Mastitis affects both dairy and meat/wool sheep industries with losses due to reductions in milk quality and quantity, increased treatment costs and restricted lamb growth. Effective vaccines would be important tools for mastitis control. However, the development of vaccines against mastitis has proved challenging due to the failure to target protective immunity to the mammary gland. In order to target responses to the mammary gland, this study tested whether local administration directly into the gland through the teat canal or in the udder skin confers protection against an intramammary infection. In this study, we tested a vaccine that confers protection against respiratory disease caused by Mannheimia haemolytica to determine if it also protects against intramammary infection by the same organism. No evidence of protection was observed in animals that received a subcutaneous immunisation in the udder skin, however, intramammary immunisation provided almost complete protection against an experimental challenge administered 7 days post immunisation but not if the challenge was delivered 14 days post immunisation. To investigate further the nature of this variation in response, the somatic cell count and concentration of cytokines Interleukin-1β, Interleukin-10 and Interleukin-17A was determined in milk over the course of each study. Intramammary immunisation induced an inflammatory response within the mammary gland, characterised by increases in SCC and in the production of cytokines IL-1β, IL-10, and IL-17A. This response was similar to that observed in un-vaccinated control animals post challenge. The SCC and cytokine levels had returned to levels comparable with un-vaccinated controls prior to challenge at both 7 and 14 days post immunisation. The transient nature of the protective effect is consistent with the priming of an innate antibacterial response within the mammary gland which provides protection against challenge at 7 days but is diminished by 14 days post-vaccination. Further studies are planned to determine the nature of the innate immune mechanisms associated with the protective effect described here to determine whether it may be exploited to improve ruminant udder health.

Chlamydia is a significant pathogen for many species, including the much-loved Australian marsupial, the koala (Phascolarctos cinereus). To combat this situation, focused research has gone into the development and refinement of a chlamydial vaccine for koalas. The foundation of this process has involved characterising the immune response of koalas to both natural chlamydial infection as well as vaccination. From parallels in human and mouse research, it is well-established that an effective anti-chlamydial response will involve a balance of cell-mediated Th1 responses involving interferon-gamma (IFN-γ), humoral Th2 responses involving systemic IgG and mucosal IgA, and inflammatory Th17 responses involving interleukin 17 (IL-17) and neutrophils. Characterisation of koalas with chlamydial disease has shown increased expression within all three of these major immunological pathways and monitoring of koalas’ post-vaccination has detected further enhancements to these key pathways. These findings offer optimism that a chlamydial vaccine for wider distribution to koalas is not far off. Recent advances in marsupial genetic knowledge and general nucleic acid assay technology have moved koala immunological research a step closer to other mammalian research systems. However, koala-specific reagents to directly assay cytokine levels and cell-surface markers are still needed to progress our understanding of koala immunology.

Objective. Inflammation is an important hallmark of all cancers and net inflammatory response is determined by a delicate balance between pro- and anti-inflammatory cytokines, which may be affected by tobacco exposure, so the present study was designed to explore the effect of various modes of tobacco exposure on interleukin-12 (IL-12) and interleukin-10 (IL-10) inflammatory cytokine levels and survival in prostate carcinoma (PCa) patients.Methods. 285 cancer patients and equal controls with 94 BPH (benign prostatic hyperplasia) were recruited; baseline levels of serum IL-12 and IL-10 were measured and analyzed in various tobacco exposed groups by appropriate statistical tool. Five-year survivals of patients were analyzed by Log-rank (Mantel-Cox) test (graph pad version 5).Results. The expression of serum proinflammatory (IL-12) and anti-inflammatory (IL-10) cytokines was correlated with tobacco exposed group as smokers, chewers, and alcohol users have shown significantly higher levels (P<0.001) with significantly lower median survivals (27.1 months, standard error = 2.86, and 95% CI: 21.4–32.62); than nonusers. Stages III and IV of tobacco addicted patients have also shown significantly increased levels of IL-12 and IL-10.Conclusions. IL-12 and IL-10 seem to be affected by various modes of tobacco exposure and inflammation also affects median survival of cancer patients.

