The antioxidant defense system controls and inhibits all stages of free radical formation reactions, which start from their initiation and end with the formation of hydroperoxides and malonic dialdehyde. Disorders of the body's antioxidant defense system can be caused by both endogenous and exogenous factors and can lead to the oxidative stress development. For the purpose of the negative impact minimizing and correction of the existing disorders, both natural and synthetic origin antioxidants are used. Preference is given to antioxidants of natural origin, due to their mild action and minimal side effects. Shiitake mushrooms are known in China and Japan as a product that has nutritional and medicinal value, for thousands of years. They contain an extremely rich complex of various biologically active substances that can have antioxidant, hepatoprotective effect and influence on the liver diseases pathogenesis. The aim of the work was to investigate the effect of shiitake mushrooms thick extract on the activity of free radical processes and indicators of the antioxidant system under the conditions of paracetamol hepatitis in rats. The experimental work was being conducted on the white male rats, which were divided into 10 groups of 6 animals each. Acute hepatitis was simulated by intragastric administration of paracetamol in a dose of 1 250 mg/kg 1 time per day (for 2 days) as a suspension in 2% starch gel solution. We investigated the effect of shiitake mushrooms thick extract on the induced pathology, which was administered intragastrically 2 hours before the paracetamol introduction and daily after the lesion in a dose of 150 mg/kg of the rat’s body weight. Silibor was selected as the comparison drug, which was administered according to the same scheme like the investigated extract in a dose of 20 mg/kg of the animal’s body weight. Euthanasia was conducted on the 3rd, 7th and 10th day after the onset of the lesion. Liver homogenate and animal serum were examined. The effectiveness of shiitake mushrooms thick extract was evaluated under the conditions of the caused pathology by superoxide dismutase and catalase activity, as well as by the content of TBA-active and proteins oxidative modification products. An increasing of the TBA-active products content and the products of proteins oxidative modification in the serum and liver of animals after the toxic paracetamol exposure testifies to the liver damage and the development of acute hepatitis in rats. Decreased activity of antioxidant enzymes such as catalase and superoxide dismutase was also noted. An investigation, which was conducted by us, allowed us to reveal a positive effect of shiitake mushrooms thick extract on the activity of lipoperoxidation and oxidative modification of proteins under the conditions of white rats paracetamol lesion.

Introduction. Wound healing according to modern concepts is a coordinated process that passes through certain stages with the participation of different cells and products of its vital functions that regulate the healing process. It is well known that in patients with diabetes there is a violation of the process of wound healing, although until now, all the features of diabetes underlying such an inclination have not been fully understood.The aim of the study – to identify the features of oxidative stress on the background of diabetes mellitus in the rat's homogenate when different methods of closing wounds had been used. Research Methods. The experiment was conducted on 60 male rats weighing 280–320 g, which pre-simulated diabetes mellitus and surgical wounds. To detect the activation of free radical oxidation processes, the content of active forms of oxygen (AFC), lipid hydroperoxides (HPL), diene conjugates (DK) and trienic conjugates (TC) and oxidation modifications of proteins (OMP370 and OMP430). To study the antioxidant defense system, activity of superoxide dismutase was determined. Also, the activity of catalase and sulfhydryl groups (SH-groups) was determined.Results and Discussion. The research conducted by us showed that during the wounded process, on the background of diabetes, the activity of free radical oxidation processes increased. However, in the animal skin homogenates, which are labeled "Dermabond", all indices are significantly lower than in the group of animals that have been sewed over the course of all research periods. In the early stages of scar formation (3 days), an increase in the activity of the antioxidant defense system in both groups was observed. At the same time, in the experimental group of animals, which imposed the nodal seams, the activity of SOD and catalase significantly exceeded the skin index of animals, which applied glue and made (199.81±7.59) UM and (175.02±8.31) cat/kg, respectively.Conclusion. The use of skin glue reduces the intensity of the course of free radical oxidation in the cells of the scar tissue of the postoperative wound in conditions of diabetes mellitus in comparison with the imposition of nodal sutures on it: in the skin of animals, which is applied to the "Dermabond" glue, all indicators are significantly lower than in the group of animals, which imposed seam joints, throughout all research periods.

