Background: Rats are commonly used animals in development of newer drugs, rectification of toxicity and to record the various alterations in physiological parameters following pharmacological and non pharmacological interventions.Aim and Objectives: The purpose of this study was to identify the best physiological window during anesthesia. Therefore, we compared the effect of anesthesia using combination of ketamine and xylazine (KX) and thiopental sodium (intraperitoneally) on blood pressure and heart rate in adult male Wistar rats. Material and Methods: Twelve, male Wistar rats with a mean body weight of 260 ± 15 g were acquired. Thiopental sodium and cocktail of ketamine and xylazine (KX) were administered (ip) in group- I and group-II respectively. The systolic blood pressure and heart rate was recorded in both the groups till the awakening phase.Results: We found that there was a constant SBP and HR in Ketamine/Xylezine groups that are from 30 to 90 minutes after injection of anesthesia while this window was not observed in thiopental group.Conclusion: Our study concludes that the best time to observe the effect of newer drug during period between 30- 90 minutes after anesthesia.Asian Journal of Medical Sciences Vol.9(6) 2018 37-39

Aims: To study the protective effect of hydroalcoholic bark extract of Terminalia arjuna against DBTC induced pancreatic cancer in male wistar rats. Study design: Healthy male Wistar Albino rats weighing 150-200 g were segregated into four groups (n=6). Group I was considered as normal control, received normal saline (0.9%w/v, 1 ml/kg body weight, orally). Group II rats were treated with DBTC (6 mg/kg body weight, i.p.) which served as negative control. Group III and IV received Terminalia arjuna Linn bark hydroalcoholic extract at doses of 250 mg/kg body weight, per oral and 500 mg/kg body weight, per oral respectively. Place and Duration of Study: University College of Pharmaceutical Sciences, Andhra University, Visakhapatnam, Andhra Pradesh, India, between May 2020 and July 2020. Methodology: The experimental animals were segregated into four groups of six rats each. According to acute toxicity data, 250 mg/kg as low dose and 500 mg/kg as high dose of the test compound have been chosen for administration. All the drugs were given for 28 consecutive days to all the respective groups with standard pellet diet and water ad libitum. The assessment of serum parameters such as α-Amylase, Lipase and blood glucose levels were carried out on 1st day, 14th day and 28th day to the respective groups. Results: Pretreated groups of Terminalia arjuna Linn bark hydroalcoholic extract (250 mg/kg and 500 mg/kg body weight, orally) showed significant (‘#’p<0.001) decrease in the levels of α-Amylase, Lipase and glucose in the blood when compared to DBTC (6 mg/kg body weight, i.p.) induced group which served as negative control. Conclusion: This study suggests that Terminalia arjuna may have a protective role against DBTC induced pancreatic cancer in male wistar rats and further investigation may be required to confirm its therapeutic potentials clinically.

Background : The 1st and 2nd degree hemorrhoids is managed non-operatively with the anti-inflammatory and plebothropic drugs. Graptophyllum pictum extract (GPE) has already been used widely in Indonesia to treat hemorrhoid with good result, however, the mechanism is not supported by the molecular research.Objective : This study is intended to study the phlebothropic effects of GPE by measuring the degree of edema and extravassal leucocytes of experimental wistar hemorrhoid.Methods : Experimental study with Randomized Controlled Trial Post-test only design in male wistar rats, weight around 200 gr, induced for the development of a disease-like condition of hemorrhoids by 6% croton oil on the anus for 3 days. Random allocation was performed to divide the 14 wistar rats in 2 groups. Group I as control got normal saline solution, while group II was treated with GPE 100mg/kgbw, started on day 4th for 5 consecutive days. On 9th day blood was extracted from retroorbital fossa and anus was resected up to 2 cm from anal verge and weighted. The degree of anal edema was measured by rectoanal coefficient, that is rasio between anal weight (miligrams) and body weight (grams).Results : Until the end of study, all wistar rat were still alive. The mean (±SD) of body weight   of control group was 173.84 ­(±13.37) and the treatment group was 171.70  (±13.10), and there was no significant differences (p = 0.833). The mean (±SD) of rectoanal coefficient in the treatment group was 2.46 (±0.41) and it was significantly lower than control group  (3.13 ± 0.85) (p = 0.029). The mean (±SD) of extravassal leukocytes in the treatment group was 900.14(±48.09) and it was significantly lower than the control (1003.28 ± 99.30) (p = 0.042).Conclusions : Graptophyllum pictum extract at doses of 100 mg/Kgbw have phlebotonic effect  in decreased edema and extravassal leukocytes.

