Abstract Monosodium glutamate is one of the additives used a food flavoring and is widely known by society. The consumption of MSG in large quantities leads to the lesions of the hypothalamus nucleus, leading to some changes in the reproductive system. The aim of this study is to find out the influence of green tea extract against Apoptosis cells Granulose ovarian follicle Rattus norvegicus Dipajan MSG. The research method of using a true experimental research approach post test only control group Design. The sample used was a female white rat (Rattus norvegicus) the number of 25 tails divided into 5 groups. Exposure of monosodium glutamate administered at a dose of 0.7 mg/grBB per oral and green tea extract a variety of doses administered 2 hours thereafter during 30 days after the rats were obtained in the Proestrus phase. Apoptosis examination uses In Situ Cell Death Detection Kit, POD catalog number 11684817910 of Roche brand with Tunnel method. Data were analyzed using the One Way ANOVA test. Results showed that the lowest average apoptosis rate was seen in the MSG Administration treatment group and green tea extract dose 1.4 mg/tail/day (18,686 ± 2,247) but increased again on the group's MSG administration treatment and green tea extract dose 2.8 mg/tail/day (23,752 ± 3,206). Background: The type of injectable contraceptive that is currently widely used is Depo Medroxyprogesterone Acetate (DMPA) which contains the hormone progestin, which is a synthetic material from progesterone. The use of progestin contraceptives can reduce endometrial proliferation and increase apoptosis in endometrial tissue. Cell apoptosis is triggered by an imbalance between positive signals (cell growth factors) and negative signals (DNA damage and increased levels of oxidants) under conditions of oxidative stress. Apoptosis of oocytes in large numbers will cause tissue damage which is characterized by atrophy of the ovaries and causes decreased reproductive function. Oxidative stress can be overcome with endogenous and exogenous antioxidants to reduce free radicals in the body. Green tea is one of the herbal plants that act as antioxidants and phytoestrogens, through the polyphenol compounds contained therein.Purpose: To know the role of green tea in providing protective effects on the ovaries, endometrium and aorta in rats.Methods : The research design used in this study was a true experiment with a post test only control group design approach. In this study, the researchers treated green tea extract at a dose of 10.8 mg/rat/day, 21.6 mg/rat/day, and 43.2 mg/rat/day and exposure to DMPA at a dose of 2.7 mg/rat/day. Green tea extract was administered orally for 28 days. The research hypothesis is that green tea can increase the number of follicles, the number of endometrial glands and reduce the arteriosclerosis score of blood vessels. The preparations were stained by the Hemotoxin-Eosin (HE) method and using the OlyVIA software.Result: There was a significant decrease in the number of corpus luteum, endometrial glands and arteriosclerosis scores due to DMPA exposure but not on the number of follicles. Green tea extract was able to increase the number of endometrial glands at a dose of 10.8 mg/day, but there was no significant difference between doses. Green tea extract was able to reduce arteriosclerosis scores, but the most effective dose was 43.2 mg/day.Conclusion: DMPA exposure was able to significantly reduce the number of corpus luteum, the number of endometrial glands and arteriosclerosis score, but there was no significant difference in the number of primary, secondary, tertiary and de Graff follicles, there was a difference in the number of corpus luteum in the group exposed to DMPA, green tea extract was able to increase the number of endometrial glands and decrease the arteriosclerosis score.Suugestion It is necessary to determine the phase of the reproductive cycle before