Objective: phytochemical investigation of the ethyl acetate fraction (EAF) of 80% aqueous methanol extract (AME) of Dypsis leptocheilos leaves, in addition to evaluation of the antioxidant, cytotoxic and antimicrobial activities of the AME and EAF. Docking was used to predict and understand cytotoxicity of the isolated compounds. Methods: The ethyl acetate fraction (EAF) of Dypsis leptocheilos leaves was subjected to different chromatographic separation techniques. Structures of the isolated compounds were established by different spectroscopic techniques (1H/13C NMR). Antioxidant activity was evaluated by DPPH assay, while cytotoxicity was evaluated by MTT cell viability assay. Antimicrobial activity was evaluated by agar diffusion method. The docking study was conducted using Auto Dock Vina; the estrogen receptor (PDB 5t92) was used as a receptor for the docking. Results: Chromatographic separation techniques were led to the isolation of five phenolic compounds; these compounds were identified to be apigenin 8-C-β-D-glucopyranoside (Vitexin) (1), apigenin 6-C-β-D-glucopyranoside (Isovitexin) (2), luteolin 7-O-β-D-glucopyranoside (3), luteolin 8-C-β-D-glucopyranoside (Orientin) (4), luteolin 6-C-β-D-glucopyranoside (Isoorientin) (5). They were isolated and identified for the first time from this plant species. The AME and EAF showed moderate activity against Gram positive and Gram negatvie bacteria, while both of them showed similar and powerful antioxidant activity with SC50 = 12.8±0.56 µg/ml and SC50 = 17±0.77 µg/ml respectively, compared to ascorbic (reference drug) SC50 = 14.2±0.35 µg/ml. The EAF showed higher cytotoxic activity on the MCF-7 cells (human breast cancer cell line), with IC50 = 12.3 ± 1.82 µg/ml, compared to Vinblastine Sulfate (reference drug). All isolated compounds showed good binding affinity to the estrogen receptors existed in the MCF-7 cell. Conclusion: Five phenolic compounds were isolated for the first time from the EAF of Dypsis leptocheilos leaves. The AME and EAF extracts showed variable antioxidant, antimicrobial and cytotoxic activities.

Antioxidants are compounds that can inhibit oxidation reactions by binding to free radicals and highly reactive molecules. One form of reactive oxygen compounds is free radicals, these compounds are formed in the body and are triggered by various factors. Noni fruits have been used by the community as a medicine for high blood pressure, beriberi, urinating, inflammation of bile, inflammation of the intestine, dysentery, constipation, spleen pain, liver pain, diabetes and lumbago. Nevertheless, research on the identification of the active compounds of noni fruit which act as antioxidants yet. This study aims to extract and fractionation of Noni fruit, determine the antioxidant activity of Noni fruit extract, and identify the types of bioactive compounds that act as antioxidants. Noni fruit was extracted used maceration technique with 96% ethanol and then fractionated using n-hexane, chloroform and ethyl acetate solvents. Antioxidant activity from the results of noni fruit fractionation was measured by the DPPH (1.1 diphenyl-2-picrylhydrachyl) method and identification of bioactive compounds was carried out by GCMS. The results showed the greatest antioxidant activity was found in chloroform fraction with a percentage of inhibition of 78.19%. For the first time this study revealed the types of bioactive compounds from noni fruit ethyl acetate fraction which act as antioxidants are n-hexadecanoic acid, squalene, pyridin-3-carboxamide, oxime, n- (2-trifluoro methyl phenyl), and beta-sitosterol

