EFFECT OF INSULIN SHOCK ON OXIDATIVE MODIFICATION OF PROTEINS IN THE NERVOUS TISSUE OF THE BRAIN OF LABORATORY RATS

The rationale for the study of the formation of products of oxidative modification of proteins (OMP) in the nervous tissue in an experimental study of the effects of insulin shock was the literature data indicating that, oxidative destruction of proteins is one of the early indicators of tissue damage. The OMP considered as a factor in the pathogenesis of nervous tissue in many diseases. In some studies, there is information about the effect of experimental GP on oxidative processes in blood serum, red blood cells, adrenal glands and some other tissues, but the effect of this pathology on the processes of free radical oxygenation in the nervous tissue not studied. The purpose of this article was to evaluate changes in the level of OMP in the nervous tissue of laboratory rats under the influence of insulin shock. The experiment was performed on 32 sexually mature mongrel male rats weighing 380–400 g (age 6 months), divided into 4 groups of 8 individuals: control (intact) group, group 1-laboratory animals exposed to a single insulin shock (1 time per day, 1 day); group 2 – laboratory rats exposed to a double insulin shock (1 time per day, 2 days); group 3 – laboratory rats exposed to triple exposure to insulin shock (1 time per day, 3 days). Rats of the experimental groups insulin (3.5 Units) subcutaneously were injected. The presence of hypoglycemic coma determined by the appearance of convulsions. Animals that are intact at this time remained in their cells. Experimental animals slaughtered by decapitation on a guillotine. The material for research was a homogenate of nervous tissue obtained by the method Of Kutlubaev M. A. and co-authors. The study of the level of OMB in nervous tissue was performed using the Dubinina E. E. method, based on the reaction of oxidized amino acid residues of protein with 2,4-dinitrophenylhydrazine (2,4 – DNFG) with the formation of anhydrous 2,4 – DNFG. Measurements of supernatant extinction at wavelengths (λ) of 356, 370, 430 and 530 nm performed using a 5400-UV spectrophotometer (Ekros-Analytica, Russia). Statistical processing of the experiment results performed using the student’s parametric t-test, since the experimental data obeyed the law of normal distribution. It was found, that insulin shock and experimental hyperinsulinemia (HI) contributed to a decrease in free radical protein oxidation in rat nervous tissue homogenate, reducing the total level of OMP carbonyl products in all the study groups exposed to insulin shock: in the 1st group – by 37.6 % (n=8; p≤0.01), in the 2nd group – by 40.4 % (n=8; p≤0.01), in the 3rd group – by 40.8 % (n=8; p≤0.01). Experimental HI causes significant changes in the level of OMP products in the nervous tissue of laboratory animals, which expressed in a decrease in aldehyde and ketone derivatives of both the main and neutral characters in comparison with the indicators of the intact group. In all three groups with experimental HI, compared with the intact group, there was a significant decrease in primary and secondary carbonyl products of OMP, which indicates that free radical oxidation processes associated with both fragmentation of protein molecules and their aggregation suppressed under conditions of insulin shock. At one- and three-time exposure to HI, primary markers of oxidative stress, formed mainly due to fragmentation of protein molecules, prevailed among the carbonyl products of OMP in the nervous tissue. The content of primary and secondary carbonyl products was almost equal when the HI applied twice, which indicates that the processes of fragmentation and aggregation of protein molecules were almost the same under these conditions. In all three experimental groups, compared with the intact group, there was a significant decrease in carbonyl products of both neutral and basic nature due to a decrease in free radical damage to neutral and basic amino acids of protein molecules. In all the study groups, including the intact group, the level of OMPo was significantly lower than the level of OMPn. The most pronounced differences between the level of carbonyl products of OMP and OMPo were observed with three-fold HI, and the least pronounced-with a single one. With increasing duration of insulin shock, the spectrum of carbonyl products of OMP shifted towards increasing the content of the main products, as the role of the main amino acids as targets for damage to protein molecules by free radicals increased.

Laboratory rats' basic behavioral characteristics were studied in the experimental "Open field" test. A rat renal carcinoma cell line RA was used to model tumor growth. Free radical processes were assessed in normal condi-tions of organism and with tumor progression. Correla-tions were revealed between the levels of behavioral ac-tivity, the intensity of tumor growth, and free radical pro-cesses. The obtained data will serve as the basis for the development of risk management systems for the oncology and development of oncopathology in the population.

The paper shows, that patients with purulent meningitis on the background of severe clinical and laboratory changes had a disturbance of prooxidant-antioxidant regulation with a shift towards activation of free radical oxidation. In the dynamics of the disease, after 7-10 days of conventional treatment, against the background of a reduction of cephalic and meningeal syndromes intensity, a normalization of liquor and hemogram indices, in most patients there was a further decrease in catalase activity and an increase of nitrites, of parameters of spontaneous and induced oxidative modification of blood proteins that remained when the patients were discharged, compared with healthy people.

