This study is designed to find out the present status of PbB level and its adverse effect on oxidative stress and antioxidant  of Silver Jewellery Workers (SJW). Forty-two SJW of having an age range of 20-45 years were included for this study and compared with 50 healthy male subjects of the same age-matched. All biochemical investigations were determined by using standard methods. In this study, we found significantly increased PbB level (325%), serum lipid peroxide (191%) and significantly decreased antioxidant enzymes like superoxide dismutase (-37.83%), Catalase (-27.64%), Ceruloplasmin (-10.2%), Nitric oxide (-36.54%) in silver jewellery workers as compared to control subjects. PbB level positively correlated with lipid peroxide (0.46) and negatively correlated with superoxide dismutase (-0.31), Catalase (-0.30), Ceruloplasmin (-0.20) and Nitric oxide (-0.28) were observed in study subjects.  Therefore we have planned this study to know the oxidative stress and antioxidant status of silver jewellery workers and we found that blood lead level results in oxidative stress and alters antioxidant enzymes of silver jewellery workers and to reduce this oxidative stress it is indispensable to decrease PbB level of silver jewellery workers by taking vitamins, minerals and using activated carbon fabric mask which can prevent the health hazards of lead.

Effect of use of Activated Carbon Fabric (ACF) mask on blood lead (PbB), lipid peroxide, erythrocytes-Superoxide Dismutase (SOD), Catalase, Ceruloplasmin, and liver functions tests of battery manufacturing workers (BMW) were studied. For this study, 36 male battery manufacturing workers, those who are regularly using ACF mask for two months were included. A blood sample was collected before and after using ACF masks from battery manufacturing workers and estimated the blood lead level, liver function tests, oxidative stress, and antioxidant status parameters by using the standard method. After two months use of ACF mask by BMW, the blood lead level (P<0.01, -15.76%), total serum bilirubin (P<0.05, -28.09%), alanine transaminase (P<0.05, -20.84%), lipid peroxide (P<0.001, -33.33%) were significantly decreased and erythrocytes-superoxide dismutase (P<0.05, 22.38%), catalase (P<0.05, 38.39%), ceruloplasmin (P<0.05, 13.13%) were significantly increased in battery manufacturing workers as compared to before using the mask. This study reflects the use of two months ACF mask of high lead-exposed workers are useful to decrease lead absorption, which results in decreases oxidative stress and improves the antioxidant status and prevents liver toxicity.

To see the present scenario of blood lead (BPb) levels of Silver Jewellery Workers (SJW) and its effects on haematological and heme biosynthesis parameters. Forty-two SJW of having an age range of 20-45 years were included for this study and compared with 50 healthy male subjects of the same age. Blood lead, RBC δ- aminolevulinic acid dehydratase (δ-ALDH), urinary δ- aminolevulinic acid (U- δ-ALA), urinary porphobilinogen (U-PBG) and haematological parameters were measured. Blood lead level of silver jewellery workers was significantly elevated (p<0.001, 325%) and non-activated δ-ALAD (p<0.01), activated δ-ALAD (p<0.05) were notably reduced and the ratio of A/NA δ-ALAD (p<0.01) was considerably enlarged in SJW as compared to control group subjects. Urinary excretion of δ-ALA (p<0.001) and PBG (p<0.001) levels were extensively increased in SJW as compared to subjects from the control group. Hb (p<0.001), PCV (p<0.001), MCV (p<0.001), MCH (p<0.05) and RBC (p<0.001) were notably reduced in SJW, while total WBC count (p<0.001, 29.8%) was considerably elevated as compared to subjects from the control group. Conclusions: Increased BPb level in SJW indicates the more absorption of lead from the GIT, which inhibits heme biosynthesis, and alters haematological parameters.

