In the present study, we evaluated the hypoglycemic, hypolipidemic and anti-peroxidative effects of yam rice powder (YRP) derived from Tremella aurantialba on streptozotocin (STZ)-induced diabetic mice and diabetic patients. Diabetic mice were treated with ethanol extract of YRP (EEYR) at three doses or YRP for 30 days. Diabetic patients were treated with ethanol extract solution of YRP (ESYR) for 4.5 months. Results indicated that EEYR administration significantly reduced levels of fasting blood glucose (FBG), serum fructosamine, total cholesterol, total triglyceride and malondialdehyde, and increased glucose metabolism velocities and superoxide dismutase level in the serum compared with diabetic controls (p<0.01). Moreover, ESYR could replace chemical drugs to control the FBG and postprandial blood glucose levels of diabetic patients. Our results suggested that YRP could enhance hypoglycemic, hypolipidemic and antioxidant properties in STZ-induced diabetic mice and diabetic patients. Collectively, YRP might be used as food to prevent or manage diabetes.

ABSTRACT This study aims to determine the antidiabetic activity of papaya flower ethanol extract (Carica papaya L.) against male white rats (Rattus norvegicus) induced by alloxan. This research is experimental. Fifteen rats were divided into 5 treatment groups, each group consisted of 3 rats. The first group was the negative control group given Aquades, the second group was positive control given Glibenklamid, the three groups were 200 mg dose variation groups, four groups were 400 mg dose variations, and the five groups were 800 mg dose variations. Previously, rats were examined fasting blood glucose levels, then mice were induced by an alloxan dose of 120 mg / kgBW intraperitoneally. On the 3rd day blood glucose levels were examined and then treated according to groups for 7 days, measurement of blood glucose levels after the treatment was carried out once every 3 days namely day 3, day 7, and day 10. Data obtained were then analyzed statistically using SPSS , including normality test (Shapiro-Wilk), homogeneity test (Levene), ANOVA test (One way). The results of this study indicate that Papaya Flower (Carica papaya L.) Terhadap Tikus Putih Jantan (Rattus norvegicus) Yang Diinduksi Aloksan. Penelitian ini bersifat eksperimental. Sebanyak 15 ekor tikus dibagi ke dalam 5 kelompok perlakuan masing- masing kelompok terdiri dari 3 ekor tikus. Kelompok pertama kelompok Kontrol negatif yang diberikan Aquades, Kelompok kedua Kontrol Positif yang diberikan Glibenklamid, kelompok tiga kelompok variasi dosis 200 mg, kelompok empat kelompok variasi dosis 400 mg, dan kelompok lima kelompok variasi dosis 800 mg. Sebelumnya tikus dilakukan pemeriksaan kadar glukosa darah puasa, selanjutnya tikus diinduksi Aloksan dosis 120 mg/kgBB secara intraperitoneal. Pada hari ke 3 diperiksa kadar Glukosa darah kemudian diberikan perlakuan sesuai kelompok selama 7 hari, pengukuran kadar glukosa darah setelah perlakuan dilakukan 3 hari sekali yaitu hari ke 3, hari ke 7, dan hari ke 10. Data yang diperoleh kemudian dianalisis secara statistik menggunakan SPSS, meliputi uji normalitas (Shapiro-Wilk), uji homogenitas (Levene), uji ANOVA (One way). Hasil penelitian ini menunjukkan bahwa Ekstrak Etanol Bunga Pepaya (Carica papaya L.)  dapat memberikan efek penurunan kadar glukosa darah yang paling baik yaitu pada dosis 800 mg. Kata kunci : Antidiabetik, Ekstrak Bunga pepaya (Carica papaya L.), Diabettes Mellitus, TikusPutih Jantan (Rattus norvegicus), Aloksan.

This Research was conducted to determine the effect of ethanol extract from red chilli (Capsicum annuum L)in lowering blood glucose levels of hyperglycemic male white mices. This study used 30 malewhitemices and divided into 6 groups: negative control group, the positive control group, the treatmentgroup withdosage of 200 mg / kgweight, 400 mg / kgwieght, 600 mg / kg weight and a comparison group with glibenclamide with dosage of 5 mg / kgweightadministered orally for 21 days. The level of fasting blood glucose was checked 6 days after dexamethasone induced, and after the ethanol extract of red chilies on day 7th, 14th, and 21st. The data was analyzed statistically with one –way and two-way Anova by usingSPSS16 program and proceed with the test Duncan to look the significant difference between treatments. The results showed that the ethanol extract from red chilies with a dosage of 200mg / kgweight, 400mg / kgweight, 600 mg / kgweight showed the effect in lowering blood sugar levels in male white mices hyperglycemia significantly (p <0.05). The duration ethanol extract of red chili affected blood glucose levels. The most effective duration in lowering blood glucose levels is the administration of a preparation within 21 days.

