Objective: The objective of this study was undertaken to estimate the total phenolic contents (TPCs), in vitro antioxidant of different solvent extracts of M. oleifera leaves, oral acute toxicity and LD50 determination of the 85% methanolic extract as well as the chromatographic isolation and identification of the extract constituents.Methods: The antioxidant activity of different solvent extracts of Moringa oleifera leaves were estimated using three antioxidant assays and the total phenolic contents (TPCs) were also evaluated using Folin-Ciocalteu’s assay. The n-BuOH extract undergoes further chromatographic isolation owing to the high antioxidant activity using 2, 2'-diphenyl-1-picrylhydrazyl radical (DPPH) method, which resulted in the isolation of seven compounds.Results: The results showed that the TPCs values of the tested extracts were varied from 309.52 to 43.28 mg gallic acid equivalent/g dry extract. The reducing power antioxidant activities (RPAA) were 0.434, 0.402, 0.395, 0.149, 0.143 and 0.124, while the total antioxidant capacity (TAC) values were 316.43, 203.35, 181.56, 86.70, 76.62 and 50.83 mg ascorbic acid equivalent/g dry extract; for n-BuOH, EtOAc, 85% MeOH, H2O, CH2Cl2, and pet. ether extracts, respectively. The oral acute toxicity study of the 85% methanol extracts of M. oleifera and M. peregrina revealed that; their LD50 values were 3458.3 and 4125 mg/kg respectively, thus the two plants could be classified as slightly toxic in the scale of Hodge and Sterner which reflected their nutrient values as edible plants. The isolated compounds were identified on the basis of their 1H and 13C-NMR spectra as; cis-p-coumaric acid 4-O-(2'-O-β-D-apiofuranosyl)-β-D-glucopyranoside (1), chlorogenic acid (2), niazirin (3), 3,4-dihydroxy-β-phenylethoxy-O-α-L-rhamnopyranosyl-(l→2)-α-L-rhamnopyranosyl-(1→3)-4-O-caffeoyl-β-D-glucopyranoside (4), gallic acid (5), taxifolin (6), and benzyl-carbamo-thioethionate (7).Conclusion: The M. oleifera leaves showed promising antioxidant activities and slightly toxic behavior.

Abstract The objective of this work was to investigate the variability of physicochemical parameters, phenolic compounds and in vitro antioxidant activity of propolis collected from different apiaries in southeastern Mexico. A high variability was found in the moisture content (1.96–8.26%), ash (0.66–5.50%) and sensory characteristics of raw propolis from southeastern Mexico, but the raw propolis samples met the requirements of the quality regulations. In the same way, most of the ethanolic extracts also complied with the quality regulations. Of all the extracts, PE2 obtained from Santa Cruz showed the highest values for dry extract, content of total phenolic compounds (TPC), total flavonoids (TF) and antioxidant activity (DPPH and ABTS). The content of the individual phenolic compounds varied according to the geographical location of the apiary, but the PE2 extract resulted in the highest pinocembrin and chrysin content. A positive correlation was obtained between TPC and TF with antioxidant activity. Propolis extracts were classified into two groups through principal component analysis (PCA). These results indicate that the apiary location in southeastern Mexico influenced the characteristics of propolis.

Cordia macleodii, an important medicinal plant traditionally used for the treatment of various disorders, including wound healing, jaundice, gastric ulcer, hepatoprotective activity and antivenom potential, thus the plant showed the high antioxidant properties. Successive extraction of leaf and barks powder with methanol and water was tested for quantitative determination of total phenolics, total flavonoids and various in vitro antioxidant activities. Phytochemical screening of crude plant extracts revealed the presence of sterols, alkaloid, flavonoid, phenolic, Reducing sugar, glycosides, tannins and saponins. All extracts showed the significant total phenolic and flavonoid contents and as well as radical scavenging and iron-chelating activities. Among all, methanol leaf extract was observed to be higher antioxidant activity than that of other extract. The methanol extracts showed greater antioxidant activity by DPPH scavenging the free radical with IC50 values of 7.63 ± 0.38 μg gm-1 along with scavenged free radical in a concentration depended manner, which is compare to the standard ascorbic acid.  Our results revealed that the presence of grater antioxidant activity of C. macleodii extract may be due to the high level of flavonoid and phenolic content in the plant. These results clearly indicated that C. macleodii is an effective natural antioxidant. Key words: Cordia macleodii, antioxidant, radical scavenging, phenolics, flavonoid, phytochemical.