Different parts of Pistasia atlantica have been used in traditional medicine for various purposes in Iran. The aim of this study was to measurement and compare antioxidant activity and polyphenolic compounds of crude ethyl alcohol extract and four fractions of P. atlantica leaf. Crude ethyl alcohol extract of P. atlantica leaf was prepared using maceration method and subjected to fractionation with different polarity. The antioxidant potential of all these fractions was evaluated by the 2,2 diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity method. The total phenolic, flavonoid, and flavonol components were measured with Folin-Ciocaltiue and Chlorid Aluminum methods. According to the radical scavenging capacity, the ethyl acetate fraction exhibited the highest antioxidant activity with IC50 value 1.54±0.12 µg/ml, followed by the chloroform fraction with higher percent inhibition of the DPPH with 3.4±0.11 µg/ml. The results are represented relative to a reference standard, butylated hydroxytoluene (BHT), with IC50 value of 33.5±3.67μg/ml. Among these fractions, the ethyl acetate fraction and chloroform fraction had the highest amount of total phenolic compounds with value of 532.73 and 355.14 mg GAE/g, respectively. The results of this study showed that some fractions of P. atlantica leaf extract could be used as easily accessible source of natural antioxidants

<p>In this work, we are interested in both the study of the phenolic compounds and the evaluation of the antioxidant properties of the extracts of <em>Ammi visnaga L. </em>collected from the Middle Atlas in Morocco. Phytochemical screening was carried out to highlight the qualitative composition of the secondary metabolites contained in this plant. Also, the extraction of total polyphenols was carried out by maceration using hydromethanolic and hydroacetonic mixtures. Then, the fractionation of the crude extracts was conducted by liquid-liquid extraction using successively two organic solvents with different polarities: ethyl acetate and n-butanol. The polyphenols contents were spectrophotometrically estimated using the Folin-Ciocalteau method. It showed that the ethyl acetate fraction is more abundant in phenolic compounds than the other fractions. The qualitative analysis was performed by high-pressure liquid chromatography with U.V. and electrospray ionization coupled to mass spectrometry (HPLC/UV-ESI-MS), which confirmed the richness of this species in polyphenols and particularly in flavonoids, especially by the identification of kaempferol, rhamnetin, biochanin A, ammiol. These molecules belong to flavones, flavonols, and furochromones, which are also identified both in the form of genin and of heteroside.<strong> </strong>The antioxidant activity of the different fractions was evaluated by diphenyl-picryl-hydrazyl (DPPH<sup>●</sup>). The IC<sub>50</sub> is equal to 12.87 μg / mL for the ethyl acetate fraction, against 8.8 μg / mL for ascorbic acid and 10.8 μg / mL for BHA which are used as reference compounds.</p>

Abstract “Nanicão Corupá” (Musa acuminata) comes from Southern Brazil. The tropical climate in the region provides unique characteristics, including a sweeter flavor. This difference resulted in a Geographical Indication Recognition and Designation of Origin, recognized by the National Institute of Industrial Property (INPI) in Brazil. Considering that “Nanicão Corupá” has some peculiarities related to the climate and there are no studies evaluating this banana cultivars, the purpose of the present study was to investigate the qualitative and quantitative phenolic composition of the aerial parts of “Nanicão Corupá” by HPLC-ESI-MS/MS in comparison to 46 commercial standards of phenolic compounds. Aerial parts (flower, leaves, fruit and stem) of “Nanicão Corupá” were collected and macerated in methanolic extracts, which were partitioned with solvents of different polarities (dichloromethane and ethyl acetate). The HPLC-ESI-MS/MS analysis was performed using the sample pre-treatment, chromatographic and mass spectrometer parameters. Results demonstrated that a total of 11 phenolic compounds were identified in the analyzed samples. The majority of compounds was identified in the ethyl acetate fraction (BFEF) of banana flowers: rutin (36.06 ± 0.23) and isoquercetin (28.83 ± 5). The compounds isoquercetin, naringerin and myricitrin were identified for the first time in the Musa genus.