The goal of our study was to examine the effect of feed supplement Humilid on the antioxidant defense system, free radical processes, and histomorphological changes in the tissues of rats affected by Chromium (VI). Rats were divided into 4 groups. Animals of groups D2 and D3 received Humilid in a dose of 2 ml per kg of body weight for 28 days. From the 14th day of the experiment, rats of groups D1 and D2 were injected by K2Cr2O7 in a dose of 2 mg Cr per kg of body weight daily for 14 days. Control animals were injected with saline solution for 14 days. Results showed that Cr (VI) causes oxidative stress in rat tissues, accompanied by an increase in superoxide dismutase activity, lipid peroxidation products, and a decrease in glutathione peroxidase and glutathione reductase activity. Chromium also caused histopathological changes in the liver and kidney of rats. Watering of rats affected by Cr (VI) with Humilid leads to decreased oxidative processes, activation of the antioxidant defense system, and reduced pathological effects on animal tissues. Our results indicate the positive influence of Humilid on the rat’s organism, inhibiting free radical processes and exhibiting antioxidant, hepatoprotective, and adaptive properties.

The aim. To study the effect of a dry extract from reishi mushrooms on the activity of lipoperoxidation and oxidative modification of proteins under the conditions of a simulated paracetamol hepatitis in rats. Materials and methods. The study was performed on white male rats. The animals were divided into 10 groups, each included 6 animals. Acute hepatitis was simulated by the intragastric administration of paracetamol in the dose of 1250 mg/kg once per day (for 2 days). Correction of the pathology induced was performed with a dry extract of reishi mushrooms in the dose of 100 mg/kg of the body weight. The reference drug “Silybor” was administered in the dose of 20 mg/kg of the animal body weight. On Day 3, 7 and 10 from the beginning of the lesion, rats were euthanized using sodium barbamyl. The liver homogenate and blood serum were used for the studies. The activity of free radical oxidation processes under the conditions of acute toxic hepatitis and after the introduction of corrective factors was assessed by superoxide dismutase, catalase activity, the content of TBA-AP and OMP products. Results and discussion. The development of acute paracetamol hepatitis in rats and damage of hepatocyte membranes are indicated by an increase in the content of TBA-active products, products of oxidative modification of neutral and basic proteins in the serum and liver of animals. Simultaneously, a decrease in the activity of catalase and superoxide dismutase was observed. After correction of the pathology induced with a dry extract of reishi mushrooms a significant increase in the activity of antioxidant enzymes, a decrease in the content of lipid peroxidation products and oxidative modification of proteins in the serum and liver of the affected animals were observed. Conclusions. It has been experimentally proven that the use of dry extract of Reishi mushrooms in paracetamol hepatitis in rats caused a significant decrease in catalase and superoxide dismutase activity, a decrease in TBA-AP, neutral and basic 2,4-DNPH in the serum and liver of animals. The results of the research indicate an effective impact of reishi mushrooms dry extract on the normalization of lipoperoxidation, oxidative modification of proteins and antioxidant protection

Aim: Cancer is one of the leading causes of death in the world. The aim of this research was to study the indices of pro- and antioxidant systems in rats with dimethylhydrazine (DMH)-induced colon carcinogenesis on the background of the enterosorbent AUT-M use. Materials and methods: The study was performed on 70 white male rats weighing 200–250 g. Adenocarcinoma of the colon was simulated by subcutaneous injection of the DMH (Sigma-Aldrich Chemie, Japan) at a dose of 7.2 mg/kg once a week during 7 months. Enterosorbent AUT-M was administered intragastrically daily for 21 days after simulation of carcinogenesis at a dose of 1 ml of suspension per 100 g of animal body weight. The state of the pro- and antioxidant systems was studied by the content of oxidative modification of proteins products (OMP), the activity of superoxide dismutase (SOD), catalase (CAT), contents of ceruloplasmin (CP) and reduced glutathione (GSH). Results: It was found that DMH-induced colon carcinogenesis in rats is accompanied by disorders in the antioxidant defense system and activation of free radical oxidation processes. Enterosorbent AUT-M provides a significant reduction in the content of OMP370 and OMP430 in both blood serum and liver homogenate of rats. Moreover, the use of enterosorbent AUT-M demonstrated a significant increase in the activity of SOD, CAT, content of GSH and a decrease in CP content in investigated tissues. Conclusion: The use of enterosorbent AUT-М demonstrated prominent potential suppression for oxidative stress and positive effect on antioxidant defense system in rats with DMH-induced colon carcinogenesis.

