Antibiotic residues when present in animal tissues, through food chain, can enter human body, causing allergic reactions or facilitating the development of resistant bacterial strains. In order to determine the presence of antibiotics in animal tissues, it is appropriate to use convenient, reliable and sensitive methods. Microbiological methods applied for the detection of antibiotic residues in primary products of animal origin are based on the sensitivity of specific bacterial strains to a particular group of antibiotics. Regulatives on the amount of pesticides, metals and metalloids and other toxic substances, chemotherapeutics, anabolics and other substances which can be found in food ("Off. Gazette", No. 5/92, 11/92 - corr. and 32/02), state that milk and milk products can be used in commercial purposes only if not contain antibiotics in quantities that can be detected by reference methods. The applied method is modified STAR (Screening test for detection of antibiotics) protocol, regulated by the CRL (Community Reference Laboratory) Fougeres, France, in which the initial validation of the method had been carried out. In accordance with the demands of Regulative Commission EC No657/2002, the sensitivity of modified STAR protocol for beta lactam antibiotics group was examined , that is, there was carried out a contracted validation of the method, which initial validation had been performed at CRL. In a couple of series of experiments, 20 blank samples of raw cow milk originating from animals not treated by antibiotics, had been examined. By the beginning of the experiment samples were stored in a freezer at -20?C. Samples of raw cow milk enriched by working solutions of seven beta-lactam antibiotics, in order to obtain concentrations at the level of 0.5, 1 and 1.5 MRL (Maximmum Residue Limit) for each given antibiotic (Commission Regulation EC No. 37/2010). For detection of beta-lactam antibiotics, there was used Kundrat agar test with previously inoculated G.stearothermophilus ATCC 10149 strain. Aliquots of 30 _l of working solution at 0.5, 1 and 1.5 MRL concentration level, for each antibiotic, were inflicted on two paper disks placed on inoculated Kundrat agar surface. Petri plates with Kundrat agar previously inoculated with G.stearothermophilus , on which the samples were deposited, were incubated for 12-15h at 55oC. The obtained width of microorganisms growth inhibition zone, that is supposed to be at least 2.0 mm, measured from the disc edge, demonstrated the capability to detect all the tested 7 antibiotics from the beta lactam group at a level below the MRLs. Consequently, this proves that use of this method it is possible to meet the demands of Regulative Commission EC No. 37/2010.

Introduction. Pseudomonas aeruginosais the most frequently isolated organism as it acts as the opportunistic pathogen and can cause infections in immunosuppressed patients. The production of different types of beta-lactamases renders this organism resistant to many commonly used antimicrobials. Therefore, the aim of this study was to document the antibiotic resistance rate inPseudomonas aeruginosaisolated from different clinical specimens.Methods. Pseudomonas aeruginosarecovered was identified by standard microbiological methods. Antibiotic susceptibility testing was performed by modified Kirby-Bauer disc diffusion method following Clinical and Laboratory Standard Institute (CLSI) guidelines and all the suspected isolates were tested for the production of ESBLs, MBLs, and AmpC.Results.Out of total (178) isolates, 83.1% were recovered from the inpatient department (IPD). Majority of the isolates mediated resistance towards the beta-lactam antibiotics, while nearly half of the isolates were resistant to ciprofloxacin. Most of the aminoglycosides used showed resistance rate up to 75% but amikacin proved to be better option. No resistance to polymyxin was observed. ESBLs, MBLs, and AmpC mediated resistance was seen in 33.1%, 30.9%, and 15.7% isolates, respectively.Conclusions. Antibiotic resistance rate and beta-lactamase mediated resistance were high. Thus, regular surveillance of drug resistance is of utmost importance.

