PERBEDAAN UJI AKTIVITAS ANTIOKSIDAN ANTARA EKSTRAK BUAH KIWI DAN APEL SECARA IN VITRO

Radikal bebas yang berbahaya jika konsentrasi ROS akan meningkat sehingga akan mengakibatkan stress oksidatif. Stress oksidatif biasanya disebabkan oleh konsentrasi tinggi dari radikal bebas dalam sel dan jaringan yang dapat diinduksi oleh beberapa faktor negatif seperti gamma, UV, radiasi sinar-X, tekanan psiko-emosional dan makanan yang tercemar.Untuk meminimalisir radikal bebas dalam tubuh diperlukan antioksidan alami seperti ekstrak buah kiwi dan ekstrak buah apel. Uji aktivitas antioksidan pada masing- masing ekstrak dilakukan dengan metode DPPH (1,1- difenil-2-pikrihidazil). Hasil menunjukkan bahwa nilai IC50 dari ekstrak buah kiwi lebih rendah daripada IC50 ekstrak buah apel. Hal ini menunjukkan bahwa ekstrak buah kiwi mempunyai aktivitas antioksidan lebih kuat dari ekstrak buah apel. Oxidative stress is usually caused by high concentrations of free radicals in cells and tissues which can be induced by several negative factors such as gamma, UV, X-ray radiation, psycho-emotional pressure and contaminated food. To minimize free radicals in the body natural antioxidants such as kiwi fruit extract and apple extract. The antioxidant activity test on each extract was carried out by the DPPH method (1,1-diphenyl-2-picrihydazyl). The results showed that IC50 value of kiwi fruit extract was lower than IC50 of apple fruit extract. This shows that kiwi fruit extract has stronger antioxidant activity than apple extract. AbstrakStress oksidatif pada tubuh dapat memicu berbagai penyakit. Stress oksidatif ini disebabkan karena radikal bebas berlebih. Tubuh memerlukan antioksidan untuk mengurangi pengaruh radikal bebas dan meredam dampak negatifnya. Penelitian ini bertujuan untuk mengevaluasi daya antioksidan ekstrak kulit buah jamblang (Syzigium cumini (L.) Skeels) dengan menggunakan metode DPPH. Berdasarkan hasil pengujian daya antioksidan ekstrak kulit buah jamblang diketahui bahwa ekstrak kulit buah jamblang memiliki kandungan antioksidan yang tergolong aktivitas sedang dengan nilai IC50 sebesar 169.3. Hasil penelitian ini menunjukkan bahwa kulit buah jamblang berpotensi untuk dikembangkan sebagai sumber antioksidan alami bagi manusia.Kata Kunci : antioksidan, DPPH, ekstraksi, jamblang, radikal bebas, stress oksidatif AbstractOxidative stress can induce many diseases. Various diseases can be triggered by the condition of oxidative stress in the body. Oxidative stress is a condition that occurs in cells when free radicals are present in excess amounts. To reduce the negative effects and negative effects of these free radicals, the body needs antioxidants. The aim of this research is to know the antioxidant potential of black plum leaf extract (Syzigium cumini (L.) Skeel) using DPPH method. Based on the results of antioxidant potential testing in black plum leaf extract known that it has antioxidant content that is classified as very active with an IC 50 value of 8.85. The results of this study indicate that the leaves of black plum (Syzigium cumini (L.) Skeel have the potential to be developed as a source of natural antioxidants for humans.

The predominance of synthetic and genetically modified foods in the diet of a modern person often leads to disruption of natural metabolic processes and the accumulation of toxic free radicals in the body, which provokes the development of various diseases. Natural antioxidants from medicinal plants can help maintain health and stop the processes of peroxidation. In our opinion, Аstragalus onobrychis can become a promising source of natural antioxidants. The aim of this study was to establish the antioxidant activity in extracts from the herb Astragalus onobrychis. In order to obtain objective and reliable results during our research, we used two methods: the method of titrimetry and spectrophotometry of free radicals. The titrimetric method is based on the chemical interaction between an oxidizing agent (potassium permanganate) and natural antioxidants present in extracts from plant raw materials, since they exhibit the properties of reducing agents. The spectrophotometric method makes it possible to assess the value of antioxidant activity and the duration of the reaction by assessing the interaction of 2,2-diphenyl-1-picrylhydrazil with plant antioxidants. To establish the relationship between the antioxidant activity and the presence of phenolic compounds in the plant, the content of flavonoids and hydroxycinnamic acids in the herb Astragalus onobrychis was determined. As a result of the studies, the total antioxidant activity of extracts from the herb Astragalus onobrychis was established and it was revealed that its value depends on the type of extractant used, since this factor is associated with the content of various groups of natural antioxidants in the extraction. The data obtained indicate the prospects of using the herb Astragalus onobrychis as a source of natural antioxidants.