AbstractBackgroundThe aim of this study was to know the present scenario of blood lead level and its effect on serum lipid peroxide and antioxidant parameters of spray painters.MethodsWe included 42 male spray painters and 50 control subjects with an age range of 20–45 years. Blood lead level, serum lipid peroxide and antioxidant parameters, i.e. erythrocyte catalase, superoxide dismutase (SOD), plasma ceruloplasmin, nitric oxide, uric acid and bilirubin, were quantified by standard methods.ResultsWe observed the significant elevated blood lead (p < 0.001, 458%), lipid peroxide (p < 0.001, 170%), uric acid (p < 0.001, 25.6%) and bilirubin (p < 0.01, 24.5%) and the significant decrease in antioxidant enzymes like SOD (p < 0.001, −50.4%), catalase (p < 0.001, −34.33%), ceruloplasmin (p < 0.001, −32.7%) and nitric oxide (p < 0.001, −39.58%) in the study group as compared to control. A significant positive correlation coefficient (r) of blood lead level with lipid peroxide (r = 0.44, p < 0.001), uric acid (r = 0.33 p < 0.05) and bilirubin (r = 0.35, p < 0.05) and a negative correlation with SOD (r = −0.32, p < 0.05), catalase (r = −0.33, p < 0.05), ceruloplasmin (r = −0.27, p < 0.05) and nitric oxide (r = 0.30, p < 0.05) were observed.ConclusionsElevated blood lead level induces serum lipid peroxide and alters the antioxidant enzymes of spray painters. Therefore, it is necessary to reduce the blood lead level by taking proper precautions while spraying the paints, and additional antioxidant supplementation like vitamin C, multivitamin and multiminerals will be useful in reducing oxidative stress.

Endometriosis is described as a gynecological disorder characterized by the presence of endometrial tissue outside the uterus; extensively explored because of its increasing incidency, with an indubitable diagnostic only after invasive surgery, with no efficient treatment, it has still many aspects to be elucidated. A growing body of facts sustain oxidative stress as a crucial factor between the numerous incriminated factors implicated in endometriosis ethiopathogeny. Reactive oxygen species(ROS) act to decline reproductive function. Our study intends to determine if an experimental model of endometriosis may be useful to assess the impact of oxidative stress on endometrial cells; we have used a murine model of 18 adult Wistar female rats. A fragment from their left uterine horn was implanted in the abdominal wall. After 4 weeks, a laparatomy was performed, 5 endometrial implants were removed, followed by biochemical tissue assay of superoxide dismutase(SOD) and catalase(CAT). At the end of the experiment, the rats were sacrificed, the implants were removed for histopathological exam and biochemical assay of antioxidant enzymes. The results revealed decreased levels of antioxidant enzymes, pointing on significant oxidative stress involvement.

Abstract The aim of the this study was to evaluate the effects of fresh, dried and freeze-dried Centaurea depressa M. Bieb. (Asteraceae) on the oxidant and antioxidant status of the model organism D. melanogaster Meigen (Diptera: Drosophilidae) experimentally. The study was carried out from 2016 to 2019, and plant leaf extracts (0-50 mg/l) were added to insect standard artificial diets. The total protein, protein carbonyl content and glutathione-S-transferase, superoxide dismutase and catalase activities were quantified at the insect’s third larval stage. Our data showed that protein carbonyl content varied from 2.70 nmol/mg protein in the control group to 59.11 nmol/mg protein in the group fed with fresh leaf extract signifying induction of oxidative stress. All extracts increased the levels of all antioxidant enzymes and decreased the amounts of total protein. Meanwhile, the group fed with the freeze-dried extract showed no significant difference in the levels of total protein and protein carbonyl content except at the 50 mg/l concentration of the extract. Moreover, this group had superoxide dismutase and catalase activities 4 to 5 times higher than in the control group. In conclusion, induction of oxidative stress indicates that the fresh form of C. depressa leaves may have potential as a natural pesticide, whereas induction of endogenous antioxidant enzymes by the freeze-dried extract suggest its potential as an antioxidant.

SummaryLong-term heat stress (HS) induced by testicular insulation generates oxidative stress (OS) on the testicular environment; consequently activating antioxidant enzymes such as superoxide dismutase (SOD), glutathione reductase (GR) and glutathione peroxidase (GPx). The aim of this work was to immunolocalize antioxidant enzymes present in different cells within the seminiferous tubule when rams were submitted to HS. Rams were divided into control (n = 6) and treated group (n = 6), comprising rams subjected to testicular insulation for 240 h. After the testicular insulation period, rams were subjected to orchiectomy. Testicular fragments were submitted to immunohistochemistry for staining against SOD, GR and GPx enzymes. We observed immunolocalization of GPx in more cell types of the testis after HS and when compared with other enzymes. In conclusion, GPx is the main antioxidant enzyme identified in testicular cells in an attempt to maintain oxidative balance when HS occurs.