calculating the number of follicles and examining the level of the hormone estrogen to determine whether the experimental animal has become a hypoestrogen model or not. Keywords : Progesterone, Contraception, Green Tea, Antioxidant ABSTRAK Latar Belakang : Jenis kontrasepsi suntik yang saat ini banyak digunakan adalah Depo Medroxyprogesterone Acetate (DMPA) yang mengandung hormon progestin yaitu bahan sintestis dari progesteron. Penggunaan kontrasepsi progestin dapat menurunkan proliferasi endometrium dan meningkatkan apoptosis pada jaringan endometrium. Apoptosis sel dipicu karena adanya ketidakseimbangan antara sinyal positif (faktor pertumbuhan sel) dan sinyal negatif (kerusakan DNA dan peningkatan kadar oksidan) pada kondisi stress oksidatif. Apoptosis pada oosit dalam jumlah yang besar akan mengakibatkan kerusakan jaringan yang ditandai dengan terjadinya atrofi pada ovarium dan menyebabkan fungsi reproduksi menurun. Stress oksidatif dapat diatasi dengan antioksidan endogen dan eksogen untuk menurunkan radikal bebas dalam tubuh. Teh hijau merupakan salah satu tanaman herbal yang berperan sebagai antioksidan dan fitoestrogen, melalui senyawa polifenol yang terkandung didalamnya.Tujuan : Mengetahui peran teh hijau dalam meberikan efek protektif terhadap ovarium, endometrium dan aorta pada tikus.Metode : Rancangan penelitian yang digunakan dalam penelitian ini adalah true experiment dengan pendekatan post test only control group design. Dalam penelitian ini perlakuan peneliti adalah pemberian ekstrak teh hijau dengan dosis 10,8 mg/tikus/hari, 21,6 mg/tikus/hari, dan 43,2 mg/tikus/hari dan paparan DMPA dosis 2,7 mg/tikus/hari pada tikus. Ekstrak teh Hijau diberikan secara oral selama 28 hari. Hipotesis penelitian adalah teh hijau mampu meningkatkan jumlah folikel, jumlah kelenjar endometrium dan menurunkan skor arteriosklerosis pembuluh darah. Preparat diwarnai dengan metode Hemotoxin-Eosin (HE) dan menggunakan software OlyVIA.Hasil : Terjadi penurunan jumlah korpus luteum, kelenjar endometrium dan skor arteriosklerosis akibat paparan DMPA secara bermakna namun tidak pada jumlah folikel. Ekstrak teh hijau mampu meningkatkan jumlah kelenjar endometrium pada dosis 10.8 mg/hari, namun tidak ada perbedaan yang signifikan antar dosis. Ekstrak teh hijau mampu menurunkan skor arteriosklerosis, namun yang paling efektif adalah dosis 43.2 mg/hari.Kesimpulan : Paparan DMPA mampu menurunkan jumlah korpus luteum, jumlah kelenjar endometrium dan skor arteriosklerosis secara bermakna, namun tidak ada perbedaan yang bermakna pada jumlah folikle primer, sekunder, tersier dan de Graff, ada perbedaan jumlah korpus luteum pada kelompok yang dipapar DMPA, ekstrak teh hijau mampu meningkatkan jumlah kelenjar endometrium  dan menurunkan skor arteriosklerosis.Saran Perlu menentukan fase siklus reproduksi sebelum dilakukan perhitungan jumlah folikel dan pemeriksaan kadar hormon estrogen untuk menentukan apakah hewan coba sudah menjadi model hypoestrogen atau belum. Kata Kunci :  DMPA, Kontrasepsi, Teh Hijau, Antioksidan

Abstract: This study was aimed to evaluate the effectiveness of 20% green tea extract in preventing the increase of melanin in guinea pig skin exposed to UV-B and to compare the effectiveness of this treatment to 4% hydroquinone cream. This was an experimental study with the post-test only control group design. Subjects were four groups; each consisted of 7 male guinea pigs. Group 1, the control group, was exposed to UV-B only. Group 2 was exposed to UV-B and applied with cream based substance. Group 3 was exposed to UV-B and applied with 4% hydroquinone cream. Group 4 was exposed to UV-B and applied with 20% green tea extract. The total UV-B intensity was 390 mJ/cm2 consistently exposed for two weeks. Histopathological slides of the skin tissue were stained with Masson Fontana technique. The total amount of melanin was calculated in percentage of pixel compared to