Five different wines (standard Graševina, macerated Graševina with and without sulfur, rosé, and standard Plavac Mali), all typical Croatian wines, were tested to determine the antimicrobial activity against two Escherichia coli bacterial strains (ATCC® 25922 and ATCC® 8739) in vitro and using sea bass (Dicentrarchus labrax) fillets as food matrix. The chemical composition of wines (pH, acidity, alcohol, total phenolics, anthocyanins, tannins, and sulfur content) and antimicrobial activity (minimal inhibitory concentration (MIC), agar-well diffusion method) were determined. The total phenolic content of the wines ranged from 305–3210 mg gallic acid equivalents per liter (GAE/L), and did not correlate to antimicrobial activity. The two wines with the lowest phenolic content (standard Graševina and rosé) had the lowest MIC values (122 and 429 mg GAE/L). A specific relation between the winemaking process and a particular MIC value was not established. There was also no relation found between the pH value, ethanol content, sulfur, or phenolics in regards to the antimicrobial effect. In fish fillets marinated in wine + water mixture (v/v = 1:1) and inoculated with 7 log colony forming units (CFU)/25 g the growth of bacteria was reduced after three days of storage at 4 °C. Subsequent storage resulted in the growth of bacteria in all samples, with the lowest growth of E. coli ATCC® 25922 in macerated Graševina and E. coli ATCC® 8739 in standard Graševina. All wines showed the capacity to reduce the number and growth of heavily infected sea bass filets, but correlation with specific wine constituents was not found. Taking into account the numerous reactive mechanisms between food and wine, all in vitro studies in controlled laboratory conditions should be further verified in the relevant environment, and additional research is needed to clarify the role of individual wine components in the mechanism of antimicrobial activity.

ABSTRAK                                                                        Imbuhan pakan unggas dapat berasal dari bahan herbal yang mengandung berbagai komponen aktif yang bermanfaat bagi pertumbuhan ternak.Temu putih dan jahe gajah dapat dimanfaatkan sebagai imbuhan pakan karena mengandung minyak atsiri yang dapat berperan sebagai agen antibakteri. Penelitian ini bertujuan untuk mengetahui komponen penyusun minyak atsiri dan aktivitas antimikroba pada rimpang temu putih dan jahe gajah. Penelitian dilakukan dengan percobaan in vitro menggunakan temu putih dan jahe gajah yang diolah menjadi bentuk ekstrak minyak atsiri temu putih dan jahe gajah sebagai materi uji komposisi penyusun minyak atsiri serta bentuktepung dan enkapsulasi sebagai materi uji aktivitas antimikroba. Komposisi minyak atsiri temu putih terdiri dari lima komponen penyusun dengan cis-1,7-octadien-3-yl acetat sebagai komponen utama. Komposisi minyak atsiri jahe gajah terdiri dari tujuh komponen dan benzene,1-(1,5-dimethyl-4-hexenyl)-4-methyl-(CAS) ar-curcumene sebagai komponen utama. Minyak atsiri yang terkandung pada temu putih dan jahe gajah mempunyai peran dalam menghambat mikroba. Uji komposisi penyusun minyak atsiri menggunakan alat GC-MS dan uji aktivitas antimikroba menggunakan metode disc diffusion dan. Hasil dari uji aktivitas antimikroba menunjukkan bahwa temu putih dan jahe gajah dalam bentuk tepung dan enkapsulasi memiliki perbedaan yang sangat nyata (P<0,01) terhadap aktivitas antimikroba pada bakteri asam laktat, Escherichia coli dan Salmonella sp. Campuran temu putih dan jahe gajah (1:1) menunjukkan kemampuan terbaik dalam menghambat pertumbuhan bakteri patogen dengan diameter zona hambat 5,70±0,14 mm  (Escherichia coli) dan 6,88±0,45 mm (Salmonella sp.).Kata Kunci : antimikroba, fitobiotik, jahe gajah, minyak atsiri, temu putihABSTRACTThe poultry feed additives can contain herbal ingredients that contain various beneficial components for livestock growth. White turmeric and giant ginger can be used as feed additives because they contain essential oils that can be used as antibacterial agents. This study aims to determine the constituent components of essential oils and antimicrobial activity in white turmeric and giant ginger rhizomes. The study was carried out by in vitro experiments using white turmeric and giant ginger which were processed into the form of essential oil extract as material for the composition of essential oils test, and powder and encapsulation form as antimicrobial activity test material. The composition of essential oils of white turmeric consists of five constituent components with cis-1,7-octadien-3-yl acetate as the main component. The composition of giant ginger essential oil consists of seven components with benzene, 1- (1,5-dimethyl-4-hexenyl) -4-methyl- (CAS) ar-curcumene as the main component. Essential oils contained in the white turmeric and giant ginger have a role in inhibiting microbes. The composition of the essential oil tested using GC-MS and the antimicrobial activity test used the disc diffusion method. The results of the antimicrobial activity test showed that white turmeric and giant ginger in powder and encapsulation form had significant differences (P <0.01) on antimicrobial activity in lactic acid bacteria, Escherichia coli and Salmonella sp. The mixture of white turmeric and giant ginger (1: 1) showed the best ability to inhibit the growth of pathogenic bacteria with inhibitory zone diameters of 5.70 ± 0.14 mm (Escherichia coli) and 6.88 ± 0.45 mm (Salmonella sp.).Keywords: antimicrobial, essential oil, giant ginger, phytobiotic, white turmeric

