Peculiar Response in the Co-Culture Fermentation of Leuconostoc mesenteroides and Lactobacillus plantarum for the Production of ABE Solvents

Two bacterial strains (CL11A and CL11D) that are capable of ABE fermentation, identified as Leuconostoc mesenteroides and Weissella cibari, were isolated from the soil surrounding the roots of bean plants. Another strain (ZM 3A), identified as Lactobacillus plantarum, which is capable of purely ethanolic fermentation was isolated from sugarcane. Glucose was used as a standard substrate to investigate the performance of these strains in mono—and co-culture fermentation for ABE production. The performance parameters employed in this study were substrate degradation rates, product and metabolite yields, pH changes and microbial growth rates. Both ABE isolates were capable of producing the three solvents but Leuconostoc mesenteroides had a higher specificity for ethanol than Weissella cibari. The co-culturing of Leuconostoc mesenteroides and Lactobacillus plantarum enhanced ethanol production at the expense of both acetone and butanol, and also influenced the final substrate consumption rate and product yield. The experiments indicated the potential of these niche environments for the isolation of ABE-producing microorganisms. This study contributes to the formulation of ideal microbial co-culture and consortia fermentation, which seeks to maximize the yield and production rates of favored products.

This paper deals with the minimal-cost control of the modified activated sludge process with varying level of wastewater in the aerator tank. The model-based adaptive controller of the effluent substrate concentration, basing on the substrate consumption rate and manipulating the effluent flow rate outcoming from the aerator tank, is proposed and its performance is compared with conventional PI controller and open loop behavior. Since the substrate consumption rate is not measurable on-line, the estimation procedure on the basis of the least-square method is suggested. Finally, it is proved that cooperation of the DO concentration controller with the adaptive controller of the effluent substrate concentration allows the process to be operated at minimum costs (low consumption of aeration energy).

Soybeans and soy-based products contain isoflavones which can be used for nutraceutical and medical applications. In soybeans and in unfermented soy foods, isoflavones are normally present as glycosides. Isoflavone glycosides can be enzymatically converted to isoflavone aglycones, thus releasing the sugar molecule. The effective absorption of isoflavones in humans requires the bioconversion of isoflavone glycosides to isoflavone aglycones through the activity of the enzyme β-glucosidase. The objective was to assess the capacity of 42 bacterial strains (belonging to Lactobacillus, Streptococcus and Enterococcus) to produce β-glucosidase activity. The strain that showed the highest β-glucosidase activity (Lactobacillus plantarum 128/2) was then used for the optimization of the bioconversion of genistin and daidzin present in commercial soymilk to their aglycone forms genistein and daidzein. The contribution of process parameters (temperature, inoculum size, time) to the efficiency of such bioactivation was tested. Lactobacillus plantarum 128/2 was able to completely bioactivate soymilk isoflavones under the following conditions: 25 °C temperature, 2% inoculum size and 48 h process time. These results confirm the suitability of lactic acid bacteria for the bioactivation of isoflavones present in soymilk and provide an interesting candidate (L. plantarum 182/2) for food industries to perform this transformation.

The microbiota in the esophageal sac of voles fed either cubed alfalfa hay or concentrate pellets were assayed to determine their capacity to anaerobically degrade mimosine in vitro. Differences (P < 0.01) were found between the two diets during the growth phase. The sac contents of voles fed concentrate pellets degraded mimosine and 3-hydroxy-4-(1H)-pyridone (DHP) rapidly, but inocula from voles fed cubed alfalfa hay only hydrolyzed mimosine to DHP. Degradation of the pyridine ring occurred at the early stage of incubation, concurrently with microbial growth. Thereafter, degradation rates appear to have been almost negligible and very similar for both diets. These results agree with previous data obtained with ruminal microorganisms, where highly active inocula were also associated with animals on concentrate diets. In this paper, the nonlinear dynamic system of microbe continuous fermentation products 1,3-propanediol (1,3-PD) is rewritten by improving the specific cellular growth rate, specific substrate consumption rate and specific product formation rate. Firstly, under the condition of substrate glycol excess and active trans-membrane transport, according to the dynamic behavior the fermentation process, we consider the glycerol and 1,3–PD concentration within the cell, and improve the specific cellular growth rate, specific substrate consumption rate and specific product formation rate, then rewrite the dynamic system of microbial continuous fermentation process. Secondly, taking the dynamic system as main constraint condition, we establish the parameter identification model and prove the existence of the optimal solution. Lastly, the numerical results calculated by particle swarm algorithm show that the improved model is suitable for describe the dynamic behavior of 1,3-PD, but is not accurate enough for by-products.