pixel of melanin in total epidermis. The results showed that the highest amount of melanin was in group 1 (24.44%) and the lowest amount of melanin was in group 3 (1.04%) meanwhile the amount of melanin in group 4 (1.34%) was nearly the same with group 3. There were significant differences between group 1 to group 3 and 4 (P < 0.05). There was no significant difference between group 3 and group 4 (P > 0.05). Conclusion: The 20% green tea extract was as effective as 4% hydroquinone cream in inhibiting the increase of melanin in guinea pigs’skin exposed to UV-B.Keywords: green tea extract, melanin, ultraviolet BAbstrak: Penelitian ini bertujuan untuk mengetahui efek pemberian krim ekstrak teh hijau 20% terhadap peningkatan jumlah melanin pada kulit marmut yang dipajan sinar UVB serta membandingkan efektivitasnya dengan krim hidrokuinon 4%. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Subyek penelitian terdiri dari empat kelompok dengan 7 ekor marmut jantan setiap kelompok. Kelompok 1 (kelompok kontrol) diberi pajanan sinar UV-B saja. Kelompok 2 diberi pajanan sinar UV-B dan diolesi krim dasar. Kelompok 3 diberi pajanan sinar UV-B dan krim hidrokuinon 4% sedangkan kelompok 4 diberi pajanan sinar UV-B dan krim ekstrak teh hijau 20%. Dosis total UVB yaitu 390 mJ/cm2 diberikan selama 2 minggu. Sediaan histopatologik jaringan kulit dipulas dengan pewarnaan Masson Fontana. Jumlah melanin dihitung dengan persentase pixel luas area melanin dibandingkan dengan pixel seluruh jaringan epidermis. Hasil penelitian menunjukkan jumlah melanin tertinggi pada kelompok 1 (24,44%) dan terendah pada kelompok 3 (1,04%). Jumlah melanin pada kelompok 4 (1,34%) hampir sama dengan kelompok 3. Terdapat perbedaan bermakna antara kelompok 1 dengan kelompok 3 dan 4 (P < 0,05). Perbandingan antara kelompok 3 dan 4 tidak berbeda bermakna dalam mencegah peningkatan jumlah melanin (P > 0,05). Simpulan: Pemberian krim ekstrak teh hijau 20% sama efektif dengan krim hidrokuinon 4% dalam mencegah peningkatan jumlah melanin kulit marmut yang dipajan sinar UV-B.Kata kunci: krim ekstrak teh hijau, melanin, ultraviolet B

Background: Zinc oxide nanoparticles (ZnO NPs) are increasingly utilized in both industrial and medical applications. Therefore, the study was aimed to investigate the effect of green nanoparticle complex (green tea extract/zinc oxide nanoparticles complex, GTE/ZnO NPs) on oxidative stress induced by monosodium glutamate (MSG) on the liver of rats. Methods: Wistar male rats (n=64) weighing between 200-250 g were divided randomly into eight groups: control group was given physiological saline (1 mg/kg), two groups were treated with two different doses of MSG (MSG-LD, MSG-HD; 6 and 17.5 mg/Kg, respectively), GTE was given 1 mg/mL, 5th group was treated with ZnO NPs and 6th group was treated with GTE/ZnO NPs complex while, 7th and 8th groups were treated with MSG-LD + GTE/ZnO NPs complex and MSG-HD + GTE/ZnO NPs complex, respectively. All substances were given orally for 30 consecutive days. At the end of the study, the liver was homogenized for measurement of the oxidative stress status and anti-inflammatory biomarkers as well as histological and transmission alternations. Results: Results showed that the antioxidant enzymes activity and glutathione level were significantly decreased in MSG groups than control in a dose-dependent manner. Conversely, the malondialdehyde and inflammatory cytokines levels were significantly increased in MSG groups than the control group. The liver indicated no evidence of alteration in oxidative status, anti-inflammatory and morphological parameters in GTE, ZnO NPs and GTE/ZnO NPs complex groups. Conclusion: In conclusion, MSG at both doses caused oxidative stress and inflammation on liver after 28 days of exposure that supported histological analysis and transmission view of hepatic parenchyma. GTE/ZnO NPs act as partial hepato-protective against MSG.

