Rice Straw Hydrolysis and Cultivation of Xanthophyllomyces dendrorhous for Astaxanthin Production and Antioxidant Properties

Astaxanthin is a natural pigment with strong antioxidant activity and is widely supplied as dietary supplement. The red yeast Xanthophyllomyces dendrorhous is one of the potential sources for astaxanthin production. Rice straw was considered for utilization on cultivation and astaxanthin production. Rice straw was treated by autoclave-assisted alkaline pretreatment. and hydrolyzed by cellulase hydrolysis. X. dendrorhous TISTR5730 was cultured and accumulated as the astaxanthin on rice straw hydrolysate. The highest reducing sugar concentration of rice straw hydrolysate was 89.82±0.39g/L (0.71±0.01g/g) when using rice straw (3.3%) and cellulase loading (3 mL). High cell density X. dendrohous TISTR5730 cultivation on rice straw hydrolysate with 20 and 40 g/L of initial reducing sugar was investigated. 40 g/L was appropriate for biomass production while 20 g/L was suitable for astaxanthin accumulation. The highest astaxanthin content was 417.28±50.89 μg/g cell basis at 192 h. Astaxanthin productivity and yield coefficient were 0.01±0.00 mg/L/h and 0.11±0.01 mg/g sugar consumed. The antioxidant activities of astaxanthin were determined by DPPH and ABTS scavenging and FRAP reducing power. The produced astaxanthin represented the high antioxidant activities with IC50 of 9.30 and 1.67 μg/mL of DPPH and ABTS scavenging respectively and FRAP reducing power of 5.31±0.07 μg TEAC/mL. This research indicated that rice straw hydrolysate could be an alternative medium for astaxanthin production. Antioxidant activity of astaxanthin was proved and was feasible for further applications.

The antioxidant properties and total phenolic contents of four fractions of ethanolic extract from Ramulus mori were examined. Various experimental models including superoxide radical, hydroxyl radical, 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH) scavenging activity, metal chelating activity, and reducing power were used for characterization of their antioxidant activity. The four fractions showed various degrees of efficacy in each assay in a dose-dependent manner. The third fraction with the highest amount of total phenolics was the most potent antioxidant in all assays used. In addition, the most powerful compound (oxyresveratrol) was isolated and identified followed by on-line HPLC method and characterized by different spectral analysis. Oxyresveratrol exhibited impressive antioxidant activities in scavenging the superoxide radical, hydroxide radical, and DPPH. On the basis of the results obtained, Ramulus mori may serve as a potential source of natural antioxidant due to its significant antioxidant activity and oxyresveratrol may be the most powerful antioxidant in ethanolic extracts of Ramulus mori.

Some antioxidant activities of extracts of irradiated black pepper (Piper nigrum) were evaluated. The ground black pepper was exposed to gamma-irradiation at doses from 5 to 30 kGy. The effect of irradiation on antioxidant properties of black pepper extracts was investigated by these methods: radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, determination of reducing power, thiobarbituric acid reactive substances assay and electron paramagnetic resonance spectroscopy (EPR). Irradiation resulted in a decrease of DPPH antiradical activity of black pepper extracts according to dose of irradiation. Reducing power was not changed by action of irradiation. Very important and sensitive differences in antioxidant activity of irradiated black pepper were investigated by EPR spectrometry and thiobarbituric acid method.

The objective of this study was to evaluate the phenolic content, in vitro antioxidant activity and the gastroprotective effect of the ethanolic extract of pear fruits (Pyrus communis L.). The amount of polyphenols and flavonoids contents of ethanolic extract were found to be 77,50 ± 2,50 mg GAE/g of dry extract and 0,31 ± 0,003 mg QE/ g of dry extract, respectively. Whereas the amount of tannins was 61,43 ± 0,009 mg TAE/ g of dry extract. Hydroxyl radical scavenging and reducing power tests were used to evaluate the antioxidant activities of this extract. The scavenging effect of ethanolic extract against hydroxyl radicals showed IC50 value of 2.71 ± 0.02 mg/ ml. Reducing power test indicated the ability of ethanolic extract of pear fruits to reduce Fe+2 to Fe+3  with EC50value of 1.043±0.01 mg/ml. The treatment of rats with the ethanolic extract at  dose of 200 mg/kg reduced the ulcerogenic effect of ethanol with value of protection rate of 94.44%, but the protection rate decreased in 600 mg/kg treated group  to 90.17%.  In conclusion, natural antioxidants derived from pear fruits can prevent gastric ulcer through their antioxidant activity  Keywords: Pyrus communis L., Polyphenols, Antioxidant activity, Ulcer, Rat.