Background: There exist natural antioxidants in plants that scavenge harmful free radicals from the body. Free radicals are species of chemical origin with an unpaired electron and play a pivotal role in combating against health-related problems like lung damage, inflammation, and cardiovascular ailments etc. Antioxidants halt the development of these free radicals called the reactive oxygen species either by chelating the trace elements or by enzymes inhibition. Objectives: The aim of present review was to collect information about Pakistani medicinally important plants with the exploration of their antioxidant potential. Methodology: Total 206 papers were looked over, which were obtained from numerous sources like; Google Scholar, Medline, PubMed, Research Gate, Science Direct, Scopus and Web of Science. Results: Overall, 93 plants representing 44 families with potential antioxidant activity reported from Pakistan have been presented in this review. Maximum number of species from Asteracea, Poaceae and Rutaceae familes were scrutinized for their potential antioxidant activity from Pakistan. Conclusion: The present review clearly designates that the presence of phytochemicals and antioxidant activity of medicinal plants from Pakistan vary with the species of the plants and material/extracts used. From this review, it is recommended to perform comprehensive experimental investigations based on toxicology and ethnopharmacology on these precious plants from Pakistan. It will be advantageous in the provision of trustworthy information to patients and determine further innovative compounds for safer and new drugs development with fewer side effects.

Crataegusfruit is one of most important fruits in Tunisian flora. Some fruits of this genus are edible. This study was undertaken in order to examine the benefits of these fruits in human health and their composition of antioxidants including total polyphenol, flavonoids, proanthocyanidins content, and total anthocyanins. The antioxidative properties of the ultrasonic methanolic extract were assessed by different in vitro methods such as the FRAP, DPPH, andβ-carotene/linoleic acid assay. We concluded that peel fraction of red fruits possessed relatively high antioxidant activity and might be a rich source of natural antioxidants in comparison with the pulp and seed fruit extract. The results also showed that hawthorn yellow fruit presents lower amounts of phenolic content, absence of anthocyanins, and less antioxidant capacity. Most of peel and seed fractions were stronger than the pulp fractions in antioxidant activity based on their DPPH IC50, FRAP values, and results ofβ-carotene/linoleic acid. The total phenolic compounds contents were also highly correlated with the DPPH method and the FRAP assay.

Antioxidants have an important role in delaying the oxidation process and preventing the occurrence of degenerative diseases in the body. Antioxidants consist of synthetic and natural; the use of synthetic antioxidants in a relatively long period is hazardous so that the use of natural antioxidants is considered more potential and safe for the body. The process of extracting secondary metabolites from the leaves of bangun-bangun (Coleus amboinicus L.) is carried out by the maceration method with methanol, then partitioned in stages with n-hexane, chloroform, and ethyl acetate. This research intends to identify secondary metabolites and test antioxidant activity from the ethyl acetate fraction with the DPPH method. Phytochemical screening results of ethyl acetate fraction showed the different secondary metabolite groups such as alkaloids, phenolics & polyphenols, flavonoids, coumarin, and triterpenoids. Antioxidant activity (IC50) obtained is 64.97 with a healthy category, so that the potential to be used as a natural antioxidant.

Free radicals are constructed by natural physiological activities in the human cells as well as in the environment. They may be produced as a result of diet, smoking, exercise, inflammation, exposure to sunlight, air pollutants, stress, alcohol and drugs. Imbalanced redox status may lead to cellular oxidative stress, which can damage the cells of the body, resulting in an incidence of various diseases. If the endogenous antioxidants do not stop the production of reactive metabolites, they will be needed to bring about a balance in redox status. Natural antioxidants, for example plants, play an important part in this context. This paper seeks to report the available evidence about oxidative stress and the application of plants as antioxidant agents to fight free radicals in the human body. For this purpose, to better understand oxidative stress, the principles of free radical production, the role of free radicals in diseases, antioxidant defense mechanisms, and the role of herbs and diet in oxidative stress are discussed.