The aim of this study was to determine the effect of green tea (Camelia sinensis) extract on hemoglobin and hematocrit level of Wistar rat (Rattus norvegicus). Sample animals used were 20 rats strain Wistar aged 2.5-3.0 months. Green tea extract was given for 30 days consecutively. Blood collection was conducted via orbitalis sinus for 2 ml using capillary pipette. Rats were fed with standart commercial food (Turbo Feed T.79-4) and aquadest as drinking water ad libitum. This study used completely randomized design (CRD) pattern with 4 treatments and 5 replications. The control group (P0) without any treatment, P1 was given green tea extract 0.045 g/ml/day, P2 was given green tea extract 0.060 g/ml/day, and P3 was given green tea extract 0.075 g/ml/day. Data were analyzed by oneway analysis of variance (Anova). The result showed that mean±SD of hemoglobin levels (g/dl) on each treatment group (P0, P1, P2, and P3) were 11.86±0.55, 12.58±1.40, 11.82±0.69, and 12.50±0.84 respectivey. Mean±SD of hematocrit levels (%) on each treatment group (P0, P1, P2, and P3) were 44.00±4.30, 43.40±1.14, 44.00±1.58, and 42.80±1.79. To conclude, the administration of green tea extract has no effect (P0.05) on haemoglobin and hematocrit levels of Wistar rat. ____________________________________________________________________________________________________________________ Key words: green tea, haemoglobin, haematocrit, Wistar rat

AbstrakVaksinasi BCG merupakan metoda pencegahan infeksi M. tuberculosis dengan rentang efektivitas antara 0 – 80%. Vaksin ini bekerja dengan meng-induksi sistem imunitas seluler, terutama sel limfosit T. Beberapa faktor yang mempengaruhi efektivitas vaksin, seperti bahan dasar, rute pemberian, penyimpanan dan lain sebagainya.penelitian ini bertujuan untuk menilai pengaruh rute vaksinasi BCG, per oral dan subkutan terhadap imunitas seluler dan humoral, yang meliputi pembentukan antibodi, kosentrasi sel limfosit B, T dan penilaian kemampuan makrofag.Penelitian eksperimental yang dilaksanakan dengan mengunakan desain post test only control group design. Jumlah sampel 10 ekor Rattus norvegicus galur Wistar. Antibodi diperiksa dengan menggunakan ELISA, serta dilakukan uji statistik mengunakan t-test.Didapatkan kadar antibodi kelompok subkutan pada hari ke-10 adalah OD 1.714 ± 0.124, sedangkan kelompok oral adalah 1.256 ± 0.243 dan pada kontrol kadar antibodi adalah 0.975 ± 0.253 (p < 0.05). Jumlah limfosit B pada kelompok subkutan adalah 4.5 x 103 sel/ml, kelompok oral 2.6 x 103 sel/ml dan kontrol 2.2. x 103 sel/ml (p < 0.05). Jumlah limfosit T kelompok subkutan adalah 3.8 x 104 sel/ml sedangkan pada kelompok oral dan kontrol, masing-masing adalah 1.1 x 104 sel/ml dan 0.8 x 104 sel/ml (p < 0.05). Kemampuan fagositosis makrofag subkutan adalah 62.5 ± 6.8 lebih tinggi dibanding kelompok oral 48.7 ± 5.8 dan kontrol 35.6 ± 4.4 (p < 0.05). Fenomena yang sama ditemukan pada hari ke-20. Namun demikian tidak ditemukan perbedaan konsentrasi seluruh variabel seluler dan humoral antara hari ke-10 dan 20 (p > 0.05).Vaksinasi BCG akan meningkatkan respon imunitas seluler dan humoral dan rute vaksinasi subkutan lebih baik dibanding per oral ditinjau dari aspek respon imun.Kata kunci : Vaksinasi BCG, seluler, humoral, makrofagAbstractBCG vaccine is a method to prevent the infection of M tuberculosis with effectiveness range between 0 – 80%. The amount of Lymphocyte B in sub cutaneous group is 4.5 x 103 cell/ml, 2.6 x 103 cell/ml in oral and 2.2 x 103 cell/ml (p < 0.05) in control. The amount of Lymphocyte T in sub cutaneous is 3.8 x 104 cell/ml, while in oral and control is 1.1 x 104 cell/ml and 0.8 x 104 cell/ml (p < 0.05). The phagocytosis ability of sub cutaneous’s macrophage is 62.5 ± 6.8 which is higher than oral group 48.7 ± 5.8 and control 35.6 ± 4.4 (p < 0.05). The same phenomenon also found in day 20th. However, the difference concentrations of the entire cellular and humeral variable between day 10th and day 20th ( p < 0.05 ) has not been found.Based on the result of this experiment’s it can be concluded that BCG vaccination will increase the cellular and humeral immunity response and sub cutaneous’s vaccination route is better that oral, from immunes response aspect side.Key word : BCG Vaccination, cellular, humeral, macrophage.

