Research on the evaluation of the spermatozoa quality of Brahma, Cochin and Bangkok chickens as the basis for the application of artificial insemination technology in native chickens has been carried out. The purpose of this study was to evaluate the spermatozoa quality of Brahma, Cochin and Bangkok rooster as the basis for determining the spermatozoa dose for artificial insemination in local chickens. method research was used a completely randomized design (CRD) with three treatments, that is: Brahma, Cochin and Bangkok rooster. Each treatment was repeated five times. Cement storage is carried out by inserting an artificial vagina into the cloaca of the rooster and stimulating it to climb the hen. The collected sperm were analyzed at the Animal Reproduction Laboratory, Faculty of Animal Science, Mataram University. The variables observed included motility, viability and morphology of spermatozoa. The results of studied that the spermatozoa motility of Brahma, Chicin and Bangkok rooster were 72 ± 2.74%. 74 ± 2.24% and 73 ± 2.74% (P> 0.05) respectively; The spermatozoan viability of Brahma rooster and Bangkok rooster was 99.2 ± 1.30%, 99.4 ± 0.55% and 99.2 ± 1.10% (P>0.05)respectively. The spermatozoan morphology of brahma rooster, chicin rooster and Bangkok rooster was 94.6 ± 2.30%, 94.4 ± 2.70% and 97.4 ± 2.07% (P> 0.05), respectively. Cochin rooster and Bangkok rooster respectively, 62.6 ± 7.92 x 107, 57 ± 5.83 x107 , and 65.2 ± 12.28 x107, respectively. The results of the study concluded that Brahma, Cochin and Bangkok rooster had good sperm quality and could be able to used in artificial insemination of Kampung hens.

<p class="MDPI17abstract"><strong>Objective: </strong>The purpose of this research was to find out the rise of spermatozoa motility of Waringin sheep which was given additional amount of PGF2α in commercial semen diluents (Andromed).</p><p class="MDPI17abstract"><strong>Methods: </strong>A total of 3 Waringin sheep aged 2-3 years were used in this study. Semen samples were collected using an artificial vagina and evaluation of the quality of the spermatozoa was carried out macroscopically and microscopically. This study used a unidirectional completely randomized design. After evaluated, semen samples were added with Andromed diluents afterwards divided into three groups treatment, K1; K2; and K3 where each group was then added with physiologic NaCl, 37,5 µg PGF2α, and 75 µg PGF2α. All of the samples were equilibrated in refrigerator for 4 hours and spermatozoa motility was examined, then after the freezing process was carried out and stored for 24 hours, the post thawing spermatozoa motility was examined. The data obtained was analyzed using one-way analysis of variance (ANOVA) and followed by Duncan test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The result of the research showed that motility of post equilibration spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 43,67±1,20; 75,00±4,00; and 82,33 ±3,18 (P&lt;0,05); and motility of post thawing spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 25,33±1,85; 49,33±3,17; dan 54,00 ±5,50 (P&lt;0,05).<strong></strong></p><strong>Conclusions: </strong>In conclusion that was the rise in motility of prefreezing spermatozoa and motility of post thawing spermatozoa of Waringin sheep which given additional amount of PGF2α on Andromed diluents.

The purpose of this research was to examine the effectivity of Tris, sodium citrate, and lactose extenders in maintaining viability of etawa crossbreed (EC) goat spermatozoa preserved at 3–5oC.  Semen was collected using artificial vagina once a week for six times as replicate.  Fresh semen was divided into three tubes then diluted with Tris, sodium citrate, and lactose extenders, respectively.  Diluted-semen were stored in refrigerator at 3–5oC.  Quality of diluted-semen including percentages of motile spermatozoa (MS) and live spermatozoa (LS) were evaluated every day during storage at 3–5oC for four days.  Data were analyzed using completely randomized design with three treatments and six replicates.  Results of this study showed that mean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS, and spermatozoa abnormal of PE goat fresh semen were 1.05 ml, cream, thick, 6.9, ++ – +++, 3,756.67 million cell/ml, 76.67%, 85.83%, and 4.5%, respectively.  Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  At day-4 storage, percentages of MS and LS for Tris (44.17 and 57.83%) was no difference significant with sodium citrate (43.33 and 56.67%), and lactose (43.33 and 54.17%).   In conclusion, Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  Tris, sodium citrate, and lactose extenders could be maintaining 40% in percentage of motile spermatozoa for three days after preserved at 3–5oC, and suitable for use in artificial insemination (AI) programKeywords: Tris, sodium citrate, lactose, spermatozoa viability, PE goat.