Objectif : L’objectif de ce travail était d’évaluer in vitro l’activité antimicrobienne des extraits de feuilles et tige de Jatropha multifida sur la croissance de Candida albicans, Escherichia coli et Staphylococcus aureus, puis d’évaluer in vivo la toxicité de cette plante. Méthodologie et résultats : Les méthodes de diffusion en milieu gélosé et de microdilution en milieu liquide ont été utilisées pour évaluer l’effet antimicrobien. Une étude en subaigüe était réalisée afin d’explorer les effets toxiques de l’extrait aqueux des feuilles. Les résultats des tests antimicrobiens montrent une activité des extraits de feuilles et tige de J. multifida sur la croissance des souches utilisées avec des diamètres de zones d’inhibition allant de 8 à 25 mm et des concentrations minimales inhibitrices (CMI) variant de 0,039 mg/mL à 1,25 mg/mL à l’exception des souches de E. coli qui sont résistantes aux extraits de la tige. L’administration en subaigüe de l’extrait aqueux des feuilles de J. multifida à la dose de 600 mg/kg entraîne une perte significative de poids chez les souris. Conclusion et applications des résultats : Les extraits aqueux, éthanolique et hydroéthanolique des feuilles et tige de J. multifida possèdent d’activité antimicrobienne et pourraient être utilisés dans le traitement des Candidoses à C. albicans et des infections à S. aureus. Mais l’essai de toxicité subaigüe montre que l’extrait aqueux de la plante serait toxique. Des études toxicologiques approfondies restent donc nécessaires sur ces extraits afin de mieux élucider leur inocuité. Conclusion and application of findings : The aqueous, ethanolic and hydroethanolic extracts of the leaves and stem of J. multifida have antimicrobial activity and could be used in the treatment of Candidiasis and bacterial infections due respectively to C. albicans and S. aureus. But the subacute toxicity test shows that the aqueous extract of the plant would be toxic. Extensive toxicological studies therefore remain necessary on these extracts in order to better elucidate their safety. AbstractObjective: Scientists are looking for new resources which have biological activities. The present study was planned to evaluate the antioxidant and antimicrobial activity of ethanol and acetone extracts, as well as the volatile compounds of two freshwater green algal species Spirogyra spp. and Zygnema stellinum (Vaucher) C. Agardh belonging to Zygnemaphyceae (Conjugatophyceae) obtained from Ordu University Campus wetlands.Methods: The extracts were tested in vitro for their antimicrobial effects using disc diffusion method. Total phenolic content and the antioxidative activity according to FRAP and DPPH methods of the extracts were also determined. The secondary metabolites from the investigated extracts were identified using GC-MS.Results: The extracts dramatically inhibited almost all tested microorganisms. The antimicrobial activity of the acetone extract of the Zygnema against C. albicans was found as more higher than positive control Nystatin. In accordance with antimicrobial activity, the highest total phenolic content was also determined in the presence of the acetone extract of the Zygnema. Furthermore the highest FRAP value and the lowest EC50 (mg/mL) value were calculated for the same extract. The main components of the all consisted of dimethyl and diethyl ester of 1,2-benzenedicarboxylic acid. However, the abundance of these metabolites in the extracts was not associated with antimicrobial or antioxidant activity. Biological activities of these algal species could be attributed to chemicals such as 1-Pentadecene and 1-Tetradecene which were present in smaller amounts.Conclusion: Investigated algal species can be evaluated to use in biotechnological applications such as food industry and medicine.

