Levels of amino acids were determined in the grains of wheat, Triticim durum Desf. The raw sample was best in Arg, Ile, Leu, Thr, Val, Ala, Asp, Glu and Pro but similar to germinated sample in Lys whereas germinated sample was best in His, Met, Cys, Phe,Gly, Ser and Tyr. The total amino acid contents were; raw 12[74.1 g/100 g crude protein (c. p.)], steeped 63.4 g/100g c. p.) and germinated (70.6 g/100 g c. p.) with respective essential amino acids of 32.9 g/100 g c. p., 30.0 g/100 g c. p. and 32.4 g/100 g c. p. The Predicted Protein Efficiency Ratio (P-PER) levels were 2.46 (raw), 2.27 (steeped) and 2.19 (germinated). Only germinated samples enhanced the quality of the wheat amino acid levels up to the tune of 41.2 %. However, significant differences occurred between raw/steeped and raw/germinated at r = 0.05. Three hundred and twenty-four DeKalb White laying hens aged 42 weeks were distributed in a completely randomised design with nine treatments and six replicates of six birds in each treatment. The experiment lasted 112 days. Diets were: T1 = 16.02% crude protein - CP [Met + Lys + Thr + Trp + Val]; T2 = 14.02% CP [Met + Lys + Thr + Trp + Ile + Val]; T3 = 14.02% CP [no amino acid supplementation]; T4 = 14.02% CP [Met + Lys + Thr + Trp]; T5 = 14.02% CP [Met + Lys + Thr]; T6 = 14.02% CP [Met]; T7 = 14.02% CP [Lys]; T8 = 14.02% CP [Thr]; T9 = 14.02% CP [Trp]. Regarding the quality of the eggs, the percentage of yolk and albumen, shell thickness and Haugh unit were not affected by the different diets. The percentage of shell, specific gravity and albumen height showed significant differences. We found that supplementation of only one amino acid in the diet (T7, T8 or T9), with the exception of methionine (T6), worsened performance relative to the control. Supplementation of three amino acids (methionine, lysine and threonine; T5) or four amino acids (methionine, lysine, threonine and tryptophan; T4) worsened egg production and conversion per mass and per dozen eggs; however, feed intake and egg weight and mass were similar to the control treatment. When all amino acids (methionine, lysine, threonine, tryptophan, isoleucine and valine; T2) were supplemented performance was similar to the control treatment in all variables. Supplementation of methionine, lysine and threonine is essential for birds in the laying phase; however the addition of six essential amino acids (lysine, methionine, threonine, tryptophan, valine and isoleucine) to the diet of laying hens is important for a good productive performance comparable with the control treatment T1. However, the inclusion of the latter two (isoleucine and valine) is justified only if the production cost is lower.

AbstractThe prime purpose of this review is to explore the pathways whereby progress towards reduced-crude protein (CP) diets and sustainable chicken-meat production may be best achieved. Reduced-CP broiler diets have the potential to attenuate environmental pollution from nitrogen and ammonia emissions; moreover, they have the capacity to diminish the global chicken-meat industry’s dependence on soybean meal to tangible extents. The variable impacts of reduced-CP broiler diets on apparent amino acid digestibility coefficients are addressed. The more accurate identification of amino acid requirements for broiler chickens offered reduced-CP diets is essential as this would diminish amino acid imbalances and the deamination of surplus amino acids. Deamination of amino acids increases the synthesis and excretion of uric acid for which there is a requirement for glycine, this emphasises the value of so-called “non-essential” amino acids. Starch digestive dynamics and their possible impact of glucose on pancreatic secretions of insulin are discussed, although the functions of insulin in avian species require clarification. Maize is probably a superior feed grain to wheat as the basis of reduced-CP diets; if so, the identification of the underlying reasons for this difference should be instructive. Moderating increases in starch concentrations and condensing dietary starch:protein ratios in reduced-CP diets may prove to be advantageous as expanding ratios appear to be aligned to inferior broiler performance. Threonine is specifically examined because elevated free threonine plasma concentrations in birds offered reduced-CP diets may be indicative of compromised performance. If progress in these directions can be realised, then the prospects of reduced-CP diets contributing to sustainable chicken-meat production are promising.

AbstractAmino acid deprivation or supplementation can affect cellular and organismal lifespan, but we know little about the role of concentration changes in free, intracellular amino acids during aging. Here, we determine free amino-acid levels during chronological aging of non-dividing fission yeast cells. We compare wild-type with long-lived mutant cells that lack the Pka1 protein of the protein kinase A signalling pathway. In wild-type cells, total amino-acid levels decrease during aging, but much less so in pka1 mutants. Two amino acids strongly change as a function of age: glutamine decreases, especially in wild-type cells, while aspartate increases, especially in pka1 mutants. Supplementation of glutamine is sufficient to extend the chronological lifespan of wild-type but not of pka1Δ cells. Supplementation of aspartate, on the other hand, shortens the lifespan of pka1Δ but not of wild-type cells. Our results raise the possibility that certain amino acids are biomarkers of aging, and their concentrations during aging can promote or limit cellular lifespan.

