Abstract Stallions experience transient fluctuations in fertility throughout the breeding season. Considering pregnancy diagnoses cannot be ascertained until ~14 days post-breeding, the timely detection of decreases in stallion fertility would enhance industry economic and welfare outcomes. Therefore, this study aimed to identify the proteomic signatures reflective of short-term fertility fluctuations, and to determine the biological mechanisms governing such differences. Using LC–MS/MS, we compared the proteomic profile of semen samples collected from commercially “fertile” stallions, during high- and low-fertility periods. A total of 1702 proteins were identified, of which, 38 showed a significant change in abundance (p ≤ 0.05). Assessment of intra- and inter-stallion variability revealed that caseins (namely κ-, α-S1-, and α-S2-casein), were significantly more abundant during “high-fertility” periods, while several epididymal, and seminal plasma proteins (chiefly, epididymal sperm binding protein 1 [ELSPbP1], horse seminal plasma protein 1 [HSP-1] and clusterin), were significantly more abundant during “low-fertility” periods. We hypothesised that an increased abundance of caseins offers greater protection from potentially harmful seminal plasma proteins, thereby preserving cell functionality and fertility. In vitro exposure of spermatozoa to casein resulted in decreased levels of lipid scrambling (Merocyanine 540), higher abundance of sperm-bound caseins (α-S1-, α-S2-, and κ-casein), and lower abundance of sperm-bound HSP-1 (p ≤ 0.05). This study demonstrates key pathways governing short-term fertility fluctuations in the stallion, thereby providing a platform to develop robust, fertility assessment strategies into the future.

One of the most commonly encountered challenges in equine breeding is endometritis, which can be difficult to resolve and causes considerable economic losses to the industry. It is a multifactorial condition, developing as an exaggerated form of the normal physiological response to breeding. Seminal plasma proteins, spermatozoa, bacteria and debris initiate an inflammatory response; the resulting fluid and neutrophils are then cleared from the uterus along with the debris. However, in some mares, the response is prolonged or exaggerated, with much fluid formation and neutrophil infiltration leading to acute endometritis. A bacterial cause has been implicated, although in some cases no pathogenic organisms can be isolated on culture. It has been postulated that any one of a variety of bacteria could be involved, or dysbiosis of the uterine microbiome could be responsible. Repeated episodes of acute endometritis may lead to the pathology associated with chronic endometritis, with mucociliary dysfunction, vascular degeneration and plasma cell infiltration. This review examines the information that is currently available about equine endometritis, particularly about the role of the inseminate in the uterus, and its current treatment. There are some promising lines of research into treatment or prevention that may help to resolve the issue.

Psoriasis is a chronic inflammatory condition associated with atherosclerotic cardiovascular disease (CVD). Systemic anti-psoriatic treatments mainly include methotrexate and biological therapies targeting TNF, IL-12/23 and IL-17A. We profiled plasma proteins from patients with moderate-to-severe psoriasis to explore potential biomarkers of effective systemic treatment and their relationship to CVD. We found that systemically well-treated patients (PASI < 3.0, n = 36) had lower circulating levels of IL-17 pathway proteins compared to untreated patients (PASI > 10, n = 23). Notably, IL-17C and PI3 were decreased with all four examined systemic treatment types. Furthermore, in patients without CVD, we observed strong correlations among IL-17C/PI3/PASI (r ≥ 0.82, p ≤ 1.5 × 10−12) pairs or between IL-17A/PASI (r = 0.72, p = 9.3 × 10−8). In patients with CVD, the IL-17A/PASI correlation was abolished (r = 0.2, p = 0.24) and the other correlations were decreased, e.g., IL-17C/PI3 (r = 0.61, p = 4.5 × 10−5). Patients with moderate-to-severe psoriasis and CVD had lower levels of IL-17A compared to those without CVD (normalized protein expression [NPX] 2.02 vs. 2.55, p = 0.013), and lower IL-17A levels (NPX < 2.3) were associated with higher incidence of CVD (OR = 24.5, p = 0.0028, 95% CI 2.1–1425.1). As a result, in patients with moderate-to-severe psoriasis, we propose circulating IL-17C and PI3 as potential biomarkers of effective systemic anti-psoriatic treatment, and IL-17A as potential marker of CVD.

