AbstractBlack Sigatoka, caused by the leaf fungus Mycosphaerella fijiensis Morelet, is a major constraint to banana production around the world. In Ecuador, the biggest banana-exporting country in the world, this disease has become increasingly aggressive. This has resulted in more fungicide applications, which have significantly increased costs in production and for the environment. Consequently, many banana growers have shifted to organic production, which produces greater economic returns as a result of higher sale prices. In addition, production costs are lower as no fungicides are applied. These organic bananas receive substantial amounts of organic products. This study describes the black Sigatoka disease and nutrient status in an organic banana plantation and compares it with a conventionally fertilized and fungicide-treated plantation. Black Sigatoka symptoms were evaluated in the vegetative and flowering stages under both production conditions and in vitro conditions. Univariate and multivariate descriptive statistics were used to analyze the parameters. Disease symptoms were more severe in leaves from the organic field than in leaves from the inorganic field, but the nutrient status (soil and foliar) did not differ between the two farms. Banana plants from the organic farm had 12 functional leaves at flowering and eight functional leaves at harvest. Average banana yields were over 40% lower for organic versus inorganic management; however, the average price received for organic bananas was over two times higher. Profit–cost analysis has shown that the organic banana farm was substantially more profitable than the inorganic one during the time period analyzed. These results indicated that bananas can be grown commercially without fungicides, and the lower productivity levels are compensated by higher prices of organic fruits in international markets. In addition, organic production has beneficial impacts on social and environmental issues.

Pseudomonas fluorescens is an opportunistic, psychotropic pathogen that can live in different environments, such as plant, soil, or water surfaces, and it is associated with food spoilage. Bioactive compounds can be used as antimicrobials and can be added into packaging systems. Quercetin and lactoferrin are the best candidates for the development of a complex of the two molecules absorbed on bio combability structure as hydroxyapatite. The minimum inhibiting concentration (MIC) of single components and of the complex dropped down the single MIC value against Pseudomonas fluorescens. Characterization analysis of the complex was performed by means SEM and zeta-potential analysis. Then, the synergistic activity (Csyn) of single components and the complex was calculated. Finally, the synergistic activity was confirmed, testing in vitro its anti-inflammatory activity on U937 macrophage-like human cell line. In conclusion, the peculiarity of our study consists of optimizing the specific propriety of each component: the affinity of lactoferrin for LPS; that of quercetin for the bacterial membrane. These proprieties make the complex a good candidate in food industry as antimicrobial compounds, and as functional food.

Leuce-poplars (mostly white poplar and its natural hybrid grey poplar) are native tree species trough Hungary. They are covering more than 4.0 per cent of the Hungarian forested area (77 000 ha). The white (grey) poplars play a significant role in the forestation under sandy soil site conditions as well as they are of importance from nature conservation point of view as well. Long-term selection breeding work is going on at the National Agricultural Research and Innovation Centre Forest Research Institute (NARIC FRI), involving selection of fast-growing Leuce-poplar clones under dry site conditions. Micropropagation technology is relatively quickly spreading in forestry. In vitro multiplication of trees is applied mainly in fruit growing in Hungary, in case of forestry it is used mainly for selection breeding. This paper presents a short overview on the micropropagation trials with different Leuce-poplar clones and the early evaluation of the seedlings growing of the micropropagated clones/varieties.

This study was designed to assess the effect of glyphosate and mancozeb on growth of Vicia faba rhizobia isolates in vitro and on their N2-fixation performance. Hence, ten isolates were isolated using plant-soil trap method from soil samples collected from farm lands. Those isolates were morphologically characterized using YEMA medium and authenticated as nodulating rhizobia using sand culture. These isolates were treated with 100, 150, and 200 μg a.e. L−1 glyphosate, 100, 150, and 200 mg L−1 mancozeb, and their combinations. The result showed that almost all isolates were affected (only 4–10% survival) at lower (100 mg L−1) concentration of mancozeb. However, 80% of isolates treated with higher concentration (200 μg a.e. L−1) of glyphosate for 72 h formed colonies on YEMA medium. Moderate (40%) isolates also showed better (31–50% and 17–45%) survival within 100 : 100 and 150 : 150 combinations of glyphosate and mancozeb, respectively. For in vivo experiment, faba bean seedlings in sand culture were inoculated with four relatively in vitro test resistant and one sensitive isolates. The inoculated isolates were treated with field recommended concentration of glyphosate, mancozeb, and combinations. Thus, experimental plants almost all showed normal (61–124 nodule plant−1) nodulation and N2-fixation (90–109%) performance as compared to the control.