Effect of storage time on the quality of smoked heteroclarias was studied. 108 samples of heteroclarias (average weight 210 + 15g) was used. Analysis carried out include: proximate, mineral composition (Ca, Na, Fe and Mg), biochemical, amino acid and sensory evaluation. Data obtained was subjected to Analysis of Variance (ANOVA) while the sensory data was subjected to nonparametric test (Kruskal Wallis test). Smoked heteroclarias have good nutritional quality in terms of proximate, mineral and amino acids all of which decrease with increase in duration of storage at ambient temperatures. Glutamic acid  was the most predominant amino acid and the highest non-essential amino acid (NEEA), lysine was the most predominant EAA. There was higher concentration of non-essential amino acids than essential amino acids, EAA/NEAA ratio (0.86 – 0.93) recorded indicates that the fish have excellent protein quality; its the predicted protein efficiency ratio (P-PER) ranged between 3.44-3.61 and its biological value ranged between 79.84 -75.04. Its chemical score and TEAA decrease with increase in storage time. Its texture quality reduced significantly (χ2 = 12.207, p<0.01) with increased storage period. It is therefore recommended that smoked heteroclarias be consumed as soon as it is smoked and regularly for good healthy conditions especially among children, aged and other vulnerable groups.

1. Free amino acid concentrations in the plasma have been compared with those in liver and quadriceps muscle, in rats fed on diets containing 209 (control) and 31 (low-protein) g protein/kg. The effects of the low-protein diet on diurnal variations in these values were also measured.2. In the plasma, the total amino acid concentration was significantly lower in animals given the low-protein diet, at all times of day except 12.00 hours. In the liver, and to a lesser extent the muscle, total amino acid concentration was maintained.3. In the control animals, diurnal variation in the concentrations of both essential and non-essential amino acids was very similar in plasma, liver and muscle. In animals given the low-protein diet, although the same diurnal pattern was maintained for non-essential amino acids, that occurring among the essential amino acids had virtually disappeared.4. In plasma, the mean 24 h concentration of essential amino acids decreased from 24· mmol/l in control animals to only 1·29 mmol/l in the low-protein-fed animals. Concentrations in muscle and liver were reduced by a similar proportion (from 8·6 to 5·56 μmol/g and from 8·67 to 5·05 μmol/g respectively). Conversely the concentrations of non-essential amino acids in animals given the low-protein diet were increased in plasma (from 1·53 to 2·00 mmol/l), muscle (from 12·5 to 14·3 μmol/g), and liver (from 16·8 to 20·5 μmol/g), muscle showing the lowest increase.5. With the exceptions of lysine, threonine, cystine and tyrosine, the concentrations of all other essential amino acids were reduced more in liver than in muscle. The relationship between this and the failure to maintain plasma albumin concentrations is discussed.

AbstractThis research focused on the effect of distant hybridization to improve the flesh quality of offspring from Oreochromis aurea (♀)×Siniperca chuatsi (♂). The proportions of different tissues and percentages of nutrient components were compared between fish of the F3 generation and O. aurea and S. chuatsi. The results showed that the proportion of flesh in F3 hybrids was significantly lower than that in O. aurea. Crude lipid content was lower and crude protein content significantly higher in the F3 generation. Concentrations of four kinds of amino acids important for flavour (DAA) were higher in the F3 generation than in O. aurea, and the total DAA was significantly higher. Furthermore, the other amino acid contents and essential amino acid index of the F3 generation were intermediate between O. aurea and S. chuatsi. We conclude that some effects of hybridization have induced flesh quality improvement.

High protein wheat was the sole cereal in 20 and 25 per cent crude protein broiler starter diets. On the. 25 per cent protein diet, performance was maximized without amino acid supplementation. Using high protein wheat in 20 per cent protein diets, growth rate was improved by l-lysine supplementation of 0.3 per cent. However, this growth rate was not at a maximum level. Some other dietary factor was necessary, and this did not appear to be essential amino acids, singly or in combination.

