The current study was intended to explore the phytochemical profiling and therapeutic activities of Putranjiva roxburghii Wall. Crude extracts of different plant parts were subjected to the determination of antioxidant, antimicrobial, antidiabetic, cytotoxic, and protein kinase inhibitory potential by using solvents of varying polarity ranges. Maximum phenolic content was notified in distilled water extracts of the stem (DW-S) and leaf (DW-L) while the highest flavonoid content was obtained in ethyl acetate leaf (EA-L) extract. HPLC-DAD analysis confirmed the presence of various polyphenols, quantified in the range of 0.02 ± 0.36 to 2.05 ± 0.18 μg/mg extract. Maximum DPPH scavenging activity was expressed by methanolic extract of the stem (MeOH-S). The highest antioxidant capacity and reducing power was shown by MeOH-S and leaf methanolic extract (MeOH-L), respectively. Proficient antibacterial activity was shown by EA-L extract against Bacillus subtilis and Escherichia coli. Remarkable α-amylase and α-glucosidase inhibition potential was expressed by ethyl acetate fruit (EA-F) and n-Hexane leaf (nH-L) extracts, respectively. In case of brine shrimp lethality assay, 41.67% of the extracts (LC50 < 50 µg/mL) were considered as extremely cytotoxic. The test extracts also showed mild antifungal and protein kinase inhibition activities. The present study explores the therapeutic potential of P. roxburghii and calls for subsequent studies to isolate new bioactive leads through bioactivity-guided isolation.

The aim of this study is to reveal allelopathic effects and antioxidant activities of methanolic extract of Ganoderma lucidum in Zorkun plateau (Osmaniye). It was determined that methanolic extract of G. lucidum has significantly phytotoxic effect by increasing the doses (1, 2, 4, 8 mg/mL) on Hordeum vulgare and Triticum aestivum. Total phenol and flavonoid amounts of G. lucidum were 114.55 mg/kg and 8.95 mg/kg, respectively. 2,2-diphenyl-1-picrylhydrazyl (DPPH), scavenging activity (60%, 91% and 92%), reducing power (0.83, 1.43 and 2.23 Abs), and nitric oxide (NO) scavenging activity (48%, 55% and 70%) of this mushroom at the different concentrations (1.25, 2.5 and 5 mg/mL) were determined, respectively. However, H2O2 scavenging activity of the different concentrations (0.0156-0.125 mg/mL) was found as 18%, 56%, 60% and 86%, respectively. Based on these findings, it can suggest that G. lucidum methanolic extract has significantly phytotoxic effect and antioxidant activity. The present study is the first report on the phytotoxic effects and antioxidant activities of G. lucidum which consumed and collected from Amanos Mountains.

Solubility of phytoconstituents depends on the polarity of the extraction medium used, which might result in the different pharmacological responses of extracts. In line with this, ethnomedicinally important food plant (i.e., Caralluma tuberculata extracts) have been made in fourteen distinct solvent systems that were then analyzed phytochemically via total phenolic amount estimation, total flavonoid amount estimation, and HPLC detection and quantification of the selected polyphenols. Test extracts were then subjected to a battery of in vitro assays i.e., antioxidants (DDPH scavenging, antioxidant capacity, and reducing power estimation), antimicrobial (antibacterial, antifungal, and antileishmanial), cytotoxic (brine shrimps, THP-1 human leukemia cell lines and normal lymphocytes), and protein kinase inhibition assays. Maximum phenolic and flavonoid contents were computed in distilled water–acetone and acetone extracts (i.e., 16 ± 1 μg/mg extract and 8 ± 0.4/mg extract, respectively). HPLC-DAD quantified rutin (0.58 µg/mg extract) and gallic acid (0.4 µg/mg extract) in methanol–ethyl acetate and methanol extracts, respectively. Water–acetone extract exhibited the highest DPPH scavenging of 36 ± 1%. Total reducing potential of 76.0 ± 1 μg/mg extract was shown by ethanol chloroform while maximum total antioxidant capacity was depicted by the acetone extract (92.21 ± 0.70 μg/mg extract). Maximal antifungal effect against Mucor spp., antileishmanial, brine shrimp cytotoxicity, THP-1 cell line cytotoxicity, and protein kinase inhibitory activities were shown by ethyl acetate-methanol (MIC: 50 µg/disc), n-hexane (IC50: 120.8 ± 3.7 µg/mL), ethyl acetate (LD50: 29.94 ± 1.6 µg/mL), distilled water–acetone (IC50: 118 ± 3.4 µg/mL) and methanol–chloroform (ZOI: 19 ± 1 mm) extracts, respectively. Our findings show the dependency of phytochemicals and bioactivities on the polarity of the extraction solvent and our preliminary screening suggests the C. tuberculata extract formulations to be tested and used in different ailments, however, detailed studies remain necessary for corroboration with our results.

