The methanolic extract obtained from the leaves of Tabernaemontana divericata (Family: Apocynaceae) was evaluated for in vitro antioxidant potential by determination of total antioxidant capacity, assay of nitric oxide scavenging activity and reducing power test. The extract was also screened for its cytotoxic effect using brine shrimp lethality bioassay. The results revealed potent antioxidant property in all antioxidant assays compared to the reference antioxidant, ascorbic acid in a dose dependent manner. Further, the methanolic extract of Tabernaemontana divericata showed significant cytotoxic effect (LC50: 3.12μg/ml) compared with positive control, Vincristine Sulphate (LC50: 0.331μg/ml).Key Words: Tabernaemontana divaricata; Apocynaceae; Antioxidant; Cytotoxicity.DOI: http://dx.doi.org/10.3329/icpj.v1i2.9446International Current Pharmaceutical Journal 2012, 1(2): 27-31

The present study was designed to investigate in vitro anthelmintic and cytotoxic activities of crude methanolic extract of two plants(Polygonum viscosum and Aphanamixis polystachya) grown in Bangladesh. Evaluation of cytotoxic activity was done using the brine shrimp lethality bioassay. The crude methanolic extract of Polygonum viscosum showed significant cytotoxic potential (LC50 value of 6.34 ?g/ml) among all the fractions comparing with that of standard vincristine sulphate (0.825 ?g/ml). Besides, the LC50 values of crude methanolic extract, pet ether and chloroform extracts of Aphanamixis polystachya showed good cytotoxic activities 11, 10.36, and 16.45 µg/ml, respectively. The other study was undertaken to evaluate anthelmintic activity (using Pheretima posthuma model) where piperazine was used as reference standard. The crude methanolic extract of Polygonum viscosum leaves produced a significant anthelmintic activity in dose dependent manner and the activity of crude extract was comparable with that of standard drugs. Besides, the Aphanamixis polystachya extract revealed moderate anthelmintic activity. Here, the anova testing was done with the P < 0.05. Objective: The purpose of this work is to evaluate the antimicrobial, antibacterial, and hypoglycemic effects of methanolic and ethanolic extracts of Embelia ribes leaves using in vitro studies.Methods: Antibacterial activities of the methanolic and ethanolic extract of E. ribes leaves against Escherichia coli, Staphylococcus aureus, Enterococci, and Klebsiella pneumoniae at different concentrations ranging from 10, 25, 50, and 75 μg/mL and their antibacterial activities were compared to those of the reference controls such as ciprofloxacin and clindamycin. Furthermore, the effect of leaf extracts on α-amylase and α-glucosidase enzymes was assayed.Results: The methanolic and ethanolic extract of E. ribes leaves effectively inhibited the activity of α-amylase and α-glucosidase in a dose-dependent manner. The effect of the methanolic extract was more prominent than that of ethanolic extract. At the same time, both the extracts showed markable inhibition of bacterial growth at a concentration of 75 μg/mL compared to the other three doses (10, 25, and 50 μg/ml) and also commercially available antibiotic drugs ciprofloxacin and clindamycin that were used as positive control drugs. The antibacterial activity of methanolic extract is significantly higher than that of ethanolic extract.Conclusion: The preliminary results of this study have put forward E. ribes into promising herb with respect to its therapeutic potential although further studies are needed to evaluate its mechanism of action.

The methanolic extract of Litsea glutinosa (Lauraceae) was evaluated for in vitro antioxidant activity by determination of hydrogen peroxide scavenging activity, total antioxidant capacity, assay of nitric oxide scavenging activity and reducing power test and in vivo antinociceptive effect in acetic acid induced writhing model in swiss albino mice. The results revealed the presence of pronounced antioxidant property as compared with ascorbic acid used as standard and a dose-dependent (250 and 500 mg/kg) analgesic effect. The antioxidant and antinociceptive properties observed seem to be in good accordance with the traditional uses of Litsea glutinosa. Bangladesh J. Sci. Ind. Bambusa bambos Linn., a herbal medicine belonging to the family Poaceae, is locally known as Kanta bans or only bans. The crude methanolic extract of leaves of B. bambos and its different partitionates were evaluated for in vitro cytotoxicity, membrane stabilizing and anti-diarrheal activities. In the cytotoxicity screening, the dichloromethane and pet ether soluble fractions displayed the highest lethality to brine shrimps with LC50 of 3.91 and 8.45?g/ml, respectively, whereas the standard vincristine sulphate had LC50 value of 0.45?g/ml. In the membrane stabilizing assay, the crude methanolic extract exhibited highest inhibition of haemolysis of human RBCs by 71.08 ± 0.43% and 49.44 ± 0.73% in heat- and hypotonic solution-induced haemolysis, respectively. The extract exhibited significant (p < 0.05) anti-diarrheal effect at a dose of 400 mg/kg body weight in the castor oil induced anti-diarrheal assay.Bangladesh Pharmaceutical Journal 19(2): 161-165, 2016