Previous freeze fracture results on the intact giant, amoeba Chaos carolinensis indicated the presence of a fibrillar arrangement of filaments within the cytoplasm. A complete interpretation of the three dimensional ultrastructure of these structures, and their possible role in amoeboid movement was not possible, since comparable results could not be obtained with conventional fixation of intact amoebae. Progress in interpreting the freeze fracture images of amoebae required a more thorough understanding of the different types of filaments present in amoebae, and of the ways in which they could be organized while remaining functional.The recent development of a calcium sensitive, demembranated, amoeboid model of Chaos carolinensis has made it possible to achieve a better understanding of such functional arrangements of amoeboid filaments. In these models the motility of demembranated cytoplasm can be controlled in vitro, and the chemical conditions necessary for contractility, and cytoplasmic streaming can be investigated. It is clear from these studies that “fibrils” exist in amoeboid models, and that they are capable of contracting along their length under conditions similar to those which cause contraction in vertebrate muscles.

Early in spermiogenesis the manchette is rapidly assembled in a distal direction from the nuclear-ring-densities. The association of vesicles of smooth endoplasmic reticulum (SER) and the manchette microtubules (MTS) has been reported. In the mouse, osmophilic densities at the distal ends of the manchette are the organizing centers (MTOCS), and are associated with the SER. Rapid MT assembly and the lack of rough ER suggests that there is an existing pool of MT protein. Colcemid potentiates the reaction of vinblastine with tubulin and was used in this investigation to detect this protein.

The differentiation and maintenance of many specialized epithelial structures are dependent on the underlying connective tissue stroma and on an intact basal lamina. These requirements are especially stringent in the development and maintenance of the skin and oral mucosa. The keratinization patterns of thin or thick cornified layers as well as the appearance of specialized functional derivatives such as hair and teeth can be correlated with the specific source of stroma which supports these differentiated expressions.

Some authors using immunofluorescent techniques had already suggested that some hepatocytes are able to synthetize several plasma proteins. In vitro studies on normal cells or on cells issued of murine hepatomas raise the same conclusion. These works could be indications of an hepatocyte functionnal non-specialization, meanwhile the authors never give direct topographic proofs suitable with this hypothesis.The use of immunoenzymatic techniques after obtention of monospecific antisera had seemed to us useful to bring forward a better knowledge of this problem. We have studied three carrier proteins (transferrin = Tf, hemopexin = Hx, albumin = Alb) operating at different levels in iron metabolism by demonstrating and localizing the adult rat hepatocytes involved in their synthesis.Immunological, histological and ultrastructural methods have been described in a previous work.

The influence of small variations in the composition on the corrosion behavior of Co-Cr-Mo alloys has been studied using scanning electron microscopy (SEM), energy dispersive x-ray analysis (EDX), and electrochemical measurements. SEM and EDX data were correlated with data from in vitro corrosion measurements involving repassivation and also potentiostatic anodic polarization measurements. Specimens studied included the four alloys shown in Table 1. Corrosion tests were conducted in Hanks' physiological saline solution which has a pH of 7.4 and was held at a temperature of 37°C. Specimens were mechanically polished to a surface finish with 0.05 µm A1203, then exposed to the solution and anodically polarized at a rate of 0.006 v/min. All voltages were measured vs. the saturated calomel electrode (s.c.e.).. Specimens had breakdown potentials near 0.47V vs. s.c.e.

The in vitro human tumor cloning assay originally described by Salmon and Hamburger has been applied recently to the investigation of differential anti-tumor drug sensitivities over a broad range of human neoplasms. A major problem in the acceptance of this technique has been the question of the relationship between the cultured cells and the original patient tumor, i.e., whether the colonies that develop derive from the neoplasm or from some other cell type within the initial cell population. A study of the ultrastructural morphology of the cultured cells vs. patient tumor has therefore been undertaken to resolve this question. Direct correlation was assured by division of a common tumor mass at surgical resection, one biopsy being fixed for TEM studies, the second being rapidly transported to the laboratory for culture.