Abstract Background The Himalaya hotspot, habitat to numerous indigenous species, harbours an endemic and endangered, cucurbit climber Herpetospermum darjeelingense (C.B.Clarke) H.Schaef. & S.S.Renner [Edgaria darjeelingensis C.B.Clarke]. Seeds of this plant have traditionally been used for ethno-veterinary treatment. The present probe aims to provide an insight into the antioxidant-related biochemical attributes of this phytochemically unexplored plant. Results DPPH scavenging activity of solvent extracts showed more potential towards the polar end with its maximum activity in methanolic extract (84.64%). Acetone extract showed more ferric reducing power than the ethanol fractions. Maximum flavonoid quantification was recorded in ethanol extract. GC–MS of methanolic extract detected 39 compounds of which 22 have preventive and curing properties for several ailments. Thirteen compounds were reported with antioxidant activity and cover an area of 63.92% among the total detected compounds by GC–MS. 1,3,4,5-Tetrahydroxy-cyclohexanecarboxylic acid; Hexadecanoic acid, methyl ester; ß sitosterol and Phytol are present in significant quantity and are the prime candidates conferring antioxidant potential. Chromatographically fractionalized methanol extract with non-polar to polar solvents and their subsequent DPPH scavenging analysis exhibited a major peak in ethanol fraction due to the presence of Phytol in considerable high quantity and some other compounds. Further sub-fractionation of ethanolic fraction provides a supportive evidence of ethanol solubility of the compounds. Biosynthetic lineage reveals fatty acid synthesis, intermediates of Shikimate pathway and terpenoid metabologenesis schemes lead to the several antioxidant molecules detected in our extract. Conclusion Metabolomics of antioxidants will provide a better understanding in its intracellular free radical quenching activity and isolation of these bioactive compounds can boost pharmaceutical industry.

Ipomoea reptans Poir or kangkung is a popular leafy vegetable, a favorite to people in Asian countries. However, limited information is available on their bioactivities. In the present study, the antioxidant, antidiabetic, and anti-obesity potential of I. reptans leaves were investigated. Different fractions (ethanol, ethyl acetate, and hexane) of I. reptans leaves were evaluated for their scavenging activity on DPPH radicals, whereas their reducing potential was investigated by cupric reducing antioxidant capacity (CuPRAC), total antioxidant, and reducing power assays. The antidiabetic potential was investigated by their inhibition effect on a-glucosidase. Total phenolic and flavonoid contents of I. reptans leaves were solvent dependent. Ethyl acetate contained the highest phenolic content, followed by ethanol and hexane fractions. However, for flavonoid content, the order was ethanol > ethyl acetate > hexane. All fractions showed DPPH scavenging activity in a concentration-dependent manner, with activities weaker than standards ascorbic acid and BHT, in the order of ethanol > ethyl acetate > hexane. All fractions showed reducing capacity, but only hexane and ethanol fractions of I. reptans leaves showed inhibition on a-glucosidase, with hexane showed more potent inhibition compared to acarbose. The study also found that fractions of I. reptans inhibit lipase and trypsin, enzymes related to lipid metabolism. Findings in this study offer a prospect for I. reptans leaves as a functional food source for antioxidant, antidiabetic, and anti-obesity purposes.

