The purpose of this research was to examine the effectivity of Tris, sodium citrate, and lactose extenders in maintaining viability of etawa crossbreed (EC) goat spermatozoa preserved at 3–5oC.  Semen was collected using artificial vagina once a week for six times as replicate.  Fresh semen was divided into three tubes then diluted with Tris, sodium citrate, and lactose extenders, respectively.  Diluted-semen were stored in refrigerator at 3–5oC.  Quality of diluted-semen including percentages of motile spermatozoa (MS) and live spermatozoa (LS) were evaluated every day during storage at 3–5oC for four days.  Data were analyzed using completely randomized design with three treatments and six replicates.  Results of this study showed that mean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS, and spermatozoa abnormal of PE goat fresh semen were 1.05 ml, cream, thick, 6.9, ++ – +++, 3,756.67 million cell/ml, 76.67%, 85.83%, and 4.5%, respectively.  Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  At day-4 storage, percentages of MS and LS for Tris (44.17 and 57.83%) was no difference significant with sodium citrate (43.33 and 56.67%), and lactose (43.33 and 54.17%).   In conclusion, Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  Tris, sodium citrate, and lactose extenders could be maintaining 40% in percentage of motile spermatozoa for three days after preserved at 3–5oC, and suitable for use in artificial insemination (AI) programKeywords: Tris, sodium citrate, lactose, spermatozoa viability, PE goat.

Research on the evaluation of the spermatozoa quality of Brahma, Cochin and Bangkok chickens as the basis for the application of artificial insemination technology in native chickens has been carried out. The purpose of this study was to evaluate the spermatozoa quality of Brahma, Cochin and Bangkok rooster as the basis for determining the spermatozoa dose for artificial insemination in local chickens. method research was used a completely randomized design (CRD) with three treatments, that is: Brahma, Cochin and Bangkok rooster. Each treatment was repeated five times. Cement storage is carried out by inserting an artificial vagina into the cloaca of the rooster and stimulating it to climb the hen. The collected sperm were analyzed at the Animal Reproduction Laboratory, Faculty of Animal Science, Mataram University. The variables observed included motility, viability and morphology of spermatozoa. The results of studied that the spermatozoa motility of Brahma, Chicin and Bangkok rooster were 72 ± 2.74%. 74 ± 2.24% and 73 ± 2.74% (P> 0.05) respectively; The spermatozoan viability of Brahma rooster and Bangkok rooster was 99.2 ± 1.30%, 99.4 ± 0.55% and 99.2 ± 1.10% (P>0.05)respectively. The spermatozoan morphology of brahma rooster, chicin rooster and Bangkok rooster was 94.6 ± 2.30%, 94.4 ± 2.70% and 97.4 ± 2.07% (P> 0.05), respectively. Cochin rooster and Bangkok rooster respectively, 62.6 ± 7.92 x 107, 57 ± 5.83 x107 , and 65.2 ± 12.28 x107, respectively. The results of the study concluded that Brahma, Cochin and Bangkok rooster had good sperm quality and could be able to used in artificial insemination of Kampung hens.

Semen was collected from four rams, using artificial vagina and viability%, motility% and plasma membrane integrity% were measured. Fresh ejaculates (n = 32) were separated by modified swim-up separation using modified human tubal fluid medium. Four fractions of supernatant were collected at 15-minute intervals. The mean volume, mass activity, concentration, motility%, viability%, normal morphology and membrane integrity% (HOST +ve) of fresh semen were 1.0 ± 0.14, 4.1 ± 0.1 × 109 spermatozoa/ml, 85.0 ± 1.3, 89.4 ± 1.0, 85.5 ± 0.7, 84.7 ± 0.5 respectively. There was no significant (P>0.05) difference in fresh semen quality parameters between rams. The motility%, viability% and HOST +ve % of first, second, third and fourth fractions were 53.4 ± 0.5, 68.2 ± 0.3, 74.8 ± 0.3 and 65.5 ± 0.4; 55.5 ± 0.4, 66.2 ± 0.4, 74.5 ± 0.3 and 73.6 ± 0.3 and 66.7 ± 0.5, 66.8 ± 0.5, 65.2 ± 0.4 and 74.7 ± 0.5 respectively. The motility%, viability% and membrane integrity% of separated semen samples differed significantly (P<0.05) between four fractions. The mean motility% and viability% were significantly higher (P<0.05) in third fraction (74.8 ± 0.3%), whereas the mean HOST +ve% was significantly higher (P<0.05) in fourth fraction (74.7 ± 0.5). All quality parameters of separated spermatozoa were significantly (P<0.05) lower than that of fresh semen. ABSTRACT The aim of this research was to determine the proportion, viability and motility of X and Y ram spermatozoa separated with egg white albumin. Sperm samples derived from Garut ram, which was collected by using an artificial vagina. Observations were made on spermatozoa fraction above and below each medium fraction treatment. There are treatment egg white albumin as separation medium, each medium consisting of fractions top and bottom fraction with different concentration: 1) P0 = sperma before separation (control); 2) P1 = 10% above fraction + 30% lower fraction; P2 = 25% + 45%; P3 = 25% + 75%. Data proportion of X and Y, viability and motility were analyzed statistically by Completely Randomized Design patern in the direction followed by Duncan’s Multiple Range Test for data with a real difference. Separation with egg white albumin affect significantly increased the proportion of spermatozoa X and Y (P≤0.05), but tends to decrease the viability and motility of spermatozoa.The proportion of spermatozoa X and Y was highest in treatment P3,76.76% of spermatozoa X (fraction above 25%) and 79.81% spermatozoa Y (75% lower fraction), with an average viability obtained respectively 68,9% (fraction above) and 59,7% (bottom fraction), motility 77,5% (fraction above) dan 84,0% (bottom fraction). It was concluded that the egg white albumin is very effective in changing the proportions of X and Y ram sperm with the quality of spermatozoa after separation feasible for applications insemination or processed into frozen semen.  (Keywords: Garut ram, White egg albumin, Spermatozoa X and Y)