Background. The study of new methods of treating frostbite is feasible using an experimental model, the effectiveness of which is confirmed by morphological results. Purpose of the study. Morphological substantiation of the effectiveness of the developed device for simulating contact frostbites of varying severity. Material and methods. In 30 laboratory rats, the designed device was used to simulate cold trauma of varying severity. The material for morphological research was collected at different time intervals. Results. Morphological argumentation of the effectiveness of the created device for reproducing both superficial and deep frostbites is presented. Conclusions. The developed device is not difficult to manufacture; it is safe and allows reproducing standardized contact frostbites of varying severity in laboratory animals.

Two especially valuable animal models of post-traumatic stress disorder (PTSD) have been developed, including brief exposure of laboratory rats to a predator (a cat) or its odor, and the single prolonged stress paradigm. In each of these models, laboratory animals are evaluated for behavioral changes several days to several weeks following the stressful experience and are compared to unstressed controls. In both of these models, stressed animals display behavioral changes consistent with a PTSD-like phenotype. Using these models, investigators have explored central and peripheral neural and endocrine changes associated with the onset of PTSD-like symptoms and approaches to prevent or block the effects of the traumatic stressor on behavioral changes. Two particularly effective treatments that have been described include administration of a protein synthesis inhibitor and intra-nasal administration of neuropeptide Y. Animal models also provide an opportunity to study transgenerational transmission of PTSD risk.

The activity of free radical processes in liver mitochondria was investigated in rats kept on high-sucrose and low protein/high-sucrose diets. Excess of dietary sucrose caused intensification of free radical processes in liver mitochondria as evidenced by increased hydroxyl radical generation, accumulation of primary (conjugated dienes, ketodienes) and secondary products (TBA-reactive products) of lipid peroxidation, increased cholesterol/phospholipids ratio and also accumulation of oxidative modification products of proteins (carbonyl derivatives). Additional nutritional protein deficiency (low protein/high-sucrose diet) enhanced destructive changes in liver mitochondria. This suggests a critical role of nutrient protein supplementation for maintaining the functional activity of mitochondria. The established changes can be considered as one of possible mechanisms of functional liver activity violation in conditions of nutrient imbalance.

Introduction: Applying a coating on hernia endoprosthesis prevents recurrent anterior abdominal wall hernias, reduces inflammatory response and stimulates reparative processes in the area of its implantation. The aim of investigation was to study the effect of Solcoseryl and Vitamin C in a collagen-stimulating coating of hernioendoprosthesis on a morphological picture in anterior abdominal wall plastic surgery. Materials and methods: The study was performed on 180 laboratory mice divided into three groups of 60 animals each: the first group animals were implanted with polypropylene endoprostheses without a collagen-stimulating coating, the second group animals – polypropylene endoprostheses with a collagen-stimulating coating with Vitamin C, and the third group animals – polypropylene endoprostheses with a collagen-stimulating coating with Solcoseryl. The laboratory animals were withdrawn from the experiment on the 10th, 30th, 60th, and 90th days. The excised sections of the abdominal wall were stained with G+E to determine the nature of inflammation and the number of cell elements. Results and discussion: When using endoprostheses with a collagen-stimulating coating, the stages of inflammatory process proceeded more quickly, which was confirmed by a reliable (р ≤ 0.05) decrease in the number of neutrophils, macrophages and lymphocytes at all stages of the study. By the 90th day of the experiment, the number of fibroblasts in the control group was by 22.64% less than in the study groups with a coating. Conclusion: A cytological and histological analysis in the control group determined a consistent decrease in an exudative phase of inflammatory reaction. When using endoprosthesis with coatings, its acceleration and lower intensity was noted throughout the study. In the group with Solcoseryl, the formation of a dense connective capsule around the endoprosthesis indicates its quality and better adaptation of the endoprosthesis in body tissues.

Complex research into biochemical aspects of the functioning of the antioxidant system for the protection of sperm of the breeding boars has established that the intensity of the flow of peroxide oxidation of lipids in germ cells is much higher than in the extracellular space. Reactions of free radical peroxide oxidation of proteins and lipids in the semen of healthy breeding boars are characterized by a stable level of activity that is necessary for the normal course of implementation of processes of the reproductive function. All components of the antioxidant system stay in mutual compensatory ratios under physiological conditions. As a rule, reducing the concentration or activity of some antioxidant enzymes leads to corresponding changes in others. The main antioxidants of the germ cell genomes are superoxide dismutase and ceruloplasmin. Catalase is a key enzyme that neutralizes H2O2 in the semen fluid of breeding boars. The content of total proteins in the semen of the synthetic line SS23 animals is greater (p < 0.05) than in the breeding boars of the large white breed. The processes of the oxidative modification of proteins in the animals body of the synthetic line runs more intensely, as it is evidenced by the higher content of the aldehyde and ketodinitrophenyl hydrazones of the main and neutral character in the sperm cytoplasm.