It is a well known medicinal plant in Sudan with different traditional uses. To date there are no chemical or biological records reported on C. zambesicus seeds. Air dried ground seeds and leaves of C. zambesicus were extracted using 80% methanol. The methanolic extract was sequentially fractionated with petroleum ether, chloroform and ethyl acetate. The obtained extracts of C. zambesicus seeds leaves were screened antimicrobial activities against four standard bacteria (Bacillus subtitles, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and two fungi (Aspergillus niger and Candida albicans) using in vitro agar diffusion method. The cytotoxicity evaluation of the bioactive fractions was carried out in vitro using brine shrimp lethality assay. The MIC of the seeds crude extract was 12 mg/ml against B. subtitles, E. coli, and P.aeruginosa. After fractionation of the seeds methanolic extract the antibacterial activity was mainly accumulated in the ethyl acetate phase with an MIC of 50 mg/ml against Bacillus subtitles. The MIC of the leaves crude extract was 6.25 mg/ml against S. aureus, Escherichia coli, and Pseudomonas aeruginosa. After fractionation of the leaves methanolic extract the antibacterial activity was mainly accumulated in the chloroform phase with an MIC of 12.5 mg/ml against Bacillus subtitles. Terpenoids were mainly detected in the petroleum ether fraction in addition to four compounds which were suggested to be phenolics. Phenolic acids and flavonoids were detected in the ethyl acetate fraction giving typical blue, yellow and orange colours with NPR at 366 nm. Terpenoids and polyphenols, mainly flavonoids, detected in C. zambesicus active extracts were believed to be responsible of the antimicrobial and antioxidant activities mentioned above. All extracts were quite safe in brine shrimp lethality assay. These results justify the traditional use of C. zambesicus seeds and leaves.

Background:: The main aim of the study was to evaluate the bioactive properties of ethyl acetate crude extract of Streptomyces sp. VITASP with a view to assess their therapeutic potential. Methods: The morphological, physiological and the biochemical properties of the strain Streptomyces sp. VITASP were confirmed by conventional methods. The present study evaluated the antibacterial, antioxidant and cytotoxic activities. Results: The isolate was identified to be Streptomyces sp. (Genbank accession number: KR233807). The ethyl acetate extract of Streptomyces sp. VITASP showed maximum antibacterial activity of two Gram- positive and Gram- negative bacteria at 0.5mg/mL. The antioxidant potential of the crude extract exhibited strong reducing power activity at 0.5 mg/mL with 93±0.05% inhibition. The antiinflammatory and anti-diabetic properties were identified at 0.5 mg/mL concentration. The cytotoxic effect was found with an IC50 of 500μg/ mL on HeLa cell lines. The GC-MS analysis and the chromatogram patterns revealed major peak at 18.485 which corresponds to Pyrrolo[1,2-a]pyrazine-1,4-dione, Hexahydro-3-(2-methylpropyl. IR spectra showed the functional groups. HPLC Retention time of the peak was found to be 2.414 min. Conclusion: This work demonstrates that the extract should be considered as a useful source as an antioxidant and antimicrobial agent and gives further insight into the potential use of the compounds as drugs for various other bioactivities.

Objective: This study was designed to evaluate the antioxidative potential of tubers of Habenaria pectinata. Methods: The tubers of H. pectinata were extracted using hexane, ethyl acetate, methanol, and water as solvents. The anti-oxidant potential of extracts was evaluated using free radical scavenging and reducing power assays. The most active methanolic extract was then fractionated into four fractions using the above-mentioned solvents. Results: The phenol and flavonoid content was found to be maximum in the methanol extracts. All the extracts and fractions showed significant levels of antioxidant activity except hexane extract. Conclusion: The tubers of H. pectinata were found to possess a significant antioxidant potential and can be explored further for isolation and preclinical investigation for the ailment of various diseased states and disorders.