<strong><em>Flavanoid, steroid, and terpenoid contents of 20 Manokwari medicinal plants were analyzed.  The antioxidant activity, total phenolic contents and antitumor activity of 20 Manokwari medicinal plants were also evaluated. The result shows that 83% positif to flavanoid test, 59% positif to terpenoid test and only 25% positif steroid. Antioxidant activity and total phenolic contents evaluated using Ferric Thiocianate (FTC) and Folin-Ciocalteu methods respectively. Antioxidant activity and total phenolic contents of medicinal plants were extracted by the traditional method, boiling in water and also in 80% methanol. Twenty plants evaluated in both exstracts have significantly varies of antioxidant activities and phenolic contents,  A significant and linier correlation coefficient between the antioxidant activity and the total phenolic content was found in both aqueous (R<sup>2</sup>= 0,77) and methanol (R<sup>2</sup> = 0,85). Antitumor activity was tested using cell maurine P-388 and only 2 of medicinal plants are active to inhibit cell maurine P-388. Comparing extraction efficiency of the two methods, the methanol extracted phenolic compounds more efficiently, and antioxidant activity of the extract was higher.</em></strong>

The objective of the current study was to evaluate the antioxidant activity and enzyme inhibitory effect of different parts of medlar including fruit, leaf and flower bud by using various in vitro methods, and also determination of total phenolic and flavonoid content in the samples. Ethanol extracts of medlar parts were prepared and their antioxidant activities were determined using 1,1-diphenyl-2-picryl-hydrazil (DPPH•) scavenging and β-carotene bleaching methods. The leaf extract showed the strongest antioxidant activity. DPPHradical scavenging activity was in the order of BHA > leaf > bud > fruit. This ordering was the same for β-carotene bleaching activity, tocopherol > leaf > bud > fruit. The highest total phenolic (60.3 ± 1.69 mg GAE g-1 extract) and flavonoid (14.77 ± 1.15 mg QE g-1 extract) content were determined in leaf extract. For possible antidiabetic effects of extracts, α-amylase and α-glucosidase inhibitory activities were investigated, the bud extract showed the highest inhibition activities among the all extracts.

Attempts regarding the improvement and development of novel rice with better quality and higher productivity have been increasing. Among approaches, mutation is a direct alteration on the genome and considered as one of the most beneficial routes to acquire new beneficial traits in rice. An experiment was carried out to explore the effects of N-methyl-N-nitrosourea (MNU) mutation on the antioxidant activities, phytochemical compounds, and momilactones A (MA) and B (MB) in rice. Two rice cultivars, K1 (an original cultivar DT84) and K2 (mutated DT84), were examined. Antioxidant activities, phenolic compounds, and momilactones of the rice grain, husk, and straw portions were measured and quantified. Antioxidant activities were higher in grain and straw of K2, whereas K1 showed greater antioxidant activity in rice husk. Additionally, K2 displayed higher total phenolic contents (TPC) in grain and straw as well as lower of it in the husk, but these variations significantly differed only in the straw portion. An increase in total flavonoid contents (TFC) was observed in the husk of K1, while K2 significantly enhanced TFC in straw. Both MA and MB, two compounds obtaining antidiabetes, anticancer, antimicrobial, antigout, and antiobesity properties, were detected and quantified in grain, husk, and straw of K1 and K2 samples. Generally, the contents of MA were higher than MB in all tested portions of rice crop. MA and MB were higher in straw followed by those in husk and grain, respectively. K2 contained higher amounts of MA and MB in straw and husk, but lower contents in grain compared with those in K1. This study illustrates that MNU mutation can improve grain quality and enhance bioactive compounds in straw, husk, and grain of rice. This approach has the potential to develop functional foods from rice, and therefore help farmers in developing countries to improve value in rice production.