Background. Diabetes mellitus is accompanied by oxidative-nitrative stress, which is caused both by an increased generation of the Reactive Oxygen Species and the Reactive Nitrogen Species under the conditions of this pathology and by the disorders of the antioxidant defense system, especially its enzymatic part. This leads alterations in the morpho-functional state of cells, organs, and the whole organism. Free radicals are involved in the destruction of pancreatic cells in type 1 diabetes mellitus, which leads to even greater inhibition of insulin secretion, worsening of the course of the disease and occurrence of diabetic complications. The complications of diabetes mellitus include often diagnosed cardiovascular diseases. Therefore, the use of treatments characterized not only by hypoglycemic properties for normalization of blood sugar level in diabetes mellitus, but also by antioxidant properties for normalization of oxidative/antioxidant balance of the organism in the studied pathology can be promising. Scientists pay a great attention to the study of substances with such properties, especially natural phenolic compounds of grape, which are also characterized by immunomodulatory properties. Materials and Methods. The study was performed on cardiac tissues of control rats, control animals treated with wine polyphenolic complex concentrate, animals with streptozotocin-induced diabetes mellitus and animals with experimental diabetes mellitus treated with a concentrate of wine polyphenolic complex. The activity of catalase, superoxide dismutase, glutathione peroxidase and glutathione reductase were detected to examine the corrective effect of the concentrate of red wine natural polyphenolic comp­lex on the state of the enzymatic part of the antioxidant defense system. Results. The results have shown the normalization of activity of catalase, superoxide dismutase, and changes in the activity of enzymes of glutathione cycle after oral administration of polyphenolic complex concentrate for 14 days to rats with streptozo­tocin-induced diabetes mellitus. Conclusions. The results confirm a hypothesis about the antioxidant effect of the studied concentrate and the ability of natural polyphenolic complexes to serve as the basis for new drugs for treatment of diabetes-induced disorders.

Background. Despite numerous studies, the pathogenesis of gastroesophageal reflux disease remains unclear. Aim of research: assessment the activity of antioxidant defense system in young patients with GERD based on expression of biomarker associated with mitochondrial function. Material and methods. The study included 45 patients with gastroesophageal reflux disease. The examined contingent was presented by students age from 18 to 25 years. 20 healthy persons were included as control group. Levels of manganese superoxide dismutase were determined in blood serum of study persons with enzyme immunoassays (ELISA, Elabscience, USA). Statistical data processing by the Statistica Basic Academic 13 for Windows En local was made. Results. Gastroesophageal reflux disease in young patients is characterized by significantly increasing of manganese superoxide dismutase as compare to control group (7.1700 ng/ml vs 4.4720 ng/ml respectively, p<0.01). Presence of erosion in esophagus mucous doesn't accompanied by significant changes of evaluated parameter as compare with non-erosion form of disease in patients. Conclusion. The elevation in young patients with GERD the biomarker of mitohondrial antioxidant defense system we may speculate as adaptive response contributing to non-specific citoprotection. Taking to account the publishing facts about dual role of manganese superoxide dismutase it is necessary to monitoring antioxidant enzyme in patients with gastroesophageal reflux disease for prediction of possible complications and outcome.

AbstractThis study examined the effects of chronic restraint stress (CRS: 2 hours × 14 days) on gene expression of three antioxidant enzymes, copper, zinc superoxide dismutase (SOD 1), manganese superoxide dismutase (SOD 2) and catalase (CAT) in the rat hippocampus. Also, we examined changes in the activities of SOD 1, SOD 2 and CAT in the hippocampus of chronically stressed rats. Investigated parameters were quantifi ed by using real-time RT-PCR, Western blot analysis and assay of enzymatic activity. We found that CRS did not change mRNA and protein levels of SOD 1 and CAT, but increased mRNA and protein levels of SOD 2. However, CRS treatment increased the enzyme activities of SOD 1, SOD 2 and CAT. Our fi ndings indicate that the increased activity of antioxidant enzymes (SOD 1, SOD 2 and CAT) in the hippocampus may be an important adaptive phenomenon of the antioxidant defense system in chronically stressed rats.