The objective of this work was to perform a phytochemical screening of Juncus acutus and to evaluate its antioxidant activity and antilithiasis activity. Phytochemical screening has demonstrated the presence of catechin, tannins and alkaloids in the various extracts studied. The antioxidant activity of the methanolic and ethyl acetate extracts by ultrasound and maceration was evaluated by the method of DPPH, FRAP and phosphomolybdate. The ethyl acetate extract by ultrasound, the methanolic extract by maceration and ultrasound respectively showed IC 50 with DPPH of 1,449mg / ml, 1,535mg / ml and 4,771mg / ml and a reducing power was observed at concentrations of 419,561mgBHA /100g, 207,143mgBHA /100g and 142,20mgBHA /100g by FRAP and at concentrations of 839,470mgAAE/100g, 283,260mgAAE/100g and 141,018mgAAE/100g by the phosphomolybdate test. The determination of the total polyphenols of the ethyl acetate extract by ultrasound, the methanolic extract by maceration and by ultrasound presented respectively contents of 2,428 mg GAE / g, 1,960mg GAE / g and 1,172mg GAE / g, likewise, the flavonoid assay showed respectively concentrations of 14,469mg EQ/g, 6,466 mg EQ/g and 3,143 mg EQ/g. Antilithiasis activity was evaluated on carbapatite, cystine and uric acid stones. The mass loss of uric acid, carbapatite and cystine stones was respectively 42%, 7% and 16%.

The antioxidant activity, total phenolic and flavonoid content of ethanolic, acetone and ethyl acetate extracts of Ruscus hypoglossum L. and Ruscus aculeatus L. (aerial parts) from Serbia were investigated in this paper. The best total antioxidant capacity (23.329 ?g AA g-1) and the highest DPPH scavenging activity (IC50 = 182.54 ?g mL-1) were found in acetone and ethyl acetate extract of R. aculeatus L. Ethanolic extract of R. hypoglossum L. showed the highest ABTS radical cation scavenging activity (IC50 = 3.04 ?g mL-1) as well as reducing power (IC50 = 0.143 ?g mL-1). The best inhibitory activity against lipid peroxidation (IC50 =651 ?g mL-1) and the best ferrous ion chelating ability (IC50 = 110 ?g mL-1) were found in acetone and ethyl acetate extract of R. hypoglossum L. The highest total phenolic (8.569 mg GAE g-1) and flavonoid content (0.136 mg RU g-1) were found in ethanolic and acetone extract of R. hypoglossum L. and R. aculeatus L, respectively.

Objective: The objective of the study was collection of plant materials, Extraction of phytoconstituents using a different solvent, to carry out preliminary phytochemical screening of different extracted solvent, to perform fluorescence analysis, to estimate the proximate composition of the leaves Lindernia ruellioides (Colsm.) Pennell, and to determine the presence of in vitro anti-oxidant of the methanolic extract of the plant. Method: Preliminary phytochemical screening of the methanolic extract of Lindernia ruellioides (Colsm.) Pennell, estimation of proximate composition of the leaves, fluorescence analysis, total phenolic content, total flavonoids content, and in vitro antioxidant activity of the methanol extract (DPPH scavenging activity, reducing power assay, and nitric oxide scavenging activity). Results: The result of phytochemical screening of methanolic extract of Lindernia ruellioides (Colsm.) Pennell contents the presence of amino acid, flavonoids, tannins, steroids, and triterpenoids. The moisture content and Ash value were found to be appropriate and the in vitro antioxidant activity of the methanolic extract showed potential antioxidant activity in terms of DPPH scavenging activity, reducing power assay, and nitric oxide scavenging activity. Conclusion: The work presented here suggests that the methanolic extract of Lindernia ruellioides (Colsm.) Pennell possesses potential antioxidant activity.
