The current study aims to investigate the influence of five rare earth elements (REEs) (i.e., lanthanum (La), cerium (Ce), praseodymium (Pr), neodymium (Nd), and gadolinium (Gd)) on the growth of Sprague–Dawley (SD) rats, and to explore the accumulation characteristics of REEs in tissues and organs with different doses as well as the detoxification and elimination of high-dose REEs. Fifty healthy male SD rats (140~160 g) were randomly divided into five groups and four of them were given gavage of sodium citrate solution with REEs in different doses, one of which was the control group. Hair, blood, and bone samples along with specific viscera tissue samples from the spleen and the liver were collected for detection of REEs by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). Treated rats expressed higher concentrations of REEs in the bones, the liver, and spleen samples than the control group (P < 0.05). Few differences were found in relative abundance of La, Ce, Pr, Nd, and Gd in the hair and the liver samples, although different administration doses were given. The relative abundance of Ce in bone samples was significantly lower in the low-dose group and control group, whereas the relative abundance of La and Pr in the bone samples were highest among all groups. Although in the REEs solution, which was given to rats in high-dose group, the La element had a higher relative abundance than Ce element, it ended up with higher Ce element relative abundance than La element in the spleen samples. REEs had a hormetic effect on body weight gain of SD rats. The accumulation of the measured REEs were reversible to low concentrations in the blood and hair, but non-reversible in the bones, the spleen, and the liver. Different tissues and organs can selectively absorb and accumulate REEs. Further inter-disciplinary studies about REEs are urgently needed to identify their toxic effects on both ecosystems and organisms.

This study was carried out to investigate the effect of two different doses of oxytocin on weight. Adult male rabbits (15) were weighed and provided with food twice daily for 3 weeks to determine the amount of food consumed daily and the time spent feeding by each rabbit.  After 3 weeks the rabbits were weighed and divided randomly into 3groups: the control group, the low dose group, and the high dose group. The animals were injected daily for 3 weeks. During that time the amount of food consumed and the time spent feeding in both periods were determined. After the end of the treatment period the rabbits were weighed, and sacrificed. The results of this study showed that before treatment the rabbits consumed more food in the evening period than they did in the afternoon period. The mean time spent feeding in the evening period was slightly higher than that spent in the afternoon period; however, this difference was not statistically significant. After treatment, there was still significant difference between the means of the consumed food in the afternoon and the evening period for the control group. The mean amounts of food consumed in both periods by the treated groups were slightly reduced, but this reduction was not statistically significant. Furthermore, the mean time spent feeding in the evening period was slightly higher than that of the afternoon period for the 3 groups; however, these differences were not significant. The mean weight of the control group was slightly increased after treatment with the hormone; and the mean weights of the treated groups were slightly reduced after treatment. However, changes in body weighs were not statistically significant.

Objective To determine the influence of chronic iron dextran administrations into the peritoneal cavity of rats on function and anatomy of the peritoneal membrane, as well as on erythropoiesis and serum iron. Design Prospective randomized animal study. Setting Animallaboratory. Animals: 36 Sprague-Dawley rats. Interventions The rats were divided into three groups (n = 12). The animals were given standard 1.5% Dianeal (control group) or 1.5% Dianeal containing iron dextran in a concentration of 2 mg/L [Iow-dose group (LDG)] or 10 mg/L [high-dose group (HDG)]. Main outcome measures On the 8th day, at 3 months, and at 6 months a 2-hour peritoneal equilibration test (PET) and blood tests including hematocrit, serum iron, and total iron-binding capacity (TIBC) were done. After the final PET at 6 months, the peritoneal membrane was evaluated by gross inspection and by light microscopy. Results Hematocrit and serum iron levels increased only in the HDG and LDG. Peritoneal transport of small solutes decreased significantly in the HDG compared to baseline. All cases of the HDG group revealed peritoneal adhesions and fibrosis around the peritoneal catheter as well as massive iron deposits on the peritoneum. Similar but less pronounced changes were found in the LDG. Conclusions These findings suggest an efficient absorption of iron from the peritoneal cavity of rats, however, dialysate iron dextran concentrations of 2 mg/L or greater are toxic to the peritoneal membrane. Therefore, future studies should be performed to determine the minimal effective and nontoxic iron dextran concentrations for intraperitoneal administration.