<p class="MDPI17abstract"><strong>Objective: </strong>The purpose of this research was to find out the rise of spermatozoa motility of Waringin sheep which was given additional amount of PGF2α in commercial semen diluents (Andromed).</p><p class="MDPI17abstract"><strong>Methods: </strong>A total of 3 Waringin sheep aged 2-3 years were used in this study. Semen samples were collected using an artificial vagina and evaluation of the quality of the spermatozoa was carried out macroscopically and microscopically. This study used a unidirectional completely randomized design. After evaluated, semen samples were added with Andromed diluents afterwards divided into three groups treatment, K1; K2; and K3 where each group was then added with physiologic NaCl, 37,5 µg PGF2α, and 75 µg PGF2α. All of the samples were equilibrated in refrigerator for 4 hours and spermatozoa motility was examined, then after the freezing process was carried out and stored for 24 hours, the post thawing spermatozoa motility was examined. The data obtained was analyzed using one-way analysis of variance (ANOVA) and followed by Duncan test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The result of the research showed that motility of post equilibration spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 43,67±1,20; 75,00±4,00; and 82,33 ±3,18 (P&lt;0,05); and motility of post thawing spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 25,33±1,85; 49,33±3,17; dan 54,00 ±5,50 (P&lt;0,05).<strong></strong></p><strong>Conclusions: </strong>In conclusion that was the rise in motility of prefreezing spermatozoa and motility of post thawing spermatozoa of Waringin sheep which given additional amount of PGF2α on Andromed diluents.

This study was conducted to examine the comparison the quality of spermatozoa in four genetic groups of poultry (Pelung chicken, Nunukan chicken, Sentul chicken, and Bangkok chicken). The parameters of this study were the concentration of spermatozoa, motility, viability, semen volume, color, and consistency. The study was designed using a Completely Randomized Design (CRD) with four local chicken nations and four replications. The results showed that the volume of Nunukan chicken semen was 0.32±0.01 mL/ejaculation, Bangkok chicken was 0.31±0.01 mL/ejaculate, Sentul chicken was 0.15±0.02 mL/ejaculate and Pelung chicken was 0.23±0.02 mL/ejaculation. the color of fresh semen is white (Pelung chicken, Nunukan chicken and Sentul chicken) except chicken Bangkok has the color of cream semen. Pelung chicken has a concentration of spermatozoa 5,043.33±51 million/mL, Nunukan chicken 3,250.22 ±45 million/mL, Sentul chicken 3,002.87±67 million/mL, and Bangkok chicken 3,002.87 ± 67 million/mL. Motility of Pelung chicken 84.,69 ± 1.12%, Bangkok chicken 82.35±1.85%, Nunukan chicken 77.74±1.57% and Sentul chicken 77.64±1.65%. Viability spermatozoa of Sentul chicken was 90.35 ± 1.21%, Bangkok chicken was 90.64 ± 1.16%, Pelung chicken was 89.17 ± 1.23% and Nunukan chicken was 86.29 ± 1.15%. It can be concluded that the motility spermatozoa of Pelung chicken and Bangkok chicken is better than the spermatozoa motility of Nunukan chicken and Sentul chicken. The viability of Sentul chicken and Bangkok chicken was higher compared to the viability semen of chicken Pelung and chicken Nunukan.

