The objective of the study was to study the state of oxidative-antioxidant homeostasis under the conditions of the formation of experimental fluoride intoxication.24 white rats of the Wistar population were daily injected with sodium fluoride solution (20 mg/kg of animal weight). The continuation of intragastric xenobiotic intake was 1.5 months. The control group was monitored for 2 weeks toanalyse their condition. After 2 weeks, and The study program was conducted after 2 weeks when healthy mature (3 months) males weighing 180-200 grams were selected . At the end of the experiment, the control and experimental groups were clogged by decapitation. Evaluation of oxidative-antioxidant homeostasis was doneon the basis of the following indicators: the intensity of biochemiluminescence (BCL) of homogenates of internal organs and blood serum, the blood levels of SH-groups, glutathione, haptoglobin, diene conjugates (DC), malondialdehyde (MDA), cysteine, catalase activity, peroxidase, ceruloplasmine, glutathione peroxidase, superoxide dismutase. Vitamin C was determined in the adrenal glands. Analyzing the effect of fluoride intoxication on the state of oxidative and antioxidant system, it was shown that stimulation of free radical lipid peroxidation, the accumulation of peroxides, hydroperoxides, free radicals and inhibition of the antioxidant system are characteristic in this process. The obtained results confirm that all these mechanisms underlie the formation of structural and metabolic mechanisms of the pathogenesis of fluoride intoxication.

We calculated studied the state of the hormonal status in experimental animals under the influence of sodium fluoride in 45 adult rats (males and females) of the Wistar population, which were orally administered a solution of sodium fluoride at a rate of 20 mg / kg of weight daily in the morning on an empty stomach, under the model conditions of intoxication. The duration of the subacute experiment was 1.5 months, after which the animals were euthanized by decapitation. Comprehensive assessment of the hormonal status in white rats was carried out by radioisotope methods in the serum. The results showed reduced levels of folliculotropin and progesterone, increased levels of thyroxine, adrenocorticotropin, glucagon, triiodothyronine, thyrotropin and calcitonin, while insulin, somatostatin and serum glucose levels decreased. There was no change in the dynamics of the content of sex hormones - luteotropin, prolactin, testosterone compared with the control (P> 0.05). Furthermore, in the blood serum of rats, there was an increase in the levels of prostaglandin PGE2, prostacyclin (6 keto-PGF1a), leukotriene B4 and a decrease in the concentrations of prostaglandins PGE1, PGF2a and leukotriene C4. At all levels of the study of the endocrine system, deep shifts in the functional activity of the hypothalamus-pituitary-cortical substance of the adrenal glands, thyroid gland, and sympatho-adrenal structures have been identified. Analysis of the detected changes in the hormonal status allows judging the nonspecific reaction of the body to fluoride intoxication and reflects the state of the protective-adaptive mechanisms, in which the hypothalamus, the thyroid gland and the adrenal glands play a large role. Changes in the dynamics of concentrations of hormones and histohormones reflected a significant stress of the protective-adaptive mechanisms, which, in terms of structural and functional units, led to disruption of metabolic processes, including those associated with the development of fluoride intoxication. Disruption of the balance of hormones and functioning of the endocrine organs and systems entails profound changes in the metabolic processes and the immunobiological reactivity of the organism, weakening the action of the protective-adaptive mechanisms in maintaining the homeostatic function.

The study was performed at the Department of Clinical Pathophysiology, Topographical Anatomy and Operative Surgery at Kharkiv Medical Academy of Postgraduate Education as a part of research project "Radiotoxins' pathochemical mechanisms and methods of early diagnostics and correction", state registration No. 0117U000589. We studied the subtoxic effect of small doses of sodium fluoride on the activity of microsomal hepatocytes on 30 Wistar rats' populations in subacute experiment. The intensity of lipid peroxidation (LPO) in liver of rats which were administered sodium fluoride orally for a long time at doses of 1/10 and 1/100 LD50, was evaluated by the content of its molecular products - diene conjugates (DC), and MDA-reagents Schiff bases. We found that oral administration of sodium fluoride to rats at doses of 1/10 and 1/100 LD50 promotes a statistically significant increase (r≤0,002) relative to the control group of animals in DC content during the entire period of observation. At a dose 1/10 LD50 we observed the most significant increase in this indicator on the 10th day of the experiment - at 265%, and at a dose 1/100 LD50 – on the 20th day an average of 234%. In rats’ liver by the action of subtoxical dose of sodium fluoride at a dose of 1/10 LD50, starting from the 20th day, we detected a gradual increase (r≤0.001) of TBA-reagents relative to control - for 27, 41, 78, 133%. Secondary end products and lipid peroxidation, which are defined under the long-term of sodium fluoride, somehow contribute to the disruption of the microstructure of hepatocytes membranes, their permeability, reduce their division, regeneration and inhibition of mitochondrial respiratory chain enzymes and microsomal monooxygenase system.

