Silver nanoparticles synthesized from aqueous extract of mushroom Pleurotus ostreatus exhibited inhibitory effect at the concentration of 12.5, 25, 50, and 100 mg/ml against some pathogenic bacteria and fungi such as Candida albicans, Candida guilliermondii, Candida krusei, Candida zeylanoides, Geotrichum klebahnii, Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. The maximum inhibition zone was observed against C. zeylanoides at the concentration of 100 mg/ml was 24.5 mm, while the minimum inhibition zone was observed against Geotrichum at the concentration of 25 mg/ml was 8 mm and the concentration of 12.5 mg/ml was not effective against some species.

Background: This study for the first time presents an eco-friendly and room temperature procedure for biologically synthesizing silver (Ag) nanoparticles from waste banana plant stems.Methods: A simple and straightforward green chemistry based technique used waste banana plant stems to act as both reducing agent and capping agent to produce Ag nanoparticles, which were subsequently characterized. In addition, antibacterial studies were conducted using the Kirby-Bauer sensitivity method.Results: Advanced characterisation revealed the Ag nanoparticles had a variety of shapes including cubes, truncated triangular and hexagonal plates, and ranged in size from 70 nm up to 600 nm. The gram-negative bacteria Escherichia coli showed the maximum inhibition zone of 12 mm.Conclusions: The study has shown that waste banana plant stems can generate Ag nanoparticles with antibacterial activity against Escherichia coli and Staphylococcus epidermis.

Bacterial strains with inhibitory effect on Salmonella Hartford, Listeria monocytogenes, Yersinia enterocolitica, and Escherichia coli, respectively, were isolated. Out of the 64 bacteria originated from food processing environments, 20 could inhibit at least one of the tested pathogens, and it was proved that growth decline of the pathogenic bacteria was more remarkable by co-culturing than by using cell-free supernatants of the isolates. Seven different genera (Pseudomonas, Bacillus, Paenibacillus, Macrococcus, Staphylococcus, Serratia, and Rothia) reduced the pathogens’ growth during the time period of analysis, and the strongest inhibitory effect was observed after 24 h between 15 and 30 °C. Sensitivity of the tested human pathogenic bacteria against the inhibitory strains was distinct, as Y. enterocolitica could be inhibited by numerous isolates, while S. Hartford proved to be the most resistant. Our results reveal that the isolated bacteria or their excreted metabolites could hinder pathogen growth when used in sufficient quantities.

ABSTRACT Introduction The main goal of endodontics is the prevention of apical periodontitis. This is due to the presence of persistent pathogenic microorganisms, such as Enterococcus faecalis, and its ability to directly cause acute and chronic inflammation in the periapical tissues. Lactobacillus has been shown to promote health in the intestines as well as to inhibit the growth of certain problematic oral bacteria. This study explores shifting the established paradigm of endodontic treatment, which has focused on eliminating all bacteria from the canal system and on elimination of the problematic bacteria through introducing probiotics. A preliminary work was performed to evaluate the possible effectiveness of probiotics in preventing the growth of E. faecalis. Materials and methods Two probiotic cultures Lactobacillus plantarum ATCC 8041 and Lactobacillus rhamnosus ATCC 7408 were selected to check their antimicrobial activity against E. faecalis ATCC 29212 by two methods: agar cup/well diffusion method and deferred antagonism test. Agar cup method A total of 0.5 mL of requisite test pathogen culture was inoculated into 20 mL of molten sterile Mueller and Hinton agar and cooled to 45 ± 2°C. Circular wells of diameter 10 mm were punched in each of the poured plates. Appropriately diluted test samples were added to the above-punched wells. The plates were incubated upright position at 37°C for 24 hours in aerobic conditions. Postincubation, zone of inhibition was measured. The cell-free supernatant of Lactobacillus species was also evaluated for antimicrobial activity. Deferred antagonism test The test probiotic strain was standardized to 0.1 optical density (OD) at 600 nm and inoculated in a 1 cm wide diametric streak across the surface of trypticase soy agar + yeast extract + calcium carbonate (TSYCa) agar using a sterile cotton swab. Then, the plate agar containing the test strain was incubated at 37°C for 24 hours under aerophilic condition and then standardized to 0.1 OD at 600 nm for overnight (18 hours, 37°C). A purified culture of indicator strain (pathogen) was streaked at right angles to the line of original producer growth. Postincubation plates were observed for the inhibition zone width of the indicator strain. Results Under the conditions of this study, Lactobacilli had an inhibitory effect on the growth of E. faecalis by agar cup method but not by deferred antagonism test. Conclusion This pilot study demonstrated that probiotics show a potential in root canal therapy. Clinical Significance If probioitcs are effective against endodontic pathogens, they can be potentially used as intracanal medicaments. This will be a novel concept of introducing bacteriotherapy in endodontics and replacing pathogenic bacteria by healthy bacteria, normal flora. How to cite this article Bohora AA, Kokate SR. Abstract: The aim of this study is to investigate the antibacterial effect of endophytic fungi originated from the root of mangrove Rhizophora stylosa growing on Mangrove Plantation Area around DAS Simpang 5 Jl. Piere Tendean Manado. The method of this research was using combination of PDA and NA media. Two species of endophytic fungi were extracted from the root of R. stylosa, black mycelium fungi as isolate A and brown mycelium fungi as isolate B. The activities of both isolates were tested against pathogenic bacteria Staphylococcus aureus and Escherichia coli. The result showed the same inhibition zone of isolate A and B to S. aureus, while, isolate A showed bigger the inhibition zone in comparison to isolate B against E. coli. As a conclusion, the endophytic fungi taken from the root of R. stylosa have antibacterial activity toward S. aureus and E. coli. Piere Tendean Manado. Metode penelitian dilakukan dengan menggunakan kombinasi media Potato Dextrose Agar (PDA) dan Nutrient Agar (NA). Dari akar bakau R. stylosa diperoleh dua  jenis jamur endofit yaitu isolat A dengan karateristik miselium jamur berwarna hitam dan isolat B yang dengan karakteristik miselium berwarna coklat. Kedua isolat ini selanjutnya diujikan aktivitasnya terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan isolat A dan B memberikan  zona hambat yang sama terhadap bakteri Staphylococcus aureus dan pada bakteri Escherichia coli isolat A menunjukkan diameter zona hambat yang lebih besar dibandingkan isolat B. Kesimpulan, Jamur endofit yang diisolasi dari akar bakau R. Stylosa memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan E. coli. Kata kunci: jamur endofit, Rhizophopra stylosa, antibakteri, Staphylococcus aureus, Escherichia coli.