Research on the antioxidant potential of the secondary metabolite compound, ethyl acetate from ebony bark (Diospyros celebica Bakh.) has been conducted. This study aims to characterize and determine the antioxidant activity of secondary metabolites of ethyl acetate fraction from ebony bark. Ethyl acetate extract was fractionated using Liquid Vacum Chromatography (KVC) and Gravity Column Chromatography (KKG). The isolated compounds were characterized using UV-Vis and IR spectrophotometers. Data on maximum absorption of λmax 273 nm from UV-Vis analysis showed the presence of chromophore phenol. IR spectral data showed several specific peaks, among others, at wave number (ʋ) of 3425.58 cm-1 indicating the presence of –OH group, C-H aliphatic bond at ʋ of  2924.09 cm-1  and 2854.65 cm-1, and the peak at ʋ of 1604.77 -1465.90 cm-1 and 3078.39 cm-1 indicated the aromatic group, reinforcing the assumption that the isolates produced were phenolic compounds. IC50 value of 75.47 µg/mL from antioxidant activity testing showed that the isolates obtained were antioxidant compounds with strong antioxidant categories. Abstract: An isolation and identification of phenolic compounds from Halban Leaves (Vitex pinnata Linn), a traditional medicinal plant in Aceh, has been done. Halban leaves were extracted through methanol maceration followed by isolation. The methanol extract went into n-hexane and ethyl acetate partition consecutively resulting in 3 different fractions of ethyl acetate and n-hexane extract.  The ethyl acetate fraction demonstrated the most promising antioxidant and cytotoxic activity. Therefore, purification of ethyl acetate fraction was done by column chromatography [SiO2; (i) n-hexane-ethyl acetate 10:1, 1:1 ; (ii) n-hexane-ethyl acetate = 2 : 1); (iii) n-hexane-acetone = 1:1]. The pure isolate as identified by using UV, IR, NMR, and MS spectra afforded 2 phenolic compounds, namely (1) 4-hydroxymethyl benzoate and (2) p-hydroxymethyl benzoic acid (PHBA). The antioxidant activity of compound 1 and cytotoxicity activity of 2 expressed in IC50  and LD50 was 41.08 ppm and 59.41 ppm, respectively.Abstrak: Isolasi dan identifikasi senyawa fenolik dari Daun Halban (Vite xpinnata Linn) tanaman obat tradisional asal Aceh telah dilakukan. Isolasi ini dilakukan dengan cara mengekstraksi daun halban dengan cara maserasi menggunakan pelarut metanol, Ekstrak metanol kemudian dipartiisi dengan n-heksan dan etilasetat secara berturut-turut  sehingga diperoleh  tiga  ekstrak yaitu ekstrak etil asetat, n-heksan dan air. Ekstrak etil asetat memiliki bioaktivitas sebagai antioksidan dan juga toksisitas. Pemurnian dilanjutkan dengan  cara kromatografi kolom [SiO2; (i) n-heksan-etilasetat   10:1,  1:1 ; (ii) n-heksan-etilasetat = 2 : 1); (iii) n-heksan-aseton = 1:1] dan diperoleh isolat  murni. Isolat murni tersebut diidentifikasi berdasarkan interpretasi data spektra UV, IR, NMR dan MS. Hasil interpretasi data menunjukkan daun halban (V. pinnata Linn) mengandung dua senyawa fenolik yaitu (1) 4-hidroksi metil benzoat  dan (2) asam para hidroksi benzoat (PHBA). Senyawa 1  memiliki aktivitas antioksidan dengan nilai IC50 = 41,08bpj dan senyawa 2 memiliki aktivitas toksisitas dengan nilai LC50 = 59,41.

