Objectives: Herbs are in use as medicine worldwide from the time immemorial. Many drugs derived from plant, animal or minerals are in use as medicine till date. This is a study on leaf extract of Putranjiva roxburghii Wall. (Euphorbiaceae) involving pharmacognosy, phytochemistry, activity against microbes and oxidation to substantiate its use. The leaf extracts were collected by successive soxhlation using solvents like petroleum ether, chloroform, benzene, acetone & ethanol. Chemicals present in the crude leaf extracts and isolated constituents were analyzed for both quality and quantity followed by TLC, UV–Visible Spectrophotometric analysis, HPTLC, Phytochemical tests and TLC examination confirmed to contain flavonoids in acetone, chloroform, aqueous and ethanol extract. The activity of Petroleum-ether, chloroform, acetone, alcoholic and aqueous extracts against the microbes was assessed by cup plate method. Okada & Okada method was followed to evaluate the DPPH free radical for aging property. The reducing, total antioxidant and peroxide radical for aging action of the extracts were assessed. The study confirmed significant antimicrobial and antioxidant property of the Putranjiva roxburghii Wall leaves may be for the presence of constituents like flavonoids, saponins, Phytosterols, favoring its traditional usage as medicine.

The current study was intended to explore the phytochemical profiling and therapeutic activities of Putranjiva roxburghii Wall. Crude extracts of different plant parts were subjected to the determination of antioxidant, antimicrobial, antidiabetic, cytotoxic, and protein kinase inhibitory potential by using solvents of varying polarity ranges. Maximum phenolic content was notified in distilled water extracts of the stem (DW-S) and leaf (DW-L) while the highest flavonoid content was obtained in ethyl acetate leaf (EA-L) extract. HPLC-DAD analysis confirmed the presence of various polyphenols, quantified in the range of 0.02 ± 0.36 to 2.05 ± 0.18 μg/mg extract. Maximum DPPH scavenging activity was expressed by methanolic extract of the stem (MeOH-S). The highest antioxidant capacity and reducing power was shown by MeOH-S and leaf methanolic extract (MeOH-L), respectively. Proficient antibacterial activity was shown by EA-L extract against Bacillus subtilis and Escherichia coli. Remarkable α-amylase and α-glucosidase inhibition potential was expressed by ethyl acetate fruit (EA-F) and n-Hexane leaf (nH-L) extracts, respectively. In case of brine shrimp lethality assay, 41.67% of the extracts (LC50 < 50 µg/mL) were considered as extremely cytotoxic. The test extracts also showed mild antifungal and protein kinase inhibition activities. The present study explores the therapeutic potential of P. roxburghii and calls for subsequent studies to isolate new bioactive leads through bioactivity-guided isolation.

Putrajeevak (Putranjiva roxburghii Wall.; synonym Drypetes roxburghii (Wall.) Hurus) seeds have been used since ancient times in the treatment of infertility in the Ayurvedic system of medicine in India. In this study, the oil component of Putrajeevak seeds (PJSO) was extracted using the supercritical fluid extraction (SCFE) method using liquid CO2 and the constituents were analyzed using gas chromatography-flame ionized detectorand high-performance thin-layer chromatography. PJSO contained trace amounts of β-sitosterol with oleic and linoleic acids as the major fatty acid constituents. Male and female zebrafish were mutagenized with N-ethyl-N-nitrosourea (ENU) and fish that produced less than 20 viable embryos were selected for the study. SCFE oil extracts from the P. roxburghii seeds were used in this study to reverse fertility impairment. The mutant fish were fed with PJSO for a period of 14 days and the rates of fertility, conception, and fecundity were determined with wild-type healthy fish as a breeding partner. Treatment with PJSO increased the ovarian follicle count as well as the number of mature eggs, while reducing the number of ovarian cysts. Sperm count as well as sperm motility were greatly enhanced in the ENU-mutagenized male zebrafish when treated with PJSO. The results obtained in this study demonstrate the effectiveness of P. roxburghii seed oil in reversing impaired fertility in both male and female zebrafish models.

Background: Putranjiva roxburghii Wall is traditionally known to cure many pathological conditions including epilepsy. Objective: The present study is aimed at determining bioactive compounds in ethanolic extract of Putranjiva roxburghii test extract (PRTE) seeds by GCMS analysis and to assess its anti-epileptic potential using various experimental models of epilepsy. Methods: The ethanolic extract of seeds of Putranjiva roxburghiiwas subjected to GC-MS analysis to detect the bioactive phytoconstituents. Acute oral toxicity of the extract was performed using OCED guideline 420. Pentylenetetrazol (PTZ) kindling model of epilepsy and Maximal electroshock epilepsy (MES) model of epilepsy were used to determine antiepileptic potential. Results: The GC-MS analysis of the extract revealed the presence of 20 phytoconstituents. The major phytoconstituents included n-Propyl heptyl ether (25.25%), 5-Ethyl hydantoin (8%), octadec-9-enoic acid (16-25%) and 1, 2-Benzene dicarboxylic acid (11.86%). The PRTE (50 mg/kg and 100 mg/kg) afforded significant and dose - dependent protection against PTZ – induced kindling epilepsy and MES induced epilepsy (p<0.001 and p<0.01). Conclusion: Based on the above finding, it is evident that Putranjiva roxburghii seeds contain biologically active compound. It can also be concluded that the extract possesses anti-epileptic potential.
