Defatted by supercritical CO2, Viburnum opulus berry pomace (VOP) was subjected to consecutive extraction with pressurized ethanol (E) and water (W) and yielded 23% of VOP-E and 8% of VOP-W, respectively. The major phytochemical groups covering 42 identified and quantified constituents in VOP extracts were organic and phenolic acids, iridoids, quercetin and (epi)catechin derivatives, flavalignans, procyanidins, and anthocyanins. The on-line HPLC-DPPH•-scavenging assay revealed the presence of numerous antioxidants. VOP-E had a higher total phenolic content, was a stronger antioxidant (equivalent to 0.77, 0.42, and 0.17 g trolox/g in oxygen radical absorbance capacity (ORAC), ABTS, and DPPH assays, respectively), and recovered the major part of phenolics from the pomace; however, both extracts demonstrated similar antioxidant activity in the cellular assay. VOP-E inhibited HT29 cancer cells at non-cytotoxic concentrations. The results of this study revealed that VOP contains valuable phytochemicals possessing antioxidant and antiproliferative activities. Consequently, extracts from VOP substances may be of interest in developing functional ingredients for healthy foods, nutraceuticals, and cosmeceuticals.

This study aimed at valorisation of sea buckthorn pomace (SBP) for the production of extracts containing valuable bioactive compounds. For this purpose, SBP defatted by supercritical CO2 was subjected to consecutive fractionation with pressurized ethanol and water, which yielded 11.9% and 4.8% of extracts, respectively. The extracts were evaluated for their antioxidant potential, phytochemical composition and antiproliferative effects against cancer cells. Water extracts exhibited remarkably higher values in Folin-Ciocalteu assay of total phenolic content, oxygen radical absorbance capacity (ORAC), ABTS●+/DPPH● scavenging and cellular antioxidant activity (CAA) assays and more efficiently inhibited proliferation of HT29 cells at non-cytotoxic concentrations measured in non-tumoral Caco2 cells. Among 28 detected and 21 quantified phytochemicals, flavonols with the structures of isorhamnetin (five compounds), quercetin (three compounds), kaempferol (three compounds) glycosides and catechin (six compounds) were the most abundant in the extracts. In conclusion, the applied method of fractionation of SBP produces promising natural antioxidant complexes with antiproliferative properties that could find potential applications in nutraceuticals, functional foods and cosmeceuticals.

Objectives: The objectives of this research were to evaluate antioxidant activity from different polarities rice bran extract of three varieties of rice using two methods of antioxidant testing which were FRAP (Ferric Reducing Antioxidant Power) and DPPH (2,2-diphenyl-1-picrylhydrazyl), and correlation of total phenolic, flavonoid and carotenoid content with their EC50 of FRAP and IC50 of DPPH antioxidant activities. Methods: Extraction was conducted by reflux using different polarity solvents. The extracts were evaporated using rotary evaporator. Determination of total phenolic, flavonoid and carotenoid content, antioxidant activities using FRAP and DPPH assays were performed by UV-visible spectrophotometry and its correlation with EC50 of FRAP capacities and IC50 of DPPH scavenging activities were analyzed by Pearson’s method. Results: Ethanolic rice bran extract of black rice showed the lowest EC50 of FRAP capacity 64.35 µg/ml and IC50 of DPPH scavenging activity 23.92 µg/ml. The highest phenolic content, flavonoid content and carotenoid content were also given by ethanolic rice bran extract of black rice. There were significantly negative correlation between total phenolic content and carotenoid content in rice bran extract of red rice and black rice with their IC50 of DPPH. Conclusions: All of rice bran extracts (except n-hexane rice bran extract of black rice and ethanolic rice bran extract of white rice) were very strong antioxidant, by DPPH assay. Phenolic and carotenoid compounds in rice bran extracts of red rice and black rice were the major contributor in antioxidant activity by DPPH assay. Rice bran extracts of black rice had linear results by FRAP and DPPH assays.

To evaluate the antioxidant potential of less valued green leafy vegetables of Bangladesh, the methanolic extracts of Alternanthera sessilis, A. philoxeroides, A. paronychioides, Glinus oppositifolius and Portulaca oleracea were subjected to assay for total phenolic content, total flavonoid content and DPPH free radicle scavenging activity. Among the studied materials, A. sessilis showed the highest phenolic and flavonoid content. However, G. oppositifolius presented the lowest phenolic content (14.53 ± 0.48 mg of GAE/g of sample) and P. oleracea exhibited minimum (7.005 ± 0.25 mg of RUE/g of sample) flavonoid content. DPPH radical scavenging activity was found to increase with increase in the concentration of each individual material. A. philoxiroides (IC50 = 6.07 ± 0.27 μg/ml) was found as the most active in DPPH radical percentage inhibition, which was followed by A. sessilis (IC50 = 7.17 ± 0.67 μg/ml). Interestingly, A. paronychioides, in spite of having good amount of total phenolic and flavonoid content, showed less DPPH scavenging activity. No linear relationship was found to exist in case of the total phenolic, flavonoid content and DPPH scavenging activity among G. oppositifolius, P. oleracea and A. paronychioides. The results from different methods may differ substantially because each complex chemical reaction generates unique values. However, all the five plants exhibited creditable antioxidant activity and thus consumption of these vegetables may play important role in combating oxidative stresses and thereby help in retaining good health.

