Objective: This research aims to analyze the ability of robusta coffee leaves fraction extract to inhibit the growth of Staphylococcus aureus and Escherichia coli and also determine the minimum inhibitory concentration (MIC). Methods: Antibacterial activity evaluated by the disc diffusion method observed in four types of fraction of extract robusta coffee leaves (n-hexane, ethyl acetate, ethanol, and water). Each extract divided into three various concentrations, 5%, 10%, and 15%. Determination of antimicrobial activity in vitro by the disk diffusion method. Results: Ethyl acetate fraction of coffee leaves extract produced the largest diameter zone of inhibition of bacterial growth compared to other extraction fractions of 17.28 mm in E. coli and 18.58 mm in S. aureus. The MIC of coffee leaves extract fraction water, ethyl acetate, and n-hexane on E. coli and S. aureus is 5%, while the fraction ethanol MIC is 10%. Conclusion: The antibacterial effect of ethyl acetate fraction of coffee leaves extract showed an antibacterial effect that was better than the fraction of n-hexane, ethanol, and water.

This study aims to determine the antibacterial activity and TLC-bioautography profile of the active fractions of Muntingia calabura L. leaves against Staphylococcus aureus. Muntingia calabura L. leaves were macerated with ethanol 96% then fractionated with n-hexane, ethyl acetate, and methanol solvent, respectively. The antibacterial activity was tested by the Kirby-Bauer method to determine the most active fraction and the lowest concentration that inhibited the growth of Staphylococcus aureus. TLC-bioautography was tested using chloroform: ethyl acetate (2:8) as the mobile phase and silica gel F254 as the stationary phase. Antibacterial activity test of n-hexane and ethyl acetate fractions at a concentration of 10% w/v showed activities with inhibition zone diameter of 0.33±0.288 and 9.66±5.77 mm, respectively. At the same time, the methanol fraction showed no activity. The lowest concentration of ethyl acetate fraction which still showed the inhibition zone was 0.312% w/v. The TLC-bioautography profile showed active spots with an Rf value of 0.82 and had an inhibitory zone diameter of 4.013±0.864 mm. It can be concluded that the ethyl acetate fraction was the most active fraction that inhibited the growth of Staphylococcus aureus and had one active spot on the bioautography test.

ABSTRACT Noni is a nutritious plant because it has several ingredients that are important for health. Antibacterial components in noni leaves include glycosides, acubins, saponins and flavonoids. The purpose of this study was to determine the fraction of the extract of noni leaf ethanol whether it has an antibacterial effect and know the class of compounds indentifed as having antibacterial activity after TLC-Bioautografi testing was carried out. The samples were extracted using maceration method with 96% ethanol solvent and fractionation using liquid-liquid fractionation method with methanol, ethyl acetat and n-hexane solvents. Antibacterial activity testing was carried out with concentration variants of 20%, 30% and 40% with agar diffusion method (Kirby and Bauer). The results showed that methanol fraction, ethyl acetat fraction and Noni hexane leaf fraction effectively inhibited Staphylococcus aureus and Escherichia coli bacteria. The largest fraction of inhibiting zone was the concentration of 40% methanol which was categorized as strong. TLC monitoring was carried out using the mobile phase of chloroform : n-hexane (2:1). Antibacterial activity testing carried out by contact bioautography method showed that there were spots on the TLC choromatogram, which produced inhibitory zones. Characteristics of blotches were performed with AlCl3 spotting and it was thought that the spots were flavonoids.  Keywords : Noni, Antibacterial, Staphylococcus aureus, Escherichia coli, TLC-Bioautografi.  ABSTRAK Mengkudu merupakan tanaman yang berkhasiat karena mempunyai beberapa kandungan senyawa yang penting bagi kesehatan tubuh. Objective: Colletotrichium capsici endophytic fungi isolated from Jamblang plant (Eugenia cumini  L).  The secondary metabolites of this plant has the potent antibacterial efficacy  as well as diarrheal and anti-diabetic. This research focussed on isolation of the endophytic microbes from branches of Jamblang plants and their secondary metabolites. Methods: Isolation of endophytes were performed in PDA(potato dextrose agar) using direct seed plant. Endophytic fungi isolates with strongest antimicrobial activity against the bacteria Staphylococcus aureus, Escherichia coli were fermented in Potato Dextrose Yeast (PDY) to produce  large scale of the metabolites.  Supernatant was extracted with ethyl acetate solvent. Ethyl acetate extract fractionated by column chromatography (SiO3, n-hexane- ethyl acetate = 50:1 ~ 1:1) and obtained three fractions. Further, agar diffusion method was performed to assess their anti-microbial activity. Results:  Antibacterial  test  results  indicated that fraction III had  the antibacterial  activity Staphylococcus aureus with  inhibition zone diameter  of 10.7 mm but no observed antibacterial activity against Escherichia coli.  Furthermore, identification  by GC-MS showed that compounds present in fraction III was mainly fatty acid and phenolic compounds. Conclusion:  In conclusion, secondary metabolites isolated from Jamblang plants branches contained predominantly fatty acid and phenol related compounds that could be responsible for its potent anti microbial activity. Keywords : Endophytic fungi, Jamblang (Eugenia cumini L.), antimicrobial activity

North Sumatera is one of the central areas of purple passion fruit production in Indonesia. Processing passion fruit into beverage products (passion fruit juice) produces peel  has not been utilized. The use of passion fruit skin needs to be studied so that it can be useful as a raw material for antibacterial drug preparations. The aim of this study was to determine the phytochemical constituent screening  and antibacterial activity of ethyl acetate fraction of purple passion fruit peel against Staphylococcus aureus and Escherichia coli. Simplicia and ethyl acetate fraction were determinated  its phytochemical properties.  The extraction process by percolation method using ethanol  96% and continue to fractionation process by liquid liquid extraction method using n-hexane and ethyl acetate. The antibacterial activity were tested  against Staphylococcus aureus and Escherichia coli using agar diffusion method with paper discs. The result showed  that  the simplicia characteristic of passion fruit peel  were water content of  8.64%, water soluble extract of  31.69%, ethanol soluble extract of 13.02%, ash total of 7.89%, and insoluble ash in acid of 0.816%. The phytochemical screening simplicia and ethyl acetate fraction showed the presence of flavonoids, glycosides, saponins and tannins. The antibacterial activity test showed that the ethyl acetate fraction has an effective inhibition at the concentration of 100 mg/ml against Staphylococcus aureus and Escherichia coli, it showed dose dependent manner. The ethyl acetate fraction of passion fruit peel (Passiflora edulis Sims) has an antibacterial activity  on Staphylococcus aureus and Escherichia coli