1. Male, 4–5-week-old, genetically diabetic mice (C57BL/KsJ db/db) and non-diabetic heterozygote litter-mates (C57BL/KsJ db/+)were fed on a diet containing 1 mg zinc/kg (low-Zn groups) or 54 mg Zn/kg (control groups) for 27 d. Food intakes and body-weight gain were recorded regularly. On day 28, after an overnight fast, animals were killed and blood glucose and insulin concentrations, liver glycogen, and femur and pancreatic Zn concentrations were determined.2. The consumption of the low-Zn diet had only a minimal effect on the Zn status of the mice as indicated by growth rate, food intake and femur and pancreatic Zn concentrations. In fact, diabetic mice fed on the low-Zn diet had a higher total food intake than those fed on the control diet. The low-Zn diabetic mice had higher fasting blood glucose and liver glycogen levels than their control counterparts. Fasting blood insulin concentration was unaffected by dietary regimen.3. A second experiment was performed in which the rate of loss of 65Zn, injected subcutaneously, was measured by whole-body counting in the two mouse genotypes over a 28 d period, from 4 to 5 weeks of age. The influence of feeding low-Zn or control diets was also examined. At the end of the study femur and pancreatic Zn and non-fasting blood glucose levels were determined.4. All mice fed on the low-Zn diet showed a marked reduction in whole-body 65Zn loss compared with those animals fed on the control diet. In the low-Zn groups, the loss of 65Zn from the diabetic mice was significantly greater than that from heterozygote mice. This difference was not observed in the control groups. Blood glucose levels were elevated in the low-Zn groups. Possible reasons for these observations are discussed.5. The present study demonstrates an adverse effect of reduced dietary Zn intake on glucose utilization in the genetically diabetic mouse, which occurred before any significant tissue Zn depletion became apparent.

Objectives: To determine the frequency of impaired fasting glucose in first degree relatives of people with Type-II diabetes and its association with BMI. Methods: This cross-sectional study was conducted in Diabetic clinic of Shaikh Zayed Hospital, Lahore from July to December 2017. Individuals aged ≥35 years, first degree relatives of people with Type-II diabetes, were selected and their fasting blood glucose levels were checked twice a week apart. Study participants were divided into 3 groups. Group-I were those with normal fasting blood glucose (FBS: <100mg/dl), Group-II were those with impaired fasting glucose (100-125mg/dl), considered as high risk and Group-III included those who turned out to be having frank diabetes (FBS: ≥126mg/dl). Exclusion criteria were known diabetes and pregnancy. Proportions of impaired fasting glucose levels versus BMI were compared using Chi-square test. Significance was considered at P <0.001. Results: A total of hundred subjects were included in the study with the mean age of 44.27 years. Sixty percent participants had normal FBS, 31% showed impaired FBS and 09% had frank diabetes (P <0.001). Significant association was found between impaired fasting glucose and BMI, as with increasing BMI the frequency of impaired fasting glucose increases. Abstract Purpose Although the effects of cadmium (Cd) on the development of diabetes have been extensively investigated, the relationship between Cd exposure and the severity of established diabetes is unclear. Herein, we investigate the effects of long-term exposure to Cd in a streptozotocin-induced mouse model of type 2 diabetes and the underlying mechanism. Methods C57BL/6 Mice were divided into the following four groups: 1) control group; 2) Cd-exposed group; 3) diabetic group; 4) Cd-exposed diabetic group. Cd exposure was established by the administration of 155 ppm CdCl2 in drinking water. After 25 weeks of treatment, serum fasting glucose and insulin were measured. Meanwhile, the liver and pancreas specimens were sectioned and stained with Hematoxylin and eosin. Gluconeogenesis, glycolysis, lactate concentration and fibrosis in liver were evaluated. Results Clinical signs attributable to diabetes were more apparent in Cd-exposed diabetic mice. Interestingly, Cd exposure significantly decreased fasting blood glucose levels in diabetic group. We further demonstrated that the glycolysis related hepatic enzymes, pyruvate kinase M2 (PKM-2) and lactic dehydrogenase A (LDHA) were both increased, while the gluconeogenesis related hepatic enzymes, phosphoenolpyruvate-1 (PCK-1) and glucose-6-phosphatase (G6Pase) were both decreased in Cd exposed diabetic mice, indicating that Cd increased glycolysis and inhibited gluconeogenesis in diabetic model. Moreover, lactate accumulation was noted accompanied by the increased inflammation and fibrosis in the livers of diabetic mice following Cd exposure. Conclusions Cd exposure disturbed glucose metabolism and exacerbated diabetes, providing a biological relevance that DM patients are at greater risk when exposed to Cd.

