Although Bacteroides fragilis is a bacterium present within gut microbiota, the toxin producer strain, known as enterotoxigenic B. fragilis (ETBF), is associated with diarrhea in children less than 5 years of age. This study includes 69 diarrheal and 29 non-diarrheal (control) samples collected from children less than 5 years old. DNA was extracted directly from stool specimens and directed to conventional PCR targeting beta-isopropylmalate dehydrogenase (leuB) gene, used for detection of B. fragilis,  and Bacteroides fragilis toxin (bft) gene, used for the detection of ETBF. The results showed that the prevalence of leuB gene was 78 (79.6%) including 56 (81.2%) in diarrheal and 22 (75.9%) in non-diarrheal subjects, while that of bft gene was only 3 (3.1%) including 2 (2.9%) in diarrheal and 1 (3.4%) in non-diarrheal subjects. Based on sequencing of bft-positive specimens, both bft-1 and bft-2 isoforms were represented in diarrheal specimens, whereas only bft-1 was found in the control specimens. In conclusion, this study examined for the first time the leuB and bft gene in a specimen of Iraqi children with diarrhea and showed no significant differences between diarrheal and control groups in both genes.

The cryopreservation of immature oocytes at the germinal vesicle (GV) stage would create an easily accessible, non-seasonal source of female gametes for research and reproduction. The present study investigated the ability of ovine oocytes vitrified at the GV stage using a cryoloop to be subsequently matured, fertilised and cultured in vitro to blastocyst-stage embryos. Selected cumulus–oocyte complexes obtained from mature ewes at the time of death were randomly divided into vitrified, toxicity and control groups. Following vitrification and warming, viable oocytes were matured in vitro for 24 h. Matured oocytes were either evaluated for nuclear maturation, spindle and chromosome configuration or fertilised and cultured in vitro for 7 days. No significant differences were observed in the frequencies of IVM (oocytes at the MII stage), oocytes with normal spindle and chromatin configuration and fertilised oocytes among the three groups. Cleavage at 24 and 48 h post insemination was significantly decreased (P < 0.01) in vitrified oocytes. No significant differences were observed in the proportion of blastocyst development between vitrified and control groups (29.4% v. 45.1%, respectively). No significant differences were observed in total cell numbers, the number of apoptotic nuclei or the proportion of diploid embryos among the three groups. In conclusion, we report for the first time that ovine oocytes vitrified at the GV stage using a cryoloop have the ability to be matured, fertilised and subsequently developed in vitro to produce good-quality blastocyst embryos at frequencies comparable to those obtained using fresh oocytes.

MHC-DRB1 exon2 was amplified by polymerase chain reaction (PCR) from 126 healthy and 67 Brucellosis-infected Chinese Merino sheep. PCR products were analyzed using the SSCP technique, and then cloned to allow sequencing of the different alleles. For each SNP, allelic and genotypic frequencies were compared between case and control samples, in addition the association with Brucellosis susceptibility was determined. Haplotypes and their frequencies were established and analyzed by SHEsis online software. There were forty-one single nucleotide polymorphisms (SNPs) in the 270 bp DNA sequence. The distribution of C>T alleles at locus 109 was significantly different between case and control samples. The linkage disequilibrium (LD) analysis showed that there were nine LD blocks in MHC-DRB1 exon2 and strong LD between SNPs existed in every Block. Haplotype analysis identified nine haplotypes with strong LD, but only Hap8 and Hap9 in case-control groups were significantly different (P<0.05); neither haplotype contained the C>T allele at locus 109. In conclusion, genetic variants of MHC-DRB1 gene exon2 demonstrated associations with Brucellosis susceptibility, indicating that further research is warranted.

