BACKGROUND: Recently, new calcium silicate bioceramic sealers were introduced to the market. The selection of root canal sealers should not only be based on the different physical parameters but also on local biocompatibility and tissue tolerance. AIM: This study aimed to evaluate and compare the in-vivo biocompatibility of a BioRoot RCS in parallel to MTA Fillapex and AH Plus sealers. METHODS: Polyethylene tubes containing the freshly mixed test materials were implanted in the subcutaneous tissue of 32 Wistar rats. Empty tubes served as negative controls. After 7, 14, 30, and 60 days, the animals were sacrificed, and the implants with surrounding tissues were processed for routine histological analysis. Histological sections were analyzed under light microscopy. The tissue response was determined by the inflammatory cell infiltration intensity and the fibrous capsule thickness. RESULTS: Results revealed a statistically significant decrease of the inflammation intensity by time within each group for all tested sealers and control. A well-defined thin capsule was observed for all tested sealers at 60 days. CONCLUSION: BioRoot RCS exhibited rapid recovery of inflammation similar to controls. Thus, within the limitations of this study, it can be considered a biocompatible sealer with acceptable tissue tolerance.

The purpose of this study was to compare the cytotoxic effects and mineralization activity of three calcium silicate-based root canal sealers to those of a conventional resin-based sealer. Experiments were performed using human dental pulp stem cells grown in a monolayer culture. The root canal sealers tested in this study were EndoSequence BC Sealer (Brasseler), BioRoot RCS (Septodont), Endoseal MTA (Maruchi), and AH Plus (Dentsply DeTrey). Experimental disks 6 mm in diameter and 3 mm in height were made and stored in a 100% humidity chamber at 37 °C for 72 h to achieve setting. The cytotoxicity of various root canal sealers was evaluated using a methyl-thiazoldiphenyl-tetrazolium (MTT) assay. To evaluate cell migration ability, a scratch wound healing method was used, and images of the scratch area were taken using a phase-contrast microscope. Cell morphology was evaluated by a scanning electron microscope after direct exposure for 72 h to each sealer disk. In the cell viability assay, there were no significant differences between the EndoSequence BC, BioRoot RCS, Endoseal MTA, and control groups in any experimental period (p > 0.05). In the cell migration assay, there were no significant differences between the EndoSequence BC, Endoseal MTA, and control groups in any experimental period (p > 0.05). BioRoot RCS exhibited slower cell migration relative to EndoSequence BC and Endoseal MTA for up to 72 h (p < 0.05). Conversely, it showed a similar wound healing percentage at 96 h (p > 0.05). In an evaluation of cell morphology, cells in direct contact with EndoSequence BC, BioRoot RCS, and Endoseal MTA disks showed superior spreading compared to those in contact with the AH Plus disk. In an Alizarin red staining assay, EndoSequence BC, BioRoot RCS, and Endoseal MTA showed a significant increase in mineralized nodule formation compared to the AH Plus group (p < 0.05). In conclusion, all calcium silicate-based root canal sealers tested in this study showed good biological properties and mineralization activity compared to conventional resin-based sealer.

Abstract Aim The purpose of this in vitro study was to analyze the antimicrobial activity of root canal sealers by using the agar diffusion test (ADT). Methods and Materials Three categories of root canal sealers were included in the study: resin-based sealers (4), zinc oxide-based sealers (3), and calcium hydroxide-based sealers (2). The microbial strains used were: S. aureus (2 strains), C. albicans (2 strains), and E. faecalis (1 strain). Statistical analysis was conducted using a one-way analysis of variance (ANOVA). Tests of differences were analyzed using the Tukey's test with a value of p < 0.05 considered statistically significant. Results The antimicrobial activity of root canal sealers was ranked in descending order as follows: Sealite Regular, Cortisemol, Dentalis KEZ, AH26, Sealapex, Acroseal/Topseal, and Endorez/AH plus. Conclusion Root canal sealers showed different inhibitory effects depending on their types and the bacterial strains tested. Root canal sealers containing formaldehyde and eugenol proved to be effective against the microorganisms studied. Clinical Significance The incorporation of antimicrobial components into root canal sealers may become an essential factor in preventing the re-growth of residual bacteria and control of bacteria re-entry into the root canal system. ABSTRACT Objective: The objective was to evaluate the subcutaneous biocompatibility of two root canal sealers. Materials and Methods: Thirty female rats were distributed into two groups of 15 animals. Each received subcutaneous dorsal implants: Silicone tubes filled with AH Plus or Obtuseal. After the 11th, 14th, and 45th days, tissues were collected for biopsy and fixed and processed for histologic evaluation. Observations of the cellular inflammatory components, such as lymphocytes and macrophages, were made. The data were analyzed using Kruskal–Wallis and Connover tests (P < 0.05). Results: By the 7th day, lymphocyte infiltration increased in both groups (P = 0.0104). However, macrophage infiltration increased only in the AH Plus group (P = 0.0011). By the 14th and 45th days, lymphocyte and macrophage infiltrations were reduced. At the end of the experimental period, no statistically significant differences were found between the AH Plus group and the Obtuseal group (P > 0.05). Conclusion: After 45 days, Obtuseal yielded a satisfactory tissue reaction; it was biocompatible when tested in subcutaneous rat tissue. On the basis of the obtained results, it was concluded that Obtuseal root canal sealer caused a satisfactory tissue reaction as AH plus, because it was biocompatible when tested in the subcutaneous tissue of rats.

