Monosodium glutamate (MSG) is widely used as a food additive. Excessive consumption of monosodium glutamate has also been shown to affect the liver and kidneys, causing damage to these tissues because of oxidative stress leading to increased production of reactive oxygen species (ROS). The purpose of the study described in this paper was to find out how the liver and kidney toxicity caused by monosodium glutamate can be mitigated using pectin. To this end, 30 albino mice females were divided into four groups. The animals were distributed in special cages. 12-15 weeks with an average body weight of 60 grams. The animals were divided into four groups: the experimental control group (1) comprising 5 female mice were given normal drinking water and the treated group (2) comprising 10 female mice were given monosodium glutamate at a dose of 3 g/kg body weight in drinking water. For three weeks, the treatment group (3) comprising 10 female mice was given pectin at a dose of 300 mg/70 kg body weight in drinking water immediately after the monosodium glutamate dose for three weeks and the pectin group (4) comprising 5 female mice were given Pectin at a dose of 300 mg/70 kg body weight in drinking water for three weeks. The mice were then anesthetized, dissected, and liver and kidney samples were taken from female mice and kept in a 10% neutral formalin solution to make tissue segments. The results showed many histological changes in the liver, such as congestion of the central vein, widening of the sinuses, and the appearance of signs of the death of most hepatocytes, infiltration of the central vein and an invasion of inflammatory cells around the central vein with the emergence of several gaps within the cells. Many of them cavity with the death of most of the tubule cells, the closure of some of them and the expansion and infiltration in others and bleeding inside the tissue. Pectin therapy has led to the disappearance of most of these changes and the emergence of a clear improvement in hepatic and renal tissue.

There are two ways, first, during the sterilization of Bro3 bromate is an anion of the element bromine has the formula of water ozone and the second addition of bromate during manufacturing, we tried through this research to shed light on the substance bromate and its risk to human health by studying the changes in some of the tissues of mice as a result of exposure to bromate by Drinking water and the use of this research (30) of female albino mice albino mice The animals were distributed in special cages with feeding bottles to drink water in a room that is ventilated subject to natural factors and provided the animal with the appropriate food. The average age of the body was 60 grams. Bromate was obtained from SATEC and the animals were divided into control groups. Five female mice were given normal drinking water. The treated group included 15 female mice were given sodium bromate at a dose. 200 mg/ kg bw in drinking water for 2 weeks after which the mice were anesthetized and dissected and liver and kidney samples were taken from female mice and kept in 10% neutral formalin solution to make tissue sections. The results showed several histological changes in the liver, such as congestion of the central vein, widening of the sinuses, the appearance of signs of death for most hepatocytes, such as cloud swelling, chromatin condensation of some nuclei, infiltration of the central vein and invasion of inflammatory cells around the central vein. With the death and decomposition of most of the tubules with tightness in the lumen of these tubules and closed some of them and expansion and infiltration in others and bleeding inside the tissue with the emergence of death and decomposition of most of the tubules cells. We conclude from this study that exposure to bromate led to a lot of damage to the liver and kidney tissue, and the kidney was more effective than the liver because the bromate pass on the kidney through the blood, which leads to the damage of kidney cells "nephrons" This proves the toxicity of the bromate of the kidney and its ability to cause Kidney failure.