The literature review summarizes the data on the classification and characteristics of the antioxidant defense system. This model combines a number of different substances. Each of the components of the antioxidant system acts in close connection with its other structural elements, harmoniously complements, and in many cases – enhances the action of each other. The functional basis of the system of antioxidant protection is formed by the glutathione system, the constituent elements of which are actually glutathione and enzymes that catalyze the reactions of its reverse transformation (oxidation ↔ reduction). Most researchers conventionally divide the system of antioxidant protection into enzymatic and non-enzymatic. The enzyme links of the antioxidant defense system include: catalase, superoxide dismutase, glutathione peroxidase, glutathione reductase, glutathione transferase and other enzymes. The non-enzymatic system includes fat-soluble vitamins A, E and K, water-soluble vitamins C and PP, biogenic amines, glutathione, carotenoids, ubiquinone, sterols. Both enzymatic and non-enzymatic antioxidant defense systems are present in the bloodstream. The activity of the enzymatic antioxidant system is very well regulated and depends on age, physiological condition, hormone dynamics, intensity of antioxidant enzyme synthesis, pH, presence of coenzymes, inhibitors, activators and other factors. The non-enzymatic part of the antioxidant system does not require as many regulators as the chemical itself - the antioxidant - reacts chemically with the radical. Only the reaction rate can change. The lungs are directly exposed to oxygen and oxidants of polluted air, they are most sensitive to oxidative damage, so they have an increased possibility of free radical reactions. Protection against the damaging effects of reactive oxygen species and free radicals is provided by anti-radical protective systems, especially the glutathione system and its enzymes. Numerous papers have obtained clinical and experimental data on the important role of free radical oxidation of lipids (FROL) and antioxidant systems (AOS) protection in the development of many diseases, including pulmonary tuberculosis (TB). The review presents modern views on the state of the system of lipid peroxidation (LPO) and antioxidant protection in TB. The main topic of the review is information on the generation of free radical compounds by different populations of leukocytes with further enhancement of LPO and secondary deepening of functional disorders. The article substantiates the feasibility of identifying LPO products as non-specific markers of aseptic inflammation in TB and the need to develop new generations of antioxidants. One of the universal mechanisms of damage to cell membranes is LPO, the excessive activation of which is normally prevented by factors of the antioxidant defense system. Membrane-bound enzymes involved in the formation of LPO products include lipoxy and cyclooxygenases. Lipoperoxidation processes change the structure and phospholipid composition of cell membranes, which negatively affects the cellular immune response due to damage to the mechanisms of information transfer from extracellular regulators to intracellular effector systems.

Background. The problem of gastric cancer remains unresolved throughout the world, while chronic atrophic gastritis (CAG) increases the likelihood of its development by 15 times. In the Russian Federation, the incidence of gastric cancer (GC) is among the highest, with it prevailing among males. One of the leading mechanisms in molecular pathology of membranes is lipid peroxidation (LPO). The severity of oxidative membrane damage depends on concomitant diseases, contributing to emergence and progression of pathological processes and development of cancer. Currently, the problem of LPO is unsolved in biological systems.The aim of this study was to investigate the state of LPO and antioxidant defense system in CAG and GC. Materials and methods. The parameters were studied in 45 patients with CAG and 50 patients with GC. The control group included 50 practically healthy volunteers without gastrointestinal complaints, who did not have changes in the gastric mucosa according to the fibroesophagogastroduodenoscopy (FEGDS) findings.Results. In patients with CAG, an increase in malondialdehyde, superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase was found in the blood plasma compared with the control group. In patients with CAG, lipid peroxidation was activated, and the malondialdehyde level increased by 3.5 times relative to normal values. At the same time, the body fought against oxidative stress by increasing the activity of antioxidant enzymes, such as superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase. All patients with GC showed pronounced oxidative stress in the blood plasma in the form of a 45-fold increase in malondialdehyde. The activity of the main antioxidant enzyme superoxide dismutase was reduced in GC. Catalase was activated, which indicated pronounced oxidative stress, significant damage to blood vessels, and massive cell death. Glutathione-related enzymes (glutathione S-transferase and glutathione peroxidase) and the antioxidant protein ceruloplasmin were activated, which also indicated significant oxidative stress and severe intoxication in patients with GC.Conclusion. Depending on the stage and type of cancer, an in-depth study of lipid peroxidation and factors of the antioxidant defense system can be used to correct therapy and prevent cancer and can serve as markers of progression and prognosis in gastric cancer.