This study aimed to find out the quality of fresh semen from Sumba Ongole (SO) bulls which was diluted with Bestvile Thawing Solution (BTS) diluent modified with soybean milk (SKD). The study used a completely randomized design consisting of four treatments and 5 replications, P0 (100 % BTS), P1 (95 % BTS + 5 % SM), P2 (90 % BTS + 10 % SM), and P3 (85 % BTS + 15 % SM). The parameters observed were the colour, smell, consistency, pH, volume, motility, viability, and abnormalities of spermatozoa. The results showed that the characteristics were in the normal, creamy white semen colour, semen volume 3,6 ml, medium consistency, pH 6,3, typical SO bulls smell, sperm motility 85 %, mass activity +++, sperm concentration 1276 million/ml, live sperm 90,05 % and sperm abnormalities 6,15 % and quality there was no significant difference between the four treatments. The spermatozoa motility and viability in each treatment was able to which achieved on day 1. The characteristics of the spermatozoa of SO bulls were in the normal category. Meanwhile, BTS diluent with the addition of SKD was unable to maintain sperm quality.

Abstract: Zinc is a cofactor of more than 100 enzymes that are essential for the metabolism of nucleic acid and protein synthesis. Zinc plays an important role in growth and development, immune response, neurological and reproductive functions. Monosodium glutamate (MSG) is the sodium salt of glutamic acid , a food flavor enhancer in the form of L-glutamic acid. The purpose of this study was to determine the effect of zinc on sperm quality adult male wistar (Rattus norvegicus), following administration of monosodium glutamate (MSG). This study is experimental with a completely randomized design were conducted from November to Desember 2014 using a sample of 6 male wistars as a control group (P0) with 2 mg MSG being administered. The P1 group consist of 2 male wistars with 2 mg of MSG and zinc at a dose of 10 mg as treatment were administered to this group. The last group P2 also consist of 2 male wistars with the administration 2 mg of MSG and 20 mg doses of zinc as treatment.This study results in an increasement of spermatozoa consentration in the treatment group (P2), accounted for 78,25x106 as well as the increased of spermatozoa motility of 43,5 % for the treadment group (P2). Morfologically, 83,5 % of normal spermatozoa were accounted from the same group. Conclusion: Administration of zinc can improve the quality of male wistar spermatozoa following the administration of MSG, which includes concentration, motility and morphology. The increasing dosage of zinc being administered showed higher impact in improving the quality of spermatozoa.Keywords: zinc, MSG, male wistar, the quality of spermatozoa.Abstrak: Zink adalah kofaktor lebih dari 100 enzim yang penting untuk metabolisme asam nukleat dan sintesis protein. Zink berperan penting dalam pertumbuhan dan perkembangan, respon kekebalan, fungsi neurologis dan reproduksi. Monosodium glutamat (MSG) adalah garam natrium dari asam glutamat (glutamic acid), sebagai penambah rasa makanan dalam bentuk L-glutamic acid. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian zink terhadap kualitas spermatozoa wistar jantan dewasa (Rattus norvegicus) yang telah diberi monosodium glutamat (MSG). Penelitian ini bersifat eksperimental dengan rancangan acak lengkap (completely randomized design) yang dilakukan pada bulan November sampai Desember 2014 dengan menggunakan sampel sebanyak 6 ekor terdiri dari 2 ekor wistar pemberian MSG dengan dosis 2 mg sebagai kontrol (P0), 2 wistar pemberian MSG dengan dosis 2 mg dan zink dengan dosis 10 mg sebagai perlakuan (P1), dan 2 wistar pemberian MSG dengan dosis 2 mg dan zink dengan dosis 20 mg sebagai perlakuan (P2). Hasil penelitian ini di dapatkan peningkatan konsentrasi spermatozoa pada kelompok perlakuan P2 sebesar 78,25 x 106, peningkatan motilitas spermatozoa kategori A pada perlakuan kelompok P2 sebesar 43,5% dan morfologi normal spermatozoa pada kelompok perlakuan P2 sebesar 83,5%. Simpulan: Pemberian zink dapat menyebabkan perbaikan pada spermatozoa wistar jantan yang sebelumnya telah di beri MSG meliputi konsentrasi, motilitas, dan morfologi. Peningkatan dosis zink yang diberikan menunjukkan adanya perbaikan kualitas spermatozoa yang semakin meningkat juga.Kata kunci: zink, MSG, wistar jantan, kualitas spermatozoa.