AbstractIn vitro antimicrobial activity of 21 crude extracts obtained from seven taxa of the genus Teucrium (T. chamaedrys, T. montanum, T. arduini, T. polium, T. scordium subsp. scordium, T. scordium subsp. scordioides and T. botrys) was tested against bacterial and fungal species. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using a microdilution analysis method. Total phenolic content and flavonoid concentrations were measured spectrophotometrically. Total phenols were determined using Folin-Ciocalteu reagent and their amounts ranged from 28.49 up to 159.84 mg CA/g of extract (chlorogenic acid equivalent). The amounts of flavonoids ranged from 38.17 up to 190.45 mg RU/g of extract (rutin equivalent).The plant extracts showed greater potential of antibacterial than antifungal activity. A relationship was found between total phenolics and biological activity. The highest level of total phenols was measured in the methanol extracts, which demonstrated higher antimicrobial activity than acetone and ethyl acetate extracts. Staphylococcus aureus ATCC 25923 appeared to be the most sensitive organism. Our results indicate that Teucrium spp extracts are rich sources of phenolic compounds and are promising candidates for further development as natural antimicrobial agents.

In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

Background and aim: The Cameroonian flora abounds in plants with multidimensional therapeutic virtues which can play an important role in the fight against resistance to antibiotics and the search for new antimicrobials. This study aimed to screen the antimicrobial potential of aqueous and ethanolic extracts of thirteen (13) samples (Bark, leaf, seed) of eight (8) plants from Cameroon against 3 reference pathogens and to evaluate their synergy with conventional antibiotics against eleven (11) multiresistant uropathogenic (MRU) bacteria. Method: Bioactive compounds were extracted from leaves of Leucanthemum vulgare, Cymbopogon citratus (DC.) Stapf, Moringa oleifera Lam and Vernonia amygdalina Delile; barks of Cinchona officinalis and Enantia chlorantha Oliv; barks and seeds of Garcinia lucida Vesque and leaves and seeds of Azadirachta indica (Neem) using distilled water and ethanol as solvents. The extracts were tested against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 6538 and Candida albicans 10231 using the well diffusion method and the microdilution method. The synergistic effect was assessed (using disc diffusion method and the checkerboard method) against MRU bacteria namely Achromobacter xylosoxidans 4892, Citrobacter freundii 426, Enterococcus avium 1669, Escherichia coli 1449, Klebsiella oxytoca 3003, Kocuria rizophilia. 1542, Moraxella catarrhalis 4222, Morganella morganii 1543, Pseudomonas aeruginosa 3057, Staphylococcus aureus 1449 and Streptococcus agalactiae 3984. Results: We found that distilled water extracted a more important mass of phytochemical compounds (7.9-21.2%) compared to ethanol (5.8-12.4%). Except C. officinalis and G. lucida leaves, the rest of extracts were active with inhibition diameters (ID) ranging from 5 to 36 mm. Both ethanolic (EE) and aqueous extract (AE) of E. chloranta bark (ECB) were the most active against all pathogens with the mean ID of 17 and 36 mm vs S. aureus ATCC 6538, 23 and 14 mm vs E. coli ATCC 25922 and 36 and 19 mm vs C. albicans ATCC 10231. Only the EE of E. chloranta bark (ECB) had a strong activity against all the microorganisms tested (MIC <2 mg / ml); L. vulgare leaves (LVL) and G. lucida seed (GLS) had moderate (average MIC of 8 mg/ml) while all other extracts showed very weak antimicrobial activity. In addition, the fractional inhibitory concentration (FIC) ranged from 0.125 to 0.750. No antagonism (FIC> 4) or indifference (1≤ FIC≤4) was noted between the extracts and the antibiotics, but the best synergies were found with ECB which well-modulated Kanamycin (FIC = 0.125 against S. aureus and 0.250 against E. coli), nitrofurantoin (FIC = 0.250 against S. aureus and 0.188 against E. coli) and ampicillin (FIC = 0.125 against E. coli). Similarly, compared to other extracts, ECB, LVL and GLS also well-modulated ampicillin, ceftazidime, tetracycline, nitrofurantoin, and trimethoprim against all the above-mentioned resistant uropathogenic bacteria with important increase in fold area (IFA). Conclusion: This study show that E. chlorantha bark, L. vulgare leaves G. lucida seed, have good antimicrobial activity against both bacteria (Gram positive and Gram negative) and fungi (C. albicans); and should be more investigated for their possible use to the fight against MDR and MRU microorganisms.

