Zusammenfassung Ziel der Untersuchung: Überprüfung der Wirksamkeit von DL-Cloprostenol vs. D-Cloprostenol in der Östrusinduktion bei Milchrindern. Probanden und Methoden: Das Probandenkollektiv umfasste 134 Rinder im Durchschnittsalter von 4,0 ± 0,5 Jahren (99 in Laktation, 35 Färsen). Vor alternierender Injektion zweier Cloprostenolpräparate (Gruppe A: DL-Cloprostenol, 500 μg, n = 70; Gruppe B: D-Cloprostenol, 150 μg, n = 64) wurde die Progesteronkonzentration im Serum bestimmt. Gynäkologische Kontrollen erfolgten 0–3 d post injectionem (p. inj.), wobei die als inseminationsfähig beurteilten Probanden (n = 123) am dritten Tag besamt wurden. Ergebnisse: In beiden Gruppen kam es bis zum dritten Tag p. inj. zu einer deutlichen Konsistenzänderung oder Regression der Corpora lutea (p ≤ 0,001). Die Lysis eines C. l. periodicum verlief bei Kühen markanter als bei Färsen (p ≤ 0,017). Insgesamt war D-Cloprostenol dem DL-Cloprostenol hier leicht überlegen. Am dritten Tag p. inj. wiesen 67,1% (A) bzw. 71,9% (B) der Probanden gut ausgeprägte Östrusanzeichen auf. Als inseminationsfähig wurden 94,3% (A) und 89,1% (B) der Tiere eingestuft. Ein geringer Präparateunterschied bestand hinsichtlich des Graviditätsresultates. Bei einem C. l. persistens verlief die Regression weniger progressiv als bei einem C. l. periodicum (p ≤ 0,024). Signifikante Wechselwirkungen zwischen den Einflussfaktoren Präparat und Indikation einerseits sowie für die übrigen gynäkologischen Kriterien andererseits ergaben sich nicht. Bei den Probanden mit prostaglandininduziertem Zyklus nach einem C. l. persistens lag die Graviditätsrate nach der 1. KB deutlich niedriger (31,9%) als bei Tieren nach Lysis eines C. l. periodicum (52,4%, p = 0,08). Schlussfolgerungen und klinische Relevanz: Der Einsatz von D-Cloprostenol erbringt im Wesentlichen die Resultate wie der von DL-Cloprostenol. Eine Überlegenheit konnte jedoch für die Progressivität der lytischen Wirkung des D-Cloprostenols festgestellt werden. Kühe mit C. l. persistens reagierten präparateunabhängig weniger intensiv als solche mit C. l. periodicum.

Anesthesia for laboratory animals is a matter of biomedical concern and one of the most present dilemmas in the current bioethical debate. The use of anesthetic agents in experimental surgery aims at analgesia and restraining the animal, in order to achieve a reasonable degree of muscle relaxation and to produce sufficient analgesia. This practice requires the use of protocols for the administration of safe and efficient doses. Eight New Zealand rabbits were submitted to laparotomies demonstrating the surgical technique discipline of the local medical course. For pre-anesthetic medication, acepromazine 1 mg.kg-1 associated with ketamine 15 mg.kg-1 was used subcutaneously. Anesthesia was maintained with isoflurane and oxygen under a laryngeal mask in a Mapleson D anesthesia system and under spontaneous breathing. Hydration was performed with 10 ml.kg-1 saline every hour. A thermal mattress was used. Precordial stethoscope, pulse oximetry and clinical parameters were used for monitoring. For euthanasia, ketamine 10 mg.kg-1 associated with potassium chloride 19.1% 1 ml.kg-1 was used intravenously. The average weight of the rabbits was 2721.25 ± 275.01 grams and the duration of the anesthetic procedure was 120 ± 87 minutes. Discussion. In long-term anesthesia, such as laparotomies, the use of pre-anesthetic medication and then anesthetic induction by the combination of agents is recommended. However, anesthetic management requires monitoring to prevent insufficient or excessive doses from occurring.

