The effect of aqueous extract of Basella alba (puishak) leaves on blood cell count of rats was studied. Forty rats of both sexes weighing between 100-150 gm were used. The rats were divided into four groups (7 rats in each group), with group A as the control group and experimental groups were denoted as B, C and D. With all aseptic precautions, aqueous extract of Basella alba (Puishak) leaves was administered into the three different doses (For group B: 60 mg/kg bw, group C: 80 mg/kg bw and group D: 100 mg/kg bw). Control group A also received distilled water as a placebo at the dose of 10 mg/kg bw for 14 days. At day 15, blood samples were collected and sent for haematological analysis. Data analysis of blood count profile of 28 rats revealed that there is an increased number of RBC, WBC and platelet count in experimental groups than in control group. ANOVA test revealed that increased blood cell counts following administration of aqueous leaves extract of keeves of B. alba were statistically significant (p value for each case was .0001<.05) than control group. Paired samples t test was performed to compare between baseline hematological parameters and parameters after 14 days of intervention. Then comparison between Group A & Group B, between Group A & Group C and between Group A & Group D were done. In all cases, probability (p) value < 0.05 was considered as statistically significant.This it is clearly evident that aqueous extract of B.alba has positive stimulant effect on blood cells count of rats. Moreover, it was found that increment of doses also increases the cell count that is positively correlated with the hypothesis. Bangladesh Pharmaceutical Journal 22(1): 73-78, 2019

This study aimed to identify septic phenotypes in patients receiving vitamin C, hydrocortisone, and thiamine using temperature and white blood cell count. Data were obtained from septic shock patients who were also treated using a vitamin C protocol in a medical intensive care unit. Patients were divided into groups according to the temperature measurements as well as white blood cell counts within 24 h before starting the vitamin C protocol. In the study, 127 patients included who met the inclusion criteria. In the cohort, four groups were identified: “Temperature ≥37.1 °C, white blood cell count ≥15.0 1000/mm3” (group A; n = 27), “≥37.1 °C, <15.0 1000/mm3” (group B; n = 30), “<37.1 °C, ≥15.0 1000/mm3” (group C; n = 35) and “<37.1 °C, <15.0 1000/mm3” (group D; n = 35). The intensive care unit mortality rates were 15% for group A, 33% for group B, 34% for group C, and 49% for group D (p = 0.051). The temporal improvement in organ dysfunction and vasopressor dose seemed more apparent in group A patients. Our results suggest that different subphenotypes exist among sepsis patients treated using a vitamin C protocol, and clinical outcomes might be better for patients with the hyperinflammatory subphenotype.

This study determined haematological changes in Wistar rats experimentally infected with local strains of Trypanosoma congolense and Trypanosoma brucei brucei. Forty-five Wistar rats between 10 – 12 weeks old weighing between 210 – 240 g were used. The Wistar rats were randomly divided into four groups (A, B, C and D), with the infected groups (B, C and D) having 10 rats each, while the uninfected control group (A) had 15 rats. Group A rats were not infected and served as the control, group B were infected with Trypanosoma congolense, group C were infected with Trypanosoma brucei brucei and group D were co-infected with Trypanosoma congolense and Trypanosoma brucei brucei. Infection was achieved using 0.1mL of blood containing approximately 1 × 103 trypanosomes intraperitoneally into each Wistar rat in the infected groups. Clinical signs were observed. The changes in the blood cells were assayed in the groups post-infection. Duncan’s Least Square Deviation showed significantly (p<0.05) higher parasitaemia in infected groups. However, group D showed a higher significant (p<0.05) difference in parasitaemia when compared to groups B and C. The pattern of mean parasitaemia for the infected groups, revealed a positive correlation with days of post-infection (p<0.05) before the decline. The packed cell volume, total red blood cell count and haemoglobin concentration were significantly (p<0.05) lower in infected groups B, C and D. The total white blood cell count, platelet counts and differential leucocyte count were significantly (p<0.05) lower in infected groups when compared to the uninfected group. These findings suggest that co-infection with Trypanosoma congolense and Trypanosoma brucei brucei obtained from Wurno and Ngaski in Sokoto and Kebbi States respectively produced a more damaging effect on haematological parameters.

