Small intestinal morphology and bacterial populations in ileal digesta and feces of newly weaned pigs receiving a high dietary level of zinc oxide

A study was carried out to determine the effect of dietary supplementation of 3000 mg kg–1 zinc oxide (ZnO) on the small intestinal morphology and populations of enterobacteriaceae, lactobacilli and clostridia in ileal digesta and feces of weaned pigs. At 17 d of age, 36 pigs from nine litters were fitted with simple T-cannulae at the distal ileum and after a 2-h post-surgery recovery returned to their sows. At 21 d of age, the pigs were weaned and housed in individual metabolism crates. Pigs were allocated to receive a standard starter diet supplemented with or without 3000 mg kg–1 of ZnO. Ileal digesta and fecal samples were collected immediately before weaning and then on days 2, 4, 7, 9, and 11 after weaning and were used to quantify enterobacteriaceae, lactobacilli and clostridial populations by colony enumeration on selective media. Pigs were euthanized following the final sampling, and 2 cm sections of tissue were collected from sites 25, 50 and 75% along the length of the small intestine for determination of mucosal thickness (MT), crypt depth (CD), villous height (VH) and villous width (VW). Zinc oxide supplementation altered the mucosal morphology of the small intestine. Mucosal thickness (P < 0.08) and VH (P < 0.05) were increased at sites 25 and 50% along the length of the small intestine by ZnO supplementation. Overall VW also increased (P < 0.01) with ZnO supplementation. Crypt depth decreased (P < 0.05) at 75% along the length of the small intestine with ZnO supplementation. The ratio of VH to CD was higher (P < 0.05) for ZnO-supplemented than for control-fed pigs at sites 25, 50 and 75% along the length of the small intestine. There was no effect (P > 0.05) of supplementary ZnO on bacterial populations in ileal digesta or feces. The present study indicates that supplementing ZnO in starter diets changes the epithelial morphology of the small intestine, which may affect nutrient digestion and absorption in newly weaned pigs. Thirty-two (7-8 weeks old) male rabbits that weighed between 700 and 800 g were used to determine the effect of inclusion of Turmeric powder on carcass yield and intestinal morphology of rabbits. The rabbits were randomly allotted to four diets containing varying levels of turmeric powder (0, 5, 10 and 15 g) in a complete randomized design. Data were collected on carcass yield, duodenum, jejunum, ileum, crypt depth, mucosal thickness and analyzed using ANOVA. Significant (p<0.05) differences were obtained on bled and eviscerated weights, forelimbs, hind limbs, neck, tail and loin. Highest (p<0.05) duodenum crypt depth (111.50 μm) was obtained with buck fed diet containing 15 g turmeric inclusion while buck fed with diet containing 0 g turmeric had the least value of (67.67 μm). This was similar (p>0.05) with values obtained for buck fed diets containing 5 g turmeric (73.83 μm) and those fed with 10 g turmeric inclusion (79.33 μm). There was significant difference (P<0.05) of rabbit fed with diets with turmeric on jejunum villi height. There was significant difference (P<0.05) in jejunum crypt depth and mucosal thickness. However, 10 g inclusion could be assumed as economic inclusion level for carcass yield and intestinal increase in rabbit production.

The effects of dietary chitosan-copper chelate (CS-Cu) on growth performance, diarrhea, intestinal morphology and epithelial cell apoptosis in weaned piglets was investigated. One hundred and sixty Duroc × Landrace × Yorkshire weanling barrows with an average body weight of 7.75 kg were randomly assigned to one of the following dietary treatments: (1) control, (2) 100 mg Cu/kg diet from CuSO<sub>4</sub>, (3) 100 mg Cu/kg diet from CuSO<sub>4</sub> mixed with chitosan (CuSO<sub>4</sub>+CS), (4) 100 mg Cu/kg diet from CS-Cu. The feeding trial lasted for 30 days. The results showed that the pigs receiving a diet containing CS-Cu had higher average daily gain and lower diarrhea incidence than the pigs receiving dietary CuSO<sub>4</sub> and CuSO<sub>4</sub>+CS. Villus height and the ratio of villus height/crypt depth in duodenum, jejunum, and ileum were higher and crypt depth was lower in CS-Cu treated pigs than in pigs fed dietary CuSO<sub>4 </sub>or CuSO<sub>4</sub>+CS. An apparent decrease of ileal epithelial cell apoptosis in pigs fed CS-Cu diet was found. The activities of antioxidant enzymes were higher in pigs fed dietary CS-Cu than in those fed other diets. The results indicated that dietary CS-Cu showed better biological and physiological function in improving small intestinal morphology and reducing diarrhea incidence.