Abstract Seaweeds are now recognized as a treasure of bioactive compounds. However, the seaweed of Bangladesh is still unexplored. So, this study was designed to explore the secondary metabolites and antioxidant activities of solvent extracts of Padina tetrastromatica and Gracilaria tenuistipitata. Phytochemical screening and Fourier transform infrared (FTIR) confirm the diverse type of bioactive compounds. Antioxidant activity of extracts were evaluated by 1, 1-diphenyl-2- picrylhydrazyl (DPPH), 2, 2-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power (RP), phosphomolybdenum and hydrogen peroxide assays. Here, methanolic extract of P. tetrastromatica showed the highest amount of total phenolic content (85.61 mg of GA/g), total flavonoid content (41.77 mg of quercetin/g), which showed as potent antioxidant properties confirmed by DPPH (77.07%), ABTS (77.65%), RP (53.24 mg AAE/g), phosphomolybdenum (31.58 mg AAE/g) and hydrogen peroxide (67.89%) assays followed by ethanol and water. This study concluded that bioactive compound and antioxidant activities are dose and solvent-dependent and seaweed type.

The antioxidant activity of cookies prepared by the addition of sugarbeet dietary fibers was investigated in order to estimate their influence on functional characteristics and shelf-life of cookies. Treated fiber (TF) was obtained from sugarbeet by extraction with sulfurous acid (75 °C at pH = 5.7during 60 min) and treatment with hydrogen peroxide (20 g/LH2O2 at pH = 11 during 24 h). The fiber obtained was dried (80 °C), ground and sieved. TF was investigated in comparison with commercially available Fibrex®. The cookies were prepared by the addition of 0, 7, 9 and 11% of sugarbeet dietary fiber as a substitute for wheat flour in the formulation of cookies. The antioxidant properties of cookies were tested every 7 days using a DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity test during 6 weeks of storage at room temperature (23 ± 1 ºC). The obtained results indicated that substitution of wheat flour with Fibrex® in the formulation of cookies upgraded the antioxidant activity, i.e. the functional characteristics of Fibrex®-enriched cookies and could prolong their shelf-life. In contrast, TF did not increase the antioxidant activity of TF-enriched cookies. The better antioxidant activities of Fibrex®-enriched cookies could be attributed to the presence of ferulic acid.

AbstractSample particle size is an important parameter in the solid–liquid extraction system of natural products for obtaining their bioactive compounds. This study evaluates the effect of sample particle size on the phytochemical composition and antioxidant activity of brown macroalgae Sargassum cristaefolium. The crude ethanol extract was extracted from dried powders of S.cristeafolium with various particle sizes (> 4000 µm, > 250 µm, > 125 µm, > 45 µm, and < 45 µm). The ethanolic extracts of S.cristaefolium were analysed for Total Phenolic Content (TPC), Total Flavonoid Content (TFC), phenolic compound concentration and antioxidant activities. The extract yield and phytochemical composition were more abundant in smaller particle sizes. Furthermore, the TPC (14.19 ± 2.08 mg GAE/g extract to 43.27 ± 2.56 mg GAE/g extract) and TFC (9.6 ± 1.8 mg QE/g extract to 70.27 ± 3.59 mg QE/g extract) values also significantly increased as particle sizes decreased. In addition, phenolic compounds epicatechin (EC), epicatechin gallate (ECG), epigallocatechin (EGC), and Epigallocatechin gallate (EGCG) concentration were frequently increased in samples of smaller particle sizes based on two-way ANOVA and Tukey’s multiple comparison analysis. These results correlate with the significantly stronger antioxidant activity in samples with smaller particle sizes. The smallest particle size (< 45 µm) demonstrated the strongest antioxidant activity based on DPPH, ABTS, hydroxyl assay and FRAP. In addition, ramp function graph evaluates the desired particle size for maximum phytochemical composition and antioxidant activity is 44 µm. In conclusion, current results show the importance of particle size reduction of macroalgae samples to increase the effectivity of its biological activity.