A prospective comparative study was done on lignocaine versus lignocaine with pethidine to observe the effect on cardiovascular response to laryngoscopy and endotracheal intubation. One hundred such elective surgical patients of active age group (16 - 60 years) having American Society of Anaesthesiologist (ASA) physical status I & II irrespective of surgical procedure were randomly assigned to one of the two groups of 50 each. Group I received injection lignocaine 1 mg/kg intravenously 02 minutes before induction of general anaesthesia. Patients in group II received injection pethidine 1 mg/kg body weight and injection lignocaine 1 mg/kg body weight intravenously 02 minutes before induction of general anaesthesia. Haemodynamic parameter i.e. blood pressures (systolic blood pressure, diastolic blood pressure and mean blood pressure), heart rate, rate pressure product were monitored after 1st, 3rd, 5th minutes following intubation. There were statistically significant (p<0.001) increase in blood pressures, heart rate and rate pressure product in group I i.e. pretreatment with 1 mg/kg body weight intravenous lignocaine and remained so after 5 minutes. On the other hand there were no statistically significant (p>0.05) increase in heart rate, blood pressures and rate pressure product in group II, where pretreatment done with pethidine 1 mg/kg body weight with lignocaine 1 mg/kg body weight and the values returned control level before 5 minutes. The study showed that pethidine 1 mg/kg body weight with lignocaine 1 mg/kg body weight pretreatment suppresses the cardiovascular response due to laryngoscopy and intubation. Abstract Objectives Exposure to light-at-night (LAN) has been reported to impair blood glucose regulation. The liver modulates blood glucose through mechanisms influenced by several factors that include peroxisome proliferator-activated receptor gamma coactivator-1alpha (PGC-1α) and glucose-6-phosphatase (G6Pase). This study investigated the effect of intermittent exposure to green and white LAN on some hepatic glucose regulatory factors in male Wistar rats. Methods Animals were divided into three equal groups. Group I (control) was exposed to normal housing conditions. Groups II and III were each daily exposed to either green or white LAN for 2 h (7–9 pm) for 14 days. Body weight and blood glucose was monitored on days 0, 7, and 14. Thereafter, retro-orbital sinus blood was obtained after light thiopental anaesthesia and serum insulin was determined. Liver samples were also obtained and evaluated for glycogen, PGC-1α, and G6Pase activity. Insulin resistance was estimated using the HOMA-IR equation. Results Body weight and blood glucose on days 7 and 14 increased in groups II and III compared to control. Hepatic PGC-1α and G6Pase increased in group II (2.33 ± 0.31; 2.07 ± 0.22) and III (2.31 ± 0.20; 0.98 ± 0.23) compared to control (1.73 ± 0.21; 0.47 ± 0.11). Hepatic glycogen was 71.8 and 82.4% reduced in groups II and III compared to control. Insulin in group II increased (63.6%) whiles group III values reduced (27.3%) compared to control. Insulin resistance increased in group II (0.29 ± 0.09) compared to control (0.12 ± 0.03) and group III (0.11 ± 0.03), respectively. Conclusions Exposure to 2 h green and white LAN in the early dark phase increases hepatic glycogenolysis and gluconeogenetic activities resulting in increased blood glucose. In male Wistar rats, exposure to green but not white LAN may predispose to insulin resistance.

The presence of μ-opioid receptors and endomorphins has been demonstrated in the general area encompassing the rostral ventrolateral medullary pressor area (RVLM). This investigation was carried out to test the hypothesis that endomorphins in the RVLM may have a modulatory role in regulating cardiovascular function. Blood pressure and heart rate (HR) were recorded in urethane-anesthetized male Wistar rats. Unilateral microinjections of endomorphin-2 (0.0125–0.5 mmol/l) into the RVLM elicited decreases in mean arterial pressure (16–30 mmHg) and HR (12–36 beats/min), which lasted for 2–4 min. Bradycardia was not vagally mediated. The effects of endomorphin-2 were mediated via μ-opioid receptors because prior microinjections of naloxonazine (1 mmol/l) abolished these responses; the blocking effect of naloxonazine lasted for 15–20 min. Unilateral stimulations of aortic nerve for 30 s (at frequencies of 5, 10, and 25 pulses/s; each pulse 0.5 V and 1-ms duration) elicited depressor and bradycardic responses. These responses were significantly attenuated by microinjections of endomorphin-2 (0.2 and 0.4 mmol/l). The inhibitory effect of endomorphin-2 on baroreflex responses was prevented by prior microinjections of naloxonazine. Microinjections of naloxonazine alone did not affect either baseline blood pressure and HR or baroreflex responses. These results indicate that endomorphin-2 elicits depressor and bradycardic responses and inhibits baroreflex function when injected into the RVLM. These effects are consistent with the known hyperpolarizing effect of opioid peptides on RVLM neurons.