AbstractThe aim of this study is to test two different methods for evaluating the in vitro antibacterial effect of Thymus fontanesii Boiss. et Reut. essential oil against standard and clinical bacterial strains responsible for bovine mastitis: the disc diffusion method or the aromatogram which allows the demonstration of the antibacterial power of essential oils on the bacterial strains tested, Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 and two strains isolated from bovine mastitis milk S. aureus and E. coli. The inhibition activity of the essential oil of T. fontanesii on bacterial strains by the two methods shows that the antimicrobial power of this oil is very important and is characterized by bactericidal and bacteriostatic action against gram negative and gram positive bacteria. The antimicrobial evaluation by the aromatogram showed good antibacterial activity against all the strains tested, the zones of inhibition of the bacteria were between 23,33±1,527mm and 37,5±3,535 mm. The search for minimum inhibitory concentrations MIC and bactericides CMB made it possible to quantitatively assess the antimicrobial power of this essential oil. In this work, the MIC was 0,625 µl/ml for all strains tested, and the lowest CMB was that of T. fontanesii against E. coli ATCC 25922 was 0,625 µl/ml.

Aim: The study evaluates the in-vitro antimicrobial activity of Hunteria umbellata against Escherichia coli, Staphylococcus aureus and Streptococcus sp. Place and Duration of Study: The study was carried out for three months in 2019 in Biochemistry Laboratory, Department of Chemical Sciences (Biochemistry unit), School of Pure and Applied Sciences, Lagos State Polytechnic, Ikorodu, Lagos- Nigeria. Methodology: The qualitative and GC-MS analysis of Hunteria umbellata methanolic seed extract were determined using standard procedure. The antimicrobial activity was evaluated by the disc diffusion method and agar well diffusion method. The experimental data was resampled 1000 times to allow for higher degrees of freedom in carrying out t-test to test for the difference of the effect of in-vitro antimicrobial activity of H. umbellata against E. coli, S. aureus and Streptococcus sp using mathematical software R language (3.6.1 version). Line plots, histogram and t-test are used to explain the effect of antimicrobial activity of H. umbellate on the selected bacteria. MIC and MBC were determined using standard methods. Results: The Phytochemical analysis of methanolic seed extract of Hunteria umbellata showed the presence of secondary metabolites like saponins, tannins, flavonoids, steroids, phenol among others. GC-MS assay of the H. umbellata seed extract revealed the presence of eight different compounds. Agar well diffusion method was characterized by inhibition zones of 18.36±0.87, 19.13±1.03 and 21.62±2.53 mm for E.coli, S. aureus and Streptococcus sp respectively at 300 mg/ml-1 and 21.70± 1.60, 23.83± 2.64 and 28.57± 1.52 for E.coli, S. aureus and Streptococcus sp respectively at 500 mg/ml. The results of the analysis show that there is a significant difference between the effects of in-vitro antimicrobial activity of H. umbellate on 3001 and 500 mg/ml on each bacteria tested at 5% level of significance. E.coli, S. aureus and Streptococcus sp were tested against 12 standard antimicrobial agents, of which six was sensitive and another six was resistance to E .coli, seven was sensitive, and five was resistance to S. aureus while four was resistance and eight sensitive to Streptococcus sp. The minimum inhibitory concentration (MIC) for E.coli, S. aureus, and  Streptococcus sp were 250, 125 and 31.25 mgml-1 while their minimum bactericidal concentration (MBC) were 500, 250 and 125 respectively. MIC and MBC tests showed that H. umbellata methanolic seed extract had noticeable bactericidal effects with MBC/MIC values ranging between 2 to 4. The extract has strong potency against these microorganisms with Streptococcus sp being the most susceptible. Conclusions: Hunteria umbellata has potential as natural therapeutic agents against E. coli, S. aureus and Streptococcus sp and they may prevent pathogenic diseases.