Teripang merupakan hewan jenis Echinodermata. Beberapa species teripang, misalnya H. scabra, H. fuscogilva dan T. ananas memiliki nilai jual yang tinggi. Meskipun tidak semua spesies memiliki nilai jual yang tinggi, teripang berpotensi untuk dikembangkan dalam bidang kesehatan, terutama dalam kemampuannya untuk meredam radikal bebas dan mencegah berbagai penyakit degeneratif. Tujuan dari penelitian ini yaitu untuk mengidentifikasi, menentukan aktivitas antioksidan, kandungan total fenol, serta kadar karotenoid pada ekstrak beberapa teripang yang berasal dari Perairan Karimunjawa Jepara. Hasil identifikasi dari ke empat spesies adalah Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, dan Holothuria atra. Ekstraksi dilakukan dengan teknik maserasi dinding tubuh teripang menggunakan pelarut metanol. Uji aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (1,1-difenil-pikrilhidrazil) sebagai agen radikal bebas. Penentuan total fenolik dilakukan menggunakan metode folin-ciocalteu, dan penentuan kadar karotenoid dilakukan dengan menggunakan spektrofotometer. Hasil menunjukkan bahwa aktivitas antioksidan Bohadschia vitiensis, Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, dan Holothuria atra tergolong sangat lemah dengan nilai IC50 secara berturut-turut adalah sebesar 454,28 ± 14,4; 713,51 ± 9,1; 801,57 ± 8,4 dan 1535,47 ppm. Kandungan total fenol sebesar 21,08 ± 0,49; 14,325 ± 0,21; 14,033 ± 0,33 dan 10,67 ± 0,12 mg GAE/g sampel, dan kadar karotenoidnya sebesar 23,28 ± 0,07; 25,78 ± 0,56; 11,85 ± 0,04; dan 41,44 ± 0,008 µmol/g sampel.Kata kunci: Teripang, Antioksidan, DPPH, Fenolik, Karotenoid Sea cucumbers are sea benthos from phylum Echinoderms phylum. Some sea cucumbers such as H. scabra, H. fuscogilva, T. Ananas have a high economical price. Eventhough, some sea cucumbers have the potency of being a healthy food. It it scientifically proven that some sea cucumbers have the ability to reduce free radicals and prevent various degenerative diseases caused by the excessive free radicals. The aims of this study were to identified, determine antioxidant activity, total phenol content, and carotenoid levels in sea cucumber extracts originally from Karimunjawa waters. Some of the sea cucumbers obtained are Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, and Holothuria atra. The extraction was done by macerating the body walls with methanol. The antioxidant activity test were tested using the DPPH method as free radicals, the total phenolic content tested using the folin-ciocalteu method, and determined the carotenoid levels spectrophotometrically. Based on the results, antioxidant activity of  B. vitiensis, S. cf. quadrifasciatus, P. graeffei, and H. atra were  classified as very weak with IC50 454.28 ± 14.4; 713.51 ± 9.1; 801.57 ± 8.4 dan 1535.47 ppm. Total phenolic content were 21.08 ± 0.49; 14.325 ± 0.21; 14.033 ± 0.33 dan 10.67 ± 0.12 mg GAE / g samples, while carotenoid levels were 23.28 ± 0.07; 25.78 ± 0.56; 11.85 ± 0.04; dan 41.44 ± 0.008 µmol / g sample.Keyword : Sea cucumber, Antioxidant, DPPH, Phenolic, Carotenoid

The high number of free radicals that are not balanced with the amount of antioxidants in the body triggers oxidative stress. Oxidative stress causes impaired vascular function, damage to proteins and lipids in membrane cell, and nucleic acid (DNA) mutations. Chronic cell damage has a negative effect on tissue that triggers various diseases such as neurodegenerative diseases (Alzheimer's, Parkinson's), cardiovascular diseases (hypertension, arteriosclerosis, and others), cataracts, retinal damage, maculopathy, rheumatoid arthritis, asthma, stroke, diabetes mellitus , immunodepression, cancer, aging, hyperoxia, dermatitis, and others. The application of a healthy lifestyle for example by consuming food sources of bioactive compounds can minimize health risks. Rice is the staple food of the Indonesian people. Some types of rice contain red and black pigments which are known to have high antioxidant activity compared to white rice. The pigment comes from anthocyanin and proanthocyanidin. Various studies in vitro and in vivo prove that anthocyanin and proantocyanidine act as antioxidants and potency as a preventative for various diseases such as cardiovascular, diabetes mellitus, and etc.