The effect of antihypertensive and hypolipidemic therapy with the use of a fixed combination of amlodipine, lisinopril and rosuvastatin on the parameters of cytokines (the level of the proinflammatory cytokine - interleukin-1β, anti-inflammatory cytokine - interleukin-10, ligand CD40 (CD40L)), central aortic pressure, arterial stiffness and the value of albuminuria in patients with stage 3 chronic kidney disease, both in combination with arterial hypertension (AH) of 1-2 degrees, and without it, and in patients with coronary artery disease in combination with AH. The results of the study allow to conclude that the proposed antihypertensive and hypolipidemic therapy significantly reduces the parameters of central hemodynamics and vascular stiffness, the levels of microalbuminuria (MAU) and blood creatinine in patients with stage 3 chronic kidney disease in combination with AH of 1-2 degrees, and significantly increases the values IL-10 in patients with hypertension and CKD.

There is dearth of information on the haematological changes associated with Mannheimia haemolytica vaccination in goats, hence this report which describes some haematological changes observed following vaccination with intranasal Recombinant Mannheimia haemolytica vaccine in goats naturally infected with peste des petits ruminants (PPR) virus. Twenty one (male, n=11; and female, n=10) goats were assigned to three vaccinated groups (A, B and D) with five goats per group (male: 3, female: 2), while the control group had 6 goats. Group A was vaccinated once intranasally, group B was vaccinated intranasally twice at one week interval and group D was vaccinated intranasally twice at two weeks interval . The control group (C) was not vaccinated. The vaccinated and control groups were challenged by comingling with pneumonic goats to simulate the field experience. PPR virus infection was later diagnosed in all the groups post vaccination. An average of four animals per treatment group in post-vaccination days were bled once weekly for six weeks (every week) to evaluate some haematological changes. The PCV values were within the normal range, while there was a decline in lymphocyte count at week 5, and a steady increase in neutrophil count in group A. In Group B, there was similar decline in lymphocyte count from the sixth week, while in groups C (Control) and D, the lymphocyte count declined at the 7th week, as the neutrophil counts increased. There were no significant changes in monocyte and eosinophil counts. The degree of changes in lymphocyte and neutrophil counts was mild in group B and marked in group D. This study revealed that intranasal vaccination of recombinant Mannheimia  haemolytica vaccine in the presence of PPR virus outbreak results in mild hematological derangement when the goats were vaccinated with  Mannheimia haemolytica bacterin at a week interval.Keywords: Goats, Haematology, Intranasal Recombinant Mannheimia haemolytica, Peste des Petits Ruminants, Vaccination

In this study, we determined the anti-inflammatory effect of manual acupuncture at the Sanyinjiao or Spleen 6 (SP6) point on carrageenan-induced peritonitis in mice and investigated mechanisms that may underlie this effect. In the first set of experiments, male Swiss mice were allocated into five groups: the control (sterile saline), dexamethasone (DEXA), invasive sham-acupuncture (non-acupoint), SP6 acupuncture and carrageenan-treated groups. Ten minutes after needle retention or 30 min after DEXA treatment, mice received an intraperitoneal injection of carrageenan (750 μg/mouse). After 4 h, total leukocyte and differential cell counts (neutrophils and mononuclear), myeloperoxidase (MPO) activity, vascular permeability and cytokine levels were evaluated. In another set of experiments, adrenalectomized (ADX) mice were used to study the involvement of the adrenal gland on the therapeutic effects of acupuncture. Mice were allocated into two groups: the ADX and sham-operated animals (Sham ADX) that were subdivided into four subgroups each: the control (sterile saline), DEXA, SP6 acupuncture and carrageenan-treated groups. The SP6 and DEXA treatments inhibited the inflammatory cell infiltration, vascular permeability and MPO activity in carrageenan-injected mice. In addition, the SP6 treatment also increased interleukin (IL)-10 levels. In contrast, when the animals were adrenalectomized, the SP6 treatment failed to reduce total leukocyte and the plasma extravasation. In conclusion, this study clearly demonstrates the anti-inflammatory effect of SP6 acupuncture in a model of carrageenan-induced peritonitis. Our results demonstrated that SP6 acupuncture depends of the adrenal glands and increased IL-10 levels to produce its anti-inflammatory action.