Herein, we report the neuroprotective and antioxidant activity of 1,1′-biphenyl nitrones (BPNs) 1–5 as α-phenyl-N-tert-butylnitrone analogues prepared from commercially available [1,1′-biphenyl]-4-carbaldehyde and [1,1′-biphenyl]-4,4′-dicarbaldehyde. The neuroprotection of BPNs1-5 has been measured against oligomycin A/rotenone and in an oxygen–glucose deprivation in vitro ischemia model in human neuroblastoma SH-SY5Y cells. Our results indicate that BPNs 1–5 have better neuroprotective and antioxidant properties than α-phenyl-N-tert-butylnitrone (PBN), and they are quite similar to N-acetyl-L-cysteine (NAC), which is a well-known antioxidant agent. Among the nitrones studied, homo-bis-nitrone BPHBN5, bearing two N-tert-Bu radicals at the nitrone motif, has the best neuroprotective capacity (EC50 = 13.16 ± 1.65 and 25.5 ± 3.93 μM, against the reduction in metabolic activity induced by respiratory chain blockers and oxygen–glucose deprivation in an in vitro ischemia model, respectively) as well as anti-necrotic, anti-apoptotic, and antioxidant activities (EC50 = 11.2 ± 3.94 μM), which were measured by its capacity to reduce superoxide production in human neuroblastoma SH-SY5Y cell cultures, followed by mononitrone BPMN3, with one N-Bn radical, and BPMN2, with only one N-tert-Bu substituent. The antioxidant activity of BPNs1-5 has also been analyzed for their capacity to scavenge hydroxyl free radicals (82% at 100 μM), lipoxygenase inhibition, and the inhibition of lipid peroxidation (68% at 100 μM). Results showed that although the number of nitrone groups improves the neuroprotection profile of these BPNs, the final effect is also dependent on the substitutent that is being incorporated. Thus, BPNs bearing N-tert-Bu and N-Bn groups show better neuroprotective and antioxidant properties than those substituted with Me. All these results led us to propose homo-bis-nitrone BPHBN5 as the most balanced and interesting nitrone based on its neuroprotective capacity in different neuronal models of oxidative stress and in vitro ischemia as well as its antioxidant activity.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

Amomum subulatum, commonly known as large or black cardamom, is a commonly used spice in Indian kitchens and is traditionally used to treat various ailments. To add more knowledge about the medicinal values of the fruit, this study was conducted to evaluate the in vitro antioxidant activities of aqueous, methanol, ethanol, hydro alcohol, ethyl acetate, acetone and chloroform extracts of the fruit. Preliminary assessment was done to detect the presence of phytoconstituents using identification tests. The antioxidant activity was measured by employing methods such as diphenylpicrylhydrazyl (DPPH) radical scavenging assay, total antioxidant activity equivalent to ascorbic acid, reducing power assay and superoxide anion scavenging assay. The antioxidant activities were compared with their respective phenol and flavonoid contents. Preliminary assessment revealed that large cardamom fruit is a good source of all the bioactive constituents as well as phenol and flavonoid essential for medicinal values. The extract obtained by polar solvents showed the highest antioxidant efficacy in relation to its phenol content. Also, all the solvent-soluble fractions showed a concentration-dependent antioxidant effect. Results from our study prove that large cardamom can alleviate oxidative stress, suggesting the potential of large cardamom as a functional food

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.