Objectives: To study the impact of anti-inflammatory agent like Bryophyllum pinnatum aqueous extract and diclofenac on blood pressure and creatinine clearance. Study Design: Experimental study. Setting: Sargodha Medical College, Sargodha, Department of Pharmacy, University of Sargodha. Period: 1st January 2019 to 30th June 2019. Material & Methods: 24 Sprague Dawley rats were obtained and were then divided into four groups. Negative control group (A) contained animals received normal diet while Group B, C and D received diet containing sucrose (20% w/w) to induce hypertension. After that group B (positive control group) received distilled water 0.5 ml was by oral route, group C (Bryophyllum pinnatum group) received Bryophyllum pinnatum aqueous extract 300 mg/ml intraperitoneally and group D received diclofenac 12 mg/kg intraperitoneally as a single morning dose for two weeks. Blood pressure of animals was recorded at baseline and then weekly throughout the study using tail cuff using non-invasive blood pressure controller (ML125R). Animals were anesthetised with chloroform and two ml blood was drawn through cardiac puncture at 0, 4and 6 weeks. Blood was tested for haematocrit. Serum sodium and potassium levels were estimated by flame photometer. Urinary creatinine levels were estimated by kinetic Jaffé method. The data collected was processed by using Statistical Package for Social Sciences (SPSS 20). Results: Mean and standard deviation of systolic blood pressure (BP) of group A rats did not change with time, while that of group B, C and D increased till 4th week. Creatinine clearance of group A, B and C did not change much with time but that of group D decreased towards end of study period. Conclusion: Bryophyllum pinnatum leaf aqueous extract is an effective anti-hypertensive agent with minimal renal effects.

Abstract A suboptimal values of the haematological variables has often been recorded in young athletes involved in intense physical activity. There have even been reports of “sports anemia” associated with intensive physical training. There are profound studies describing the haematological changes of the soccer players after training or 90 min match. Our previous studies demonstrated lower values of red blood cell count (RBC), packed cell volume (HCT), haemoglobin concentration (HGB) in highly trained football players than in untrained controls of the same pubescent age. THE AIM: It was a matter of interest to evaluate the haematological status of 2314 football players (15.11±2.89 yrs old) and to compare the results for pre-pubescent, pubescent and mature boys recruited in the study and also to relate the values obtained to the individulal aerobic functional capacity. MATERIAS: A total of 2314 highly trained football players were included in the study. Their mean±SD age, weight, and BMI were: 15.11±2.89 yrs, 56.33±13.28 kg, and 19.62±2.58 kg/m2. The sportsmen were divided into three groups in accordance with their age: pre-pubescent (9–14 years) group A (n=1178) − 13.22±1.05 yrs (158.6±12.6 months); pubescent (15–19 years) group B (n=1029) − 16.57±1.04 yrs (198.9±12.6 months); and mature (20–35 years) group C (n=107) − 24.30±3.78 yrs (291.6±45.3 months) old. Profound clinical examination of all sportsmen was performed at the day before obtaining the blood samples. Haematological variables including RBC, HCT, HGB, mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin content (MCHC), white blood cell count (WBC) and platelet count (PLT) were analysed on a Sysmex K800 (Japan) analyser. Each football player passed also PWC170-test for assessment the working capacity on AT-104 Spiro-Ergo equipment (Schiller, Switzerland). RESULTS: All haematological variables in the groups were found in normal ranges, but the RBC was highest in group B (5.17±0.28 *1012/l vs. 5.03±0.28 *1012/l for group A, and 5.08±0.41 *1012/l for group C). Group A demonstrated not only lowest values for the RBC, but also for the HGB (144.7±6.4 g/l vs. 149.1±6.0 g/l for group B, and 151.8±7.5 g/l for group C, P&lt;0.001), and for HCT (0.437±0.02 vs. 0.451±0.02 for group B, and 0.451±0.03 for group C, P&lt;0.001). Group A demonstrated similar values for the above basic red blood cell parameters to these measured for great male group of same age. The pre-pubescent football players (group A) had lower MCV (87.1±2.8 fl), than the players of group B (87.5±2.9 fl) and than the players from group C (88.1±3.0 fl). No evidences for blood dilution as a result of long-term training had been found in groups B and C. There was not difference in the WBC and PLT between pre-pubescent, pubescent and mature footballers. The mature players (C-group) demonstrated higher PWC/kg than pre-pubescent (20.6±2.3 vs. 19.2±2.3 kgm/min/kg), and than pubescent boys (19.8±2.7 kgm/min/kg). Significant correlations were obtained between the age and HGB, and between the age and HCT in groups A (0.19 and 0.19 respectively) and B (0.24 and 0.15 respectively). Good correlations were found between RBC and PWC170/kg in all groups investigated. CONCLUSIONS: Pre-pubescent highly trained football players have lower values of the red blood cell variables than pubescent and mature footballers, which correlates with lower aerobic working capacity.