Xeroderma is a frequent complication in diabetic patients. In this study, we investigated the mechanism underlying the onset of diabetic xeroderma, focusing on aquaporin-3 (AQP3), which plays an important role in water transport in the skin. Dermal water content in diabetic mice was significantly lower than that in control mice. The expression level of AQP3 in the skin was significantly lower in diabetic mice than in control mice. One week after streptozotocin (STZ) treatment, despite their increased blood glucose levels, mice showed no changes in the expression levels of AQP3, Bmal1, Clock, and D site-binding protein (Dbp) in the skin and 8-hydroxydeoxyguanosine (8-OHdG) in the urine. In contrast, two weeks after STZ treatment, mice showed increases in the blood glucose level, decreases in AQP3, Bmal1, Clock, and Dbp levels, and increases in the urinary levels of 8-OHdG. The results of this study suggest that skin AQP3 expression decreases in diabetes, which may limit water transport from the vessel side to the corneum side, causing dry skin. In addition, in diabetic mice, increased oxidative stress triggered decreases in the expression levels of Bmal1 and Clock in the skin, thereby inhibiting the transcription of Aqp3 by Dbp, which resulted in decreased AQP3 expression.

Abstract Dexamethasone is the corticosteroid most commonly used for the management of vasogenic edema in patients with brain tumors. The occurrence of hyperglycemia following corticosteroid administration is well known to physicians and health care givers. The purpose of the study is to investigate late onset of hyperglycemia and associated factors in patients with brain tumors. The inclusion criteria are as follows; 1) brain tumor patients underwent craniotomy; 2) steroid administered for at least 3 days longer perioperatively or postoperatively; 3) serum glucose levels examined for at least 3 months postoperatively The exclusion criteria are as follows; 1) steroid administered for less than 3 days; 2) patients diagnosed with type 1 diabetes, renal failure, hepatic failure or autoimmune diseases; 3) other medical illness treated with corticosteroid. A total of 101 patients were enrolled for the analysis. Fasting blood glucose of diabetes patients and non-diabetes patients was 160±93 mg/dl and 114±43 mg/dl, respectively. There was no difference of type of brain tumors, body mass index, hypertension, smoking history, hyperlipidemia, GOT, GPT, ESR, CRP, and Cr between diabetes patients and non-diabetes patients. The peak of Fasting blood glucose was noted on day 1 postoperatively in non-diabetes patients. Two peak of Fasting glucose levels were found on day 1 and 6 postoperatively in diabetes patients. Late hyperglycemia was determined as in patients with fasting blood glucose of >125 mg/dl during 3~6 months postoperatively. Fifteen patients out of 86 non-diabetes patients developed late hyperglycemia. There were significant differences of duration and total dose of dexamethasone administration between late hyperglycemia patients and non-late hyperglycemia patients. Steroid-induced hyperglycemia can develop late postoperatively even in non-diabetic patients with brain tumor treated with dexamethasone. Fasting blood glucose during the first week postoperatively can predict late hyperglycemia. Duration and total dosage of dexamethasone administered are associated with late hyperglycemia.

Background: Diabetes mellitus is a heterogeneous group of diseases in the form of disorders in the body's metabolism clinically. Peperomia pellucida herbs have phytochemical containing which is antidiabetic potential development. Objectives: This study was conducted to compare the antidiabetic activity of ethanol extract and n-hexane fraction of Peperomia pellucida. Material and Methods: This research was conducted by make diabetic mice with 50 mg/kg.bw of streptozotocin induction, which was then treated with ethanol extract and n-hexane fraction of Peperomia pellucida with doses 250 mg/kgbw for 7 days.  Results: The results showed that the ethanol extract and n-hexane fraction of Peperomia pellucida reduced blood glucose levels in diabetic mice due to streptozotocin induction. The n-hexane fraction of Peperomia pellucida can lower blood glucose levels as much 244.00 ± 18.99 mg/dL better than the ethanol extract, which is 99.50 ± 28.17 mg/dL. Conclusions: Peperomia pellucida herb has the potential to be developed as an antidiabetic agent.