ABSTRACTSponges are one of the biota components that make up coral reefs which are quite widely distributed. The metabolite content in the sponge can ward off and inhibit the pathogenic bacteria that interfere with it. This study aims to determine the activity of inhibiting bacterial growth from the extract and fraction of Leucetta chagosensis sponge against the growth of Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli bacteria. The samples were extracted using the maceration method with 95% ethanol solvent and then fractionated using 3 solvents with different polarity levels, namely methanol, n-hexane and chloroform. Activity test using the disk diffusion agar method of Kirby and Bauer. Only the MeOH fraction was able to inhibit the growth of E. coli bacteria with an average inhibition zone of 6.88 mm. Whereas for S.aureus bacteria extracts and all fractions showed activity to inhibit bacterial growth with an average inhibition zone of EtOH (6.61 mm), CHCI3 (6.68 mm), n-hexane (7.83 mm) and MeOH (8.00 mm), respectively. Kandungan metabolit yang ada di dalam spons dapat menangkal dan menghambat bakteri patogen pengganggunya.  Penelitian ini bertujuan untuk melihat aktivitas menghambat pertumbuhan bakteri dari ekstrak dan fraksi spons Leucetta chagosensis. terhadap pertumbuhan bakteri Gram positif Staphylococcus aureus dan Gram negatif Escherichia coli. Sampel di ekstraksi menggunakan metode maserasi dengan pelarut etanol 95% lalu di fraksinasi dengan menggunakan 3 pelarut dengan tingkat kepolaran yang berbeda yaitu metanol, n-heksan dan kloroform. Uji aktivitas menggunakan metode disk diffusion agar Kirby dan Bauer.  Hanya fraksi MeOH yang mampu menghambat pertumbuhan bakteri E.coli dengan zona hambat rata-rata 6,88 mm. Sedangkan terhadap bakteri S.aureus ekstrak dan semua fraksi menunjukan aktifitas menghambat pertumbuhan bakteri dengan rata-rata zona hambat masing-masing EtOH (6,61 mm), CHCl3 (6,68 mm), n-Heksan (7,83 mm), dan MeOH (8,00 mm). Semua aktivitas yang ditunjukan dikategorikan lemah (weak).Kata kunci : Antibakteri, Leucetta chagosensis, Staphylococcus aureus,  Escherichia coli