Abstract BackgroundMite infestations in laying hens can cause losses to producers due to stress, reduced egg production and even death of birds. A new species of mite, Allopsoroptoides galli (A. galli), Analgoidea: Psoroptoididae, was recently identified in commercial laying farms in Brazil, causing damage due to its highly aggressive infestation that results in a sharp drop in egg production and culling. The present study evaluated the acaricidal action of a formulation containing fluralaner (Exzolt®) against A. galli.Methods The study was carried out from November 2019 to January 2020. Thirty-four laying hens naturally infested with A. galli were equally divided into a fluralaner-treated group and an untreated control group. The fluralaner-treated group received Exzolt® in drinking water at a dose of 0.05 mL/kg body weight (equivalent to 0.5 mg fluralaner/kg body weight), twice, seven days apart. Both groups were followed for 70 days evaluating the level of infestation by counting mites in skin scrapings and assessment of skin lesions.Results The average mite count of the treated group decreased significantly, dropping from 61.6 to 3.8 mites (D+7 to D+70). The efficacy progressively increased on subsequent days, reaching 98.8% on day +56 post-treatment and 96.9% on day +70. Recovery of skin lesions was observed after administration of Exzolt®, showing a marked remission in the degree of lesions (2.5 on day -14 to 0.2 on day +70). The mean number of mites in the untreated control group ranged from 79.3 to 124.1 and the lesion score from 2.6 to 2.9, thus remaining stable throughout the study.Conclusion The results obtained in the present study demonstrated that Exzolt® administered at a dose of 0.05 mL of product/kg body weight (equivalent to 0.5 mg of fluralaner/kg body weight), twice at a seven-day interval, in drinking water was effective in the treatment of the mite Allopsoroptoides galli in naturally infested laying hens.

This study was carried out to investigate the immunomodulating activity of β-glucanwhich extracted from the cell wall of Saccharomyces cerevisiae.β-glucan of 225µg/ml wassupplemented in drinking water of broiler chicken vaccinated with Newcastle disease virus(NDV) vaccine. The parameters of the immunomodulating activity employed were the bodyweight, Phagocytic activity and determination of antibody titer to NDV vaccine. Chicks oneday old (120), were divided into four equal groups, G1 treated with β-glucan for six weeksand vaccinated with NDV vaccine; G2 treated with β-glucan for three weeks and vaccinatedwith NDV vaccine; G3 not given β-glucan but vaccinated with NDV vaccine (control group);while G4 was not treated with β-glucan and not vaccinated with NDV vaccine (secondcontrol). The results of the body weight indicated that; there were significant differences (P<0.05) between treated groups (G1 and G2) compared to the control groups (G3 and G4) at21,28 and 35 day of age. The results of phagocytic activity showed that treated group hadsignificantly (P< 0.05) higher clearance of carbon particles from blood circulation than did thecontrol groups; and the antibody titer to NDV showed significant differences (P<0.05)between treated and non treated groups at 14d and 28d. The data presented in this studycontribute for the first time in Iraq; that β –glucan given via drinking water to chicks from dayone for 35 days improves the immune responses and body weight.

Carbamazepine (CBZ) is one of many anticonvulsants used to treat trigeminal neuralgia and epilepsy. Antiepileptic drugs (AED`s) are the second most important class of medications that lead to hepatotoxicity and induced liver injury, this study was conducted to evaluate the effects of CBZ on the liver. A total of 40 female mice were taken and divided into four groups (A/treated for 14 days, B/ control, C/ treated for 30 days, D/ control), the drug was given as an oral suspension formula 100mg/5ml at dose 20 mg/kg/mouse via gastric gavage daily for 14 and 30 days. Statistical analysis revealed that there were no significant differences in the white female mice body weight (P>0.05) in the treated group for 14 days as well as the treated group for 30 days, but there were significant differences between the treated groups studied. Statistical analyses result of liver weight, hepatocytes diameter, central vein and portal vein diameters showed significant differences in the treated group for 14 days (P<0.05) as well as the treated group for 30 days, and there were significant differences between all treated groups studied. The study concluded that carbamazepine (CBZ) can had no effect on body weight but it can induce several hepatic changes if it used on a short or long terms, therefore, it was advised to take cautions when describing this drug.

The present study aimed to investigate the impact of perinatal potassium bromate (KBrO3) exposure on the development of sensorimotor reflexes and redox status, and on the histological architecture of the brain, liver, and kidney of newborn mice. Pregnant mice received 1-ml bottled drinking water daily by oral intubation and served as the control group. Another group of pregnant mice were supplemented orally with 200 mg/kg body weight KBrO3 dissolved in drinking water from gestation day 5 to postnatal day 21. KBrO3 induced a decrease in the postnatal body weight in the newborn mice. KBrO3-exposed newborn mice showed poor performance and delayed development of the sensorimotor reflexes. Histological changes, increased lipid peroxidation, and altered antioxidants were reported in the cerebrum, cerebellum, medulla oblongata, liver, and kidney of the KBrO3-exposed newborn mice. In conclusion, these findings demonstrated that perinatal exposure to bromate induced oxidative stress, histological and behavioral alterations, and was a potential teratogen in newborn mice.