Objectives: To observe the effect of lagenaria siceraria on inflammation and fibrosis brought about by arsenic in liver of Sprague Dawley rat. Study Design: Laboratory based randomized control trial. Place and Duration of Study: This experiment was performed at Department of Anatomy, Army Medical College Rawalpindi in co-operation with National Institute of Health (NIH) Islamabad for eight weeks (1st March 2017 to 25th April 2017). Material and Methods: Fifty Sprague Dawley rats (both male and females housed separately) were carefully chosen and distributed randomly into five groups, each consisting of 10 animals. A and B were the control groups whereas C, D and E served as experimental groups. During the first four weeks, experiment groups C, D and E were given a dosage of 5milligram/kilogram body weight of sodium arsenite. At the end of four weeks, animals from control group A and experimental group C were dissected and liver samples were processed for microscopic studies. In the next 4 weeks, group D animals were set aside without any further intervention. At that time, sodium arsenite at a dose of 5 milligram/kilogram body weight and lagenaria siceraria at a dose of 100 milligram/kilogram bodyweight were administered to group E animals. Group B animals served as control for experimental groups D and E. At the end of these 4 weeks animals of groups B, D and E were dissected. Liver was processed, fixed and stained for microscopic study. Area of portal triad as well as liver lobules were studied for inflammation and fibrosis and results were analysed. Statistical tool used to analyse the data was SPSS v 22. Results were considered to be significant when p-value is ≤ 0.05. Results: Experimental group C developed moderate grade fibrosis and inflammation (grade 2 to 3) as compared to group A. Degree of inflammation and fibrosis was mild to moderate (grade 1 to 2) in group D. There was no inflammation and fibrosis (Grade 0 to 1) in group E. Group B served as a control for group D and E. Conclusion: Inflammation and fibrosis developed in the liver of adult rats when they were subjected to sodium arsenite even for a brief calculated period. Simultaneous administration of lagenaria siceraria can shield and diminish the toxic effects of arsenic. Oxidative potential and immunomodulatory properties and presence of flavonoid like substances renders lagenaria siceraria to act as ameliorative against this fibrosis and inflammation in liver lobules and surrounding area of portal triads.

Background: A well-known disease, Acute Respiratory Distress Syndrome (ARDS) presents a daunting challenge to the medical community with alarmingly high mortality rates. Initiation of hypertonic saline (HTS) nebulization therapy for patients with early ARDS appears to be tolerable and may be beneficial. However, using a nebulizer is cumbersome and less efficient. This study aims to assess the efficacy of using HTS3% in a nebulizer in comparison with intravenously injecting it to attenuate the manifestation of ARDS. The study analyses factors such as the severity of the disease, need and duration of mechanical ventilation (MV), ICU stay, and mortality rate in the comparative analysis. Materials and Methods: The randomized, comparative, and controlled study included patients of both genders from an age bracket of 18-60 years. The patients fulfilled the Berlin definition of ARDS. Additionally, the whole sample was divided into four groups (26 patients each): Group A received standard pharmacotherapy [methylprednisolone 1mg/kg/day intravenously and salbutamol nebulizer 2.5 mg (1 ml) + 3 ml normal saline/8hr] and normal saline 0.9% (5ml) nebulizer /8hr: Group B received standard pharmacotherapy + HTS2.7% (5ml) nebulizer/8hr: Group C also received standard pharmacotherapy and 500ml normal saline intravenously over 24 hours: Group D received standard pharmacotherapy and 500 ml HTS 3% intravenously over 24 hours. The following parameters were recorded: Lung injury score (Murray Score, calculated every 24 hours), the incidence of the need for MV, duration of MV, length of ICU stay, and mortality rate. Results: Group B (HTS nebulizer group) and group D (HTS IV group) showed the most rapid improvement in the Murray score with a 50% decrease in the score from its initial value (D50%) by day 4 with P-values of 0.013 and 0.022 respectively. Subjects from Group D (HTS IV) and Group B (HTS Nebulizer) were the least in need of MV as only 38.5% needed MV with a P-value <0.001. In comparison, 69.2% subjects from group A (control) and 73.1% from group C required MV, with a P-value <0.001. The shortest duration of MV care was found in group D (median 3.12 days), followed by group B (median 4.21 days). The results were highly significant, with P-value 0.004 when compared with group A (control group) and group C. In addition, the longest duration of MV care was reported in group A (median 5.37 days). There was significant variation in all groups regarding the length of ICU stay: group D required the least number of days (median 6.76 days), followed by group B (median 7.08 days). The result was statistically significant, with a P-value of 0.004 when compared with the control group (median 9.1 days) and group C(longest duration of MV with a median of 10.13 days). However, no significant difference was found in the mortality rate (P-value >0.05). Conclusion: The use of hypertonic saline 3% intravenously has a comparable effect as the use of hypertonic saline 3% via nebulizer to attenuate the manifestations of (ARDS) and even superior in the mechanically ventilated patient.