Abstrak Penelitian ini bertujuan untuk mengetahui kualitas semen segar Kambing Kacang pejantan pada penyimpanan suhu 5oC dengan lama simpan yang berbeda menggunakan pengencer dan tanpa pengencer. Penelitian ini dilakukan di Laboratorium Fakultas Peternakan Universitas Kanjuruhan Malang pada tanggal 6 sampai 11 Februari 2012. Metode penelitian yang digunakan adalah metode percobaan dengan menggunakan Rancangan Acak Lengkap (RAL) Faktorial. Materi penelitian yang digunakan adalah semen segar Kambing Kacang. Berdasarkan hasil penelitian menunjukkan bahwa kualitas spermatozoa Kambing Kacang pada suhu 5oC dengan lama simpan yang berbeda menggunakan pengencer dan tanpa pengencer menunjukkan adanya perbedaan yang sangat nyata (P<0.01). Pada pengamatan motilitas dan viabilitas spermatozoa kambing Kacang pada suhu 5oC baik yang menggunakan pengencer maupun tanpa pengencer menunjukkan adanya perbedaan yang sangat nyata (P<0.01). Tetapi pada pengamatan abnormalitas tidak menunjukkan adanya pengaruh penggunaan pengencer (P>0.01) serta tidak terdapat pengaruh interaksi antara lama simpan dan penggunaan pengencer terhadap abnormalitas spermatozoa kambing Kacang (P>0.01). Namun terdapat pengaruh yang sangat nyata lama simpan terhadap abnormalitas spermatozoa kambing Kacang pada suhu 5oC baik yang menggunakan pengencer maupun tanpa pengencer (P<0.01). Kesimpulan yang diperoleh dari hasil penelitian ini yaitu  bahwa penyimpanan semen segar kambing Kacang pada suhu 50C tanpa menggunakan pengencer mempunyai kualitas yang paling baik yaitu mampu bertahan sampai lama simpan ke 42 jam.   Abstract  This research aimed to determine the quality of a fresh semen Kacang goat at 50C temperature the various preservings time within and withouth extender.  This research had been performed in Animal Husbandry Faculty of  Kanjuruhan University Malang since 6 until 11 February 2012. The research method used was Completely Randomized Design (CRD) Factorial. Research used was fresh semen Kacang goat. The result of the research indicated that spermatozoa quality of Kacang goat at 50C in different preservings time within and without extender showed the significant differentcy (P<0,01). In motility and viability observation spermatozoa of Kacang goat at 50C either with and without extender showed the significant differntcy  (P<0,01). But at the abnormality observation it did not show the influence of the usage extender (P>0,01) and there was no interaction influence between the preserving time and the extender usage forwards the abnormality of Kacang goat spermatozoa (P>0,01). But there was a significant influence of preserving time to wards the abnormality of spermatozoa at 50C temperature either with extender and withoud extender (P<0,01). This research result concludes that the Kacang goat fresh semen preserving without using extender is the best quality that it is able to stay freshly until 42 hours.

The purpose of this study was to observe the macroscopic and microscopic quality of Bronze turkey fresh semen on various ages. The observed toms were at the ages of 9 to 10 months (P1), 13 to 14 months (P2), and 17 to 18 months (P3), with each taken from 3 different toms. The collection was done by abdominal massage, and collected for 3 times a week in triplicate. This data was analyzed by analysis of variance (ANOVA) in a completely randomized design, followed by Duncan's New Multiple Range Test (DMRT) if there was any difference. The results showed that the average volume of turkey fresh semen were P1 (0.16 ± 0.04 ml), P2 (0.13 ± 0.02 ml), and P3 (0.10 ± 0.02 ml). The average pH of fresh semen were P1 (7.19 ± 0.05), P2 (7.12 ± 0.10), and P3 (7.06 ± 0.06). The average motility percentage of fresh semen were P1 (78.11 ± 1.38%), P2 (82.55 ± 1.17%), and P3 (74.55 ± 4.16%). The average viability percentage of fresh semen were P1 (78.63 ± 1.55%), P2 (83.26 ± 1.54%), and P3 (79.20 ± 1.06%). The average abnormal percentage of spermatozoa were P1 (3.01 ± 0.04%), P2 (3.04 ± 0.88%), and P3 (3.21 ± 0.44%). In conclusion, the best fresh semen quality is found on toms aged from 13 to14 months (P2). Furthermore, at the age 17 to18 months and more, the semen quality was decreased.