The use of natural food additives is currently a rising trend. In the present study, the aim was to determine the antimicrobial effects of plum, pomegranate, Seville orange and sumac sauces on E. coli O157:H7,E. coli type I,Listeriamonocytogenes, Listeria ivanovii, Salmonella Typhimurium and Staphylococcus aureus. Different concentrations (1%, 10%, 100%, v/v) of the sauces were tested on the studied bacteria in vitro using the agar diffusion and minimal inhibition concentration (MIC) methods. The results showed that the sumac sauce had the highest antimicrobial activity. The Seville orange, plum and pomegranate sauces also exerted antimicrobial activity in descending order. The antimicrobial activity of the fruit sauces was more effective at a concentration of 100% than at 10% and 1%, v/v. The most inhibitory effect was recorded for sumac sauce at a concentration of 100% (v/v) on L.monocytogenesand E. coli O157:H7. The findings of the MIC method aligned with the agar diffusion method. In addition, the in situ(food method) antimicrobial effect of the sauces on the indigenous microflora of chicken breast samples sold in stores was determined. Chicken samples hosting aerobic mesophilic bacteria, coliforms and E. coli were treated for two hours at 4 °C with plum, pomegranate, Seville orange and sumac sauces and were then monitored. The findings revealed that the Seville orange and sumac sauces were the most effective in reducing the indigenous microbial growth on the chicken samples. The plum sauce showed higher antimicrobial activity than pomegranate sauce. The phenolic content and acidity of the samples significantly (P< 0.05) affected the antimicrobial activity both in vitro (agar diffusion and MIC) and in situ (chilled chicken breast). In conclusion, the sumac and Seville orange sauces were found to be the most promising natural antibacterial agents, and their use could be recommended, for example, in catering services to reduce the risk of foodborne illness.

Devido ao aumento da resistência de patógenos humanos e animais aos medicamentos atuais, pesquisas por novos antimicrobianos têm sido incentivadas, principalmente por meio de plantas medicinais. Eugenia uniflora L., conhecida como pitangueira, é popularmente utilizada com diversas funções terapêuticas, justificando estudos nesta espécie. Assim, os objetivos deste estudo foram avaliar a atividade antibacteriana “in vitro” de extratos de folhas e cascas de Eugenia uniflora L. sobre Escherichia coli e Staphylococcus aureus. Para as etapas de triagem fitoquímica e microbiologia, foram utilizadas as metodologias usuais nessas áreas para obtenção dos extratos e avaliação da atividade antibacteriana com uso de controle positivo e negativo. Os resultados obtidos mostraram que no extrato das folhas foi verificada a presença de saponinas, flavonóides, alcalóides e taninos, enquanto na casca foram obtidos apenas saponinas e alcalóides. Os extratos da folha e da casca foram capazes de inibir o crescimento de Stapylococcus aureus apenas, promovendo halos de inibição de meio de 19,4 mm e 14,4 mm, respectivamente. As análises permitiram concluir que os extratos apresentam metabólitos secundários interessantes e de grande potencial antimicrobiano e que esses extratos foram capazes de inibir apenas as cepas de S. aureus, sendo resistentes às cepas de E. Coli.
