One of the most promising directions of solving the satisfying constantly growing problem of Ukrainians residents tourist, business and migration activity is the diversification of the rail transport activity in the international passenger transportation market. In the conducted studies, the analysis of possible variants of passenger transportation logistics from Eastern Ukraine (in particular from Kharkiv) to Krakow was made. For comparison, seven possible variants of transportation and a new project solution (direct passenger train Kharkiv - Kiev - Lviv - Mostiska II - Krakow with the perspective of continuation of the route to Prague or Vienna) is selected. Based on the SWOT analysis, as well as marketing and logistics research on project variants, it has been proved that the proposed direct rail rout between Kharkiv and Krakow can compete on an equal footing with existing variants of transport.

Rhabdomyosarcoma (RMS), is the most frequent soft tissue tumor in children that originates from disturbances in differentiation process. Mechanisms leading to the development of RMS are still poorly understood. Therefore, by analysis of two RMS RH30 cell line subclones, one subclone PAX7 negative, while the second one PAX7 positive, and comparison with other RMS cell lines we aimed at identifying new mechanisms crucial for RMS progression. RH30 subclones were characterized by the same STR profile, but different morphology, rate of proliferation, migration activity and chemotactic abilities in vitro, as well as differences in tumor morphology and growth in vivo. Our analysis indicated a different level of expression of adhesion molecules (e.g., from VLA and ICAM families), myogenic microRNAs, such as miR-206 and transcription factors, such as MYOD, MYOG, SIX1, and ID. Silencing of PAX7 transcription factor with siRNA confirmed the crucial role of PAX7 transcription factor in proliferation, differentiation and migration of RMS cells. To conclude, our results suggest that tumor cell lines with the same STR profile can produce subclones that differ in many features and indicate crucial roles of PAX7 and ID proteins in the development of RMS.

The pace and scale of the digitalization of today’s global information society open up new opportunities for business. At the same time, they set new challenges for business owners and managers in the field of marketing. Given this fact, the purpose of the study was to present the impact of digitalization on the marketing activity of the enterprise in the field of services by promoting the use of online sales via electronic distribution channels, social networks, and mobile applications. A comparative system of estimating the parameters of the influence of digitalization on the marketing activity of the enterprise was proposed as a confirmation of this impact. Based on the developed “tree of goals,” the dynamics of the digitalization of services were projected and the prospects of development of this sphere of activity were outlined. For testing the proposed methodology, the railway passenger transportation company (JSC “Ukrzaliznytsia”) was chosen as the object of the research. Research methods used in the study include: (1) statistical; (2) SWOT analysis; (3) systematization, comparative, and structural-dynamic analysis; and (4) an expert survey. As a result of revealing the impact of individual elements of digitalization on the level of marketing activity, the number of recommendations regarding the development of digitalization of electronic ticket sales services and their accounting for enterprises dealing with railway passenger transportation were proposed.

We previously demonstrated that smooth muscle (SM) 22α promotes the migration activity in contractile vascular smooth muscle cells (VSMCs). Based on the varied functions exhibited by SM22α in different VSMC phenotypes, we investigated the effect of SM22α on VSMC migration under pathological conditions. The results demonstrated that SM22α overexpression in synthetic VSMCs inhibited platelet-derived growth factor (PDGF)-BB-induced cell lamellipodium formation and migration, which was different from its action in contractile cells. The results indicated two distinct mechanisms underlying inhibition of lamellipodium formation by SM22α, increased actin dynamic stability and decreased Ras activity via interference with interactions between Ras and guanine nucleotide exchange factor. The former inhibited actin cytoskeleton rearrangement in the cell cortex, while the latter significantly disrupted actin nucleation activation of the Arp2/3 complex. Baicalin, a herb-derived flavonoid compound, inhibited VSMC migration via upregulation of SM22α expression in vitro and in vivo. These data suggest that SM22α regulates lamellipodium formation and cell migration in a phenotype-dependent manner in VSMCs, which may be a new therapeutic target for vascular lesion formation.