Stevia rebaudiana is a perennial shrub belonging to the Asteraceae family. Recently, it has been introduced to Morocco from its native Paraguay; its molecules have a sweetening power that is about 300 times sweeter than sucrose. This characteristic makes of this plant a considerable natural noncalorific sweetener in case of hypoglycemic or low carbohydrate diet. This important interest has prompted us to make further studies on this plant. The objective of this work is to show the value of Stevia rebaudiana grown in the Larache region of Morocco by the chemical characterization and evaluation of the antioxidant activity of its extracts. A phytochemical screening was carried out to highlight the qualitative composition of secondary metabolites. This analysis showed the presence of flavonoids, tannins, oses and holosides, sterols, triterpenes, and free anthraquinones. However, the absence of alkaloids and reducing compounds has been observed. In addition, extraction of the total polyphenols was carried out by maceration using a 70% methanol–water mixture. The yield is of the order of 28.6%. Subsequently, fractionation of the crude extract was carried out by successively using three organic solvents of different polarities: chloroform, ethyl acetate, and n-butanol. Polyphenol dosage with Folin-Ciocalteu’s reagent showed that ethyl acetate fraction is richer in phenolic compounds (26.4%) than the other fractions. Flavonoids dosage with aluminum trichloride showed the richness of this plant in these compounds. The antioxidant activity of different fractions was evaluated by 2,2-diphenyl-1-picrylhydrazyl-free radical scavenging method and ferric reducing/antioxidant power method; the values of the 50% inhibitory concentrations (IC50) were determined graphically. It is equal to 0.32 mg/ml for the ethyl acetate fraction, compared to 0.08 mg/ml for the ascorbic acid used as a reference. In this study, we have shown that Stevia rebaudiana is very rich in phenolic compounds and possesses a very important antioxidant power.

Introduction: The ethyl acetate fraction of the Saudi Lavandula coronopifolia Poir has been previously reported to have hepatoprotective activity against ethanol-induced oxidative stress. The aim of the current study was to investigate the chemical composition, cytotoxic effect, and antioxidant activities of ethyl acetate fraction of the aerial parts of Saudi L. coronopifolia Poir. Methods: Air dried aerial parts of L. coronopifolia were extracted using 90% ethyl alcohol. The dried extract was suspended in water, defatted with light petroleum and then fractionated with ethyl acetate. The ethyl acetate fraction was subjected to ultra performance liquid chromatography-tandem mass spectrometeric (UPLC-ESI/MS/MS) analysis in a negative ionization mode. The antioxidant activity of the fraction was determined using free radical 2,2-diphyenyl-picrylhydrazyl (DPPH) scavenging assay and its cytotoxic effect against HepG2 (human hepatocarcinoma) and MCF-7 (human breast carcinoma) cells were determined using (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) tetrazolium (MTT) cell viability assay. Results: The major components of the ethyl acetate fraction included carvacrol-O-diglucoside, (34.98%) and trihydroxy ursolic acid (12.07%). Moreover, the DPPH radical scavenging activity of ethyl acetate fraction was measured. The ethyl acetate fraction revealed an antioxidant potential with EC50 17.8 ± 1.3 µg/mL. Additionally, he ethyl acetate fraction showed cytotoxic activity against HepG-2 and MCF-7 cells with IC50=29.3 ± 0.9 µg/mL and 14.6 ± 0.3 µg/mL, respectively. Conclusion: The ethyl acetate fraction of the Saudi L. coronopifolia has antioxidant activity and also cytotoxic activity against breast and liver cancer cells.

This study aims to find the treatment of diabetes using natural materials by exploring plants in the province of East Nusa Tenggara. his research was conducted out by extracting the Strychnos nitida G.Don stem using a method of maceration by ethanol 70%. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Simplicia from maceration and fractionation results were then tested for antioxidant activity, α-glucosidase inhibition activity and identification of active compounds. The results showed that ethyl acetate fraction had the lowest IC50 value of 86.83 μg / ml. Results of the α-glucosidase activity test showed that ethyl acetate fraction and n-heksan fraction at 900 ppm had the highest percentage of inhibition of 34.23% and 33.89%. Identification using LCMS/MS method showed that ethyl acetate fraction consist of Benzenemethamine, N, N-dioctyl- as an antioxidantcompound and compound 24-methyl-5-cholestone-hexol as an antidiabetic compound. From the results of this study, we concluded that the extract of kayu ular Strychnos nitida G.Don stem has inhibition activity toward α-glucosidase enzyme.