(1) Aim: To perform a systematic review of the literature on the biocompatibility of root canal sealers that encompasses the various types of sealers that are commercially available as well as both in vitro and in vivo evidence. (2) Methods: This systematic review has been registered in PROSPERO (ID 140445) and was carried out according to PRISMA guidelines using the following databases: PubMed, Cochrane Library, ClinicalTrials.gov, Science Direct, and Web of Science Core Collection. Studies published between 2000 and 11 June 2019 that evaluated cytotoxicity (cell viability/proliferation) and biocompatibility (tissue response) of root canal sealers were included. (3) Results: From a total of 1249 studies, 73 in vitro and 21 in vivo studies were included. In general, studies suggest that root canal sealers elicit mild to severe toxic effects and that several factors may influence biocompatibility, e.g., material setting condition and time, material concentration, and type of exposure. Bioactive endodontic sealers seem to exhibit a lower toxic potential in vitro. (4) Conclusions: The available evidence shows that root canal sealers exhibit variable toxic potential at the cellular and tissue level. However, the methodological heterogeneity among studies included in this systematic review and the somewhat conflicting results do not allow a conclusion on which type of sealer presents higher biocompatibility. Further research is crucial to achieve a better understanding of the biological effects of root canal sealers.

Aim: This study investigated the sealing ability produced by AH Plus (Dentsply DeTrey, Konstaz, Germany), EndoSequence BC sealer (Brasseler USA, Savannah, GA, USA), GuttaFlow (Coltène/Whaledent, Altstätten, France) and MTA Fillapex (Angelus, Londrina, Brazil). Methdos: A total of forty-six single-root human canines were prepared and randomly divided into four experimental groups (n=10): MTA Fillapex, EndoSequence BC Sealer, AH Plus or GuttaFlow. Teeth with intact crowns served as negative controls (n=3) and teeth filled with only gutta-percha served as positive controls (n=3). Teeth were mounted in a two-chamber apparatus and exposed to Enterococcus faecalis. The number of days over a 60-days period was recorded for the appearance of turbidity in the lower chamber. Kaplan-Meier method was used to estimate the survival curves. The nonparametric log-rank test was used to compare the survival curves using a significance level of 0.05. Results: The results at the end of the observation time were further analyzed by chi-square testing. All positive controls leaked within 24 h, whereas none of the negative controls leaked after 60 days. EndoSequence BC Sealer and MTA Fillapex had significant less bacterial leakage (P<0.05) than the other tested sealers. No significant difference between AH Plus and GuttaFlow was observed (P>0.05). Conclusion: In conclusion, calcium-silicate based root canal sealers promoted improved sealing ability when compared to other endodontic sealers.