Plant-extracted triterpenoids belong to a class of bioactive compounds with pleotropic functions, including antioxidant, anti-cancer, and anti-inflammatory effects. In this work, we investigated the anti-inflammatory and anti-oxidative activities of a semisynthetic derivative of 18βH-glycyrrhetinic acid (18βH-GA), soloxolone methyl (methyl 2-cyano-3,12-dioxo-18βH-olean-9(11),1(2)-dien-30-oate, or SM) in vitro on lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages and in vivo in models of acute inflammation: LPS-induced endotoxemia and carrageenan-induced peritonitis. SM used at non-cytotoxic concentrations was found to attenuate the production of reactive oxygen species and nitric oxide (II) and increase the level of reduced glutathione production by LPS-stimulated RAW264.7 cells. Moreover, SM strongly suppressed the phagocytic and migration activity of activated macrophages. These effects were found to be associated with the stimulation of heme oxigenase-1 (HO-1) expression, as well as with the inhibition of nuclear factor-κB (NF-κB) and Akt phosphorylation. Surprisingly, it was found that SM significantly enhanced LPS-induced expression of the pro-inflammatory cytokines interleukin-6 (IL-6), tumour necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) in RAW264.7 cells via activation of the c-Jun/Toll-like receptor 4 (TLR4) signaling axis. In vivo pre-exposure treatment with SM effectively inhibited the development of carrageenan-induced acute inflammation in the peritoneal cavity, but it did not improve LPS-induced inflammation in the endotoxemia model.

The article considers the main stages of formation, the main internal and external factors of development of the Ukrainian community of Meskhetian Turks in the current contexts of socio-political realities of today. Based on the analysis of theoretical sources and applied research of problems, factors, nature and directions of ethnocultural adaptation of Meskhetian Turks in Ukraine, there are the tendencies of growth of social mobility and migration activity, democratization of marital and family relations, diversification of employment, significant changes in social, cultural and educational environment, places of compact residence of Ukrainian Meskhetian Turks. The culturological and sociological study of the problems of sociocultural adaptation and the definition of the content elements and the target direction of the system target regional programs of ethnocultural adaptation of the youth of Meskhetian Turks in Ukraine are actualized. It is justified the need to predict sociocultural trends and timely creation of adequate mechanisms and development the forms and methods of coordination of cultural policy in the field of education in the multicultural environment of the south-eastern regions of Ukraine.

Abstract Background: Posterior capsule opacification (PCO) occurs frequently following surgical treatment of cataracts, representing a common adverse outcome. PCO develops as a result of lens epithelial cell (LEC) transdifferentiation, migration, and proliferation. In the present report we sought to explore the role of microRNA (miR)-19a in this process and to establish the underlying molecular mechanisms. Methods: miR-19a was transduced into HLE-B3 LECs, with microscopy and RT-qPCR used to confirm transfection efficiency. We then employed MTT, wound healing, and transwell assay approaches to monitor the proliferation and migration of these LECs. We further assessed levels of the proteins PTEN, AKT, and phosphorylated AKT (pAKT) via western blotting in WT and miRNA-transfected cells. Results: HLE‑B3 proliferation was markedly enhanced by miR-19a transduction, as well the migration activity of these cells (both P<0.01). Furthermore, overexpressing miR‑19a failed to reduce PTEN expression whereas it did enhance pAKT levels within these LECs (P<0.05). Conclusions: This suggests that miR-19a can enhance LEC proliferation and migratory activity through a mechanism that may be linked with regulating AKT activation and signaling, thus highlighting a potential avenue for therapeutic treatment of PCO patients.

HOXB5 is significantly up-regulated in gastric cancer and its expression is closely correlated with β-catenin expression. HOXB5 binds directly to the CTNNB1 promoter and activates its transcriptional expression, which in turn promotes invasion and migration activity of gastric cancer cells.