Background: The study for the first time demonstrates an eco-friendly and room temperature procedure for biosynthesizing gold (Au) nanoparticles from waste Macadamia nut shells. Currently Australia contributes around 40% to the global market and generates around AUS $150 million of export revenue. However, a consequence of large nut production is the generation of large quantities of waste nut shells. The green chemistry-based method is clean, nontoxic and eco-friendly. The method presented in this study produced a variety of Au nanoparticle sizes and shapes.Methods: The straightforward green chemistry-based technique used waste Macadamia nut shells to generate Au nanoparticles, which were subsequently studied using several advanced characterization techniques. Furthermore, the Kirby-Bauer sensitivity method was used to evaluate the antibacterial properties of the extracts and synthesized gold nanoparticles.Results: Advanced characterisation revealed the Au nanoparticles were crystalline, ranged in size from 50nm up to 2µm, and had spherical, triangular and hexagonal morphology. The gram-negative bacteria Escherichia coli produced a maximum inhibition zone of 11mm, while Staphylococcus epidermidis produced a maximum inhibition zone of 9mm.Conclusions: The study has shown that waste Macadamia nut shell extracts have no antibacterial activity, but the synthesised Au nanoparticles did display antibacterial activity to both Escherichia coli and Staphylococcus epidermidis. Thus, the present work has demonstrated a waste valorisation strategy that can be used to produce high-value Au nanoparticles with antimicrobial properties for use in future pharmaceuticals.

Present days due to the pathogenic bacteria and fungi, the human mortality rate has increased in number. The synthetic antimicrobial and antifungal are used in the treatment of various fungal and bacterial diseases, but have various side effects which limit's their use. Traditionally Pithecellobium dulce plant has been used for treatment of antifungal and antimicrobial infection. In this article we are comparing the activity of antimicrobial and antifungal properties of Pithecellobium dulce plant using leaves extract on gram negative bacteria,gram positive bacteria and fungi.Methanolic extraction of Pithecellobium dulce leaves was carried out using Soxhlet method and the phyto constitutional analysis was carried out for presenceof alkaloids, avonoids, glycosides, saponins, phytosterols, and triterpenoids.The inhibitory effect was determined by Cup plate method, by the measurement of zone of inhibition.The outcome was the leaves extract consisted of active phytoconstituents which were phenols, glycoside, sugar and avanoid due to which the extract showed signicant zone of inhibition,by which property of antibacterial and antifungal was listed. It can be concluded that the leaves of Pithecellobium dulce consist of active phytoconstituents which can be used as potent antimicrobials

A double green approach to the synthesis of silver nanoparticles using chitosan as a reducing agent and microwave radiation has been carried out in this study. Chitosan was extracted from marine crab carapace and the morphology of the produced chitosan was characterized using Scanning Electron Microscope (SEM). The production of silver nanoparticles (AgNPs) was monitored by the formation of surface plasmon resonance (SPR) at the λmax= 420 nm, indicates the reduction of the oxidation state of Ag+ to Ag0. The XRD data confirms the crystallinity nature of obtained AgNps, while the SEM and TEM revealed the formation of near-spherical shapes, polydisperse, heterogeneous distribution of AgNps with the size range between 7 to 25 nm. The antimicrobial activities of the AgNPs were investigated using The prepared AgNPs demonstrated dual mechanism action with chitosan, which acted as a stabilizer on the surface of AgNPs, and enhance the inhibition zone against E. tarda and E. coli. Therefore, the synthesized AgNPs in this study may have excellent potential for clinical application as it is green, low-cost and eco-friendly.

The resin extract of Commiphora myrrh is Widely used in the folk medicine. The studying myrrh resin extract include moisture. minerals such as (Ca, Fe, Mg, Na, Cu and Zn), protein, total fat and crude fiber. In this study used Muffle furnace, Kjeldahl methods Soxlet and atomic absorption. HPLC using to evaluating Polyphenol constituents of myrrh different resin extract (ethanol, ethyl acetate, petroleum ether and chloroform) as Conc. (µg / g) and in all extract (ethanol, ethyl acetate and petroleum ether and chloroform) it contained Chlorogenic acid, gallic acid Catechin, Coffeic acid, caffeine, Syringic acid, Coumaric acid, Ferulic acid, Naringenin, 4`.7-Dihydroxyisoflavone, Cinnamic, Propyl Gallate Vanillin, Querectin and Acid Ellagic acid in different concentration percentage and area The effect of Commiphora myrrh (ethanol, ethyl acetate, petroleum ether and chloroform) resin extract against four different pathogenic bacteria Salmonella typhimurium, Pseudomona aeruginosa, Escherichia coli, and Bacillus cereus, were examine by Mueller Hinton Agar and measuring inhibition zone (diameter mm), show that there were significant different among bacteria and different method of extract. All different Commiphora myrrh seed extract (aqueous, ethyl acetate and petroleum ether) have high activity against Candida albicans fungus. The study was conducted to identified the Commiphora myrrh nutritive value, polyphenol Compound and the activity against bacteria and fungi.
