Isolation of anti-proliferative compounds from plants is always hindered by the complexities of the plant’s nature and tedious processes. Clinacanthus nutans (Burm. f.) Lindau is a medicinal plant with reported anti-proliferative activities. Our study aimed to isolate potential anti-proliferative compounds present in C. nutans plant. To start with, for our study, we came up with a strategy by first profiling the volatile compounds present in the leaf, stem and root of C. nutans using GC-MS. Comparing the plant’s volatile profiles greatly narrowed down our target of study. We decided to start with the isolation and characterization of a pentacyclic terpenoid, i.e., lupeol from the roots of C. nutans, as this compound was found to present abundantly in the roots compared to the leaf or stem. We developed a simple maceration and re-crystallization method, without the necessity to go through the fractionation or column chromatography for the isolation of lupeol. Characterizations of the isolated compound identified the compound as lupeol. The anti-proliferative activity of the isolated lupeol was further investigated against the MCF-7 cell line, which showed comparable anti-proliferative activity with the authentic lupeol and camptothecin. Our strategy to profile every part of the plant first, followed by selection of the most suitable plant part and targeted compound proved useful for further isolation and characterization bioactive compound from C. nutans.

The study was carried out to identify the compound responsible for hypolipidemic and hypoglycemic effects of Cajanus cajan (redgram). The methanol extract of redgram seeds was found to decrease fasting blood glucose and lipid profile (p<0.001) on streptozotocin-induced mice compared to control. This activity- guided fraction led to the isolation of a compound, substituted benzene containing polyhydroxy functions fused with lactone (CCA3) by analysis of 1H and 13C-NMR data. Biological studies of the isolated compound possessed prominent hypolipidemic activity. Although a number of hypoglycemic compounds are reported, yet not any hypolipidemic compound from redgram. The compound CCA3 seems to be the first report on hypolipidemic activity from methanol extract of redgram. TAJ 2011; 24(1): 6-10

Aristolochic Acids (AAs) are major components of plants in Aristolochia species. The plant Aristolochia indica Linn. has diverse biological actions such as analgesic, anti-diabetics, anti-bacterial, anti-fungal and treatment of malaria and fevers. Here, we report a new and simple isolation method of Aristolochic Acid-Ι (AA-I) from leaves of Aristolochia indica Linn. Defatted leaves of A. indica were extracted with chloroform by soxhlation. AA-I was further isolated from the chloroform extract by partitioning with aqueous sodium bicarbonate. AA-I was purified by repetitive washings with methanol and recrystallization in methanol yielded yellow leaflets. The purity of the isolated compound was ascertained by HPLC analysis. structural elucidation of the isolated compound was done by IR, MS and NMR spectral analysis.

Coffee capsules market is on the rise as it allows access to a wide selection of coffee, differing in taste and brand. However, few data about the chemical characterization of the capsule-brewed coffee aroma are available. In this work, an untargeted approach using headspace solid-phase microextraction (HS-SPME) coupled to gas chromatography–mass spectrometry (GC-MS) and combined to chemometrics was performed to study and compare aroma profile from 65 capsule-brewed espresso coffees (ECs) commercialized by five of the most representative brands in Italy. Volatile profiles obtained from ECs were subjected to multivariate statistical analysis, which generally did not show a significant variability among coffees belonging to the same brand, except for those modified after the addition of specific flavor additives or aromatic substances (such as caramel, chocolate, etc.). Similarities may be related to the starting coffee brew or the processing method, which is likely the same for each individual brand. Additionally, partial least squares discriminant analysis (PLS-DA) showed that capsules from a specific brand contain the highest concentration of pyrazines, thus characterized by an intense and characteristic aroma, and a stronger note than those from the other brands. This study supports that the chemical analysis in conjunction with chemometric tools is a useful approach for assessing flavor quality, even if the need remains to identify volatile markers of high-quality beverages.

The present study is aimed at the isolation and characterization of betulin from Entada africana.  A dichloromethane soluble portion of the stem bark methanol/acetone (1:1 v/v) extract was subjected to gradient elution using ethyl acetate in hexane (5 – 30 %) on an open column. A pure compound was obtained with Rf = 0.61 in hexane/ethyl acetate (8:2 v/v) after repeated washing and recrystallization from methanol and coded Enac1. The pure compound was analyzed using IR, 1H & 13C NMR and GC-MS. Clinical isolates of Escherichia coli, Klebsiella pneumoniae, Salmonella typhi and Staphylococcus aureus were used to assess the antibacterial activity of the pure compound while its preliminary Cytotoxicity was evaluated using brine shrimp nauplii. Based on the spectroscopic data obtained and in comparison with literature, the isolated compound was identified as betulin. The minimum inhibitory concentration (MIC) of betulin ranged between 62.50 - 250.00 µg/mL for all the four bacterial isolates in the study while its fifty percent lethal concentration (LC50) was 10.00 µg/mL. Significant Cytotoxicity with moderate antibacterial activity was observed. The study therefore justifies the existence of bioactive compounds in the stem bark of Entada africana and its use in traditional medicine.

The genus Nocardiopsis is well known to produce secondary metabolites especially antibacterial bioactive compound. Isolation and characterization of bioactive compounds producing novel isolates from unusual habitats are crucial. The present study was aimed to explore Didwana dry salt lake of Rajasthan state in India for the isolation and characterization of actinomycetes. The isolated actinomycetes isolates were characterized based on culture characteristics, biochemical tests and 16S rRNA gene sequencing. The 16S rRNA gene sequence analysis revealed that all the five isolates inhabiting soil of the said dry salt lake of Didwana, Rajasthan belonged to four species of Nocardiopsis viz., N. synnemataformans, N. potens, N. prasina and N. dassonvillei subsp. albirubida. The molecular identification based on 16S rRNA gene sequences was found accurate and robust. The phylogram generated through multiple sequence alignment of all the test isolates of Nocardiopsis revealed that the isolates aroused from a single branch and validated monophyletic association. The present study is the first report of exploring Nocardiopsis isolates from the dry salt lake. These characterized Nocardiopsis isolates isolated from Didwana dry salt lake habitat are novel stains and can be of significance in the detection and utilization of novel bioactive compounds.

The present work involves extraction of phytochemicals from aerial part of Phyllanthus amarus Schum. & Thonn with n-hexane and 85% methanol. The isolation and characterization of Phytoconstituents was done from the methanol extract through portioning with chloroform and ethyl acetate. Fractionation and isolation (using column and thin layer chromatography respectively) of ethyl acetate column pooled portion afforded a compound coded as E-3.3C. The structure of the isolated compound was established on spectroscopic evidences (IR, 1HNMR, MS), which revealed the compound as 1-de (oxygalloyl)-2ˈ,3ˈ,-di-methoxy-amariin a hydrolysable tannins.
