(1) Background: Observational studies have established that vitamin D-binding protein (DBP) and 25-hydroxyvitamin D3 (25(OH)D) concentrations are the major factors affecting the bioavailability of 25(OH)D. It has also been shown that poor 25(OH)D bioavailability elevates the risk of obesity and its related cardio-metabolic disorders. However, the relationship between 25(OH)D and DBP concentrations with cardio-metabolic risk factors in overweight and obese cohorts has not been established. Consequently, we evaluated the association between DBP and 25(OH)D concentrations with lipid profile, blood pressure (BP), and body composition in overweight and obese women. (2) Methods: In this cross-sectional study of 236 overweight and obese women, DBP and 25(OH)D concentrations were measured using an enzyme-linked immunosorbent assay. Body composition was assessed via bioelectrical impedance analysis. Lipid profile and BP were assessed by an auto-analyzer and digital BP monitor, respectively. The associations were examined by multivariate logistic regression. (3) Results: The indicated showed an inverse relationship between DBP and high-density lipoprotein (HDL) (p = 0.010) concentrations (where individuals with higher DBP had lower HDL) which, after adjusting for possible cofounders, remained significant (p = 0.006). Moreover, DBP concentration was positively associated with fat mass index (FMI) after adjustment (p = 0.022). No significant relationships were observed among 25(OH)D and target variables. (4) Conclusions: In conclusion, lower concentrations of HDL and higher values of FMI are associated with higher concentrations of DBP in overweight and obese women. These findings present novel awareness regarding the association of DBP with some metabolic and body composition variables in overweight and obese women. However, a two-way causal relationship between DBP and target variables should be considered.

AbstractSeveral studies have shown that rs9939609 and rs1421085 in fat mass and obesity-associated (FTO) gene rs17782313 and rs12970134 in melanocortin-4 receptor (MC4R) gene influence obesity. In the present study, we aimed to determine association between rs9939609, rs1421085, rs17782313, and rs12970134 polymorphism, and their relation with body mass index (BMI), glucose, insulin, homeostasis model assessment of insulin resistance (HOMA-IR) and lipid values in obese children. We included 100 newly diagnosed obese children and 100 healthy children. The rs1421085 (CC/CT) (p = 0.019) and rs9939609 (AA/AT) (p = 0.002) polymorphism regions were higher in the obese group. Additionally, we found that both the rs1421085 (CC/CT) and rs9939609 (AA/AT) polymorphism associated with high-density lipoprotein cholesterol (p = 0.011 and p = 0.003) and triglycerides (p = 0.01 and p = 0.004) level, respectively. Further, the rs9939609 and rs1421085 variants of FTO gene associated with HDL-cholesterol and triglycerides levels in obese children; however, updated studies with a large sample size are required to establish strong links with genetic variants and risk factors in childhood obesity.

Abstract Context Patients with lipodystrophy have dyslipidemia and insulin resistance. Leptin treatment with metreleptin in lipodystrophy decreases insulin resistance and lowers triglycerides without changing high-density lipoprotein. Detailed measurement of lipoprotein particles with nuclear magnetic resonance (NMR) spectroscopy can offer insights into cardiovascular disease (CVD) risk and lipid metabolism beyond a standard lipid panel. We hypothesized that patients with lipodystrophy would have a more atherogenic lipid profile than controls at baseline, which would be ameliorated with metreleptin treatment. Objective To characterize the lipoprotein profile in patients with lipodystrophy compared with controls and to evaluate effects of metreleptin treatment. Design, Setting, Patients, and Intervention Patients with lipodystrophy (N = 17) were studied before and after metreleptin for 2 weeks and 6 months and compared with 51 insulin-sensitive sex-matched controls. Main Outcome Measures Lipoprotein profiles were measured by NMR with the LP4 deconvolution algorithm, which reports triglyceride-rich lipoprotein particles (TRLPs), high-density lipoprotein particles (HDLPs), and low-density lipoprotein particles (LDLPs). Results Patients with lipodystrophy had elevated large TRLPs and smaller HDLPs and LDLPs compared with controls. Five patients with lipodystrophy had chylomicrons, compared with zero controls. Metreleptin decreased the size and concentration of TRLPs, eliminated chylomicrons in all but one patient, decreased LDLPs, and increased LDLP size. Metreleptin treatment did not have major effects on HDLPs. Conclusions Patients with lipodystrophy had an atherogenic lipoprotein profile at baseline consistent with elevated CVD risk, which improved after metreleptin treatment. The presence of fasting chylomicrons in a subset of patients with lipodystrophy suggests saturation of chylomicron clearance by lipoprotein lipase.