Moxidectin administered in January or February at a single dose was tested for efficacy in horses on two farms for 12 and 11 months, respectively. Horses were infected with cyathostomes naturally in the previous grazing period. Forty horses of farm 1 and 20 horses of farm 2 were used in controlled tests to evaluate the efficacy of moxidectin 2% gel formulation at the dosage 0.4 mg moxidectin per kg of live weight, ivermectin commercial paste formulation at the dosage 0.2 mg ivermectin per kg of live weight, mebendazole and fenbendazole commercial paste formulation at the dosage both 7.5 mg mebendazole and fenbendazole per kg of live weight, all applied orally. Three control groups of 10 horses each (farm 1) were treated twice a year with ivermectin and benzimidazoles, respectively. Individual faecal egg counts, faecal cultures and larval differentiation were performed. Moxidectin had more prolonged and greater suppressive effects on the post-treatment reappearance and magnitude of strongyle egg counts than did ivermectin or benzimidazoles. In the moxidectin treated group (M1) strongyle eggs were seen for the first time in April and a slight increase in the mean count of eggs per gram of faeces (EPG) was observed during the rest of the season. Litter larval counts significantly reflected levels of exposure during the tested season. Twenty animals of farm 2 were allocated into two groups of ten horses each based on pre-treatment eggs per gram (EPG) counts (moxidectin treated group and control group). In the moxidectin treated group mean egg counts remained very low throughout the study. A plateau was reached by autumn, with egg counts ranging from 74 to 145 EPG. The faecal egg counts of moxidectin treated group (M2) were significantly higher in March, April, May and June.

BackgroundWhen new mothers do not understand how to interact with their newborn babies, they would increase anxiety, even decrease the quality of parent-infant interactions. Previous studies indicate that the postpartum parenting education for first-time-mothers can improve the quality of mother-infant interactions in first two months. This study aimed to evaluate the long-term effectiveness of parenting education for postpartum women during the first year.MethodsThe study recruited 81 healthy first-time-mother infant dyads from the medical center in Taipei city. The experimental and control groups received extra education by way of a 40-minute videotape and normal postpartum care, respectively. Data from around first week (T1), followed by the third (T2), sixth (T3), ninth (T4), and twelfth (T5) month postpartum are collected. Assessment scales such as the Edinburgh perinatal depression scale (EPDS), the Chinese version of the parenting sense of competence scale (C-PSOC), and the Nursing child assessment teaching scale (NCATS) used for videotaped mother-infant interactions measurement were used in the study.ResultsThe analytical results show that the quality of mother-infant interaction increased at T2, T3, T4, and T5 in the experimental group (Fig. 1). No different change in maternal sense of competency was found in the experimental and control groups from T1 to T4. However, a positive change in sense of parenting competency at T5 was found in the experimental group (Fig. 2). No difference in postpartum depression was found between the two groups (Fig. 3).ConclusionsThe first-time-mothers parent education has long-term effectiveness in the mother-infant dyad interaction quality.Disclosure of interestThe authors have not supplied their declaration of competing interest.

Numerous scientific studies have been carried out on the predisposition of the Pro47Ser polymorphism of the TR53 gene to breast cancer in women living in most countries of the world. However, no studies have been conducted on the susceptibility of this Pro47Ser gene polymorphism to breast cancer in Uzbek women. In this article, genotyping of the Pro47Ser polymorphism of the TR53 gene was performed for the first time in 207 Uzbek women. According to the results of our study, the functionally dangerous T allele of the Pro47Ser polymorphism of the TR53 gene was statistically significantly higher than that in healthy donors in patients with breast cancer; (ch2 = 8.2; p = 0.004; OR = 11.2; 95% CI: 1.422-88.38; RR = 10.7; 95% CI: 1.382-82.82). The safe C allele, on the other hand, was more common in the control group than in the main group. The natural S / C genotype was found in the control group with a higher frequency than in the control group. However, the limit of the statistical difference reached a significant level: (ch2 = 8.4; p = 0.004; OR = 0.08; 95% CI: 0.01066-0.676; ch2 = 8.2; p = 0.004; RR = 0.1 ; 95% CI: 0.8489- 0.9723). The S / T genotype of this polymorphism significantly differs in the frequency of occurrence in the main and control groups. Relative risk according to statistical analysis: RR = 10.7; (95% CI: 1.395–82.07) and odds ratio: OR = 11.8; (95% CI: 1.479-93.77). It is known that the minor T allele and the S / T genotype of this polymorphism increase the risk of developing breast cancer by 10.7 times. It should be noted that the unique homozygous genotype of Pro47Ser polymorphism was not identified in patients and control groups studied by T / T. Thus, the T allele and the C / T genotype of the Pro47Ser polymorphism of the TR53 gene are important factors that increase the risk of breast cancer (r <0.05). The C allele and S / S genotype of this polymorphism are reliable protective markers against the development of this pathology. Therefore, we believe that the Pro47Ser polymorphism of the TR53 gene can be used as important genetic markers in determining the likelihood of developing breast cancer.

