bitter melon juice and boiled extract could improve body weight after 4 days. Additionally, the decreased of body weight percentage  afterbitter melon juice (2.68%) and boiled extract (1.89%) diet  were effective than control (8.81%). Bitter melon juice extract may effective in reduced  blood glucose levels than bitter melon boiled extract and control. The research indicate that bitter melon acts recovery body weight and regulating blood glucose level on diabetic rats.