Purpose. The aim of the work was to study certain morpho-physiological parameters of carp as a result of additional introduction of a mycotoxin sorbent to a feed with signs of spoilage. Methodology. During 2019–2020, two experiments were conducted in the laboratory. The first one included 7 groups-analogues of age-1+ Nyvky carp, 20 fish in each group, with an average weight of 22.5 g. The control group of fish was fed for 24 days with a compound feed with signs of spoilage, «Mykosorb» was additionally added to the diet of the experimental groups in the amounts of 0.05, 0.1; 0.15; 0.2; 0.25 and 0.4%, respectively. The second group included 4 groups of age-1+ fish, 12 fish in each group, with an average weight of 20.0 g. The diet of the control group was identical to that of the first experiment, and the experimental groups with the addition of 0.05, 0.075 and 0.1% of «Mycosorb», respectively. During the study, optimal conditions for keeping fish were provided. Fish after were measured and weighed at the end of the experiment and their weight gains were analyzed according to generally accepted methods in fish farming. The activity of antioxidant enzymes and the content of lipid peroxidation products depending on the composition of the diet were studied. Findings. The studies used compound feed with signs of spoilage as a result of violation of their storage technology, which identified three groups of toxins: Aflatoxin B1, Zearalenone and Deoxynivalenol. As a result of the first stage of the study, the rates of application of mycotoxin sorbent into the diet of carp at amounts of 0.05 - 0.2% were determined. At the same time, there was no clear proportional pattern of changes in the average weight of fish depending on the amount of introduction of «Mycosorb», the value increased in all experimental groups compared to the control by 0.4 – 15.2%. Muscles of age-1+ fish were found to have higher (p<0,001) catalase activity after application of 0.15% of the test drug by 1.4 times, 0.2% — by 46%, 0.25% — by 69 and 0.4% — by 79%. Superoxide dismutase activity also increased, except for the groups of 0.2, 0.25 and 0.4% of «Mycosorb». There was a tendency of 21.0 and 22.6% decrease in the content of diene conjugates as a result of feeding 0.05 and 0.1% sorbent and some increase as a result of feeding 0.15; 0.2; 0.25 and 0.4% additives. Studies of the next stage showed that addition of «Mykosorb» at the rate of 0.05, 0.075 and 0.1% to the feed, the average weight of age-1+ carp in the experimental groups increased by 45.4; 37.2 and 30.3%, the condition factor — by 41.9; 40.8 and 42.7%, the relative growth rate is 4.3; 5.9 and 4.6 times, respectively. The fish body height to length ratio was lower by 10.3, 7.0 and 7.4% relative to the control group. Addition of 0.05, 0.075 and 0.1% of «Mycosorb» increased resistance of fish due to an increase in the hepatopancreatic activity of antioxidant enzymes: superoxide dismutase by 28.3, 30.5 and 5.0% and catalase by 56.5 (p<0.001), 55.5 (p<0.001) and 46.3% (p<0.05). At the same time, addition of 0.05 and 0.1% of «Mykosorb» increased the content of diene conjugates by 11.3 (p<0.01) and 12.3% (p<0.01), TBC-active products by 19.7 (p<0.05) and 13.2%. Originality. For the first time, the feasibility of addition of the mycotoxin sorbent «Mykosorb» into the feed with signs of spoilage was studied. The effect of feeding age-1+ carp with diets of various compositions on some morpho-physiological parameters of carp in the conditions of model tests was carried out. Practical value. Additional weigh gains and improvements of the antioxidant defense system of the carp organism were observed. The use of «Mykosorb» in carp feeding allows reducing the negative impact of low quality feed on fish body.
