<p class="JITV-ReferenceIsi">The prolactin gene (PRL) is a gene that controls the incubation and egg production in laying chickens. The nature of incubation will reduce egg production and disrupt the reproductive system in local chickens. The purpose of this study was to identify the polymorphism of prolactin genes in IPB-D1 chickens using the direct sequencing method. The polymorphism of the exon 5 prolactin gene was carried out on 46 samples of IPB-D1 chicken DNA which was a collection of the Division of Animal Genetics and Breeding, Faculty of Animal Science IPB. DNA sequences as a reference for designing exon 5 primers were obtained from the National Center for Biotechnology Information (NCBI) with the GenBank access code: AF288765.2. DNA extraction was carried out using the phenol-chloroform technique. DNA amplification resulted in a PCR product with a size of 557 bp. In this study, the genotype frequency, allele frequency, heterozygosity value and Hardy-Weinberg equilibrium were calculated. The results of the study found 5 SNPs in exon 5, namely g.7823A&gt;G, g.7835A&gt;G, g.7886T&gt;A, g.8052T&gt;C, and g.8069T&gt;C. All SNPs are polymorphic and in Hardy-Weinberg equilibrium except g.8052T&gt;C. The g.7823A&gt;G, g.7835A&gt;G, g.8052T&gt;C SNPs are synonymous mutations that do not change amino acids, while the g.7886T&gt;A and g.8069T&gt;C SNPs are non-synonymous that change amino acids. Both g.7886T&gt;A and g.8069T&gt;C SNPs are potential as a marker assisted selection for the characteristics of egg production in IPB-D1 chickens.</p>

Abstract Insulin-induced gene 1 (INSIG1) encoded protein that blocked of proteolysis activity from protein sterol regulatory element binding proteins (SREBP). The role gene plays of cholesterol, fatty acid, and glucose metabolism. Indonesia has many several beef cattle which has good quality of growth trait. The aim of this study was to identify polymorphism of INSIG1 gene (SNP 4366 (A>G) and 4534 (T>C)) of three local beef cattle in Indonesia (Bali, Pasundan and Ongole Decendent). One hundred and twenty seven samples were used in this study consisting of Bali cattle (46) from Pulukan Bali Island, Pasundan (36) from UPTD BPPT Beef cattle Ciamis West Java and Ongole Grade (OD) (45). DNA was extracted from whole blood using High Salt method then genotyping used PCR-RFLP method with RsaI and TaqI enzymes. Parameters in this study were genotype and allele frequencies, heterozigosity observed (Ho), expected (He), Hardy-Weinberg Equilibrium (HWE) and Polymorphism Information Content (PIC). Result showed that a 428 bp of DNA fragment was successfully amplified and digested. Three variant of genotypes with two alleles were identified. For SNP 4366 (A>G) were AA, AG and GG with dominant of G allele while SNP 4534 (T>C) were CC, CT and TT with dominant C allele. Both of SNPs in each of breed populations was in Hardy-Weinberg Equilibrium and polymorphic. Additionally, PIC value reached moderate. In conclusion, polymorphism was found in beef cattle and it could be early study for genetic diversity

Eriani K, Rahmi R, Jamil I, Rosnizar R, Azhar A. 2019. Body size characteristics and polymorphism in GH and GHRH genes of Simeulue Buffalo of Aceh, Indonesia. Biodiversitas 20: 236-242. This study was done to determine body size characteristics, to identify the presence of polymorphism in the GH/MspI and GHRH/HaeIII genes, and to analyze the relationship between GH and GHRH gene polymorphisms to body size characteristics in Simeulue buffalo. Experimental animals used were 45 Simeulue buffalo randomly sampled from three subdistricts, i.e., Salang, Alafan and Simeulue Cut, fifteen from each subdistrict. Body size characteristics measured were withered height, body length, chest circumference, chest depth, and hip height. A 5 mL whole blood was collected from fifteen out of forty five by using EDTA as an anticoagulant. Extraction of genomic DNA and analysis of GH/MspI and GHRH/HaeIII polymorphisms was done using GeneAid DNA extraction kit and polymerase chain reaction-restriction length fragment polymorphism (PCR-RFLP) method, respectively. The data of body size characteristics which includes mean value, standard deviation, and coefficient of variation were analyzed descriptively. The molecular data represented by DNA banding pattern was used to calculate allele and genotype frequencies, expected (He) and observed (Ho) heterozygosities, and Hardy-Weinberg equilibrium. The relationship between genotype and body size characteristic was analyzed using t-test. The results showed that coefficient of variation in body size characteristics was low, ranging from 5.39-16.06% in the age category of 4-6 years old and ranging from 5.25-11.27% in the age category of 2- 4 years old. The results of the molecular analysis indicated that GH/MspI locus was monomorphic by the presence of one (+/+) genotype. GHRH/HaeIII locus was polymorphic as shown by the presence of 3 genotypes namely AA (0.13), AB (0.80) and BB (0.07). Expected heterozygosity (0.80) was higher than observed heterozygosity (0.49). The frequency of GHRH/HaeIII agreed with Hardy Weinberg equilibrium. GHRH/HaeIII polymorphism did not significantly influence (P>0.05) body size characteristics of Simeulue buffalo.

Abstract. Recently, Garfinkel and Critchley (2013) proposed to distinguish between three facets of interoception: interoceptive sensibility, interoceptive accuracy, and interoceptive awareness. This pilot study investigated how these facets interrelate to each other and whether interoceptive awareness is related to the metacognitive awareness of memory performance. A sample of 24 healthy students completed a heartbeat perception task (HPT) and a memory task. Judgments of confidence were requested for each task. Participants filled in questionnaires assessing interoceptive sensibility, depression, anxiety, and socio-demographic characteristics. The three facets of interoception were found to be uncorrelated and interoceptive awareness was not related to metacognitive awareness of memory performance. Whereas memory performance was significantly related to metamemory awareness, interoceptive accuracy (HPT) and interoceptive awareness were not correlated. Results suggest that future research on interoception should assess all facets of interoception in order to capture the multifaceted quality of the construct.

The scientific problem of economics “Managing the quality of human life” is formulated on the basis of artificial intelligence, algebra of logic and logical-probabilistic calculus. Managing the quality of human life is represented by managing the processes of his treatment, training and decision making. Events in these processes and the corresponding logical variables relate to the behavior of a person, other persons and infrastructure. The processes of the quality of human life are modeled, analyzed and managed with the participation of the person himself. Scenarios and structural, logical and probabilistic models of managing the quality of human life are given. Special software for quality management is described. The relationship of human quality of life and the digital economy is examined. We consider the role of public opinion in the management of the “bottom” based on the synthesis of many studies on the management of the economics and the state. The bottom management is also feedback from the top management.