Abstract Introduction: Parabens are used commonly as preservatives in a range of cosmetics applied to the under arm and breast area as well as popular preservatives because of their cost.Aim of the work: This study was done to evaluate the neprohepatic toxicity of parabens. Materials and methods: Thirty adult female rats were used and given paraben orally for six months at parabens at dose of 10 % of the LD50 equal to 4.6mg\kg.bw. Mushroom was given orally to at dose of 10 mg/kg/day for six months too. Results: Oral administration of BP induced biochemical and histopathological changes. Biochemical changes: BP toxicity manifested by changes in the liver and kidney function tests manifested by increase AST, ALT, Bilirubin, urea and createnine with decreases to plasma proteins in comparison to control group. Giving mushroom caused amelioration to the nephrohepatic toxicity by inducing recovery in liver and kidney functions in comparison to paraben treated group. For histopathological findings: BP induced vascular congestion in liver and kidney in association with necrotic changes in the hepatorenal epithelium which improved after mushroom treatment. Conclusion: BP induced hepatorenal toxicity which improved by mushroom treatment.

Bovine livestock is one of the most important economic and social sectors for many countries. In this sense, the development of strategies to improve reproductive bull fertility and reproduction rates is relevant. It's highlighted the role of seminal plasma proteins (SPP) in reproductive fertility, so it has found close relationships among studies on the structure and biological activity of SPP, with seminal quality, including viability, sperm motility, and morphology. In addition, they have been found to regulate sperm functions such as capacitation, acrosome reaction, and they are even related to protecting sperm against thermal and oxidative stress. Moreover, the methods of separation and protein identification and their contribution to characterizing the bovine SP proteome should be also highlighted. In this sense, the most recent studies have been directed towards developing supplements with SPP that improve quality sperm subjected to cryopreservation processes. Research has begun and should forward to establish how the networks or sets of proteins are related to the functioning and fertility of sperm, the search for biomarkers of fertility, and the use of proteins in biotechnological processes, to increase efficiency reproductive.

Abstract Two-dimensional (2D) nanomaterial Ti3C2Tx is a novel biomaterial used for medical apparatus. For its application, biosafety serves as a prerequisite for their use in vivo. So far, no research has systematically reported how Ti3C2Tx interacts with various components in the blood. In this work, we evaluated the hemocompatibility of Ti3C2Tx nanosheets which we prepared by HF etching. Effects of the concentration and size of Ti3C2Tx on the morphology and hemolysis rate of human red blood cells (RBCs), the structure and conformation of plasma proteins, the complement activation, as well as in vitro blood coagulation were studied. In general, Ti3C2Tx takes on good blood compatibility, but in the case of high concentration (>30 μg/mL) and “Small size” (about 100 nm), it led to the rupture of RBCs membrane and a higher rate of hemolysis. Meanwhile, platelets and complement were inclined to be activated with the increased concentration, accompanying the changed configuration of plasma proteins dependent on concentration. Surprisingly, the presence of Ti3C2Tx did not significantly disrupt the coagulation. In vitro cell culture, the results prove that when the Ti3C2Tx concentration is as high as 60μg/mL and still has good biological safety. By establishing a fuzzy mathematical model, it was proved that the hemocompatibility of Ti3C2Tx is more concentration-dependent than size-dependent, and the hemolysis rate is the most sensitive to the size and concentration of the Ti3C2Tx. These findings provide insight into the potential use of Ti3C2Tx as biofriendly nanocontainers for biomaterials in vivo.

The present study was carried out to evaluate the effects of partial replacement of soybean with shrimp by-products (SBp) in African catfish (Clarias lazera) diets on productive (growth, digestibility, body composition, dressing yield, blood parameters, immunity) and economic performances. Therefore, 750 fingerlings (~20.0 g) were distributed into five groups of 150 fingerlings/group with three replicates (n = 50) per group. Group 1 was fed a basal diet, while G2 and G3 were fed diets containing 25% SBp (autoclaved and acid-treated, respectively) as soybean replacer for 12 weeks, while, in G4 and G5, SBp level was 50%. All growth and nutrient utilization parameters (body weight, Wg, ADG, PER, FCR, and digestibility), plasma proteins, and immunity significantly increased in G4 and G5. Anemia screening markers recorded insignificant differences between the tested groups and control one, whereas lipid markers of plasma and fish body decreased by autoclaved SBp and increased by acid-treated SBp. The African catfish body dry matter and crude protein percentages were enriched by the addition of SBp. Replacing the soybean meal decreases diet costs and diet costs/1 kg of weight gain. In conclusion, replacing soybean with SBp (especially 50% acid-treated) positively influenced productive and economic performances with friendly effects to avoid the environmental pollution by these wastes.