Carbon tetrachloride (CCl₄) is a chemical that can cause damage to liver cells. One of the natural ingredients developed to reduce liver damage due to chemicals and infections is Chlorella sp. The research aimed to explore the hepatoprotective effect of the antioxidant superoxide dismutase (SOD) by administering Sun Chlorella in Rattus norvegicus CCl₄ induced rats. The study design used a post-test with a control group design with a completely randomized design trial on 30 male rats of Wistar strains, aged 2–3 months, 200–250 grams in weight. The research was conducted in November 2016–January 2017 in Purwokerto. Rats were divided into five groups and treated for four weeks as follows: K1 group was given aquades; K2 was given CCl₄; K3, K4, K5 were given CCl₄ and Sun Chlorella 3.6 mg/200 gBW, 7.2 mg/200 gBW, and 14.4 mg/200 gBW respectively, by the gastric probe. Statistical analysis with correlation test and one way ANOVA multivariate test showed that Sun Chlorella 7.2 and 14.4 mg/200 gBW significantly increased SOD levels in rats induced CCl₄ (p=0.004, p=0.009). SOD rates were significantly strong associated with aspartate aminotransferase/AST (r=−0.685, p=0.000) and alanine aminotransferase/ALT (r=−0.659, p=0.000). The conclusion is Sun Chlorella increases SOD levels in CCl₄-induced rats. Increased SOD levels may decrease AST and ALT levels. EFEK HEPATOPROTEKTIF SUN CHLORELLA SEBAGAI ANTIOKSIDAN PADA RATTUS NORVEGICUS YANG DIINDUKSI KARBON TETRAKLORIDAKarbon tetraklorida (CCl₄) merupakan bahan kimia yang dapat menyebabkan kerusakan sel hati. Berbagai bahan alami telah dikembangkan untuk mengurangi kerusakan hati baik akibat bahan kimia maupun infeksi, salah satunya adalah Chlorella sp. Penelitian ini bertujuan melihat efek hepatoprotektif  dengan pemberian Sun Chlorella pada tikus Rattus norvegicus yang diinduksi CCl₄. Desain penelitian menggunakan post-test with a control group dengan rancangan percobaan rancangan acak lengkap pada 30 tikus jantan galur Wistar, usia 2–3 bulan, dan berat 200–250 gram. Penelitian ini dilakukan di Purwokerto pada periode November 2016–Januari 2017. Tikus dibagi menjadi lima kelompok dan mendapatkan perlakuan per oral dengan sonde lambung selama 4 minggu sebagai berikut: kelompok K1 sebagi kontrol negatif diberikan aquades; kelompok control positif K2 diberikan CCl₄; kelompok perlakuan K3, K4, dan K5 diberikan CCl4 dan Sun Chlorella 3,6 mg/200 gBB; 7,2 mg/200 gBB; 14,4 mg/200 gBB tikus berurutan. Uji statistik dengan menggunakan uji korelasi dan ANOVA satu arah menunjukkan bahwa pemberian Sun Chlorella 7,2 mg dan 14,4 mg meningkatkan kadar SOD pada tikus yang diinduksi CCl₄ secara bermakna (p=0,004; p=0,009). Kadar SOD berhubungan kuat dengan kadar aspartat aminotransferase/AST (r=−0,685; p=0,000) dan alanin aminotransferase/ALT (r=−0,659; p=0,000). Kesimpulan penelitian ini adalah pemberian Sun Chlorella meningkatkan kadar SOD pada tikus yang diinduksi CCl₄. Peningkatan kadar SOD menyebabkan penurunan kadar AST dan ALT.