The aim of this study was to evaluate the differences in the antioxidant activity and phenolic profile of sunflower (Helianthus annuus L.) extracts obtained from the aerial parts of plants harvested at five growth stages. In vitro assays were used to determine the antioxidant activity, i.e., ABTS•+ and DPPH• scavenging activity, the ferric-reducing antioxidant power (FRAP) and the ability to inhibit β-carotene–linoleic acid emulsion oxidation. Phenolic compounds, such as mono- and dicaffeoylquinic acid isomers and caffeic acid hexose, were identified using the LC–TOF–MS/MS technique. The predominant compound during the growth cycle of the plant was 3,5-di-O-caffeoylquinic acid, whose content was the highest at the mid-flowering stage. The total phenolic content was also the highest in sunflowers at the mid-flowering stage. The main phenolic compound contents were closely correlated with ABTS•+ and DPPH• scavenging activity and FRAP. No significant correlation was found between the total phenolic content and the antioxidant activity in the emulsion system. The highest antiradical activity and FRAP were generally determined in older plants (mid-flowering and late flowering stages). In conclusion, the aerial parts of sunflowers, in particular those harvested at the mid-flowering stage, are a good plant material from which to obtain phenolic compound extracts, albeit mainly of one class (esters of caffeic acid and quinic acid), with high antioxidant activity.

Antioxidant properties of crude extract, partition extract, and fermented medium from Dendrobium sabin (DS) flower were investigated. The oven-dried DS flower was extracted using 100% methanol (w/v), 100% ethanol (w/v), and 100% water (w/v). The 100% methanolic crude extract showed the highest total phenolic content (40.33 ± mg GAE/g extract) and the best antioxidant properties as shown by DPPH, ABTS, and FRAP assays. A correlation relationship between antioxidant activity and total phenolic content showed that phenolic compounds were the dominant antioxidant components in this flower extract. The microbial fermentation on DS flower medium showed a potential in increasing the phenolic content and DPPH scavenging activity. The TPC of final fermented medium showed approximately 18% increment, while the DPPH of fermented medium increased significantly to approximately 80% at the end of the fermentation. Dendrobium sabin (DS) flower showed very good potential properties of antioxidant in crude extract and partition extract as well as better antioxidant activity in the flower fermented medium.

The demand for sprouted seeds as dietetics and exotic healthy foods has recently increased. Winged bean seeds contain both nutritional composition and antinutrients. In order to convert this seed into healthy food, the winged bean was germinated into a vegetable sprout. In this research, the influence of soaking and sprouting conditions on the physicochemical and sensory attributes of sprouts was observed. Results showed that winged bean should be soaked in water at 34oC in 7 hrs, water to solid 3/1. The soaked seed was drained for 15 mins and incubated at temperature 36oC for 18 hrs. Following these parameters, the winged bean sprout achieved extended length (46.25±0.02 mm), vitamin C (124.13±0.01 mg/100 g), total phenolic content (997.34±1.14 mg GAE/100 g) and overall acceptance (8.94±0.02). Soaking terms for sprouting could improve the nutritional attribute of the winged bean as a functional foodstuff

Four Korea-native wild vegetables belonging to the Allium genus (Allium tuberosum Rottl., Allium senescens L., Allium thunbergii G. Don., and Allium sacculiferum Maxim.) were investigated for their antioxidant and anti-obesity potential, in order to promote the utilization of wild edible plants for the development of potential new crops. In this study, we compared the antioxidant activity of the plant extracts by ABTS+ and DPPH scavenging method. We then compared the anti-adipogenic effects of the extracts in adipocytes. We quantified the phenolics in the extracts using high-performance liquid chromatography (HPLC) to identify the bioactive compounds. A. tuberosum and A. sacculiferum had a higher total phenolic content (19.0 ± 0.3 and 19.6 ± 0.7 mg GA/g extract, respectively) than A. senescens and A. thunbergii (16.0 ± 0.2 and 14.5 ± 0.7 mg GA/g extract). Caffeic acid, an antioxidant and anti-adipogenic compound, was detected in A. tuberosum and A. sacculiferum, but not in A. senescens and A. thunbergii. A. tuberosum, and A. sacculiferum showed superior antioxidative and anti-adipogenic activities compared to A. senescens and A. thunbergii. Therefore, A. tuberosum and A. sacculiferum, both of which contain antioxidant and anti-adipogenic phenolics, such as caffeic acid, present a therapeutic potential against obesity. Various phenolic compounds, which have been found to differ among Allium spp., may partially account for differences among the Allium spp. with respect to their antioxidant and anti-obesity effects.