Annotation. One of the main factors of postoperative eventration is intra-abdominal hypertension, which occurs in various surgical pathologies of the abdominal cavity. Despite the presence of a large number of scientific papers on the negative effect of intra-abdominal hypertension on the morphological state of granulation tissue in the area of the laparotomy wound, there are no publications on the impact on the strength of the postoperative scar. Therefore, the study aimed to investigate in an experiment on small laboratory animals the effect of intra-abdominal hypertension on the mechanical strength of the postoperative scar of a laparotomy wound. The experiment was performed on 120 laboratory rats, which underwent a median laparotomy and brought together the edges of the musculoaponeurotic layer of the anterior abdominal wall with simple nodal sutures. The main group consisted of 72 animals who developed intra-abdominal hypertension by inserting a container (condom) with a certain amount of Furacilin into the abdominal cavity. The comparison group consisted of 48 animals who had an empty condom inserted into the abdominal cavity after laparotomy. The mechanical strength of the postoperative scar of the laparotomy wound was determined by the method of G. V. Petrovich (2010) on the 1st, 3rd, and 5th day after the creation of intra-abdominal hypertension, by measuring the level of intra-abdominal pressure at the time of rupture of the postoperative scar of the laparotomy wound. Statistical analysis of the results was performed using Microsoft Excel spreadsheets and a package of statistical processing software PAST. Differences between study groups were determined using Mann-Whitney criteria. The results of the study indicate that the created intra-abdominal hypertension leads to a decrease in the mechanical strength of the postoperative scar of the laparotomy wound. The degree of the negative impact of intra-abdominal hypertension on the strength of the postoperative scar is inversely proportional to the level of intra-abdominal pressure.

The effect of the synthetic biguanide derivatives N-[imino(1-piperidinyl)methyl]guanidine (NIPMG) and 1,3-dimethyl-5-[(carbamimidamidomethanimidoil) amino]benzoyl-1,3dicarboxylate (DCB) on the degree of proteins oxidative modification (POM) and the DNA fragmentation, the content of the lipid peroxidation primary products – conjugated dienes (CD), and the activity of glutathione antioxidant system in the liver and heart of rats with experimental hyperglycemia was investigated. Administration of the biguanides (15.0 mg/kg) to hypoglycemic rats promoted reduction of the free radical processes intensity in the studied tissues. Data about CD and POM level changes in hyperglycemic rats treated by NIPMG and DKB correlate with the results of DNA fragmentation degree evaluation. At the same time, the activity of antioxidant enzymes (glutathione peroxidase and glutathione reductase), and the reduced glutathione content in the liver and heart of rats changed toward control values. For metformin, which was used as a comparison drug, changes in the studied parameters in the same direction were also found. These results indicate the ability of the tested biguanide derivatives to exhibit a positive regulatory effect on free radical homeostasis, reducing the degree of oxidative stress at this pathology.

Aim. To investigate the relationship between blood plasma biochemical indicators in patients with solid tumors before a treatment and after the first course of chemotherapy with objective treatment response. Methods. Blood plasma samples taken from 14 patients with cancer relapse were studied before the treatment and after the first course of specific chemotherapy (carboplatin, methotrexate, vinblastine in patients with urine bladder cancer and irinotecan, leukovorin and fluorouracil in patients with colorectal cancer). The first group included patients with colorectal cancer relapse (males - 3, females - 4) aged 57-62 years. The second group included patients with urine bladder cancer relapse (males - 5, females - 2) aged 48-64 years. Free radical activity and protein oxidative modification, as well as endogenous intoxication and major mineral and trace elements levels were studied. Results. When achieving the objective chemotherapy effect in patients after the first treatment course, an increase of alpha-1 globulin and gamma globulin level, as well as an increase of phosphorus, zinc, lithium and iron blood plasma level was observed. In case of following disease progression, the opposite dynamics of above-mentioned indicators was revealed. In patients with good objective effect of polychemotherapy after the first treatment course, a significant increase of endogenous intoxication and blood plasma free radical activity decrease were observed. If the following tumor progression was observed, the tendency of blood plasma free radical activity increase, accompanied by the total protein oxidative modification activation, was revealed after the first treatment course. Conclusion. The increase of alpha-1 and gamma globulin levels, as well as an increase of phosphorus level in blood plasma after the first course of polychemotherapy can be used as prognostic factors of anticancer therapy efficiency. High level of endogenous intoxication as well as an increase of copper, iron, zinc and lithium blood plasma concentrations can be applied as the additional markers.