Crataegusfruit is one of most important fruits in Tunisian flora. Some fruits of this genus are edible. This study was undertaken in order to examine the benefits of these fruits in human health and their composition of antioxidants including total polyphenol, flavonoids, proanthocyanidins content, and total anthocyanins. The antioxidative properties of the ultrasonic methanolic extract were assessed by different in vitro methods such as the FRAP, DPPH, andβ-carotene/linoleic acid assay. We concluded that peel fraction of red fruits possessed relatively high antioxidant activity and might be a rich source of natural antioxidants in comparison with the pulp and seed fruit extract. The results also showed that hawthorn yellow fruit presents lower amounts of phenolic content, absence of anthocyanins, and less antioxidant capacity. Most of peel and seed fractions were stronger than the pulp fractions in antioxidant activity based on their DPPH IC50, FRAP values, and results ofβ-carotene/linoleic acid. The total phenolic compounds contents were also highly correlated with the DPPH method and the FRAP assay.

Dialium angolense is used in Bagira for its various medicinal properties particularly in the management of infectious diseases. In this study, the methanol and aqueous extracts of leaves and fruits were evaluated for their in vitro antioxidant and antimicrobial properties and their in vivo toxicity on Cavia porcellus. The major phytochemical classes of extracts were screened using standard in-tube reactions. The antimicrobial study was tested on Candida albicans, Escherichia coli, Salmonella typhi, Staphylococcus aureus and Streptococcus pneumoniae using agar well diffusion and dilution methods, while the antioxidant activity was evaluated by a DPPH assay. For the acute toxicity study, animals (6/group) were orally given in a single dose 5000, 1000 or 15000 mg of extract/kg body weight (BW) then observed for 14 days. In sub-acute toxicity assays, 150 or 300 mg/kg BW/day were orally given, and animals observed for 28 days. Total phenolics and total flavonoids contents ranged 1.19 to 1.61 mg GAE.g-1 and 0.45 to 1.01 mg QEg-1, respectively. The extracts presented antioxidant activity with IC50 ranging 4.9 to 6.9 µg/mL. The minimal inhibitory concentration (MIC) on tested strains ranged from 1.9 to 500 µg/mL with the aqueous extract of fruits as a most active extract: MIC=1.9 µg/mL on E. coli and C. albicans. No signs of toxicity were noted during the acute and sub-acute toxicity assessments, suggesting a maximal tolerate doses (MDT) and LD50 > 15000 mg/kg BW. This study highlights the antioxidant and antimicrobial activities of Dialium angolense and suggests that further studies be directed towards the isolation of active compounds.