AbstractTwenty-eight New Zealand White X Californian doe rabbits weighing 3791±141 g were surgically fitted with a glass T-cannula at ileum level. Animals were given ad libitum access to either a casein-based diet (C) or a protein-free diet (PF). The only difference in the ingredient composition of the diets was the substitution of 160g/kg of maize starch by casein in the C diet. Animals were randomly allotted to each experimental diet. Seven rabbits per diet were not allowed to practice caecotrophy. The C diet resulted in higher food dry matter (DM) intake (DMI) (111-5 v. 51-5 g/day), food nitrogen intake (3-12 v. 0-20 g/day), ileal flow of DM (56-0 v. 23-8 g/day) and ileal flow of nitrogen (0-72 v. 0-24 g/day) but lower soft faeces DMI (21-7 v. 41-4 g/day) than the PF diet. A linear relationship between ileal endogenous nitrogen flow and total DMI was found for animals given the C but not for the PF diet. The ingestion ofC diet led to a higher (proportionately 0-41, on average), endogenous ileal amino acid flow than PF diet for all amino acids except for glycine. The ileal flow of glycine was proportionately 0-25 higher in animals given the PF diet than in animals given the C diet. The C diet resulted in higher content of nitrogen, threonine and proline in soft faeces than the PF diet. In contrast, the ingestion of the PF diet led to a higher content of lysine and methionine in soft faeces than the C diet. Within the C diet, animals practising caecotrophy showed higher ileal flow of DM (73-4 v. 56-0 g/day) and phenylalanine (0-834 v. 0-677 g/kg DMI) than animals not practising it. Caecotrophy led to a higher endogenous proportion of some of the most important limiting essential amino acids in rabbits, such as arginine, lysine, phenylalanine and threonine. In rabbits, the use of PF diets has limited practical application in the estimation of nitrogen endogenous losses as the low food DMI and the high soft faecesDMI leads to abnormal food: soft faeces intake ratio. Moreover, due to the important effect of caecotrophy on the amino acid composition of endogenous losses, it is important to correct the total amino acid content of the ileal chyme by the undigested amino acid residue coming from the soft faeces intake. This correction could be made, without the methodological complexity implied, by fitting animals with a wooden collar and thereby avoiding caecotrophy as in the present trial.

Cereals and legumes are staple foods in India and are limiting in lysine and sulphur amino acids, respectively. Available lysine loss, due to Maillard-type reactions that may occur during food preparation, exacerbates the problem of lysine deficiency particularly in cereals. Consequently, determining the contents of digestible essential amino acids, particularly lysine, is important. True ileal digestibilities of most amino acids (including total and reactive lysine) were determined for ten food ingredients and eleven foods commonly consumed in India. Semi-synthetic diets each containing either an ingredient or the prepared food as the sole protein source were formulated to contain 100 g kg− 1protein (75 g kg− 1for rice-based diets) and fed to growing rats. Titanium dioxide was included as an indigestible marker. Digesta were collected and the amino acid content (including reactive lysine) of diets and ileal digesta determined. Available (digestible reactive) lysine content ranged from 1·9–15·4 g kg− 1and 1·8–12·7 g kg− 1across the ingredients and prepared foods respectively. True ileal amino acid digestibility varied widely both across ingredients and prepared foods for each amino acid (on average 60–92 %) and across amino acids within each ingredient and prepared food (overall digestibility 31–96 %). Amino acid digestibility was low for many of the ingredients and prepared foods and consequently digestibility must be considered when assessing the protein quality of poorer quality foods. Given commonly encountered daily energy intakes for members of the Indian population, it is estimated that lysine is limiting for adults in many Indian diets.

The Asian citrus psyllid (ACP) Diaphorina citri is the main vector of the pathogen Candidatus Liberibacter asiaticus (CLas), which is the causal agent of citrus Huanglongbing disease. Feeding by both ACP nymphs and adults on host plants allows them to obtain nutrition. Therefore, the nutritional content within the plant phloem is of much importance for the development and reproduction of ACP. The infection by pathogenic microbiomes may affect the amino acid contents of their host plants and then indirectly affect the biology of sap-feeding insects. In this study, we investigated the amino acid contents and their proportions in both CLas-infected and CLas-free citrus plants, ACP adults, and also in honeydew produced by ACP nymphs. Results showed that infection by CLas had a large impact on the amino acid species and proportion in all the tested target plants, ACP adults, and in the honeydew of ACP nymphs. The content of total amino acids in CLas-infected citrus was much higher than that of CLas-free citrus. However, CLas infection significantly reduced the proportion of essential amino acids (EAAs) in these plants. When feeding on CLas-infected citrus plants, ACP adults absorbed less total amino acids than those adults feeding on healthy plants, but the proportion of EAAs was significantly higher when they fed on CLas-infected citrus plants. The proportion of EAAs also significantly increased in the honeydew secreted by ACP nymphs that fed on CLas-infected citrus plants. However, EAA detection in the honeydew of ACP nymphs indicated that the utilization rate of EAAs by CLas positive ACP nymphs was reduced. Our study has revealed that CLas infection significantly affects the contents, proportion, and utilization efficiency of different amino acids in citrus plants, ACP adults, and nymphs, leading to a developmental pattern of ACP that is more conducive to CLas transmission.