<p>Perubahan pola demografi di negara maju dan negara berkembang, angka kejadian infertilitas di negara maju dilaporkan sekitar 5%-8% dan di negara berkembang sekitar 30%.WHO memperkirakan sekitar 8%-10% atau sekitar 50-80 juta pasangan suami istri di seluruh dunia mengalami masalah infertilitas, sehingga membuat infertilitas menjadi masalah mendesak. Untuk itu diperlukan pengendalian infertilitas, salah satunya adalah kewaspadaan perubahan gaya hidup, perubahan ini juga mempengaruhi pola konsumsi makanan dengan lebih banyak mengkonsumsi jenis makanan cepat saji yang banyak mengandung zat aditif (penyedap rasa. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian monosodium glutamate terhadap kadar hormon estradiol dan kadar hormon progesteron pada tikus putih betina ( <em>Rattus norvegicus ).</em></p><p>Penelitian ini menggunakan metode pendekatan <em>post test only control group design,</em> terhadap tikus putih betina dengan berat 200 – 250 gr. Sampel terdiri dari 24 ekor tikus yang dibagi 4 kelompok yaitu kelompok kontrol ( K ), perlakuan I, II dan III . Kelompok perlakuan diberikan monosodium glutamat dengan dosis masing-masing : 45 mg, 54 mg dan 63 mg setiap hari diberikan peroral yang dilarutkan dengan aquabides 2 ml selama 20 hari yang dimulai pada awal fase proestrus. Setelah 20 hari perlakuan tikus di korbankan dan diambil darahnya. Pemeriksaan kadar hormone estradiol dan progesteron menggunakan Elisa Spectrophotometer.  Kemudian hasilnya dianalisa dengan menggunakan One Way <em>ANOVA</em> dan dilanjutkan dengan uji <em>Multiple Comparison</em> jenis<em> Bonferroni.</em></p><p>Hasil penelitian pemberian  monosodium glutamat dengan dosis 45 mg/ ekor/ hari, 54 mg/ekor/ hari dan 63 mg/ ekor /hari dapat menurunkan kadar hormon estradiol tikus putih betina <em>(Rattus norvegicus)</em> secara signifikan. Dan pemberian monosodium glutamate dengan dosis 45 mg/ ekor/ hari dapat menurunkan kadar hormon progesteron tikus putih betina<em> (Rattus norvegicus)</em> walaupun tidak berpengaruh secara signifikan , dan pada dosis 54 mg/ekor/ hari dan 63 mg/ ekor /hari memberikan pengaruh  terhadap penurunan kadar hormon progesteron secara signifikan.</p><p>Dapat di simpulkan ada pengaruh pemberian monosodium glutamat terhadap penurunan kadar hormon estradiol dan kadar hormon progesteron <em> </em>pada tikus putih betina (<em>Rattus norvegicus).</em> Disarankan Untuk pemeriksaan kadar hormon sebaiknya dilakukan segera setelah pengambilan darah. </p>

In last decade there has been an increase prevalence of overweight adolescents. Various strategies and products have been evaluated in effort to prevent and treat overweight using Green Tea Extract (GTE) and Senobi Breathing Exercise (SBE). However, the effect of GTE and SBE on decreasing adolescent body fat percentage in acute conditions when combined are unknown. The method used quasi experimental with control group design. The research conducted in Gymnasium laboratory of the Politeknik Unggulan Kalimantan in March 2020. The samples used were aged 18-21 years with BMI 25-29.9. Determination of samples using Federer's formula: (t-1) (n-1) ?15 obtained 4 treatment groups with a sample size of each group are 10 people that divided into: (a) overweight GTE; (b) overweight SBE; (c) overweight GTE and SBE; (d) overweight control . Measurement percentage of body fat in acute conditions shortly after treatment utilize Body Fat Monitor Scale GEA Medical EF 981. Statistical Test using Krusskall Wallis , the results obtained were P> 0.05, which indicates that there was no significant effect of GTE and SBE to reduce prevalence of adolescent body fat in acute conditions. Further research is needed to find out the correct protocol.