The efficiency of oat bran and barley bran in lowering the induced hyperlipidemia and hypercholesterolemia in blood of male Albino rats (Rattus rattus) was studied. Twenty rats were divided into four groups each consisted of five rats and fed the specified test diets for eight weeks. The first group (G1) is the negative group which was fed basal diet, the second group (G2) was fed 1.0% cholesterol, was the third group (G3) fed 1.0% cholesterol and 10% oats bran, and the fourth group (G4) was fed 1.0% cholesterol and 10% barley bran. Feeding rats on 1% cholesterol significantly increased serum total cholesterol, low density lipoprotein, and very low density lipoprotein and triglyceride and decreased serum high density lipoprotein. Furthermore, enzyme activity of alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase was increased, and lipid peroxide was increased, whereas catalase and glutathione-S-transferase were decreased. Kidney functions parameters in the cholesterol supplemented group were elevated compared with the negative control. In addition, histological alteration in kidney, liver, heart, and testes was observed, compared with the negative control. Hypercholesterolemic rats supplemented with oat bran and barley bran showed significant decrease in lipid parameters, significant increase in high density lipoprotein-cholesterol, improved antioxidant enzyme, and improved histopathology of kidney, liver, heart, and testes. In conclusion, both oat bran and barley bran had protective effects against induced hyperlipidemia and improved histological alterations. Oat bran appeared more efficient than barley bran in lowering the lipid profile levels in hypercholesterolemic rats.

Background: The increasing prevalence of overweight and related diseases has gained more scientific attention. Overweight and obesity are known as a threat to health, and low serum 25-hydroxy vitamin D levels is associated with obesity. Objectives: Therefore, we examined the effect of Pilates training on body composition, lipid profile, and serum 25-hydroxy vitamin D levels in inactive overweight women. Methods: In this clinical study, 28 overweight women were randomly divided into a training group (n = 14) and a control group (n = 14). Pilates training was performed three 60-min sessions during 12 weeks. In two stages, blood samples were collected 48 hours before and after the last protocol exercise training session. During the 12 weeks, the control group had no exercise training. For analyzing within- and between-group changes, paired t-test and ANCOVA with the significant level of P < 0.05 were used, respectively. Results: After 12 weeks of Pilates training in the training group compared to the control group, there was a significant decrease in the body mass index (P = 0.005), cholesterol (P = 0.001) and triglyceride (P = 0.001) values, and serum 25-hydroxy vitamin D levels (P = 0.005), while high-density lipoprotein (P = 0.028) increased significantly. However, no significant change was observed in low-density lipoprotein levels (P = 0.435). Conclusions: According to the results, it can be indicated that 12 weeks of Pilates training have improved serum 25-hydroxy vitamin D levels, changed anthropometry, and lipid profile in inactive overweight women.