La investigación tuvo como objetivos aislar e identificar bacterias lácticas (BAL), evaluar la capacidad probiótica in vitro e in vivo de bacterias lácticas aisladas de chicha de molle, para ello se muestreó chichas de molle elaboradas artesanalmente de las provincias de Huanta y Huamanga, aislando 55 cepas BAL e identificando a Lactobacillus plantarum, Lactobacillus maltaromicus y Leuconostoc mesenteroides en base a la coloración Gram, producción de gas, gluconato y fermentación de azúcares. Para evaluar la capacidad probiótica in vitro se realizaron pruebas de antagonismo entre BAL con cuatro microorganismos patógenos (Escherichia coli ATCC 25922, Salmonella typhimurium ATCC 14028, Staphylococus aureus ATCC 25923 y Candida albicans ATCC 90028), mostrándose que 14 de las 55 cepas BAL producen sustancias inhibitorias de amplio espectro; asimismo, se evaluó la capacidad de tolerancia a condiciones gastrointestinales de cepas BAL, realizando ensayos a diferentes pHs , diferentes concentraciones de sales biliares y extracto gástrico artificial, resultando 25 cepas BAL con capacidad de tolerancia gastrointestinal y se seleccionaron cuatro cepas con mayor diámetro de halos de inhibición y cepas tolerantes a condiciones gastrointestinales siendo las cepas: BL-1 (Lactobacillus plantarum), BL-26 (Lactobacillus maltaromicus), BL-27 (Lactobacillus plantarum) y BL-53 (Lactobacillus maltaromicus), a las cuales se evaluaron la capacidad probiótica in vivo en 20 ratas para luego realizar recuento de BAL en el intestino a los 21 días, encontrándose en el grupo de estudio con BAL a 60x1019 UFC/ mL, mientras en el tratamiento con BAL más yacón a 25x1024 UFC/mL y los tratamientos de yacón y control a 50x1014 UFC/mL de BAL obteniéndose una de ganancia de peso en ratas en el grupo de estudio de BAL más yacón de 46 g, mientras con bacterias lácticas se tuvo 24 g y 16 g en el grupo control y extracto de yacón. En consecuencia esta investigación demuestra que la toma diaria de bebidas fermentadas tradicionales favorece el incremento de Lactobacillus en la microbiota intestinal.

<strong>Objective</strong>. To design a complex culture media for the production of biomass and acid phosphatases from phosphate-solubilizing bacteria isolated from soil. <strong>Materials</strong> <strong>and methods</strong>. Phosphate-solubilizing bacteria were isolated from oil palm crop soil samples and selected on SMRS1 agar, which were then assessed with antagonism tests to verify their aptitude to form a co-culture. A Box-Behnken experimental design was applied to<br />evaluate the effect of each one of the culture media components on the production of biomass and phosphatase enzymes at a laboratory scale. Finally, microbial growth and enzyme production curves were carried out in order to determine their production times. <strong>Results</strong>. Five phosphate-solubilizing bacterial strains were isolated and three of them were selected based on their solubilization indices.These Gram negative strains with bacillus morphology were identified as A, B and C; their solubilization indices were 2.03, 2.12, and 2.83, respectively. According to the ANOVA analyses for the Box-Behnken design, the only factor which had a significant effect on the phosphatase activity (p&lt;0.01) was hydrolyzed yeast, and the formulation that generated the highest biomass concentration and phosphatase activity (p&lt;0.01) contained 10, 15 and 2.5 gL-1 of phosphoric rock, sucrose and hydrolyzed yeast, respectively. After 24 hours of incubation at 100 rpm, the highest values of biomass and phosphatase activity were obtained: 11.8 logarithmic units of CFU and 12.9 phosphatase units. <strong>Conclusion</strong>. We determined that the culture media based on phosphoric rock 10 gL-1, hydrolyzed yeast 2.5 gL-1 and commercial sucrose 15 gL-1 was ideal for the production of biomass and phosphatases by the strains evaluated; likewise, we proved that the hydrolyzed yeast was the only factor significantly influential for the production of phosphatases.<br /><br /><strong>Key words</strong>: bio-inoculants, phosphate solubilizing microorganisms, phosphatase activity, Box Behnken design.

Exopolysaccharides (EPS) produced by three Lactic Acid Bacteria strains,Lactococcus lactisSLT10,Lactobacillus plantarumC7, andLeuconostoc mesenteroidesB3, were isolated using two methods: ethanol precipitation (EPS-ETOH) and ultrafiltration (EPS-UF) through a 10 KDa cut-off membrane. EPS recovery by ultrafiltration was higher than ethanol precipitation forLactococcus lactisSLT10 andLactobacillus plantarumC7. However, it was similar with both methods forLeuconostoc mesenteroidesB3. The monomer composition of the EPS fractions revealed differences in structures and molar ratios between the two studied methods. EPS isolated fromLactococcus lactisSLT10 are composed of glucose and mannose for EPS-ETOH against glucose, mannose, and rhamnose for EPS-UF. EPS extracted fromLactobacillus plantarumC7 andLeuconostoc mesenteroidesB3 showed similar composition (glucose and mannose) but different molar ratios. The molecular weights of the different EPS fractions ranged from 11.6±1.83 to 62.4±2.94 kDa. Molecular weights of EPS-ETOH fractions were higher than those of EPS-UF fractions. Fourier transform infrared (FTIR) analysis revealed a similarity in the distribution of the functional groups (O-H, C-H, C=O, -COO, and C-O-C) between the EPS isolated from the three strains.

ABSTRACT Lactobacillus plantarum WCFS1 requires both heme and menaquinone to induce respiration-like behavior under aerobic conditions. The addition of these compounds enhanced both biomass production, without progressive acidification, and the oxygen consumption rate. When both heme and menaquinone were present, L. plantarum WCFS1 was also able to reduce nitrate. The ability to reduce nitrate was severely inhibited by the glucose levels that are typically found in L. plantarum growth media (1 to 2% [vol/vol] glucose). In contrast, comparable mannitol levels did not inhibit the reduction of nitrate. L. plantarum reduced nitrate with concomitant formation of nitrite and ammonia. Genes that encode a bd-type cytochrome (cydABCD) and a nitrate reductase (narGHJI) were identified in the genome of L. plantarum. The narGHJI operon is part of a cluster of genes that includes the molybdopterin cofactor biosynthesis genes and narK. Besides a menaquinone source, isogenic mutants revealed that cydA and ndh1 are required for the aerobic-respiration-like response and narG for nitrate reduction. The ndh1 mutant was still able to reduce nitrate. The existence of a nonredundant branched electron transport chain in L. plantarum WCFS1 that is capable of using oxygen or nitrate as a terminal electron acceptor is proposed.