Abstract: Ultraviolet B (UVB) is a source of free radicals that accelerate aging process of the skin such as activating enzymes that degrade collagen and inhibit collagen production by inducing the expression of MMP-1. Lemon peel contains vitamin C, vitamin A, tannins and phenols which possess antioxidant activity and prevent oxidative stress. This study was aimed to prove that oral administration of lemon peel extract could decrease MMP-1 levels and increase the number of collagen in the UVB-induced male Wistar rats (Rattus norvegicus). This was a true experimental study with the post test only control group design. Subjects were 30 male Wistar rats (Rattus norvegicus), aged 2-3 months, body weight 120 g, divided into 3 groups, as follows: the control group (P0) which were not exposed to UVB; the treatment group 1 (P1) given aquabidest and UVB ray exposure; and the treatment group 2 (P2) given lemon peel extract and UVB ray exposure. After 15 days of treatment, all rats were anesthetized and their skin tissues were prepared for histological examination of MMP1 and collagen. The results showed that the average expression of MMP1 in P0 group was 22.02±3.20%; in the P1 group was 29.04±6.36%; and in P2 group was 7.98±2.76% (P <0.01). In addition, the average amount of collagen in P0 group was 70.01±2.99%; in the P1 group was 57.68±4.84%; and in P2 group was 77.45±4.29% (P <0.01). Conclusion: Oral administration of lemon peel extract could decrease the expression of MMP-1 and increase the amount of collagen in the UVB-induced male Wistar rats (Rattus norvegicus).Keywords: lemon peels, collagen, MMP-1, UVB Abstrak: Ultraviolet B (UVB) merupakan sumber radikal bebas yang mempercepat proses penuaan kulit dengan mengaktivasi enzim yang mendegradasi kolagen dan menghambat produksi kkolagen melalui induksi ekspresi MMP-1. Kulit buah lemon mengandung vitamin C, vitamin A, tanin, dan fenol yang memiliki aktivitas antioksidan dan mencegah stres oksidatif. Penelitian ini bertujuan untuk membuktikan bahwa pemberian ekstrak kulit buah lemon per oral dapat menurunkan kadar MMP-1 dan meningkatkan jumlah kolagen pada tikus putih jantan galur Wistar (Rattus norvegicus) yang dipajan sinar UVB. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Subjek penelitian ialah 30 ekor tikus putih jantan, galur Wistar (Rattus norvegicus), umur 2-3 bulan, dengan berat badan 120 gr yang dibagi menjadi 3 kelompok, masing-masing berjumlah 10 ekor tikus. Kelompok kontrol (P0) tidak dipapar sinar UVB; kelompok perlakuan 1 (P1) diberikan aquabides oral dan paparan sinar UVB; dan kelompok perlakuan 2 (P2) diberikan ekstrak kulit buah lemon oral dan paparan sinar UVB. Setelah 15 hari perlakuan, seluruh tikus dianestesi kemudian diambil jaringan kulitnya untuk dibuat preparat histologik dan dihitung jumlah kolagen dermisnya sebagai data post test. Hasil analisis menunjukkan rerata jumlah ekspresi MMP1 pada kelompok P0 ialah 22,02±3,20%; kelompok P1 ialah 29,04±6,36%; dan kelompok P2 ialah 7,98±2,76% (P <0,01). Rerata jumlah kolagen pada kelompok P0 ialah 70,01±2,99%; kelompok P1 ialah 57,68±4,84%; dan kelompok P2 ialah 77,45±4,29% (P <0,01). Simpulan: Pemberian ekstrak kulit buah lemon per oral dapat menurunkan ekspresi MMP-1 dan meningkatkan jumlah kolagen pada tikus putih jantan galur Wistar (Rattus norvegicus) yang dipajan sinar UVB.