Tea Benalu (Scurrula atropurpurea [Bl.] Dans.) and Mango Benalu (Dendrophthoe pentandra L. miq.) containssecondary metabolites with potential antioxidant potential. Antioxidants can inhibit the oxidation process and reduce Total Cholesterol, Triglycerides, Low-Density Lipoprotein (LDL) and increase High-Density Lipoprotein (HDL). The safety of the preparations for the combination of parasites of tea and mango leaves was tested to obtain toxic effects after repeated treatment tests over some time. The purpose of this study was to determine the toxic effects of a combination of methanolic extracts of a combination of tea parasites and mangoes in female rats (Rattus norvegicus) subchronically for 28 days by looking at the results of the clinical biochemical examination on lipid profiles, namely total cholesterol levels, triglycerides, high-density lipoproteins (28%). HDL), and Low-Density Lipoprotein (LDL). The method of this study was experimental with a combination dose of methanol extract of tea parasite leaves and mango parasite leaves at a dose of 250 mg / KgBB, 500 mg / KgBB and 1000 mg / KgBB with 5x replications each treatment. The subjects used white rats (Rattus norvegicus) female Wistar strains aged 6-8 weeks with a minimum body weight of 100 grams. The treatment in this study was in the form of each dose given 5 times a week for 28 days (subchronic toxicity test) orally. Lipid profile levels were analyzed using the one-way ANOVA test. The results showed that the level of control blood lipid profile with a treatment dose of 250 mg / KgBW, 500 mg / KgBW, and 1000 mg / KgBW was not significantly different. The toxicity test of combined extracts of tea leaves and mango parasite leaves of female rat Rattus novergicus on subchronic exposure 28 days with those doses showed no difference tangible between treatments of controls. Metode penelitian ini adalah eksperimental dengan perlakuan dosis kombinasi ekstrak metanol daun benalu teh dan daun benalu mangga dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB dengan ulangan 5x tiap perlakuan. Subjek menggunakan tikus putih (Rattus norvegicus) betina strain wistar yang berumur 6-8 minggu dengan berat badan minimal 100 gram. Perlakuan dalam penelitian ini berupa setiap dosis  diberikan 5 kali seminggu selama 28 hari (uji toksisitas subkronik) secara per-oral. Kadar profil lipid dianalisis menggunakan uji One-way ANOVA. Hasil penelitian menunjukkan bahwa kadar profil lipid darah kontrol dengan perlakuan dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB tidak berbeda nyata. Uji”toksisitas”ekstrak kombinasi daun benalu teh dan daun benalu mangga  tikus betina Rattus novergicus pada paparan subkronik 28 hari dengan”dosis”tersebut menunjukkan hasil tidak”beda”nyata antara perlakuan terhadap”kontrol. ”Hal ini berarti aman dan tidak menimbulkan sifat toksik pada profil lipid tikus wistar betina.Kata Kunci : Profil Lipid, Scurrula atropurpurea, Dendophthoe pentandra, Subkronik, Rattus norvegicus

Objective: To compare lipid profile in patients with diabetic neuropathy and without neuropathy. Study Design: Cross-sectional comparative study. Place and Duration of Study: This study was carried out at Multidisciplinary Lab-1, Department of Biochemistry and Molecular Biology, Army Medical College, from Jan 2020 to Sep 2020 in collaboration with Pak-Emirates Military Hospital, Rawalpindi. Methodology: Eighty-four subjects were selected and divided into three groups, twenty-eight in each. Group Iincluded diagnosed patients with type 2 diabetes mellitus with peripheral neuropathy, group II included diagnosed patients with type 2 diabetes mellitus without peripheral neuropathy and group III consisted of healthy individuals. Biochemical parameters including total cholesterol, triglycerides, high density lipoprotein cholesterol and low density lipoprotein cholesterol were measured. Data was analyzed by SPSS version 22.0. One way ANOVA test was used that followed by post-Hoc Tukey test for group comparison. Results: There was significant difference in triglycerides (p-value 0.001) and high density lipoprotein cholesterol(0.003) levels among group I and III. While, between group II and III significant difference of high density lipoprotein cholesterol was present. There was no significant difference between group I and II regarding all lipid profile parameters. Conclusion: There was significant increase of triglycerides levels and decrease of high density lipoproteincholesterol in patients with diabetic peripheral neuropathy as compared to healthy controls. No significantdifference was observed for all parameters of lipid profile between patients with diabetic peripheral neuropathyand without peripheral neuropathy.

Objective: Ulcerative colitis is a type of inflammatory bowel disease is a chronic gastrointestinal disorder characterized by intestinal inflammation and mucosal tissue damage. We examined the lipid profile levels in murine model of 7,12 Dimethylbenz(a)anthracene induced ulcerative colitis.Methods: Serum was separated from whole blood and was used to determine the lipid profile such as total cholesterol (TC), phospholipids (PL), triglycerides (TG), free fatty acids, high density lipoprotein (HDL-C) and low density lipoprotein (HDL-C).Results: Ulcerative colitis rats exhibit low level of low density lipoprotein cholesterol and total cholesterol. No significant difference was observed in high density lipoprotein and triglycerides and significant difference was observed in phospholipids and free fatty acid serum levels. This communication highlights the lipid profile that occurs in ulcerative colitis.Conclusion: This study, thus, provides valuable information about the disturbances in the lipids and lipoproteins occur in ulcerative colitis.Keywords: Ulcerative colitis, 7,12-Dimethylbenz(a)anthracene, Lipoprotein, Low-density lipoprotein, Phospholipids.