Twenty mature albino rats randomly assigned to two groups (n=10) were used for the study. Group A was exposed to the sun, while group B was kept under a shade. The study was for 28 days. At the end of the study, the effects of heat stress on haematology and serum biochemistry were determined. There was a significant increase (P < 0.05) in the packed cell volume  of group A when compared to group B. Red blood cell count of group A increased significantly when compared to group B. The mean erythroid values for MCHC and MCH decreased significantly in group A when compared to group B.  The mean haemoglobin concentration of group A was significantly lower when compared to that of group B. White blood cell count of group A decreased significantly (p<0.05) when compared to group B on day 28. The result of the differential leucocyte count showed significant decrease in lymphocytes in the heat stressed group when compared to the unstressed for the entire period. Total serum protein significantly increased in group A when compared to group B on day 14. Alanine amino transferase significantly increases in group A when compared to group B on day 28. Alkaline phosphatase decreased significantly in group A throughout the study period when compared to group B. Aspartate - amino transferase increased significantly in group A when compared to group B on day 28. Serum cholesterol significantly decreased in group A on day 28 when compared to group B.

The purpose of the research: study of the influence of increased doses of fenbendazole supramolecular complex (FSMC) on sheep’s organism. Materials and methods. The experiment was carried out at the Podolsk Department of All-Russian Scientific Research Institute of Fundamental and Applied Parasitology of Animals and Plants named after K. I. Skryabin on 20 manorial invasion-free sheep aged 2-3 years old. Animals were divided according to the principle of analogues into 4 groups, 5 heads in each group. Animals of the 1, 2 and 3 group were orally administered with FSMC given as a single dose of 2, 6, 10 mg/kg, respectively, according to the active substance, i.e in therapeutic and in a dose increased by 3 and 5 times. Sheep of the fourth group didn’t receive the drug and they were as control. Study of clinical, hematological and biochemical parameters of animals from all groups was conducted 1 day before and in 1, 3, 5 days after administration of the drug by means of standard methods. Results and discussion. FSMC in therapeutic dose as well as in a dose increased by 3 and 5 times doesn’t have negative influence on clinical, hematological and biochemical parameters of the sheep. State of the sheep, which received the drug in doses of 20, 60, 100 mg/kg, was within the physiologically normal state and didn’t differ from the state before administration of the drug and from the animals of the control group. Drug security index exceeds 5. Red blood cell count, white blood cell count, hemoglobin count, leukogram parameters as well as biochemical parameters of blood: activity of alkaline phosphatase and amylase, bilirubin, creatinine, urea and glucose counts were within normal limits and didn’t differ from the parameters of the control animals.

Objective: The main purpose of this study is to evaluate the hepatoprotective effects of propolis in hepatocytes injury caused by ATT due to isoniazid and rifampicin. Methods: Healthy albino rats of with average weight of 200-250g were under this study. These rats dividing into main four groups, A group is taken a control group and then further into the group B, group C, and group D as group for experiments. The control group had 15 rats with measured weight, they were given distilled water. Group B had 15 rats, they were given with standard dose of rifampicin and isoniazid. Group c had 15 rats, they were also given with standard dose of rifampicin and isoniazid. Group D had 15 rats, they were given with standard dose of rifampicin and isoniazid and also extract of the propolis we prepared. Results: Serum ALT in the experimental group B with group C, group D were also found to be of statistically significant with p-value < 0.001. ALT serum level observed high in group B. Multiple comparison between groups revealed that group B with a significantly increase in the serum enzyme AST level in comparison to group A, group C and group D with p-value <0.001. Conclusion: This study showed that ethanolic extract of propolis prevents isoniazid and rifampicin induced hepatotoxicity in the albino rats. The toxic effects of an aqueous extract of Abrus precatorius were studied in 20 male white rats over a period of 18 days. The rats were divided into four groups of five rats per group. Those in Group A served as controls while the rats in Groups B, C and D were dosed per os with 400 mg/kg, 800 mg/kg and 1 600 mg/kg of the extract, respectively. Blood samples were collected for haematological and biochemical analysis and specimens of the liver, kidney and testes were taken for histopathological studies. The study showed that the extract of A. precatorius caused decreased levels of packed cell volume, haemoglobin concentration, red blood cell count, white blood cell count, mean corpuscular volume and mean corpuscular haemoglobin. The extract also resulted in increased levels of total serum protein, albumin, alanine amino transaminase, aspartate amino transferase, alkaline phosphatase and total bilirubin. Histologically, testicular degeneration characterized by decreased numbers of lining cells of the epithelium as well as reduction in sperm cells with presence of scattered Sertoli cells were noted. The study thus showed that aqueous extract of Abrus precatorius is toxic and caution should be exercised in its use for medicinal purpose.