We investigated the change of tryptophan-niacin metabolism in rats with puromycin aminonucleoside PAN-induced nephrosis, the mechanisms responsible for their change of urinary excretion of nicotinamide and its metabolites, and the role of the kidney in tryptophan-niacin conversion. PAN-treated rats were intraperitoneally injected once with a 1.0% (w/v) solution of PAN at a dose of 100 mg/kg body weight. The collection of 24-hour urine was conducted 8 days after PAN injection. Daily urinary excretion of nicotinamide and its metabolites, liver and blood NAD, and key enzyme activities of tryptophan-niacin metabolism were determined. In PAN-treated rats, the sum of urinary excretion of nicotinamide and its metabolites was significantly lower compared with controls. The kidneyα-amino-β-carboxymuconate-ε-semialdehyde decarboxylase (ACMSD) activity in the PAN-treated group was significantly decreased by 50%, compared with the control group. Although kidney ACMSD activity was reduced, the conversion of tryptophan to niacin tended to be lower in the PAN-treated rats. A decrease in urinary excretion of niacin and the conversion of tryptophan to niacin in nephrotic rats may contribute to a low level of blood tryptophan. The role of kidney ACMSD activity may be minimal concerning tryptophan-niacin conversion under this experimental condition.

Excess body weight is a major risk factor for type 2 diabetes (T2D) and associated metabolic complications, and weight loss has been shown to improve glycemic control and decrease morbidity and mortality in T2D patients. Weight-loss strategies using dietary interventions produce a significant decrease in diabetes-related metabolic disturbance. We have previously reported that the supplementation of low molecular chitosan oligosaccharide (GO2KA1) significantly inhibited blood glucose levels in both animals and humans. However, the effect of GO2KA1 on obesity still remains unclear. The aim of the study was to evaluate the anti-obesity effect of GO2KA1 on lipid accumulation and adipogenic gene expression using 3T3-L1 adipocytes in vitro and plasma lipid profiles using a Sprague-Dawley (SD) rat model. Murine 3T3-L1 preadipocytes were stimulated to differentiate under the adipogenic stimulation in the presence and absence of varying concentrations of GO2KA1. Adipocyte differentiation was confirmed by Oil Red O staining of lipids and the expression of adipogenic gene expression. Compared to control group, the cells treated with GO2KA1 significantly decreased in intracellular lipid accumulation with concomitant decreases in the expression of key transcription factors, peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT/enhancer-binding protein alpha (CEBP/α). Consistently, the mRNA expression of downstream adipogenic target genes such as fatty acid binding protein 4 (FABP4), fatty acid synthase (FAS), were significantly lower in the GO2KA1-treated group than in the control group. In vivo, male SD rats were fed a high fat diet (HFD) for 6 weeks to induced obesity, followed by oral administration of GO2KA1 at 0.1 g/kg/body weight or vehicle control in HFD. We assessed body weight, food intake, plasma lipids, levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) for liver function, and serum level of adiponectin, a marker for obesity-mediated metabolic syndrome. Compared to control group GO2KA1 significantly suppressed body weight gain (185.8 ± 8.8 g vs. 211.6 ± 20.1 g, p < 0.05) with no significant difference in food intake. The serum total cholesterol, triglyceride, and low-density lipoprotein (LDL) levels were significantly lower in the GO2KA1-treated group than in the control group, whereas the high-density lipoprotein (HDL) level was higher in the GO2KA1 group. The GO2KA1-treated group also showed a significant reduction in ALT and AST levels compared to the control. Moreover, serum adiponectin levels were significantly 1.5-folder higher than the control group. These in vivo and in vitro findings suggest that dietary supplementation of GO2KA1 may prevent diet-induced weight gain and the anti-obesity effect is mediated in part by inhibiting adipogenesis and increasing adiponectin level.