In the present study the cardioprotective effects of curcumin, a herbal polyphenolic compound, against sodium fluoride (NaF)-induced toxicity in rat heart was evaluated. Fifty rats were divided into five experimental groups containing 10 rats each. Group I received standard water and diet and was used as a normal group; groups II and III were pretreated with curcumin intraperitoneally for 7 days prior to NaF intoxication. Group IV was pretreated with vitamin C, a standard antioxidant, intraperitoneally for 7 days prior to NaF intoxication and used as a positive control group. The animals in group V were intoxicated with NaF for the same time and used as a control group. There was a significant increase in lipid peroxidation along with a decrease in superoxide dismutase activity in the homogenates of tissues of the NaF-treated animals. Curcumin pretreatment in animals prior to fluoride intoxication normalized the levels of biochemical parameters measured.

Objective: to assess the state of the pro-oxidant/antioxidant system in children with hereditary spherocytosis (HS) depending on its severity.Material and methods. The study involved 44 HS patients at the age from 1 to 17 who were divided into 2 groups depending on the disease severity: mild course (n = 24) and moderate or severe course (n = 20). The control group included 23 practically healthy children who were comparable with the main group by gender and age. The state of the pro-oxidant/antioxidant balance of blood plasma was assessed by the method of luminol-dependent chemiluminescence (LDCL) with the determination of the maximum luminescence intensity (Imax, %) and the light sum of chemiluminescence (S, %). The activity of superoxide dismutase (SOD) and catalase was determined in the erythrocytes of the examined children.Results. On average, the parameters of the pro-oxidant/antioxidant status in the HS patients significantly differed from those of the control group (p <0.05), which corresponded to moderately pronounced oxidative stress. The activity of SOD and catalase in the erythrocytes of the patients was higher as compared with that of the control group (p = 0.0001 and p <0.0001, respectively). The comparison of the severity of oxidative stress depending on HS severity has determined that the degree of stress was more pronounced in patients with moderate or severe course of the disease (p <0.05).Conclusion. HS patients develop oxidative stress (decreased activity of the antioxidant system associated with increased accumulation of prooxidant substances), the degree of which is higher in patients with a severe course of the disease. This allows of considering plasma LDCL indicators as an additional marker for the assessment of the severity of the disease and of justifying the necessity to include antioxidants in the HS treatment regimen.

The enhancement of free radical oxidation, characteristic of inflammatory processes, is accompanied by a violation of the properties of biological membranes, the functioning of cells and damage to surrounding tissues. The purpose and task of the research was to make correction of antioxidant protection of the body of cows during the period of dry preparations containing nanoparticles of metals. To activate the antioxidant system of the body of cows during the dry period, use the vitamin complex «Nanovit». The experiment was conducted on cows of Ukrainian dairy black-and-white breed in Agroprodservice-Invest LLP of the Kozivsky District of the Ternopil Oblast. The experiment was conducted before 35–25 days to the calves birth where the experimental and control groups of cows were formed (n = 10). Cows of the experimental group for 35–25 days to the cattle, intramuscularly injected 5.0 ml vitamin complex «Nanovit» (composition: vitamins A, D, E and nanoparticles Cu, Zn, Mn, Co). Producer of «NVP» LLC «Ecological Capital». Cows of the control group for 35–25 days to the catheter, intramuscularly administered 15.0 ml of the drug Prodevit-tetra (1 ml of the preparation contains vitamin A – 50000 IU, vitamin D – 25000 IU, vitamin E – 20 mg, vitamin F – 6 mg). Manufacturer of Product Ltd. The study found that vitamins A, D, E, and nanoparticles Cu, Zn, Mn, Co, which are part of the drug «Nanovit», activate the antioxidant system of the body of cows during the dry period. So after the application of the drug «Nanovit» in the body of cows there was a decrease of 58.5% (Р ≤ 0.01) of diene conjugates and 95.0% (Р ≤ 0.001) of TBK-active products against the background of an increase of 20.0% (Р ≤ 0.01) activity of catalase. After the application of the drug «Nanovit», the service period of the period from cows decreased by 54 days, and the index of insemination decreased by 0.2 units against the cows fed «Prodevit-tetra», indicating a positive effect of the drug containing the nanoparticles of metals Cu, Zn, Mn, Co and vitamins A, D, E during the post-period period in cows.