This study evaluated camphora-induced androgenic and histopathological changes in male Sprague-Dawley rats. Thirty-five animals weighing 200 g±20 g were used for this study and randomly divided equally into five groups, with seven rats in each group. Group A animals (normal control group) were served water and rat chow only; Groups B-D (treatment groups) were orally administered camphora in doses of 1 g/kg (Low-dose), 2 g/kg (Medium-dose) and 4 g/kg (High-dose) respectively while Group E (vehicle group) were orally administered 6 mL/kg olive oil (a solvent for camphora) per day for 56 days. There was a significant decrease (P< .05) in activity levels of Follicle-Stimulating Hormone (FSH); Superoxide Dismutase (SOD) when the treatment was compared with the control group. Also, a significant decrease (P< .05) in activity level of FSH was observed when the Medium-dose group was compared with Low-dose group. Insignificant irregular pattern in activity level of Testosterone was observed across the treatment groups when compared with the control. However, a significant increase (P< .05) in activity level of Testosterone was observed when the High-dose group was compared with the Medium-dose group. There was a significant increase (P< .05) in activity levels of Luteinizing Hormone (LH) and Malondialdehyde (MDA) when the treatment was compared with the control group. Semen analysis showed reduction in sperm concentration, motility and morphology with increasing concentration of camphora. Significant decrease was recorded in testicular weight when High-dose group was compared to Control and Low-dose groups. Histopathological changes were seen in the testes of the camphor administered groups, ranging from mild disintegrated interstitial tissues in Low-dose to severe degeneration and disintegration of both seminiferous and interstitial tissues in the testes in the Medium-dose and High-dose groups. In conclusion, camphora had androgenic and toxic effects on testis and may cause testicular tissue damage.

Background. Scutellaria baicalensis (SB) is commonly used in traditional Chinese medicine for chronic inflammatory diseases. This study aims to investigate the effects of the early intervention with SB on airway remodeling in a well-established rat model of COPD induced by cigarette smoking. Methods. COPD model in Sprague Dawley (SD) rats were established by exposing them to smoke for 6 days/week, for 12 weeks, 24 weeks, or 36 weeks. Meanwhile, rats were randomly divided into normal control group, model group, Budesonide (BUD) group, and the SB (low, middle, and high) dose groups with 8 rats in each group and 3 stages (12 weeks, 24 weeks, and 36 weeks). After treatment, the pulmonary function was evaluated by BUXCO system and the morphology changes of the lungs were observed with HE and Masson staining. The serum IL-6, IL-8, and IL-10 and TNF-α, TGF-beta (TGF-β1), MMP-2, MMP-9, and TIMP-1 levels in BALF were detected by ELISA-kit assay. The protein expression levels of AKT and NF-κB (p65) were determined by western blot (WB). Results. The oral of SB significantly improved pulmonary function (PF) and ameliorated the pathological damage and attenuated inflammatory cytokines infiltration into the lungs. Meanwhile, the levels of TGF-β, MMP-2, MMP-9, and TIMP-1 were partially significantly decreased. The levels of PI3K/AKT/NF-κB pathway were also markedly suppressed by SB. Conclusions. SB could significantly improve the condition of airway remodeling by inhibiting airway inflammation and partially quenching TGF-β and MMPs via PI3K/AKT/NF-κB pathway.