The aim of this study was to determine the prevalence of methicillin-resistant Staphylococcus (MRS) in healthy goats on the Island of Tenerife, Spain, as well as to identify the phenotypic and genotypic characteristics of the strains found. A cross-sectional prevalence study was conducted. A total of 158 goats from 15 different farms were sampled between September 2017 and January 2018. The percentage of positive samples of methicillin-resistant Staphylococcus aureus (MRSA) was 15.8% (25/158) and that of methicillin-resistant coagulase-negative staphylococci (MRCoNS) was 6.9% (11/158). All MRSA isolates from goats belonged to one clonal group showing Multi-Locus Sequence type 398. All strains studied (n = 36) were resistant to non-carbapenem beta-lactam antibiotics and susceptible to teicoplanin, linezolid, vancomycin, rifampicin, quinupristin-dalfospristin and mupirocine. In MRSA isolates, the highest percentage of resistance obtained, besides beta-lactam non-carbapenem antibiotics, was to trimethoprim-sulphamethoxazole and, in the case of MRCoNS isolates, to phosphomycin and erythromycin. A total of 12 resistance patterns were obtained, presenting differences between patterns obtained for MRSA and MRCoNS, with 7 different patterns for MRSA and 5 for MRCoNS. We therefore consider it essential to expand the epidemiological study of these strains of animal origin, as well as to increase surveillance and control measures at all stages of the food chain.

OBJECTIVE: To present a case of Rhodococcus equi (RE) pneumonia and discuss its pathophysiology and treatment. CASE SUMMARY: An HIV-positive patient presented with pneumonia. A lung biopsy was performed after sputum and thoracentesis cultures failed to identify a pathogen. The lung biopsy revealed an unidentifiable, diphtheroid-like, gram-positive rod. A bronchoscopy performed five days after the lung biopsy produced the same diphtheroid-like, gram-positive rod. The patient was treated with several injectable antibiotics, but emergence of resistance to two of the antibiotics was suspected. Two weeks after the bacterial isolate was sent to a reference laboratory, it was identified as RE. The patient was discharged on oral antibiotics and experienced no recurrence of RE pneumonia. CONCLUSIONS: RE can be difficult to identify in the microbiology laboratory, or it may be assumed to be a colonizing diphtheroid. The isolation of difficult-to-identify, gram-positive rods, or diphtheroids, from a pulmonary source in a patient with decreased cell-mediated immunity should cause one to suspect RE. RE has been noted to develop resistance to beta-lactam antibiotics during therapy. A prolonged course of combination antibiotic therapy directed at the intracellular component of infection is necessary.

Enterobacteriaceae producing extended-spectrum beta-lactamase (ESBL) enzymes are resistant to beta-lactam agents and are also commonly multidrug resistant being associated with the resistance to other classes of antibiotics.The aim of our study was to characterise resistance patterns in non-beta-lactam antibiotics of ESBL-producing Enterobacteriaceae strains isolated from faecal matter of pets and owners.The study was carried out on 63 samples of faecal matter (42 from pets and 21 from owners). The ESBL screening was carried out using the Brilliance ESBL Oxoid chromogenic medium. The isolated strains that generated characteristic presumptive ESBL-producing colonies were cultivated on 5% sheep blood medium for the extraction of bacterial DNA using the boiled preps technique. The confirmation of E. coli species was performed molecularly based on the detection of blauidA and blauspA genes. Other Enterobacteriaceae species were identified based on the minimum biochemical characteristics using the MIU and TSI medium. The phenotypical confirmation of presumptive ESBL-producing strains was carried out using the Double Disc Synergy Test (DDST) using a combination of 3rd generation cephalosporins and beta-lactamase inhibitor agents. The determination of the resistance degree in other classes of antibiotics was carried out through the Kirby-Bauer diffusimetric method, and the results were interpreted according to the CLSI standard.Following the species investigation of isolates, 60/63 (95.28%) belonged to the E. coli species and 3/63 (4.72%) to the K. pneumoniae species. Animal isolates were resistant to sulphonamides (54.76% resistance to SXT), fluoroquinolones (45.23% resistance to ENR) and tetracyclines (54.75% resistance to TE). In addition to strains of animal origin for isolates of human origin, an increased resistance has been noticed to phenicols and aminoglycosides.This study has identified a high prevalence of ESBL-producing Enterobacteriaceae strains and associated with multidrug resistance for pets and their owners.

The term "functional foods" comprises some bacterial strains and products of plant and animal origin containing physiologically active compounds beneficial for human health and reducing the risk of chronic diseases. Among the best known functional compounds probiotics, prebiotics and natural antioxidants should be given as examples. These substances can be obtained by biotechnological methods and by extraction from plant or animal tissues.