Antioxidant plays a significant role in inhibiting and scavenging free radicals, which protectshuman against infection and degenerative diseases. Nowadays, most studies focus on “natural antioxidants” from herbs due to safe therapeutic. In this study, performance on different extraction methods, i.e. Soxhlet, cold maceration and traditional extraction method assisted with ultrasonic-assisted extraction (UAE) for Asystasia gangetica leaves and Phyllanthus niruri plant are investigated by means of antioxidant, total phenolic content and antibacterial activity. The extracts were characterized for its antioxidant activity and total phenolic content by means of DPPH radical scavenging method and Folin-Ciocalteau reagent respectively. Optimization studies were carried out on combined cold-maceration and UAE extraction method by using Response Surface Method. Then, the optimized extracts were further characterized for antibacterial activity using Escherichia coliand Staphylococcus epidermidisby the agar disc diffusion method. Sample containing cold macerated A. gangetica and P. niruri indicates the highest antioxidant property as it contains the highest percentage of DPPH scavenging activity (59.57% and 51.14% respectively). While, the total phenolic content of macerated A. gangetica and P. niruri are 2.73 mg GAE/g and 2.68 mg GAE/g respectively. Consequently, after the determination of highest percentage of DPPH scavenging activity, the extracts werefurther optimized using UAE. From the optimization, the optimum condition was determined to be power of 17.57±0.01W and time of 5.83±0.01minutes for A. gangetica while, the power of 98.14±0.01W and 0.17±0.01minutes for P. niruri. Both optimized extracts showed potent antibacterial activity with high degree of antibacterial against E. coli. In conclusion, combination of cold maceration and UAE method increase the amount of antioxidant from extracts of A. gangetica and P. niruri. The finding from this study can be useful in pharmaceutical and cosmeceutical industries.

Background:Propolis contains compounds with antibacterial and antioxidant capacities, such as phenolic compounds and flavonoids. The aim of this study was to investigate the antibacterial and antioxidant effects of six propolis samples from northern Morocco and to determine their content of polyphenols and flavonoids.Methods:The total polyphenols and flavonoids of Ethanol Extracts (EEP) were evaluated by colorimetric assays. To test the antibacterial activity, the well diffusion and micro-dilution methods were adopted. The kinetic of bacterial growth in the presence of EEP was tested. The antioxidant activity was evaluated using three techniques: reducing power, β-carotene bleaching test, and DPPH scavenging capacity assay.Results:The EEPs were found to have high total phenolic content (65.09 - 122.00 mg GAE/g) and flavonoids (41.95 - 50.44 mg QE/g). The screening of the antibacterial activity showed that all tested bacteria were sensitive to EEP except Escherichia coli. The inhibitory zones varied from 11.50 ± 0.70 to 32.00 ± 0.70 mm, and MIC and MBC ranged from 0.07 to 5 mg/mL and 0.07 to >5 mg/mL, respectively. The antioxidant activity was proportional to the phenolic content of propolis. The IC50 varied from 48.70 to 156.00 mg/mL and the RAA% ranged from 75.07 to 96.68 %.

AbstractIn the present study, the objective was to evaluate the effects of barley kernel size, grind level, and extract solvent on the antioxidant activities associated with total phenolic content and phenolic acid compositions. Three barley varieties (Kindred, Azure, and Tradition), were used and the results showed that with the exception of superoxide anion radical scavenging activity, thin kernel size fraction (2.0 mm) showed much higher antioxidant activities and total phenolic content than those in the 2.4 mm and 2.8 mm fractions when averaged across barley cultivar, grind level, and extract solvent. A similar trend was found for individual phenolic acid compositions among kernel sizes. Simple correlation analysis revealed that total phenolic content showed strong correlation with DPPH· scavenging activity, ABTS + scavenging activity, and reducing power. Generally, fine grind and 80% acetone extract showed averaged highest values in antioxidant activities, total phenolic content, and individual phenolic acid compositions. Stepwise linear regression showed that extract solvent was the most important factor for DPPH· scavenging activity, ABTS + scavenging activity, reducing power, and iron chelating activity. The sum of syringic acid and caffeic acid exhibited a dominant role in explaining the major variation in antioxidant activities except for superoxide anion radical scavenging activity.