This study investigated the effect of frankincense extract on peripheral nerve regeneration in a crush injury rat model. Forty-eight Sprague-Dawley rats were randomly divided into four groups: control and frankincense extract low-, medium-, and high-dose groups. At days 7, 14, 21, and 28 following the surgery, nerve regeneration and functional recovery were evaluated using the sciatic functional index (SFI), expression of GAP-43, and the proliferation of Schwann cells (SCs) in vivo and in vitro. At day 7, the SFI in the frankincense extract high-dose group was significantly improved compared with the control group. After day 14, SFI was significantly improved in the medium- and high-dose groups. There was no significant difference in GAP-43 expression among the groups at day 7. However, after day 14, expression of GAP-43 in the high-dose group was higher than that in the control group. Histological evaluation showed that the injured nerve of frankincense extract high-dose group recovered better than the other groups 28 days after surgery. Further, S100 immunohistochemical staining, MTT colorimetry, and flow cytometry assays all showed that frankincense extract could promote the proliferation of SCs. In conclusion, frankincense extract is able to promote sciatic nerve regeneration and improve the function of a crushed sciatic nerve. This study provides a new direction for the repair of peripheral nerve injury.

Diabetes mellitus has been on a continual rise as one of the top chronic diseases to affect individuals worldwide. The goal of this study was to determine how exposure from a well-known toxicant, a polycyclic aromatic hydrocarbon called 2-aminoanthracene (2AA), could potentially lead to diabetes, damage the liver, and have negative effects to the offspring. Humans are exposed to 2AA from foods cooked in high heat and tobacco smoke, among others. To analyze the effects of 2AA, three groups of Sprague Dawley dams consumed an adulterated 2AA diet from gestation to their postnatal period. Timed-pregnant dams ingested 0 mg/kg (control group (C)), 50 mg/kg (low dose group (LD)), and 100 mg/kg (high dose group (HD)) 2AA. Hepatic gene expressions of Adam8, Bax, Ccng1, CD68, CD93, Cdkn1c, and Ddit4 indicated a significant overexpression of Bax, Ccng1, CD68, CD93, and Cdkn1c in treated groups. Although there was no significant difference in the damage to the liver architecture by 2AA, the positively stained CD68+ cells were slightly increased in treated rats. Significant decreases in the albumin and aspartate aminotransferase levels might indicate an inflammatory response from 2AA exposure in dams. Immunoglobulin A (IgA) concentration was also decreased, in contrast to studies of liver cirrhosis that reported increased serum IgA concentration. Overexpression of genes Ddit4, Cdkn1c, Ccng1, Bax, CD93, and CD68 point to hepatic inflammation and apoptosis. Overall results suggest a link between environmental 2AA exposure and adverse liver effects, which has potential to increase susceptibility to type 2 diabetes and other diseases.

Objective: to probe the interference of Ruyanneixiao cream for hemorheology and mammary microcirculation of mammary precancer rats. Methods: 48 virginal female SD rats were randomly allocated into 6 groups, A: Blank control group (8); B: Mammary precancer model group (8); C: Tamoxifen (TAM) group (8); D: High dose group of Ruyanneixiao cream (8); E: Middle dose group of Ruyanneixiao cream (8); F: Low dose group of Ruyanneixiao cream (8). The changes of hemorheology and mammary microcirculation were recorded when the rats were executed after 60d. Result: compared with the Blank control group, the whole blood viscosity and plasma viscosity of mammary precancer model group was higher (P<0.05) and the perfusion of mammary microcirculation was lower (P<0.01). Compared with mammary precancer model group, the hemorheological parameters and perfusion of mammary microcirculation of each dose group of Ruyanneixiao cream were improved (P<0.05). Conclusion: Ruyanneixiao cream can improve the status of hemorheology, increase the perfusion of mammary microcirculation. And it may be one of mechanism to treat and prevent mammary precancer disease.