Abstract Porphysomes (PS) are liposome-like nanoparticles comprising pyropheophorbide-conjugated phospholipids that have demonstrated potential as multimodal theranostic agents for applications that include phototherapies, targeted drug delivery and in vivo fluorescence, photoacoustic, magnetic resonance or positron emission imaging. Previous therapeutic applications focused primarily on photothermal therapy (PTT) and suggested that PSs require target-triggered activation for use as photodynamic therapy (PDT) sensitizers. Here, athymic nude mice bearing subcutaneous A549 human lung tumors were randomized into treatment and control groups: PS-PDT at various doses, PS-only and no treatment negative controls, as well as positive controls using the clinical photosensitizer Photofrin. Animals were followed for 30 days post-treatment. PS-PDT at all doses demonstrated a significant tumor ablative effect, with the greatest effect seen with 10 mg/kg PS at a drug-light interval of 24 h. By comparison, negative controls (PS-only, Photofrin-only, and no treatment) showed uncontrolled tumor growth. PDT with Photofrin at 5 mg/kg and PS at 10 mg/kg demonstrated similar tumor growth suppression and complete tumor response rates (15 vs. 25%, p = 0.52). Hence, porphysome nanoparticles are an effective PDT agent and have the additional advantages of multimodal diagnostic and therapeutic applications arising from their intrinsic structure. Porphysomes may also be the first single all-organic agent capable of concurrent PDT and PTT.

The present study was performed in order to evaluate, by the autoradiographic technique the sealing properties of three root canal sealers, combined with gutta - percha and silver points, in vitro. A total of one hundred and five extracted human, single root teeth were used and divided into five groups according to the filling combinations tested. The teeth were filled, using two techniques, of the single and multiple points. Autoradiographies, using S35, in all instances, were performed and the results obtained were compared with the respective dental radiograms The obtained results lead to the following conclusions: 1. Silver and gutta - percha points may be always combined with sealers. Otherwise the root canal is not adequately sealed. 2. A gutta - percha point, without sealer, produced a superior filling to silver point. 3. It seems possible to obtain a complete obturation of the rootcanal with the combination of gutta - percha and Grosman’s Sealer using the multiple - point technique. 4. It follows in effectivness, the combination of silver point and Grossman’s sealer.5. The combination of gutta - percha point with Zinc - oxide eugenol sealer and chloropercha are the least efficient. 6. Finally it must be stressed that since the conditions of the present study do not correspond exactly to those encountered in vivo, extrapolation of the results obtained, to man has to be performed with caution.

OBJECTIVE: To histologically evaluate the behavior of the subcutaneous tissue of rats after placement of polyethylene tubes filled with calcium hydroxide-based root canal sealers (SealapexTM, ApexitTM e Sealer 26TM). STUDY DESIGN: The study employed 60 rats, which were divided into eight groups, being four experimental groups with 10 rats each, and four control groups with five rats. Analysis was conducted at 48 hours, 7, 21 and 60 days. RESULTS: Histological analysis revealed that all sealers were irritant; however, the intensity varied between each sealer and as a function of time. At the initial periods (48 hours and seven days), all materials demonstrated similar outcomes, with inflammatory reaction from severe to moderate, respectively. At the final periods (21 and 60 days), the tissue reactions to the implants containing SealapexTM displayed an active status yet with regression, compared to the ApexitTM and Sealer 26TM. The control groups exhibited less inflammatory cells than the experimental groups, keeping a reaction status with regression. CONCLUSIONS: The results achieved in this study revealed that the SealapexTM sealer was the most compatible, followed by ApexitTM and Sealer 26TM.

The purpose of this study was to compare the penetration ability of calcium silicate root canal sealers and conventional resin-based sealer using confocal laser scanning microscopy (CLSM). A total of 60 recently extracted single-rooted human premolars were used in this study. The root canals were prepared to a size 40/0.06 taper with ProFile rotary instruments and irrigated with NaOCl and EDTA. After drying all canals, the specimens were randomly divided into three experimental groups (n = 20): Group 1, gutta-percha (GP)/AH Plus with continuous wave compaction; group 2, GP/BioRoot RCS with a single-cone technique; and group 3, GP/Endoseal MTA with a single-cone technique. All experimental samples were sectioned perpendicular to their long axis using a low-speed diamond wheel at the apical, middle, and coronal third levels. The penetration abilities of all samples were evaluated using CLSM. A Kruskal–Wallis analysis and a series of Mann–Whitney U post hoc tests were performed. A higher intensity level was found in the coronal area and a lower intensity level in the apical area in all the experimental groups. The AH Plus group showed higher sum fluorescence intensity in the apical and coronal thirds compared with the BioRoot RCS and Endoseal MTA groups, whereas the BioRoot RCS group showed a higher intensity level in the middle third, similar to the AH Plus group. The maximum sealer penetration depth was low in the apical area and high in the coronal area in the AH Plus and Endoseal MTA groups. In the BioRoot RCS group, maximum sealer penetration was observed in the middle third. In conclusion, there were significant differences in sealer penetration pattern and distance according to the root level and sealer type.