Змееголовник молдавский (Dracocephalum moldavica L.) в народной медицине используется в качестве противовоспалительного, ранозаживляющего, отхаркивающего и седативного средства. В ФГБНУ ВИЛАР разработан змееголовника молдавского травы экстракт сухой под условным названием «Люкатил» (сумма фенольных соединений 64,12% в пересчёте на цинарозид). Цель работы - изучение острой токсичности и противовоспалительной активности экстракта змееголовника для разработки на его основе лекарственного препарата. Методика. Проведено определение параметров острой токсичности и противовоспалительной активности экстракта. При изучении острой токсичности экстракта по методу Кербера использованы белые нелинейные мыши-самцы в количестве 30 особей. «Люкатил» вводили животным внутрижелудочно в дозах 500, 1000, 1500 и 2000 мг/кг. Для выявления противовоспалительной активности экстракта змееголовника молдавского использована in vitro ферментная биотест-система на основе индуцибельной NO-синтазы. Для выявления противовоспалительной активности экстракта in vivo использованы нелинейные мыши-самцы. Оценку влияния экстракта в дозе 200 мг/кг на экссудативную стадию воспаления проводили на модели 1% формалинового отёка. В качества препарата сравнения использовали индометацин (5 мг/кг). Формалиновый отёк вызывали однократным субплантарным введением под апоневроз задней правой лапки мыши 0,05 мл 1% формалина в качестве флогогенного агента. Величину отёка определяли по разнице в массе лапок контрольных и опытных животных и рассчитывали процент снижения степени отёка. Результаты. При однократном введении экстракт «Люкатил» не приводил к гибели животных, изменения внешнего вида и поведенческих реакций мышей не наблюдалось. В соответствии с классификацией токсичности химических веществ по ГОСТ 12.1.007-76 «Люкатил» является малотоксичным веществом. In vitro установлена высокая противовоспалительная активность экстракта, при этом остаточная активность iNOS снижалась до 25%. Экстракт в дозе 200 мг/кг in vivo обладал статистически значимым противовоспалительным эффектом. Он подавлял развитие экссудативной фазы воспаления на 33,7%, по сравнению с контрольной группой животных, уступая противовоспалительному эффекту индометацина. Заключение. Змееголовника молдавского травы экстракт сухой под условным названием «Люкатил» является малотоксичным веществом, обладает выраженным противовоспалительным эффектом в опытах in vitro, in vivo и является перспективным объектом для дальнейшего фармакологического изучения в качестве противовоспалительного лекарственного средства.Moldavian dragonhead (Dracocephalum moldavica L.) is used in traditional medicine as an anti-inflammatory, wound-healing, expectorant, and sedative means. In our Institute, a Moldavian dragonhead herb dry extract (total phenolic content, 64.12% in cynaroside equivalent) was developed and conventionally named Lyukatil. Objective. To study acute toxicity and anti-inflammatory activity of the dragonhead extract for developing a drug based on this extract. Method. Parameters of acute toxicity and anti-inflammatory activity of the extract were assessed. The study of acute toxicity of the extract was performed using the Kerber method on male white mongrel mice (n=30). Lyukatil was administered to the animals intragastrically at doses of 500 mg/kg, 1000 mg/kg, 1500 mg/kg, and 2000 mg/kg. Anti-inflammatory activity of the Moldavian dragonhead extract was determined in vitro using an enzyme Biotest system based on inducible NO synthase. Mongrel male mice were used to study the anti-inflammatory activity of the extract in vivo. The effect of the extract at a dose of 200 mg/kg on the exudative phase of inflammation was evaluated on a model of 1% formalin-induced edema. Indomethacin 5 mg/kg was used as a reference drug. Formalin edema was induced by a single subplantar injection of 0.05 ml of 1% formalin as a phlogogenic agent under the aponeurosis of the right hind leg. The degree of edema was determined by the difference in leg weights in control and experimental animals; then the decrease in edema was calculated in per cent. Results. A single administration of the extract Lyukatil did not cause death of animals, changes in the appearance or in behavioral responses, shortness of breath, or drowsiness. In accordance with the toxicity classification for chemical substances as per GOST Standard 12.1.007-76, Lyukatil is a low-toxic substance. The extract at a dose of 200 mg/kg exerted a significant anti-inflammatory effect as shown by suppression of the exudative phase of formalin-induced inflammation by 33.7% compared to the control group. However, this effect was inferior to the anti-inflammatory effect of indomethacin. Conclusions. The Moldavian dragonhead herb dry extract under the conventional name of Lyukatil is a low-toxic substance that has a significant anti-inflammatory effect both in vitro and in vivo and is a promising target for further pharmacological studies as an anti-inflammatory drug.

The study aimed to evaluate the effects of bay (B), pandan (P), citrus leaves (C) and their combinations against starch hydrolysis enzymes (α-glucosidase and α-amylase enzymes) and antioxidant activity and to examine the role of polyphenol compounds in enzyme inhibition and antioxidant activity. Three single leaves extracts and five of their combinations were applied to inhibit α-glucosidase hydrolyzing p-nitrophenyl-α-D-glucopyranosyde or α-amylase hydrolyzing starch solution as well as to scavenge free radicals. The leaf extracts and their combination showed inhibition activities against α-glucosidase and α-amylase enzymes with range of inhibition activities were between 17.63% to 26.04% and 20.14% to 35.30% respectively. There is no significantly differ among the extracts in modulation of α-glucosidase activity, but each extract exhibited different effect on α-amylase or antioxidant activities. Mixing P with B and C increases the inhibitory activity of the extract against α-amylase as seen that percent of inhibition of BPC is significantly higher than P, eventhough their total phenolic content was not different. The synergism or antagonism effect was not observed when the extracts were combined as the enzyme inhibition or antioxidant activities are not depend on the proportion of the extract in the mixtures. The role of polyphenol compounds on inhibition of the starch digestion enzymes and on antioxidant activity was not observed. Further study is required to fully elucidate the effect of the leaf or their combinations on diabetic animal models or diabetic patients.