Broccoli (Brassica oleracea var. italica), a popular vegetable crop, has one of the most exacting climatic and cultural requirements, which limit its commercial production to a few favored locations. A field experiment was conducted at Mizoram University, Tanhril, Mizoram during winter of 2013/2014 to find out the effect of time of sowing on plant performance and yield of broccoli. Six sowing time was done viz. 17 October (T1), 24 October (T2), 31 October (T3), 7 November (T4), 14 November (T5) and 21 November (T6) with plant spacing of 45cm x 45cm. Yield and yield contributing characters were significantly influenced by the planting time. Highest average weight of marketable curd per plant (199.20 g) was obtained from T2, whereas lowest average weight obtained from T6 (75 g). The influence of planting time also showed significant difference on the calculated yield (tonnes per hectare) of broccoli, in which T2 showed highest marketable yield of 9.83 t/ha.

ABSTRACT The »early-androgen« syndrome in the rat – i. e. anovulatory ovaries in adult females after a single injection of testosterone propionate (TP) within a week of birth – may not become apparent until some time after the attainment of sexual maturity. Large doses (50 or 100 μg) of TP were effective earlier than lower doses (5 or 10 μg). Rats which received 5 μg TP were ovulating at 10 weeks of age, mated but were infertile at 13 weeks of age, and were anovulatory at 21 weeks. In rats between 10 and 13 weeks old there was a marked fall in the number of corpora lutea in the ovaries of animals which had been given 5 μg TP. Hemi-spaying was followed by compensatory growth of the remaining ovary which consisted of corpora lutea in ovulating, and of follicles in anovulatory rats; little or no compensatory weight increase occurred in animals which seemed to be in the transition stage from the ovulatory to the anovulatory condition.

ABSTRACT Absence of compensatory ovarian hypertrophy in »constant oestrus rats« from lesions in the anterior hypothalamic area suggests that nervous elements localized in this region play an essential role in the stimulation of gonadotrophin output by diminution of the blood oestrogen level. The constant vaginal oestrus after unilateral ovariectomy in the majority of animals was, however, repeatedly interrupted by vaginal smears of a dioestrous type. The appearance of a dioestrous vaginal smear in the »hypothalamic constant oestrus rats« is often associated with some luteinisation. It is assumed that diminution of the blood oestrogen level by reduction of ovarian tissue in these animals may bring about a release of LH sufficient to cause formation of corpora lutea.

ABSTRACT Male newborn rats were injected with 2 mg of an antiandrogen (1,2α-methylene-6-chloro-pregna-4,6-dien-17α-ol-3,20-dione-17α-acetate = cyproterone acetate) daily from their 1st to their 14th day of life. The following effects of this treatment were observed in these animals after onset of sexual maturity: 84% of the animals are unable to reproduce. Penis: the frenulum is broadened to a lamina of triangular shape, which almost completely prevents the preputium from being pushed back. These males show a rather insufficient male sexual behaviour towards females in oestrus. After castration and ovar implantation, some of the treated animals show true corpora lutea and at attempts of cohabitation partially female sexual behaviour towards normal male animals. From these results it can be concluded, that differentiation of the penis is not completed at the time of birth. The infertility of the animals may be caused by the penile changes (difficulties with intromission) as well as by the aimless sexual behaviour. This aimless sexual behaviour, the ability to produce true corpora lutea and finally their partially female sexual attitude under the influence of the hormones from the implanted ovaries led to the conclusion, that the above described neonatal treatment apparently inhibited testosterone-depending post partum developments of sexual differentiation in hypothalamic centers.

ABSTRACT The ultrastructure of granulosal lutein cells of 13 corpora lutea in early human pregnancy was studied. The predominant cytoplasmic element was the smooth endoplasmic reticulum. No convincing signs of degeneration of the lutein cells could be demonstrated within the first 14 weeks of pregnancy, as the mitochondria as well as the rough and smooth endoplasmic reticulum were well preserved. However, lysosomes may be slightly more numerous in older specimens and the subendothelial space increases with the age of gestation. A particular type of multilaminated structure one to five micron in diameter was observed, particularly in the earliest specimens. The possible intracellular location of steroid synthesis is discussed.