Purpose To investigate variations of circulating serum levels of interleukin-6 (IL-6), tumor necrosis factor alpha (TNFα), and interleukin-10 (IL-10) during three-dimensional conformal radiation therapy (3D-CRT) in patients with non–small-cell lung cancer and correlate these variations with the occurrence of radiation pneumonitis. Patients and Methods Ninety-six patients receiving 3D-CRT for stage I to III disease were evaluated prospectively. Circulating cytokine levels were determined before, every 2 weeks during, and at the end of treatment. Radiation pneumonitis was evaluated prospectively between 6 and 8 weeks after 3D-CRT. The predictive value of clinical, dosimetric, and biologic (cytokine levels) factors was evaluated both in univariate and multivariate analyses. Results Forty patients (44%) experienced score 1 or more radiation pneumonitis. No association was found between baseline cytokine levels and the risk of radiation pneumonitis. In the whole population, mean levels of TNFα, IL-6, and IL-10 remained stable during radiotherapy. IL-6 levels were significantly higher (P = .047) during 3D-CRT in patients with radiation pneumonitis. In the multivariate analysis, covariations of IL-6 and IL-10 levels during the first 2 weeks of 3D-CRT were evidenced as independently predictive of radiation pneumonitis in this series (P = .011). Conclusion Early variations of circulating IL-6 and IL-10 levels during 3D-CRT are significantly associated with the risk of radiation pneumonitis. Variations of circulating IL-6 and IL-10 levels during 3D-CRT may serve as independent predictive factors for this complication.

An early single full-term pregnancy induces a long-lasting protective effect against mammary tumor development in humans and rodents. This protective effect can be mimicked in rats by short-term administration of estrogen and progesterone hormones prior to carcinogen administration. The hormones of pregnancy are able to induce a proliferative block upon carcinogen challenge that is not observed in the age-matched virgin. We wished to determine whether carcinogen is needed to induce a paracrine-to-autocrine shift of proliferation in steroid receptor positive cells or if such a cell population already exists in the age-matched virgin mammary gland. Here we show that estrogen receptor positive (ER+) proliferating cells are rare in the developing mammary gland of the virgin rat but represent the majority of the proliferating cells in the mature (96-day-old) mammary gland of the virgin rat. As the majority of the proliferating cells before carcinogen challenge were ER positive, the ER+ proliferating cells in the mature mammary gland may represent the target cells for carcinogen-induced transformation. Importantly, prior exposure of the mammary gland to pregnancy levels of estrogen/progesterone blocked this positive association. This ability to block the proliferation of the ER+ cells may be one factor by which pregnancy induces protection against breast cancer.

SummaryEighteen cows with a wide range of quarter peak flow rates (0·35–2·22 kg/min) were inoculated with Streptococcus agalactiae and Str. dysgalactiae 4 mm into each streak canal every 3 d for 12 d. Thirty of the 72 quarters developed intramammary infection. Mean peak flow rate and length of streak canal of those quarters that became infected were 1·26 ± 0·08 kg/min (mean ± SEM) and 11·47 ± 0·41 mm respectively, compared with 1·01 ± 0·05 kg/min and 12·05 ± 0·35 mm for those that remained uninfected. Logistic regression analysis showed that the probability of infection increased significantly with the increase in peak flow rate (P = O·O1). The influence of streak canal length on new infection, after allowing for the effect of peak flow rate, was not significant at the 5% level (P = 0·07), suggesting that there may be an inverse relationship between teat duct length and infection, but that it is of secondary importance to peak flow rate. If increased mass of milk distends the teat duct by raising intramammary pressure, then quarter susceptibility to infection is further compromised. These results strongly suggest that the benefits of reduced infection from mastitis control, achieved despite dramatic increases in milk flow rate and milk yield, are significantly underestimated.