ABSTRACT The aim of this research was to determine the proportion, viability and motility of X and Y ram spermatozoa separated with egg white albumin. Sperm samples derived from Garut ram, which was collected by using an artificial vagina. Observations were made on spermatozoa fraction above and below each medium fraction treatment. There are treatment egg white albumin as separation medium, each medium consisting of fractions top and bottom fraction with different concentration: 1) P0 = sperma before separation (control); 2) P1 = 10% above fraction + 30% lower fraction; P2 = 25% + 45%; P3 = 25% + 75%. Data proportion of X and Y, viability and motility were analyzed statistically by Completely Randomized Design patern in the direction followed by Duncan’s Multiple Range Test for data with a real difference. Separation with egg white albumin affect significantly increased the proportion of spermatozoa X and Y (P≤0.05), but tends to decrease the viability and motility of spermatozoa.The proportion of spermatozoa X and Y was highest in treatment P3,76.76% of spermatozoa X (fraction above 25%) and 79.81% spermatozoa Y (75% lower fraction), with an average viability obtained respectively 68,9% (fraction above) and 59,7% (bottom fraction), motility 77,5% (fraction above) dan 84,0% (bottom fraction). It was concluded that the egg white albumin is very effective in changing the proportions of X and Y ram sperm with the quality of spermatozoa after separation feasible for applications insemination or processed into frozen semen.  (Keywords: Garut ram, White egg albumin, Spermatozoa X and Y)

Quality of spermatozoa motility and viability from rejected limousin bull semen diluted with skim milk egg yolk sitrat added with various levels of glucose. Glucose level used were 0%, 0,5%, 1,5%, 2,5%, and 3,5%. Writer was using on four years old Limousine bull. Bull semen used in this research was bull rejected semen with bellow 70% motility. Semen observation was done at 0 hour, 24 hours and 48 hours. Research design used in this study was completely randomized design with faktorial pattern with 5 replicates. Highest result in motility this research was showed at 24 hours with 30% value in glucose 2,5% treatment and 48 hours with 10% value. The lowest result showed in glucose 0% treatment at 24 hours and 0% at 48 hours. Highest result in viability showed on glucose 2,5% treatment with 62,6% value at 24 hours and at 48 hours with 53,4% value. Lowest result in viability showed on glucose 0% treatment with 44,2% value at 24 hours and 31,4% value at 48 hours.

This study aims to determine the quality of Bali cattle spermatozoa diluted using sugarcane juice-egg yolk. This research was carried out in the stables and Laboratory of the Faculty of Agriculture, University of Timor in August-September 2020. Semen was collected from an adult male bali cattle, aged ± 4.5 years in healthy condition. The method used in this study is an experimental method using a completely randomized design consisting of 4 treatments and 5 replications. Each treatment was T1: 0.075 ml semen + 0.3 ml of egg yolk sugarcane juice diluent and stored for 24 hours, T2: semen 0.075 ml + 0.3 ml of diluent for egg yolk cane juice and stored for 48 hours, T3: semen 0.075 ml + 0.3 ml of diluent for egg yolk sugarcane juice and stored for 72 hours, T4: semen 0.075 ml + 0 ,3 ml of egg yolk sugarcane juice diluent and stored for 96 hours. The variables measured were individual motility, viability, spermatozoa abnormalities, and semen pH. The results showed that the T4 treatment on a shelf life of 96 hours still showed a good average value with individual spermatozoa motility values of 45%, spermatozoa viability of 77.3%, spermatozoa abnormalities of 12.5% and semen pH 6.6. It can be concluded that the use of cane juice-egg yolk diluent has a positive impact because it is able to maintain the viability of spermatozoa for 96 hours.