ABSTRACT The role of luteinizing hormone (LH) for the maintenance of pregnancy, parturition and lactation was investigated by immunological and histochemical methods in the rat. Neutralisation of endogenous rat-LH with Rabbit-Anti-Bovine-LH-Serum (selective hypophysectomy) from days 7-12 of pregnancy resulted in reabsorption of the foetuses and the reappearance of strong 20α-hydroxysteroid-dehydrogenase (20α-OHSD) activity in the corpora lutea (CL) of pregnancy, which normally show no such activity at that time. This effect could be prevented in part by concurrent pregnenolone administration and fully by progesterone, but was not influenced by oestrogen or prolactin. It is concluded that in early pregnancy LH is the main luteotrophic hormone in the rat even though prolactin might act synergistically with it. Antiserum treatment after the 12th day of gestation had no influence on the state or duration of pregnancy or on parturition. LH-injections during the first half of pregnancy had no luteolytic effects i. e. they did not activate 20α-OHSD activity. After day 16 they advanced the reappearance of the enzyme, but delayed parturition or resulted in stillbirths. Neither LH nor antiserum seemed to alter lactation. Since progesterone prevented both the termination of pregnancy and the recurrence of 20α-OHSD activity, it should have some regulatory properties on the enzyme. It is discussed whether the gonadotrophin-dependent progesterone level could regulate the 20α-OHSD activity rather than result from it.

ABSTRACT Activities of various hydrolases and dehydrogenases were studied during the formation, development and involution of cyclic corpora lutea and in the corpora lutea of early pregnancy. At 24 hours postovulation the luteal cells, whether of granulosal or thecal origin, contained demonstrable levels of Δ5-3β-hydroxysteroid dehydrogenase and the NADP and NADPH2 diaphorases. During the period of proliferation and cellular growth, enzymic activities in the luteal cells were moderate at first, and then increased. In the mature corpus luteum, activities of the dehydrogenases occurred in all luteal cells but were most intense in the large polymorphic luteal cells. Activities of hydrolytic enzymes, low in the immediate postovulatory period, increased with the development of the vascular system. Enzymic characteristics of corpora lutea of gestation were similar to those of cyclic corpora, except for phosphorylase activity which was observed in luteal cells in gestational corpora, but confined to the vascular walls in cyclic corpora. No increase in activities of 17β- and 20β-hydroxysteroid dehydrogenases (above those seen in pre-ovulatory follicles) were observed after incubation of sections of either mature cyclic or gestational corpora. Involution of cyclic corpora lutea began with degenerative changes in the blood vessels: pyknosis of the endothelial cell nuclei and a sudden decline in activities of hydrolytic enzymes in the vascular walls. Subsequently, the luteal cells showed a sharp decrease in activities of the dehydrogenases as well as other signs of regressive change. The cytochemical findings are discussed in relation to biochemical observations on steroid synthesis by the bovine corpus luteum.

Abstract. Immunoreactive oxytocin is detectable in the corpora lutea of women and cynomolgus monkeys by radioimmunoassay. To localize the presence of oxytocin and neurophysin I in ovarian tissues of subhuman primates, three corpora lutea and ovarian stromal tissues and two Fallopian tubes obtained during the menstrual cycle of the baboon and decidua from two pregnant baboons were examined using highly specific antisera against either oxytocin or neurophysin I and preoxidase-antiperoxidase light microscopy immunohistochemistry. Oxytocin-like as well as neurophysin I-like immunoreactivities were found in some cells of all the corpora lutea only, but could not be demonstrated in ovarian stromal tissues, Fallopian tubes and decidua. Specificity of the immunocytochemical reaction was further confirmed by immunoabsorption of the antiserum with excess oxytocin or neurophysin, after which the immunoreactivities for both oxytocin and neurophysin in the luteal tissue were negative. Similar controls using normal rabbit serum gave no positive staining for either oxytocin or neurophysin. Counterstaining of the positive immunoreactivities for oxytocin and neurophysin I with Mayer's haematoxylin and eosin demonstrated clearly that the oxytocin and neurophysin I appeared as granular material mainly within the cytoplasm of the luteal cells. The localization of immunoreactive oxytocin and neurophysin I in the corpus luteum of the baboon demonstrates directly the presence of these two neurohypophysial peptides within primate luteal cells and suggests their local production.