<p class="Abstract">The aim of this study was to evaluate the diuretic activity of <em>Eysenhardtia polystachya</em> bark aqueous extract at different doses in a rat model. Different doses of <em>E. polystachya</em> (125, 250, 500 and 750 mg/kg body weight), furosemide (4 mg/kg) and vehicle were administered per os to female rats (n=6 animals per group). After 6 hours in metabolic cages, the effect on urinary flow, glomerular filtration rate and electrolyte balance of sodium and potassium were assessed in all animals. <em>E. polystachya</em> at the doses of 500 and 750 mg/kg induced diuretic activity, since markedly increased (p&lt;0.05) the urinary flow rate, similar to that of furosemide treated group. Only the dose of 750 mg/kg produced an increment in urinary excretion of sodium but not of potassium compared with control group. These findings indicate that<em> E. polystachya</em> bark-induced diuretic activity, providing evidence for its folkloric use.</p><p> </p>

The metabolic response to nutrient ingestion and the rate of digestion and absorption of nutrient molecules in bowel physiology plays an important role in the metabolic control of some human chronic non-infectious diseases. This experimentally-controlled designed nutritional study which lasted eight weeks aimed to determine the effects of proportional high-protein/low-carbohydrate (HP/LC) formulated diet on glycemic tolerance, glycemic control, body weight, organ weight and organ morphometry in healthy and diabetic adult male Wistar rats. Twenty-four male Wistar rats purchased from a disease-free stock were randomly categorized into four groups (n = 6, each) after two weeks acclimatization period in raised stainless steel cages with 6 mm2mesh floor and replaceable numbered blotters papers placed under each cage in a well-ventilated animal house. Animal groups include: Healthy control group (HC), Healthy treated group (HT), Diabetic control group (DC) and Diabetic treated group (DT. The animals were fed according to the experimental design with water ad libitumfor eight weeks. Diabetes was inducted with freshly prepared alloxan monohydrate solution (150 mg/kg bw, intraperitoneally). Body weights and fasting blood sugar concentrations were measured twice weekly, while oral glucose tolerance test was conducted on the last day of the eighth-week study and subsequently followed by organs extraction after anesthesia for weight and gross assessment. Proportional high-protein/low-carbohydrate formulated diet caused significant reduction in mean body weight of treated diabetic (DT: 22.6%; P= .001) and healthy (HT: 5.8%; P= .007) rats while the control animals on control diet recorded significant (P< .05) increase in body weight gain (DC: 12.4%; HC: 11.2%). Glycemic tolerance and control improved significantly in diabetic treated rats over that of the healthy treated rats. Gross morphometry of the extracted organs (kidneys, liver, heart, lungs, spleen and testes) revealed sustained normal morphological features without any visible lesion. In conclusion, consumption of proportional high-protein/low-carbohydrate formulated diet enhanced body weight reduction and sustained normal organ morphological features with good glycemic tolerance and control in experimental rats, suggesting its dietary potentiality, safety and suitability to ameliorate obesity-related diabetes.

Introduction: This study evaluates the histological changes of uterine tissue in the female Sprague Dawley rats after administration of Trigonella foenum-graecum (fenugreek) seeds in comparison to combined oral contraceptive pills (COCPs). Methods: Twenty four female Sprague Dawley strain rats of 8 weeks old were divided into A, B, and C groups. Group A was the control and B was administrated with 0.05 mg/kg body weight of COCPs for 15 days. Group C was treated with 750 mg/kg body weight of fenugreek seed aqueous (FSA) extract for 15 days. The uterine tissues were isolated and studied histologically. Results: The uterine weight in the FSA extract treated group was normal, while in the COCPs treated group it was insignificantly increased. The endometrial thickness and endometrial gland density were normal in the FSA extract treated group, while in the COCPs treated group it was atrophied. Decidual-like cells were observed in the COCPs treated group only. Conclusions: Endometrial atrophy and decidual-like cells were noted after COCPs treatment while in the FSA extract and control groups they were absent. The findings suggest that fenugreek seeds could be used as an alternative natural and safe contraceptive product.