Antibiotic resistance of enterobacteria contaminating food products of animal origin has become one of the serious health problems in recent years. Increased resistance of bacteria of Klebsiella genus to antimicrobials leads to their adaptation and spread of high - and multi-resistant strains. The study of 44 samples of poultry meat, dairy products and food established that the frequency of detection of Klebsiella is 22%. Most of the isolated food isolates were sensitive to beta-lactam antibiotics, cephalosporins, fluoroquinolones and chloramphenicol. In 30% of cases resistance of Кlebsiell food isolates to penicillin and ampicillin, tetracycline antibiotics, gentamicin, streptomycin and nitrofurantoin was found. The presence of dissociation processes of Klebsiella food isolates and antibiotic resistance formation is shown.

The objective of this study was (i) to analyze antibiotic residues, two which were beta-lactam antibiotics, one tetracycline, and one quinolone in the milk of lactating animals; (ii) to evaluate the interference of the drug ceftiofur which is considered as discard-zero. The SNAPduo™ ST Plus kit was used to evaluate the presence of beta-lactam antibiotics and tetracyclines in natural milk. Medications based on penicillin, ceftiofur, enrofloxacin, and oxytetracycline were used. As expected, the milk from control animals and animals administered with enrofloxacin did not present antibiotic residue because it is not one of the classes detected by the SNAPduo™ ST Plus kit, and thus it was used to prove the efficiency of the test. Ceftiofur, known as a “discard-zero” antibiotic, tested positive for the SNAPduo™ ST Plus kit and the BetaStar Combo® “S” test. Chemical residues were detected in cow’s milk treated with different groups of antibiotics after the withdrawal time indicated in the package inserts of the medicines used.

The objective of the paper was to verify the convenience of the application of three standardized detection systems: disk diffusion method, Delvotest SP and Penzym S 100 to control the antibiotic residues in goat’s milk (β-lactam antibiotics and cephalosporins, aminoglycosides, tetracyclines, macrolides and others). It has been found that despite of certain specificity of goat’s milk versus cow’s milk the values of the majority of detection limits mutually correspond approximately to 90 %. The sensitivity of tests manifested itself in the following order: Penzym S 100 > Delvotest SP > disk diffusion method (the sensitivity was even several times lower). Inasmuch as the treatment of mastitis is carried out by using β-lactam antibiotics and cephalosporins, the above-indicated rapid methods (especially Penzym S 100 and Delvotest SP) can be recommended for the routine purposes of accomplishing a rapid hygienic control of goat’s milk.

Background: Globally, blaCTX-M-15 beta-lactamases are the most popular extended spectrum beta-lactamase alleles that are widely distributed due its mobilisation by mobile genetic elements in several compartments. We aimed to determine the conjugation frequencies and replicon types associated with plasmids carrying blaCTX-M-15 gene from Extended Spectrum Beta-lactamase producing isolates in order to understand the dissemination of resistance genes in different compartments. Material and methods: A total of 51 archived isolates carrying blaCTX-M-15 beta-lactamases were used as donors in this study. Antibiotic susceptibility tests were performed as previously described for both donors and transconjugants. Conjugation experiment was performed by a modified protocol of the plate mating experiment, and plasmid replicon types were screened among donor and transconjugant isolates by multiplex Polymerase Chain Reaction in a set of three primer panels. Results: The conjugation efficiency of plasmids carrying blaCTX-M-15 was 88.2% (45/51) with conjugation frequencies in the order of 10−1 to 10−9 and a 100% transfer efficiency observed among E. coli of animal origin. Majority of donors (n = 21) and transconjugants (n = 14) plasmids were typed as either Inc FIA or Inc FIB. Resistance to non-beta-lactam antibiotics was transferrable in 34/45 (75.6%) of events. Ciprofloxacin, tetracycline and sulphamethoxazole-trimethoprim resistance was co-transferred in 29/34 (85.3%) such events. Gentamicin resistance was transferred in 17/34 (50%) of events. Conclusions: Majority of plasmids carrying blaCTX-M-15 were conjugatively transferred by IncF plasmids along with non-beta lactam resistance. There is a need for more research on plasmids to understand how plasmids especially multi replicon plasmids interact and the effect of such interaction on conjugation. One Health approach is to be intensified to address antimicrobial resistance which is a public health threat.