Modern environmental conditions dramatically increased the level of radical forming processes in the body. Heat exposure stimulates the generation of reactive oxygen species inducing peroxidation of lipids as a result of the development of hypoxia. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the oral introduction of the phytoadaptogens that contain the complex of natural antioxidants. The animals were divided into 5 groups and each of them had 30 rats: intact animals (1) which were held in standard conditions of vivarium; the control group (2) in which rats were exposed to heat during forty-five minutes daily; the experimental groups (3, 4, 5) in which before the effects of heat animals had a daily oral intake of the extracts of Eleutherococcus, Rhodiola, liquorice in a dose of 1 ml/kg. It was found out that in the liver tissue of experimental animals a daily heat exposure during forty-five minutes contributes to the increase of lipid hydroperoxides level (by 34-41%), of diene conjugate (by 45-50%), and of malonic dialdehyde (by 62-74%) against the decrease of antioxidant system activity in the liver of intact animals. The introduction of phytoadaptogens to rats in the conditions of heat exposure contributes to the reliable decrease in the liver of lipid hydroperoxides by 22-28%, of diene conjugates by 22-32%, and of malonic dialdehyde by 30-37% in comparison with the rats of the control group. While analyzing the effect of the phytoadaptogens on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the liver of animals was reliably higher by 68-82%, of vitamin E by 42-65% in comparison with the same parameters of the rats of the control group. So, the application of phytoadaptogens in the conditions of long heat exposure of the organism of animals under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

Introduction One of the reasons for the lack of quality drinking water is the poor quality of natural water, which is constantly polluted by waste water from industrial and municipal enterprises, surface runoff from the fields and territories of settlements with which it enters a large amount of toxic substances. Among them, surface-active substances such as potassium and sodium stearate are not the last.The aim of the study to determine the specifics of lipid peroxidation and the state of the antioxidant protection under the influence of potassium and sodium stearates on the body of the experimental rats when they enter with drinking water.Research Methods. White rats were divided into 7 groups, which consumed water with stearates in quantities equal to the maximum non-active dose of the substance (MND), 1/2 MND and 1/4 MND for 25 days. The content of TBC-active lipid peroxidation products, diene conjugates and the activity of superoxide dismutase and catalase in liver homogenate were determined.Results and Discussion. In the homogenate of the liver of animals of all groups, there was a significant increase in the level of diene conjugates and TBK-active products in proportion to the concentration of the substance. It was also found that in animals that consumed water with different concentrations of potassium stearate, inhibition of antioxidant enzymes was observed. Consumption of water with different concentrations of sodium stearate caused the activation of these enzymes.Conclusions. Prolonged use by test animals of aqueous solution with  concentrations of potassium and sodium stearate in MND and 1/2 MND negatively affects the state of hepatocyte cell membranes due to the activation of the lipid peroxidation processes, namely TBK-active products and diene conjugates, the amount of which depended on the concentration of stearates in drinking water. The use of drinking water by test animals with maintenance of potassium and sodium stearates at various doses caused changes in the content of antioxidant enzymes: water with potassium stearate inhibits the activity of these indicators, and causes sodium activation with sodium stearate.

Background: Currently, there is a growing interest in new local coordination compounds (CC), which demonstrated antitumor properties, but their influence on the erythrocyte antioxidant system has not been studied. The aim of the study: to study the effects of CC, thiosemicarbazide derivatives –TIA-160, CMT-67 and CMJ-33 on indices of the antioxidant system indicators in erythrocytes peripheral blood in vivo experiments. Material and methods: The action of CC on superoxide dismutase, catalase, total antioxidant capacity was evaluated on a group of 34 white rats, randomly divided into 4 groups: the first control group was injected subcutaneously with saline. The other groups (2 – TIA-160, 3 – CMT-67 and 4 – CMJ-33) were given subcutaneously 3 times a week for 30 days, 0.1µM / kg CC. Results: It was established that the TIA-160 compound demonstrated the highest capacity to induce the expression of erythrocyte catalase that exceeded the control level of 1.8, which did not correlate with the enzymatic superoxide dismutase (SOD) activity. Thus, this study showed that there are differences in the mechanisms of action of thiosemicarbazone derivatives. Conclusions: The influence of tested CC on the indices of the antioxidant system is selective. This selectivity may be the base to their strong selective antiproliferative and cytotoxic action on tumor cells, but not on healthy ones.