Production of dabai ( Canarium odontophyllum) pulp oil by supercritical carbon dioxide extraction is still relatively new and should be investigated. The objective of this study was to investigate the effects of supercritical carbon dioxide (SC-CO2) extracted dabai pulp oil (DPO) on rats induced with hypercholesterolemia. Male-specific pathogen free (SPF) Sprague-Dawley rats were fed with diets with high cholesterol contents for 4 weeks to induce hypercholesterolemia. Afterwards, the hypercholesterolemia rats were divided into groups namely: positive control group (PG), low dose group (LG), high dose group (HG), and statin group (SG). 0.5% and 2% of SC-CO2 DPO were administered to the LG and HG groups respectively for another 4 weeks. Changes in body weight and biochemistry profiles were measured. When compared with the normal rats that were fed with a normal basal diet, the hypercholesterolemia rats had elevated body weights and major increments in total cholesterol and LDL levels(NG) ( p<0.05). Paired-samples t-tests showed that the LG group exhibited a notable reduction in total cholesterol, triglyceride and LDL levels ( p<0.05) and an 8.26% increment in HDL level. Meanwhile, diminishing levels of total cholesterol, triglyceride and LDL levels were found in the HG group. Notable differences in AST and ALT levels were not detected in LG and HG groups when compared with the NG group. These results indicated that SC-CO2dabai pulp oil contains vital elements which contribute to cholesterol-loweringeffects and which can be used as special oils for health promotion and disease prevention.


Background: Hepatocellular damage caused by physical activity or the use of supplements is one of the serious problems facing athletes in various fields. This study aimed to evaluate the effect of different doses of nitric oxide supplements on AST and ALT liver enzymes and the ratio of AST to ALT following a session of eccentric exercise in Sprague Dawley male rats. Materials and Methods: In this study, 36 Sprague Dawley male rats (two months old) were divided into three groups of control, low dose (4.8 mg/kg body weight), and high dose of NO supplements (15.4 mg/kg body weight). Supplements were given to rats for seven days. Subsequently, all three groups of rats were forced to run on a treadmill for 45 min with a speed of 20 m/min, and a slope of -15 degrees. Blood samples were taken directly from cardiac puncture of rats 24 h after the running exercise. Blood serum variables of the study were measured afterward. Results: Low dose of nitrate supplements did not change AST and ALT indices, while the high dose of nitrate supplements increased ALT serum level and decreased AST to ALT ratio, compared to a low dose of NO supplements and control group. Conclusion: Based on the obtained results, the consumption of a low dose of NO supplements does not change hepatocellular damage markers, while the high dose of NO supplements causes degeneration of hepatic cells in athletes.

Purpose: To explore the therapeutic role of amber powder in endometriosis by investigating its effect on endometrial ultrastructure, ERK1/2, p38MAPK, and NF-κB mRNA pathways and CSRC/EFR/ERK1/2 proteins. Methods: Sprague Dawley (SD) rats were randomly divided into blank group, disease model group (untreated), amber powder high-dose group, amber powder medium-dose group, amber powder lowdose group and danazol group. Morphological changes in endometrial cells were studied using transmission electron microscopy. The expression of ERK1/2, p38MAPK, and NF-κB mRNA in endometrial tissues of each group was determined using quantitative real-time polymerase chain reaction (qRT-PCR). Immunohistochemistry was utilized for the measurement of C-SRC/EFR/ERK1/2 pathway protein expression. Results: The endometriosis rats treated with a high-, medium- and low-dose amber powder showed a decrease in the volume of ectopic lesions, compared with the untreated disease model group. The expressions of ERK1/2, p38MAPK, NF-κB mRNA, and C-SRC/EFR/ERK1/2 protein were higher in the eutopic and ectopic endometrial tissues in untreated disease group than those in normal control group. Moreover, treatment of endometriosis rats with amber powder revealed a reduction in the expressions of ERK1/2, p38MAPK, NF-κB mRNA and C-SRC/EFR/ERK1/2 proteins in eutopic and ectopic endometrium tissues. Conclusion: Amber powder reduces ectopic lesions and slows down the development of endometriosis, probably via inhibition of MAPK pathway genes in eutopic and ectopic endometrial tissues.