Background: Quercus serrata Murray leaves have been used traditionally in the treatment of diabetes, dysmenorrhoea, inflammation and urinary tract infection. So, far no study had been reported on the toxicological profile and antioxidant properties of the plant. Objective: The present study was aimed to investigate the in-vivo toxicological profile and in-vitro antioxidant activities of the methanolic extract of standardized Quercus serrata leaves. Methods: Per-oral sub-acute toxicity study was performed in rats using three dose levels (200, 400 and 800 mg/kg b.w.) of the extract for 28-days. Control group received gum acacia suspended in water. Bodyweight was measured weekly. Biochemical parameters were analysed using the serum, the blood-cell count was done using whole blood. Pathological changes were also checked in highly perfused tissues. Further, in-vitro reducing power assay, nitric oxide scavenging assay, DPPH free-radical scavenging assay were performed to check the antioxidant activity of the extract. Results: There were no significant alterations in the blood-cell count and biochemical parameters analysed in the treatment group when compared with the normal control. Histopathology study of liver, kidney, pancreas, heart and brain revealed normal cellular architecture in the treatment groups alike the control group animals. Quercus serrata also showed a significant reduction of DPPH with IC50 4.48±0.254 µg/mL, in-vitro reducing power activity with IC50121.65±0.320 µg/mL and nitric oxide scavenging activity IC50 106.43±0.338 µg/mL. Conclusion: The above study showed that standardized methanolic extract of Quercus serrata leaves was safe after subacute oral administration in rats and has good antioxidant potential.

Novel imidazole derivatives were designed, prepared, and evaluated in vitro for antitumor activity. The majority of the tested derivatives showed improved antiproliferative activity compared to the positive control drugs 5-FU and MTX. Among them, compound 4f exhibited outstanding antiproliferative activity against three cancer cell lines and was considerably more potent than both 5-FU and MTX. In particular, the selectivity index indicated that the tolerance of normal L-02 cells to 4f was 23–46-fold higher than that of tumor cells. This selectivity was significantly higher than that exhibited by the positive control drugs. Furthermore, compound 4f induced cell apoptosis by increasing the protein expression levels of Bax and decreasing those of Bcl-2 in a time-dependent manner. Therefore, 4f could be a potential candidate for the development of a novel antitumor agent.

Abstract Background The objective of the study is to evaluate the hepatoprotective activity of methanolic extract fractions of Lindernia ciliata (LC) and development of qualitative analytical profile of the bioactive fraction using HPLC fingerprinting analysis. All the fractions of methanolic extract of Lindernia ciliata (LCME) are assessed for their total phenolic, flavonoid contents and in vitro antioxidant properties by using DPPH, superoxide, nitric oxide, hydroxyl radical scavenging activities and reducing power assay. Acute toxicity study was conducted for all the fractions and the two test doses 50 and 100 mg/kg were selected for the hepatoprotective study. Liver damage was induced in different groups of rats by administering 3 g/kg.b.w.p.o. paracetamol and the effect of fractions were tested for hepatoprotective potential by evaluating serum biochemical parameters and histology of liver of rats. The effective fraction was evaluated for its antihepatotoxic activity against D-Galactosamine (400 mg/kg b.w. i.p.) and in vivo antioxidant parameters viz., Glutathione (GSH), Melondialdehyde (MDA) and Catalase (CAT) levels are estimated using liver homogenate. Results Among all the fractions, butanone fraction of LCME, (BNF-LCME) has shown better hepatoprotective activity and hence it is selected to evaluate the antihepatotoxicity against D-GaIN. The activity of BNF-LCME is well supported in in vitro and in vivo antioxidant studies and may be attributed to flavonoidal, phenolic compounds present in the fraction. Hence, BNF-LCME was subjected to the development of qualitative analytical profile using HPLC finger printing analysis. Conclusions All the fractions of LCME exhibited significant hepatoprotective activity and BNF-LCME (50 mg/kg) was identified as the most effective fraction.