This study was conducted to examine the comparison the quality of spermatozoa in four genetic groups of poultry (Pelung chicken, Nunukan chicken, Sentul chicken, and Bangkok chicken). The parameters of this study were the concentration of spermatozoa, motility, viability, semen volume, color, and consistency. The study was designed using a Completely Randomized Design (CRD) with four local chicken nations and four replications. The results showed that the volume of Nunukan chicken semen was 0.32±0.01 mL/ejaculation, Bangkok chicken was 0.31±0.01 mL/ejaculate, Sentul chicken was 0.15±0.02 mL/ejaculate and Pelung chicken was 0.23±0.02 mL/ejaculation. the color of fresh semen is white (Pelung chicken, Nunukan chicken and Sentul chicken) except chicken Bangkok has the color of cream semen. Pelung chicken has a concentration of spermatozoa 5,043.33±51 million/mL, Nunukan chicken 3,250.22 ±45 million/mL, Sentul chicken 3,002.87±67 million/mL, and Bangkok chicken 3,002.87 ± 67 million/mL. Motility of Pelung chicken 84.,69 ± 1.12%, Bangkok chicken 82.35±1.85%, Nunukan chicken 77.74±1.57% and Sentul chicken 77.64±1.65%. Viability spermatozoa of Sentul chicken was 90.35 ± 1.21%, Bangkok chicken was 90.64 ± 1.16%, Pelung chicken was 89.17 ± 1.23% and Nunukan chicken was 86.29 ± 1.15%. It can be concluded that the motility spermatozoa of Pelung chicken and Bangkok chicken is better than the spermatozoa motility of Nunukan chicken and Sentul chicken. The viability of Sentul chicken and Bangkok chicken was higher compared to the viability semen of chicken Pelung and chicken Nunukan.

The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,27±2,75; 80,25±2,83; and 78,07±0,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,29±2,71; 80,63±1,87; and 80,09±3,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,23±0,27; 8,21±0,55; and 10,75±3,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC.

ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh lama thawing yang berbeda pada suhu 250C untuk mendapatkan kualitas spermatozoa semen beku sapi ongole yang optimal. Penelitian dilakukan di Laboratorium Fakultas Peternakan Universitas Kanjuruhan Malang. Metode Penelitian yang digunakan adalah metode percobaan dengan menggunakan Rancangan Acak Lengkap (RAL) Faktorial. Setiap perlakuan thawing diberikan ulangan sebanyak 10 sampel semen beku sapi ongole. Hasil penelitian menunjukkan bahwa perlakuan dengan lama thawing 7, 15, 30 detik pada suhu 25oC memberikan perbedaan pengaruh yang sangat nyata (p<0.01) pada motilitas, viabilitas dan abnormalitas spermatozoa semen beku sapi ongole. Motilitas tertinggi diperoleh pada perlakuan P3 dengan rata-rata sebesar 40,8%, viabilitas tertinggi diperoleh pada perlakuan P3 dengan rata-rata sebesar 82,39%, sedangkan abnormalitas terendah diperoleh pada perlakuan P3 dengan rata-rata sebesar 11,95%. Berdasarkan penelitian ini maka dapat disimpulkan bahwa thawing pada suhu 25oC dengan lama waktu 30 detik memberikan kualitas spermatozoa yang paling baik sehingga disarankan untuk melakukan thawing pada suhu 25oC dengan lama waktu 30 detik. Research conducted at the Laboratory of Animal Husbandry Faculty Kanjuruhan University of Malang. The method of study by using Completely Randomized Design (CRD) factorial. Any treatment given repeated thawing of frozen samples of 10 times. The study show that treatment with time thawing 7 (P1), 15 (P2), 30 (P3) seconds at 25oC gives a very significant difference (P<0.01) on motility, viability and abnormalities of ongole Bull sperm. The highest motility and viability were obtained at P3 (40,8%) and (82,39%), while the lowest abnormalities obtained on P3 (11,95%). Based on this research it can be concluded that thawing at 25oC with 30 seconds to give the best quality sperm that is recommended for thawing.