<p class="IsiAbstrakIndo">Aceh swamp buffaloes<span lang="AR-YE" dir="RTL"> </span><em>(Bubalus bubalis) </em>are decreasing their population and genetic quality. This study was conducted to determine the influence of lactose and glycerol cryoprotectants on spermatozoa of Aceh swamp buffaloes<span lang="AR-YE" dir="RTL"> </span><span lang="EN-GB">after</span><span lang="AR-YE" dir="RTL"> </span>thawing<em>.</em><span lang="AR-YE" dir="RTL"> </span><span lang="EN-GB">This study used completely</span>a factorial randomized design with nine treatments, and five replications. Fresh semen of the Aceh swamp buffalo were diluted by using a combination extender lactose cryoprotectants 0 mM (L<sub>0</sub>), 60 mM (L<sub>60</sub>), 120 mM (L<sub>120</sub>) and glycerol 3% (G<sub>3</sub>), 5% (G<sub>5</sub>), 7% (G<sub>7</sub>) with the equilibration of 4 hours. The results showed that the combination of cryoprotectants L<sub>120</sub>G<sub>7 </sub>influenced significantly (P &lt; 0.05) on the quality of spermatozoa of the Aceh swamp buffalo <em>(B. bubalis)</em><span lang="AR-YE" dir="RTL"> </span><span lang="EN-GB">after</span><span lang="AR-YE" dir="RTL">  </span>thawing<em>.</em><span lang="AR-YE" dir="RTL"> </span><span lang="EN-GB">The percentage of sperm motility L</span><sub>120</sub>G<sub>7</sub> (42.60 ± 1.14); viability L<sub>120</sub>G<sub>7 </sub>(55.00 ± 0.71);  acrosome integrity L<sub>120</sub>G<sub>7</sub> (52.00 ± 0.71); and plasma membrane integrity L<sub>120</sub>G<sub>7</sub><span lang="AR-YE" dir="RTL"> (</span>53.20 ± 1.48).  The combination of lactose cryoprotectants 120 mM (L<sub>120</sub>) and glycerol 7% (G<sub>7</sub>) was the best combination to maintain the quality of spermatozoa of swamp buffalo. This finding could be used to define a policy for the spermatozoa storage of Aceh swamp buffalo to artificial insemination (AI). </p>

The objective of this research was to investigate the effect of flamboyant flower extract as a natural antioxidant on sperm quality of Aceh cattle after dilution and equilibration. Fresh semen was collected from 2 healthy bull aged 3.5-4 years old. The research took place in BIB Laboratory. The method used was CRD (Completed Randomized Design), with 6 treatments and 5 replications each The treatments (m/v) were 0%, 0.05%, 0.10%, 0.15% and 0.20% of flamboyant flower extracts. The method used was CRD (Completed Randomized Design), with 6 treatments and 5 replications each. The treatments (m/v) were 0%, 0.05%, 0.10%, 0.15% and 0.20% of flamboyant flower extracts. The data obtained were analyzed by analysis of variance (ANOVA). Parameters evaluated in this study were motility, viability, plasma membrane integrity, and abnormality. Results showed that there was an insignificant effect (p>0.05) of flamboyant flower extract on the quality of Aceh cattle spermatozoa. The best percentage of motility and viability, 80.0% and 88.4%, respectively, were found at P3 (0.15% flamboyant flower extract). In can be concluded from the research that the addition of 0.15% (m/v) D. regia flower extract was able to maintain sperm quality of Aceh cattle to be used in the artificial insemination program.

This study aimed to find out the quality of fresh semen from Sumba Ongole (SO) bulls which was diluted with Bestvile Thawing Solution (BTS) diluent modified with soybean milk (SKD). The study used a completely randomized design consisting of four treatments and 5 replications, P0 (100 % BTS), P1 (95 % BTS + 5 % SM), P2 (90 % BTS + 10 % SM), and P3 (85 % BTS + 15 % SM). The parameters observed were the colour, smell, consistency, pH, volume, motility, viability, and abnormalities of spermatozoa. The results showed that the characteristics were in the normal, creamy white semen colour, semen volume 3,6 ml, medium consistency, pH 6,3, typical SO bulls smell, sperm motility 85 %, mass activity +++, sperm concentration 1276 million/ml, live sperm 90,05 % and sperm abnormalities 6,15 % and quality there was no significant difference between the four treatments. The spermatozoa motility and viability in each treatment was able to which achieved on day 1. The characteristics of the spermatozoa of SO bulls were in the normal category. Meanwhile, BTS diluent with the addition of SKD was unable to maintain sperm quality.