Conjugated linoleic acid (CLA) is a collective term for the positional and geometric isomers of a conjugated diene of linoleic acid (C18:2, n-6). The aims of the present study were to evaluate whether levels of hepatic α-tocopherol, α-tocopherol transfer protein (α-TTP), and antioxidant enzymes in mice were affected by a CLA-supplemented diet. C57BL/6 J mice were divided into the CLA and control groups, which were fed, respectively, a 5 % fat diet with or without 1 g/100 g of CLA (1:1 mixture of cis-9, trans-11 and trans-10, cis-12) for four weeks. α-Tocopherol levels in plasma and liver were significantly higher in the CLA group than in the control group. Liver α-TTP levels were also significantly increased in the CLA group, the α-TTP/β-actin ratio being 2.5-fold higher than that in control mice (p<0.01). Thiobarbituric acid-reactive substances were significantly decreased in the CLA group (p<0.01). There were no significant differences between the two groups in levels of three antioxidant enzymes (superoxide dismutase, glutathione peroxidase, and catalase). The accumulation of liver α-tocopherol seen with the CLA diet can be attributed to the antioxidant potential of CLA and the ability of α-TTP induction. The lack of changes in antioxidant enzyme protein levels and the reduced lipid peroxidation in the liver of CLA mice are due to α-tocopherol accumulation.

The paper describes the method for increasing the level of self-organisation in students which has been developed by the author. It also contains the method testing results and presents the prospects and risks teachers could face while applying the method in a higher education institution. The purpose of this study is to find out the prospects and risks of applying the method for increasing the level of self-organisation in students and to determine the ways of reducing the risks. Methodology. The author points out the learning approaches which were the basis of developing the method and describes diagnostic methods for determining students’ self-organisation levels. The work focused on increasing each student’s initial level consists of a theoretical and a practical part and includes project activities on creating a study guide. The results of the study. The method developed proved to be effective. It was established by diagnosing the final level of self-organisation in students in the experimental and control groups. The paper considers the advantages of the method among which there is universal character, flexibility, improvements to teacher’s and students’ professional competence, etc. At the same time it is necessary to be aware of the risks due to the increased amount of teacher’s work and the fact that students’ work within the project tends to be monotonous. In conclusion, the prospects of the method for increasing the level of self-organisation in students are related to its advantages and the final results of the work. The risks of its use can be reduced with the help of the measures proposed in the paper.

Aim. To determine the response of the populations of Erysipelothrix rhusiopathiae and Leptospira interrogans pathogenic microorganisms to the impact of broadleaf cattail (Thypha latifolia) root diffusates. Methods. Aqueous solutions of T. latifolia root diffusates were sterilized by vacuum fi ltration through the fi lters with 0.2-micron pore diameter. The experimental samples contained cattail secretions, sterile water, and cultures of E. rhusiopathiae or L. interrogans. The same amount of sterile water, as in the experimental samples, was used for the purpose of control, and the same quantity of microbial cultures was added in it. After exposure, the density of cells in the experimental and control samples was determined. Results. Root diffusates of T. latifolia caused an increase in cell density in the populations of E. rhusiopathiae throughout the whole range of the studied dilutions (1:10–1:10000). In the populations of the 6 studied serological variants of L. interrogans spirochetes (pomona, grippotyphosa, copenhageni, kabura, tarassovi, canicola), the action of broadleaf cattail root diffusates caused the decrease in cell density. A stimulatory effect was marked in the experimental samples of the pollonica serological variant of leptospira. Conclusions. The populations of E. rhusiopathiae and L. interrogans pathogenic microorganisms respond to the allelopathic effect of Thypha latifolia by changing the cell density. The obtained results provide the background to assume that broadleaf cattail thickets create favorable conditions for the existence of E. rhusiopathiae pathogen bacteria. The reduced cell density of L. interrogans in the experimental samples compared to the control samples observed under the infl uence of T. latifolia root diffusates suggests that reservoirs with broadleaf cattail thickets are marked by the unfavorable conditions for the existence of pathogenic leptospira (except L. pollonica).
