T-2 and HT-2 toxins are secondary metabolites of various species of Fusarium. These molecules can have high potential toxic effects for human and animal health. In this work, ELISA and ultra performance liquid chromatography with fluorescence detection (UPLC/FLD) were implemented and validated as screening and confirmatory tests for the detection of these two toxins in cereal samples. The developed methods were tested by analyzing 100 samples of cereals by ELISA screening for reducing costs and analysis time and then using UPLC/FLD for confirmation purposes. Both methods met the performance criteria for sensitivity, linearity, selectivity, precision, and ruggedness, as reported in the European Decision No. 2002/657/EC and in Regulation (EC) No. 401/2006. The correlation between ELISA and UPLC/FLD approaches showed good results (r = 0.9056), confirming that these two techniques should be considered to be complementary in the official control activities of cereal and derived products.

Eruca sativa and Brassica juncea belong to the Brassicaceae family and have been used traditionally for the treatment of cancer and various cardiovascular ailments. A rapid and simple ultra performance liquid chromatography (UPLC) method has been developed for the simultaneous quantification of erucin, allyl isothiocyanate and benzyl isothiocyanate in E. sativa and B. juncea oil. Eruca sativa oil contains erucin (28.93%), allyl isothiocyanate (0.06%) and benzyl isothiocyanate (0.70%). Identification of other secondary metabolites in E. sativa and B. juncea oil and aqueous methanolic extracts were also carried out using ultra performance liquid chromatography-electro spray ionization-quadrupole time of flight (UPLC-ESI-QTOF). The UPLC-ESI-QTOF based study leads to the identification of ten compounds in B. juncea oil and three compounds in E. sativa oil. The UPLC-ESI-QTOF analysis of aqueous methanolic extracts of E. sativa and B. juncea leads to the identification of eight and nine compounds, respectively.

The present study was conducted to obtain a comprehensive overview of oligosaccharides present in the milk of a variety of important domestic animals including cows, goats, sheep, pigs, horses and dromedary camels. Using an analytical workflow that included ultra-performance liquid chromatography–hydrophilic interaction liquid chromatography with fluorescence detection coupled to quadrupole time-of-flight MS, detailed oligosaccharide libraries were established. The partial or full characterisation of the neutral/fucosylated, phosphorylated and sialylated structures was facilitated by sequencing with linkage- and sugar-specific exoglycosidases. Relative peak quantification of the 2-aminobenzamide-labelled oligosaccharides provided additional information. Milk from domestic animals contained a much larger variety of complex oligosaccharides than was previously assumed, and thirteen of these structures have been identified previously in human milk. The direct comparison of the oligosaccharide mixtures reflects their role in the postnatal maturation of different types of gastrointestinal systems, which, in this way, are prepared for certain post-weaning diets. The potential value of animal milk for the commercial extraction of oligosaccharides to be used in human and animal health is highlighted.