Background Hypercholesterolemia is the predominantfactor in developing atherosclerosis and myocardial diseases.A major contributor for the progression of atherosclerosis is abnormalities in lipid and lipoprotein metabolism.Hence the objectives of the study was to estimatethe comparative efficacy of Lutein with atorvastatin on lipid profile and lipoprotein(a) and to estimate the histopahthological changes in hypercholesterolemic male wistar rats. Materials and Methods Experimental Wistar rats (male) were grouped into six. Each group contains 6 rats. Group I is control. Group II received cholesterol rich diet. Group III received cholesterol rich diet and the drug Atorvastatin 5mg/kg. Group IV received cholesterol rich diet and the drugLutein 25mg/kg. Group V received cholesterol rich diet and the drugLutein 50mg/kg. Group VI received cholesterol rich diet and the drug Lutein100mg/kg. At the end of 16 weeks, Blood samples from each rats was taken through retro-orbital puncture to evaluate serum lipoproteinsand lipoprotein(a) and thenwistar rats were sacrificed underinjection I.M Ketamine,Aortaand Liverwere dissected out and sent for histopathological studies. Results The plasma LDL, VLDL, Triglycerides, total cholesterol, lipoprotein(a) levels were reduced in all lutein treated groups and atorvastatin treated group compared to high cholesterol diet group. A significant rise in HDL levels was noted in all Lutein treated groups and atorvastatin treated group. No statistically significant difference was seen between Atorvastatin 5mg/kg body weight and Lutein 100mg/kg body weight on reduction of total cholesterol.The efficacy of the drug Lutein in progression of atherosclerosis and its cytoprotective action in liver was proved in this study. Conclusion This study indicates that Lutein has effect onreducing plasma lipoproteins&the study had shown significant antiatherogenic effect.


Several studies have shown that smoking behavior is closely related to weight and obesity. Smokers tend to prefer fatty foods with high calories which ultimately makes a smoker vulnerable to being overweight. However, there are some studies that show lower body weight was found in smokers than nonsmokers. Our aim is to evaluate the short-term effects of smoking with conventional cigarettes and e-cigarret on weight changes in Wistar rats because animals try to minimize some confounding factors that are difficult to work on human samples (for example, physical activity, diet and motivation). This research is an experimental study, using 8-week-old male Wistar rats with body weight 150-200 grams divided randomly into 5 groups, namely 1) the group was not treated, 2) the filter cigarette exposure group, 3) the clove cigarette group , 4) e-cigarette group with a dose of 3 mL with nicotine content of 0 mg, and 5) ecigarette group with a dose of 3 mL with nicotine content of 3 mg. Body weight measurements using a digital scale. The results showed that there was a significant change in body weight between clove cigarettes and filter cigarettes with p = 0.024. In the results of this study, showed weight loss in all groups given cigarette smoke. Weight loss was significantly lower in the filter cigarette group compared to clove cigarettes and there was no significant weight difference between the conventional cigarette group and e-cigarette. This is likely the cigarette filter actually creates a major problem for smokers because of the design and materials used so it does not provide a safer effect for smoker.

Experiments were carried out in pentobarbital-anesthetized, vagotomized, paralyzed, and artificially ventilated male Wistar rats with a pneumothorax. Blood pressure, heart rate, and phrenic nerve activity were recorded. Extracellular neuronal recordings were made by glass micropipettes in a midline area in the commissural subnucleus of the nucleus tractus solitarius. At the end of the experiment, the recording sites were marked. Chemoreceptors were stimulated by tracheal administration of N2 or injection of CO2-saturated saline (50 microliters) near the carotid body. The baroreceptors were stimulated by an intravenous injection of phenylephrine (1-3 micrograms). Neurons that were excited by chemoreceptor stimulation but not baroreceptor stimulation were found in the commissural subnucleus of nucleus tractus solitarius and were designated as chemoreceptor-sensitive neurons. They exhibited a continuous discharge (9.9 +/- 1.3 spikes/s) that showed no apparent relation to either phrenic nerve bursts or arterial pulses. Such neurons were not found in the rostral portions of the nucleus tractus solitarius. These results confirm and extend our previous reports in which a discrete chemoreceptor projection site was identified in the commissural subnucleus of the rat.

Newborn male Wistar rats were treated with nerve growth factor daily by subcutaneous injection for 2 weeks, and control rats were treated with either cytochrome c or buffered saline. Average body weight of the treated animals was lower than that of the controls during the 2 weeks of treatment, but became similar to that of the controls thereafter. Tissue levels of norepinephrine were elevated in the brain, adrenal glands, mesenteric arteries, and vas deferens of the treated animals immediately after the treatment, but became similar in the three groups 2 weeks after the termination of the treatment. Blood pressure and heart rate were measured beginning at 4 weeks of age until 28 weeks, when the rats were sacrificed and the mesenteric arteries sampled for morphometric measurements of vessel wall dimensions. Pretreatment with nerve growth factor did not affect blood pressure, nor heart rate. Structural alteration of the three types of mesenteric arteries was also absent in the treated animals. We conclude that even though neonatal treatment of normal Wistar rats with nerve growth factor for 2 weeks induced an elevation of the norepinephrine levels in several tissues at the end of the treatment period, it was not sufficient to produce hypertension and structural alterations in the blood vessels.Key words: nerve growth factor, sympathetic nerve, hypertension, vascular changes, morphometry.