Currently there is an active search for medicines increasing the stability of liver to pathological effects, reinforcing its neutralizing function, promoting its functional recovery after various injuries, including poisoning by industrial poisons. Therefore, the study of opportunities to reduce toxic liver damage by carbon tetrachloride introduction of synthetic and natural antioxidants is of interest. In experimental conditions the possibility to correct free radical lipid oxidation of rats’ organism membranes was studied with the introduction of the succinate containing drug called Reamberin (Polysan, St.Petersburg) and licorice syrup. The animals were divided into 4 groups and each of them had 10 rats: the group with intact animals which were held in standard conditions of vivarium; the control group in which rats were given carbon tetrachloride during 3 days daily; the experimental group in which before the introduction of carbon tetrachloride animals had a daily intra-abdominal intake of the Reamberin in a dose of 100 mg/kg (20 ml/kg); the experimental group in which before the introduction of carbon tetrachloride animals had a daily oral intake of the licorice syrup in a dose of 5 ml/kg. It was found out that in the blood and in the liver of experimental animals an introduction of carbon tetrachloride during 3 days contributes to the increase of lipid hydroperoxides level (by 24-43%), of diene conjugate (by 19-47%), and of malonic dialdehyde (by 61-81%) against the decrease of antioxidant system activity in the blood of intact animals. The introduction of the succinate containing drug to rats in the conditions of oxidative stress contributes to the decrease in the blood and in the liver of lipid hydroperoxides by 13-26%, of diene conjugates by 11-28%, and of malonic dialdehyde by 30-34% in comparison with the rats of the control group. Adding of the licorice syrup to rats contributes to the reliable decrease in the blood and in the liver of lipid hydroperoxides by 6-23%, of diene conjugates by 9-24%, and of malonic dialdehyde by 25-30% in comparison with the rats of the control group. While analyzing the effect of the antioxidants on the activity of the components of antioxidant system it was shown that the level of ceruloplasmin in the blood and in the liver of animals was reliably higher by 10-47%, of vitamin E by 13-42% in comparison with the same parameters of the rats of the control group. So, the application of the synthetic and natural antioxidants in the conditions of introduction of carbon tetrachloride of the organism of animals under experiment leads to the stabilization of the processes of peroxidation against the increase of antioxidant system activity.

BACKGROUND: The administration of doxorubicin can increase the production of reactive oxygen species (ROS) and disrupt the balance of antioxidant defenses triggered a cell damage including liver cells and renal cells. Antioxidant from medicinal plants have play role in protecting the cells against ROS. Beetroot tuber contains various phytochemical compound that has an antioxidant property. AIM: The purpose of the study was to determine the protective effect of beetroot ethanol extract on concurrent use of doxorubicin for 15 days by measuring blood levels of aspartate aminotransferase, alanine aminotransferase, Creatinine, blood urea nitrogen and histopathological changes. MATERIALS AND METHODS: Twenty-five male white rats as weighing 180–200 g were divided into five treatment groups. Group I was the normal control group, Group II was a negative control group that only injected doxorubicin with a cumulative dose of 15 mg/kg body weight (BW) for 15 days. Group III-V was the test group given beetroot extract (BE) at doses of 100, 200 and 400 mg, respectively, for 15 days with doxorubicin. RESULTS: The group treated with BE at a dose of 200 mg/kg BW could improve serum levels of liver enzymes compared with negative controls (p < 0.05). The group treated with BE at a dose of 100 mg/kg BW also showed an improved effect on creatinine and BUN levels of rats compared to negative controls (p < 0.05). All parameters showed that the protective activity of beetroot ethanol extract was dependent on increasing the dose. CONCLUSION: This research proves that the compound contained in beetroot ethanol extract has protective activity in liver and kidney of rats induced by doxorubicin.