Oxidative stress is one of several factors which contribute to the development of colorectal carcinogenesis. The aim of the study was an assessment of the activity of antioxidant enzymes in tumour and corresponding normal distal mucosa in a group of patients with colorectal adenocarcinoma. Samples of tumour and corresponding normal mucosa were obtained during a resection of colorectal cancer from 47 patients aged between 26 and 82 years. The average distance of corresponding normal distal mucosa from the tumour was 4.49 cm. Activities of antioxidant enzymes: superoxide dismutase (SOD), glutathione reductase (GR), glutathione peroxidase (GPx), glutathione S-transferase (GST), and catalase (CAT) were measured in tissue homogenates. The patients were grouped according to the tumour stage (Duke's staging), grading, localization, and size of tumour, as well as age and sex. Statistical analysis was performed. The activity of SOD and GPx was considerably increased, while the activity of GST decreased significantly in tumour as compared with normal mucosa. GR activity in colorectal cancer was evidently higher in tumours of proximal location compared with the distal ones. The distance of corresponding normal distal mucosa from the tumour was analyzed and related to all assayed parameters. A decreased GST activity was observed in corresponding normal mucosa more than 5 cm distant from the tumour in patients with CD Duke's stage. The higher activity of superoxide dismutase and glutathione peroxidase in tumour compared to corresponding normal mucosa could indicate higher oxidative stress in colorectal adenocarcinoma cells.

Superoxide radical (O2-.) combines with nitric oxide (NO) to form peroxynitrite, thereby nullifying the biological activity of NO. Superoxide dismutase (SOD) prevents this reaction by converting O2-. to H2O2. We tested the hypotheses that the antioxidant enzymes catalase (CAT), Mn SOD, and Cu/Zn SOD are present in enteric neurons of the opossum esophagus, and that O2-. alters esophageal motor function. Immunostaining demonstrated CAT, Mn SOD, and Cu/Zn SOD immunoreactivity in interganglionic nerve bundles and ganglia of the myenteric and submucosal plexuses. Western blot analysis confirmed the presence of these enzymes in homogenates of esophageal muscularis propria, and enzyme assays demonstrated Cu/Zn SOD and Mn SOD activities of 262 and 73 U/mg protein, respectively. Both diethyldithiocarbamic acid, an inhibitor of Cu/Zn SOD, and xanthine (X) with xanthine oxidase (XO), which generate O2-., shortened the latency of the nerve-mediated contraction of circular esophageal muscle, the off response, by 20.2 and 23.4%, respectively. SOD alone did not affect the latency, but it inhibited the effect of X with XO on the latency. Antioxidant enzymes found in intramural esophageal nerves may play a role in regulating NO-mediated neuromuscular communication in the esophagus.

Objective: To compare oxidative stress between primary retinoblastoma and retinoblastoma with distant metastasis. Patients and methods: Forty consecutive patients presented with primary retinoblastoma and the same number of patients presented with distant metastasis, attending the outpatient department of our hospital between August 2002 and April 2005. All the patients with retinoblastoma underwent a standard metastasis workup and were subsequently categorized into two groups (without metastasis and with metastasis).Venous blood samples were drawn from each patient. After proper centrifugation, serum was collected and antioxidant enzymes and reactive oxygen species (ROS) were assayed. Main outcome measures: Serum collected from the patients was subjected to biochemical assay of the antioxidant enzymes (superoxide dismutase, catalase and peroxidise) and ROS to determine any difference in enzyme activity between the two groups. Results: Antioxidant levels were found to be less in the metastasis group as compared to the primary intraocular retinoblastoma group(p<0.05).Mean ROS activity was found to be increased in metastatic group (p<0.05). Conclusion: The decreased antioxidant enzymes level along with increased ROS activity in patients with metastatic retinoblastoma reflect increased oxidative stress as compared to primary intraocular retinoblastoma patients.DOI: http://dx.doi.org/10.3126/nepjoph.v4i2.6543 Nepal J Ophthalmol 2012; 4 (2): 271-276