Context HepPar-1 and Arginase-1 are urea cycle enzymes used to distinguish hepatocellular carcinoma from other carcinomas. HepPar-1, but not Arginase-1, is known to be immunoreactive with normal human small intestine. Objectives To better define and compare the immunohistochemical staining patterns of HepPar-1 and Arginase-1 in adenocarcinomas arising in the small intestine, including the ampullary region. Design Staining for HepPar-1 and Arginase-1 was performed on 20 nonampullary small intestinal adenocarcinomas and 32 adenocarcinomas from the ampullary region. Ampullary adenocarcinomas were divided into intestinal morphology (15), pancreatobiliary morphology (14), and unclassifiable (3). Nonneoplastic small intestinal mucosa and colorectal adenocarcinomas were used as control groups. Results HepPar-1 stained 12 of 20 nonampullary small intestinal adenocarcinomas, with a median of 63% of cells staining in positive cases. It also stained 11 of 15 ampullary carcinomas with intestinal morphology, with a median of 75% of cells staining in positive cases. Two of 14 ampullary carcinomas with pancreatobiliary morphology were positive for HepPar-1. Arginase-1 showed positivity in 2 ampullary region carcinomas and diffuse positivity in 1 duodenal adenocarcinoma. Two of 22 colorectal carcinomas stained for HepPar-1 with none positive for Arginase-1. Conclusions HepPar-1, but not Arginase-1, usually shows positivity in small intestinal adenocarcinomas and ampullary adenocarcinomas with intestinal morphology, but only rarely shows positivity in ampullary adenocarcinomas with pancreatobiliary morphology. HepPar-1 positivity in metastatic adenocarcinoma with intestinal morphology is suggestive of an upper gastrointestinal primary site.

Intestinal resection is one of the most common surgeries performed on urgent indications, the most difficult is the decision in favor of the formation of primary anastomosis in the case of primary infection of abdominal cavity, multiple defects of the intestinal wall, as well as the general serious condition of a patient. In order to improve the methods of diagnosis and prevention of post−surgery complications, as well as personification of surgical tactics of treatment in the patients undergoing anastomotic surgeries in intestine, the results of treatment of 96 patients were analyzed. The level of serum cryoglobulins was determined by the method of A. E. Kalovidoris with modifications. The results of surgical treatment were evaluated according to the classification of D. Dindo et al. (2004). The use of cryoglobulin levels before surgery can significantly affect surgical tactics: at a low degree it is possible to perform resection of a segment of small intestine with the formation of primary small intestinal anastomoses; at average − it is possible to perform small−intestinal anastomoses with unloading intestinal stoma or with the location of the anastomosis extraperitoneally (if possible) and decompression of the anastomosis with an incubation probe; at high − it is expedient to supplement performance of an anastomosis with a variant of an enterostomy or "delayed" anastomoses, at a severe general condition of patients it is expedient to form final small intestinal stoma. Determining the level of cryoglobulinemia as a marker of the prognosis of failure of the sutures of intestinal anastomoses and the use of differentiated surgical tactics depending on the level of this index contributes to a significant improvement in direct results of surgical treatment of the patients undergoing resection of small intestine segments. The proposed tactics virtually eliminate the implementation of multi−stage surgical interventions and helps to reduce the duration of treatment of patients, reduce the level of post−surgery complications and mortality. Essential fatty acid (EFA) deficiency in mice induces fat malabsorption. We previously reported indications that the underlying mechanism is located at the level of the intestinal mucosa. We have investigated the effects of EFA deficiency on small intestinal morphology and function. Mice were fed an EFA-deficient or control diet for 8 wk. A 72-h fat balance, the EFA status, and small intestinal histology were determined. Carbohydrate absorptive and digestive capacities were assessed by stable isotope methodology after administration of [U-13C]glucose and [1-13C]lactose. The mRNA expression and enzyme activity of lactase, and concentrations of the EFA linoleic acid (LA) were measured in small intestinal mucosa. Mice fed the EFA-deficient diet were markedly EFA-deficient with a profound fat malabsorption. EFA deficiency did not affect the histology or proliferative capacity of the small intestine. Blood [13C6]glucose appearance and disappearance were similar in both groups, indicating unaffected monosaccharide absorption. In contrast, blood appearance of [13C]glucose, originating from [1-13C]lactose, was delayed in EFA-deficient mice. EFA deficiency profoundly reduced lactase activity (−58%, P < 0.01) and mRNA expression (−55%, P < 0.01) in mid-small intestine. Both lactase activity and its mRNA expression strongly correlated with mucosal LA concentrations ( r = 0.77 and 0.79, respectively, P < 0.01). EFA deficiency in mice inhibits the capacity to digest lactose but does not affect small intestinal histology. These data underscore the observation that EFA deficiency functionally impairs the small intestine, which in part may be mediated by low LA levels in the enterocytes.
